Exploration of RNA-Guided Recombinase Target Sites for Hyperactivated Recombinase Beta in Bovine Genome

The present study was conducted to determine the effect of different cooling systems; Fan Fogger (FF) and Fan Pad (FP) on micro environment of poultry house, thermal comfort, welfare, egg production and egg quality parameters of laying hens. This experiment was conducted on 210, White Leghorn laying pullets (32 weeks old) during hot-dry summer months (May - July) under deep litter system of housing. The FP and FF cooling systems significantly dropped the mean shed temperature and increased the relative humidity. Thus, better THI resulted in increase in egg production by 4.66 % and 3.32 % under FP and FF systems over the control group. However, specific gravity, H.U, egg shell thickness, yolk index and yolk color were not significantly influenced by cooling treatments. Significantly lower levels of antioxidant enzymes viz. LPO, Catalase, G6PD, GPx and SOD was registered in cooling groups. Both the cooling devices contributed towards bird welfare by altering the behavioral expression from agonistic to non-agonistic activities.RNA-guided recombinases (RGR) are potentially valuable tools for basic research and genetic modifications. The platform has been demonstrated to do genome editing efficiently. The platform operates on a typical recognition site comprised of the degenerate recombinase site, a 5 to 6-base pair spacer flanking it and this whole central region is flanked by two guide RNA-specified DNA sequences or Cas9 binding sites which is followed by protospacer adjacent motifs. In present investigation, a detailed map of target sites for RNA-guided recombinase platforms based on hyperactivated recombinase Beta throughout the bovine genome was prepared. For this, Chromosome wise whole genomic sequence data was retrieved from Ensembl followed by designing search pattern for recombinase Beta with spacer length five. By using this search pattern, RGR target sites were located by using dreg program of Emboss package. In total,436 RGR target sites were identified in bovine genome for recombinase Beta with spacer length five. These RGR target site provide potential of being utilized for specific genomic integration, deletion or inversion.

Background: Programmable nucleases are very promising tools of genome editing (GE), but they suffer from limitations including potential risk of genotoxicity which led to the exploration of safer approach of GE based on RNA-guided recombinase (RGR) platform. RNA-guided recombinase (RGR) platform operates on a typical recognition or target site comprised of the minimal pseudo-core recombinase site, a 5 to 6-base pair spacer flanking it and whole this central region is flanked by two guide RNA-specified DNA sequences or Cas9 binding sites followed by protospacer adjacent motifs (PAMs). Methods: The current study focuses on analysis of entire cattle genome to prepare a detailed map of target sites for RNA-guided hyperactivated recombinase Gin with spacer length six. For this, chromosome wise whole genomic sequence data was retrieved from Ensembl. After that search pattern for recombinase Gin with spacer length six was designed. By using this search pattern, RGR target sites were located by using dreg program of Emboss package. Result: Total number of RGR target sites identified in bovine genome for recombinase Gin was 677 with spacer length six. It was also investigated that whether these RGR target sites are present with in any gene or not and it was found that RGR target sites lies in both genic and intergenic region. Besides this, description of genes in context with these target sites was identified.

The present study aimed to evaluate the effect of dietary Moringa oleifera (M. oleifera) leaves and/or seed powder on laying Japanese quail performance in terms of egg production, egg quality, blood serum characteristics, and reproduction. In total, 168 Japanese quails (120 hens and 48 males) at eight weeks of age in laying period were randomly distributed to four treatment groups, with six replicates per group and seven birds (five hens and two males) per replicate. The first group (G1) served as a control group, while G2, G3 and G4 groups were supplemented with M. oleifera leaves (ML) and M. oleifera seeds (MS) and their combination ((1 g/kg ML; 1 g/kg MS; and 1 ML g/kg + 1 MS g/kg (MSL), respectively). From the results, feed consumption, feed conversion ratio, egg weight, fertility and hatchability from fertile eggs, egg and yolk index, and Haugh unit were not affected by dietary treatments. However, egg production, egg mass, eggshell thickness, and hatchability were significantly increased and blood aspartate transaminase (AST) and urea decreased in the MS treatment. Both triglycerides and total cholesterol were reduced (p < 0.05) in all treatments with ML, MS, and MSL, with no significant differences in alanine aminotransferase (ALT), albumin, total protein, globulin, and A/G ratio among dietary treatment. Our results clearly indicated that the inclusion of M. oleifera seeds in Japanese quail diet significantly increased egg production and improved hatchability, along with some egg quality parameters, and also lowered some blood biochemical components.

