Background and Purpose: The aim of this study was to determine the effect of acute aerobic exercise and 14-days grape seed extract supplementation on total antioxidant capacity, lipid peroxidation and muscle cell damage biomarkers in untrained males Materials and Methods: In a randomized, double-blinded, and placebo-controlled study, twenty-two male students (age 19±1 years, weight 67.44±7 kg, BMI 22±2, V̇ O2max 39±2 ml/kg-1 /min-1 ) were randomly assigned to two groups of grape seed extract and placebo (PLA) (GSE: 200 mg/day for two weeks). After subsequent 14-days of supplementation, subjects did a single session of aerobic exercise (running) on treadmill at 75% VO2max for 30 minutes. Blood samples were taken 3 times: prior to supplementation (baseline), 14-days after supplementation, and immediately after exercise (post-exercise). Total antioxidant capacity (TAC), malondialdehyde (MDA), creatine kinase (CK), lactate dehydrogenase (LDH) and lactate were measured. TAC was determined by ABTs method. The collected data were then analyzed by running analysis of variance (ANOVA) with repeated measure and Bonferroni post-hoc tests as appropriate using SPSS17 at p<0.05. Results: Malondialdehyde, CK, LDH, Lactate were significantly increased after aerobic exercise (p<0.05). Short-term GSE supplementation significantly prevented MDA and CK cascade after exercise compared to PLA (p<0.05) but, it had no significant effect on basal parameters (p>0.05). The exercise had also no significant effect on total antioxidant capacity in any of the groups (p>0.05). Conclusion: Aerobic exercise could increase blood oxidative stress biomarkers and GSE supplementation, due to influential antioxidant effect; yet, it could attenuate exercise-induced oxidative stress in men

Objective: This investigation was undertaken to determine whether a grape seed extract (GSE) that is rich in mono-, oligo- and poly- meric polyphenols would modify postprandial oxidative stress and inflammation in individuals with the metabolic syndrome (MetS).Background: MetS is known to be associated with impaired glucose tolerance and poor glycemic control. Consumption of a meal high in readily available carbohydrates and fat causes postprandial increases in glycemia and lipidemia and markers of oxidative stress, inflammation and insulin resistance.Materials/methods: After an overnight fast, twelve subjects with MetS (5 men and 7 women) consumed a breakfast meal high in fat and carbohydrate in a cross-over design. A GSE (300 mg) or placebo capsule was administrated 1 hr before the meal (-1 hr). Changes in plasma insulin, glucose, oxidative stress and inflammatory markers were measured hourly for 6 hr. Results: Plasma hydrophilic oxygen radical absorbance capacity (ORAC) measured as the positive incremental area under the curve (-1 to 5 hr) was significantly increased when the meal was preceded by GSE compared with placebo (P<0.05). Meal-induced increases in oxidized LDL were attenuated with GSE (P<0.05). Plasma glucose concentrations (area under the curve from -1 to 5 hr) were also significantly lower when the meal was preceded by GSE (P<0.05) while the insulin concentrations remained unchanged (P>0.05). No changes in inflammatory markers were evident. Conclusion: These data suggest that GSE enhances postprandial plasma antioxidant status and reduces the glycemic response to a meal, high in fat and carbohydrate in subjects with the MetS. The aim of the present study was to investigate the effects of grape seed extract (GSE) supplementation on exercise performance and oxidative stress in acutely and chronically exercised rats. A total of sixty-four male rats were used in the study. Rats were divided into six groups: control, chronic exercise control, acute exercise control (AEC), GSE-supplemented control, GSE-supplemented chronic exercise and GSE-supplemented acute exercise groups. Chronic exercise consisted of treadmill running at 25 m/min, 45 min/d, 5 d a week for 6 weeks. Rats in the acute exercise groups were run on the treadmill at 30 m/min until exhaustion. GSE were given at 100 mg/kg of body weight with drinking water for 6 weeks. Plasma was separated from blood samples for the analysis of oxidative stress markers. There was no significant difference in time of exhaustion between the acute exercise groups. Plasma malondialdehyde (MDA) levels were higher in the acute exercise groups and lower in the chronic exercise groups. GSE supplementation decreased MDA levels. Xanthine oxidase and adenosine deaminase activities were higher in the AEC group compared to all the other groups. NO levels were increased with both chronic exercise and GSE supplementation. Superoxide dismutase and glutathione peroxidase activities were lower in the acute exercised groups and higher in the chronic exercised groups. GSE supplementation caused an increase in antioxidant enzyme activities. In conclusion, GSE supplementation prevents exercise-induced oxidative stress by preventing lipid peroxidation and increasing antioxidant enzyme activities.

