The current study investigated the inhibiting effect on reactive oxygen species (ROS), reactive nitrogen species (RNS), and matrix metalloproteinase-1 (MMP-1) production in a cell-based study of standardized rosmarinic acid enriched extract (SRAEE) prepared from Thunbergia laurifolia leaves. HPLC chromatogram revealed that rosmarinic acid is a major component in prepared SRAEE, followed by caffeic acid. SRAEE exhibited antioxidant activity both in vitro and cell-based studies. SRAEE showed scavenging effects on nitric oxide and superoxide anion and inhibition effects on lipid peroxidation in vitro. SRAEE also inhibited ROS and MMP-1 production in normal human dermal fibroblast cells induced by H2O2 and UVA, respectively, without exerted cytotoxicity. Additionally, collagen degradation was protected by SRAEE induced by UVA. Nitric oxide and inducible nitric oxide synthase (iNOS) productions were also inhibited by SRAEE in RAW264.7 mouse macrophage cells induced by combined lipopolysaccharide (LPS)-interferon-γ (IFN-γ). The results indicated that SRAEE is a potential candidate as a natural pharmaceutical active ingredient for cosmeceutical product application.

In this study, nanocomposite hydrogels composed of sodium carboxymethylated starch (CMS)-containing CuO nanoparticles (CMS@CuO) were synthesized and used as experimental wound healing materials. The hydrogels were fabricated by a solution-casting technique using citric acid as a crosslinking agent. They were characterized by Fourier-transform infrared spectroscopy (FTIR), energy-dispersive X-ray spectroscopy (EDS), X-ray diffraction (XRD), field emission scanning electron microscopy (FESEM), and thermogravimetric analysis (TGA) to evaluate their physicochemical properties. In addition, swelling, antibacterial activities, antioxidant activities, cytotoxicity, and in vivo wound healing were investigated to evaluate the wound healing potential of the CMS@CuO nanocomposite hydrogels. Growth inhibition of the Gram-positive and Gram-negative pathogens, antioxidant activity, and swelling were observed in the CMS@CuO nanocomposite hydrogels containing 2 wt.% and 4 wt.% CuO nanoparticles. The hydrogel containing 2 wt.% CuO nanoparticles displayed low toxicity to human fibroblasts and exhibited good biocompatibility. Wounds created in rats and treated with the CMS@2%CuO nanocomposite hydrogel healed within 13 days, whereas wounds were still present when treated for the same time-period with CMS only. The impact of antibacterial and antioxidant activities on accelerating wound healing could be ascribed to the antibacterial and antioxidant activities of the nanocomposite hydrogel. Incorporation of CuO nanoparticles in the hydrogel improved its antibacterial properties, antioxidant activity, and degree of swelling. The present nanocomposite hydrogel has the potential to be used clinically as a novel wound healing material.

Alcea setosa A. (Malvaceae) is a wild plant that grows in Jordan and have several traditional medicinal uses. This study aims to collect and chemically analyze the methanolic extract from Alcea setosa A. from Jordan and to evaluate its cytotoxic and antioxidant activity against human breast cancer cells (T47D), colorectal adenocarcinoma cells (CACO2), and normal human fibroblasts (MRC5). The extract was extracted by methanol solvent and analyzed by liquid chromatography coupled with a mass spectrometer. Cell viability was assessed using trypan blue, neutral red, and MTT assays, and antioxidant activity was evaluated using DPPH scavenging activity assay. A total of 290 compounds, 12 among which were identified when compared to available standards, the extract contained six flavons derivatives, Two fatty acids, one ketone derivative, one flavonol derivative, one organic acid, and one coumarin derivative. The results also revealed that the IC50 values of the viability assays were higher among normal cells compared to the human cancer cell lines, and the viability inhibition was significant at higher concentrations compared to untreated cells. Nevertheless, moderate antioxidant activity was observed for the extract in the DPPH scavenging activity test. To sum up, this study indicates that samples of A. setosa collected from Jordan is likely to be an effective antioxidant, is optimistically potential to be utilized in breast and colon cancers treatment due to its preferential cytotoxicity against cancer cells.