Background: Burns trauma is associated with considerable morbidity and mortality. Resuscitation of burns patients with high-dose of Ascorbic acid has shown potential for mitigating the injury, but the optimal dose for this indication is unknown. Aim: This study aimed to evaluate the impact of adjuvant therapy with intravenous Ascorbic acid (6g over 24 hours) on indicators of oxidative stress in patients with major burns. Materials and Methods: We conducted a randomized placebo-controlled study on patients with major burns who presented at the National Orthopaedic Hospital, Enugu, Nigeria between August 2017 and July 2020. Each patient in the treatment group received intravenous Ascorbic acid, 6g over 24 hours, while those in the placebo group received Normal saline in the resuscitation fluid. Oxidative stress evaluation was based on measurement of total antioxidant capacity and malondialdehyde in the participants. The level of statistical significance was determined by a p value of <0.05. Results: The study was conducted on 37 burns patients and 15 healthy subjects. At presentation, the burns patients had significantly lower total antioxidant capacity; P=0.006, and higher serum malondialdehyde; P=0.040, compared to the healthy volunteers. The decrease in serum malondialdehyde in the burns patients treated with high-dose Ascorbic acid; 0.9±0.8 nmol/mL, was greater than that in those treated with placebo; 0.3±1.4 nmol/mL. Similarly the increase in total antioxidant capacity in the burns patients treated with high-dose Ascorbic acid; 151.7±116.5 µmol/L was greater than that in those treated with placebo; 58.4±219.1 µmol/L. However these effects weren’t statistically significant. Conclusion: Intravenous Ascorbic acid at a dose of 6g over 24 hours did not significantly alter the indicators of oxidative stress in the burns patients, under the prevailing conditions of the study.

Abstract. Physical exercise increases free radicals production; antioxidant supplementation may improve the muscle fiber’s ability to scavenge ROS and protect muscles against exercise-induced oxidative damage. This study was designed to examine the effects of all-trans resveratrol supplementation as an antioxidant to mediate anti-oxidation and lipid per-oxidation responses to exercise in male Wistar rats. Sixty-four male Wistar rats were randomly divided into four equal number (n = 16) including training + supplement (TS), training (T), supplement (S) and control (C) group. The rats in TS and S groups received a dose of 10 mg/kg resveratrol per day via gavage. The training groups ran on a rodent treadmill 5 times per week at the speed of 10 m/min for 10 min; the speed gradually increased to 30 m/min for 60 minutes at the end of 12th week. The acute phase of exercise protocol included a speed of 25 m/min set to an inclination of 10° to the exhaustion point. Superoxide dismutase (SOD), glutathione peroxidase (GPx), catalase (CAT) activity, non-enzymatic antioxidants bilirubin, uric acid, lipid peroxidation levels (MDA) and the total antioxidant capacity (TAC) were measured after the exercise termination. The data were analyzed by using one-way ANOVA. The result showed that endurance training caused a significant increase in MDA level [4.5 ± 0.75 (C group) vs. 5.9 ± 0.41 nmol/l (T group)] whereas it decreased the total antioxidant capacity [8.5 ± 1.35 (C group) vs. 7.1 ± 0.55 mmol/l (T group)] (p = 0.001). In addition, GPx and CAT decreased but not significantly (p > 0.05). The training and t-resveratrol supplementation had no significant effect on the acute response of all variables except MDA [4.3 ± 1.4 (C group) vs. 4.0 ± 0.90 nmol/l (TS group)] (p = 0.001) and TAC [8.5 ± 0.90 (C group) vs. 6.6 ± 0.80 mmol/l (TS group)] (p = 0.004). It was concluded that resveratrol supplementation may prevent exercise-induced oxidative stress by preventing lipid peroxidation.

Background: Cigarette smoke free radicals can cause cellular damage and different diseases. All the body fluids have antioxidants which protect against free radicals. Objective: The aim of this study was to evaluate salivary total antioxidant capacity and peroxidase, uric acid and malondialdehyde levels in smokers and a nonsmoking control group. Methods: Unstimulated saliva was collected from 510 males. A total of 259 subjects were current smokers and 251 were non-smokers. The levels of salivary total antioxidant capacity, uric acid, peroxidase and malondialdehyde were measured using standard procedures. Data were analyzed with t test and ANOVA. Results: The smokers were younger and dental hygiene index was higher than healthy nonsmoking controls. The mean total antioxidant capacity in smokers and nonsmokers was 0.13±0.07 and 0.21±011, respectively (P=0.001). Smokers had significantly lower peroxidase and uric acid levels than healthy controls. In addition, the mean malondialdehyde levels in the smokers and nonsmokers were 4.55 ±2.61 and 2.79 ±2.21, respectively (P=0.001). Conclusion: Cigarette smoke produces free radical and oxidative stress, causing many side effects. Salivary antioxidant levels decreased and malondialdehyde levels increased in smokers, indicating the high oxidative stress among smokers compared to nonsmokers. Cigarette smoke had deleterious effects on main salivary antioxidants levels.

Oxidative stress plays a pivotal role in the progression of chronic hepatitis B; however, it is unclear whether the status of blood oxidative stress and antioxidant components differs depending on the degree of hepatic fibrosis. To explore the relationship between oxidative stress/antioxidant capacity and the extent of hepatic fibrosis, fifty-four subjects with liver fibrosis (5.5 ≤ liver stiffness measurement (LSM) score ≤ 16.0 kPa) by chronic hepatitis B virus (HBV) were analyzed. From the analysis of eight kinds of serum oxidative stress/antioxidant profiles and liver fibrosis degrees, the level of total antioxidant capacity (TAC) reflected a negative correlation with the severity of hepatic fibrosis (Pearson correlation, r = −0.35, p = 0.01). Moreover, TAC showed higher sensitivity (73.91%) than the aspartate transaminase (AST) to platelet ratio index (APRI, 56.52%) in the receiver operating characteristic (ROC) curves. Interestingly, the TAC level finely reflected the fibrosis degree in inactive carriers (HBV DNA < 2000 IU/mL), while the APRI did in active carriers (HBV DNA > 2000 IU/mL). In conclusion, TAC is a promising biomarker for evaluating the progression of liver fibrosis in patients with HBV, and this finding may indicate the involvement of TAC-composing factors in the pathogenesis of hepatic fibrosis in chronic HBV carriers.

Abstract Purpose Over the past several decades, research has suggested reactive oxygen species act as cofactors for cervical cancer development. The aim of this study is to evaluate the antioxidant and lipid peroxidation status in cervical cancer patients in Bangladesh. Methods From December 2017 to 2018, a cross-sectional observational study was conducted on 50 cervical cancer patients and 50 controls. Plasma levels of lipid peroxidation and total antioxidant capacity were measured. The Student’s t test was used for statistical analysis. P values less than 0.05 were taken as a level of significance. Results There was a significant reduction in total antioxidant levels in patients with cervical cancer, 972.77 ± 244.22 SD µmol equivalent to ascorbic acid/L, compared to normal controls, 1720.13 ± 150.81 SD µmol equivalent to ascorbic acid/L (P < 0.001). Levels of lipid peroxidation were found to be significantly higher in cervical cancer, 7.49 ± 2.13 SD µmol/L, than in women without cervical cancer, 3.28 ± 0.58 SD µmol/L (P < 0.001). The cervical cancer patients had significantly higher levels of oxidative stress index (0.83 ± 0.31) in comparison to controls (0.19 ± 0.04) (P < 0.001). Conclusion There was an increased oxidative stress index due to imbalance between lipid peroxidation generation and total antioxidant capacity in cervical cancer patients. Further studies are needed to explore the role of oxidative stress as a cofactor for cervical carcinogenesis.