Bioassay-guided isolation was used to separate the active ingredients of the ethanolic extract of Dioscorea membranacea by testing cytotoxic activity against three human cancer cell lines, i.e. large cell lung carcinoma (COR-L23), colon cell line (LS-174T) and breast cancer cell line (MCF-7), and two normal human cell lines, keratinocytes (SVK-14) and normal human fibroblasts (HF), using the SRB assay. The DPPH test for antioxidant activity was also employed, as was a test for LDH release as an indicator of damage to the cell membrane. Eight compounds were isolated, two naphthofuranoxepins (dioscorealides A [1] and B [2]), a 1,4-phenanthraquinone (dioscoreanone [3]), three steroids (β-sitosterol [4], stigmasterol [5] and β-D-sitosterol glucoside [8]) and two steroid saponins diosgenin-(3- O-α-L-rhamnopyranosyl (1→2)-β-D-glucopyranoside [6] and diosgenin 3- O-β-D-glucopyranosyl (1→3)-β-D-glucopyranoside [7]). Cytotoxic activity of 2, 3 and 6 was shown against three cancer cell lines, and 2 showed selective cytotoxic activity against lung and breast cancer, but was less active against the two normal cells, and was not toxic to cell membranes in the LDH assay. The highest antioxidant activity was shown by 3.

Herpes-like viruses were isolated from rhesus monkey peripheral blood leucocytes when co-cultivated with WI-38 cells. The virus was originally designated rhesus leucocyte-associated herpesvirus (LAHV) and subsequently called Herpesvirus mulatta (HVM). The original isolations were from juvenile rhesus monkeys shown to be free of antibody to rhesus cytomegalic virus. The virus could only be propagated in human or simian fibroblasts. Use of specific antisera developed from HVM showed no relationship between this virus and other herpesviruses. An electron microscopic study was undertaken to determine the morphology of Herpesvirus mulatta (HVM) in infected human fibroblasts.

Type II diabetes is a metabolic disease mediated through multiple molecular pathways. Here, we report anti-diabetic effect of a standardized isolate from a fossil material - a mineraloid leonardite - in in vitro tests and in genetically diabetic mice. The mineraloid isolate stimulated mitochondrial metabolism in human fibroblasts and this stimulation correlated with enhanced expression of genes coding for mitochondrial proteins such as ATP synthases and ribosomal protein precursors, as measured by DNA microarrays. In the diabetic animal model, consumption of the Totala isolate resulted in decreased weight gain, blood glucose, and glycated hemoglobin. To our best knowledge, this is the first description ever of a fossil material having anti-diabetic activity in pre-clinical models.

Abstract. In this study, cytoprotective and antioxidant activities of Rosa canina (RC) and Salix alba (SA), medicinal plants, were studied on mouse primary splenocytes by comparing Controlled Differential Sieving process (CDSp), which is a novel green solvent-free process, versus a conventional technique, employing hydroethanolic extraction (HEE). Thus, preventive antioxidant activity of three plant powders of homogeneous particle sizes, 50–100 µm, 100–180 µm and 180–315 µm, dissolved directly in the cellular buffer, were compared to those of hydroethanolic (HE) extract, at 2 concentrations (250 and 500 µg/mL) in H2O2-treated spleen cells. Overall, compared to HE extract, the superfine powders, i. e., fractions < 180 µm, at the lowest concentration, resulted in greater reactive oxygen species (ROS) elimination, increased glutathione peroxidase (GPx) activity and lower malondialdehyde (MDA) production. Better antioxidant and preventive effects in pre-treated cells were found with the superfine powders for SA (i. e., 50–100 µm and 100–180 µm, both p < 0.001), and with the intermediate powder for RC (i. e., 100–180 µm, p < 0.05) versus HE extract. The activity levels of catalase (CAT) and superoxide dismutase (SOD) in pretreated splenocytes exposed to H2O2, albeit reduced, were near to those in unexposed cells, suggesting that pretreatment with the fine powders has relatively restored the normal levels of antioxidant-related enzymes. These findings supported that CDSp improved the biological activities of plants, avoiding the use of organic solvents and thus it could be a good alternative to conventional extraction techniques.