Abstract Background The excessive use of antibiotics in the livestock feed industry caused inevitable side effects of microbial resistance. Besides this residual antibiotics in animal-derived foodstuff imposed serious health problems for humans. So this study aimed to investigate the potential use of Bacillus velezensis to substitute antibiotics for poultry production. A total of 468, 49-week-old Hy-Line Brown chickens, were randomly divided into four groups the control group (regular diet), experiment group I (0.1% B. veleznesis), experiment group II (0.2% B. veleznesis), and antibiotic group (50 mg/kg flavomycin), with three replicates per group and trial period consisted on 42 days. Results The results showed that, compared with the control group, the average egg production rate and daily feed intake of experimental groups I and II increased significantly (P < 0.05), while the average egg weight was increased in experimental group II as compared to (I) (P < 0.01). The feed conversion ratio was decreased (P > 0.05) in group (II) Egg quality parameters such as yolk weight of the experimental group II was increased, but that of the antibiotic group and experiment group I was decreased, neither significant (P > 0.05). Moreover, the eggshell strength, yolk color, albumen height, and Haugh unit were significantly increased (P < 0.05). Compared with the control group, probiotic groups can increase the progesterone and motilin (P > 0.05) but decrease the secretin and cholecystokinin in the blood plasma (P > 0.05). Conclusions This study suggested that B. velezensis can substitute in-feed-antibiotics and improved most of the study parameters significantly. Which suggested that B. velezensis has potential future application value to replace the feed antibiotics.

In mammals, time-restricted feeding (TRF) with no caloric restriction provides health benefits and extends longevity, usually with a minor (∼3%) or no reduction in total food consumption. In the current study, a TRF regimen of 6 h free access to food (08:00–14:00 h) was applied to Leghorn chickens from 25 to 86 weeks of age; control birds ate freely during the light hours (06:00–20:00 h). Unexpectedly, the TRF-treated birds consumed, on average, 11.7% less food than the controls. This was manifested by an average reduction of 9.6% in body weight, 2.6-fold in visceral fat accumulation, and 6.5% in egg weight. Hen-housed egg production was reduced by 3.6% in the TRF group compared with the control, along the first 40 weeks of the follow-up (P &lt; 0.05), and changed into a tendency of 0.7% higher egg production thereafter. Several parameters of egg quality showed significant improvement (P &lt; 0.05) in the TRF group compared with the controls. A comparison of diurnal patterns of feed consumption revealed a higher rate of hourly consumption in the TRF group and increased consumption before dark in the control group. In conclusion, the reduced feed intake in response to the TRF treatment and loss in visceral fat accumulation supports the lack of a strong adipostat activity in chickens and different appetite regulation mechanisms compared with mammals. Therefore, future TRF studies in chickens should be adjusted by extending the ad libitum time window. The lower feed intake by the TRF-treated chickens compared with the ad libitum-fed controls seems to reduce the efficiency of egg production. Nevertheless, the improved egg quality and persistence of egg lay at the older age suggest that similarly to mammals, the TRF treatment delayed at least some of the negative impacts associated with advanced age.

The Altai Region along with the regions of the Siberian Federal District belongs to the zone with trace element deficiency including iodine deficiency. The research goal was to study the effect of iodine on edible egg quality of laying hens of the Rodonit cross. At the final stage of egg-laying, 4 groups of 4 laying hens were formed. No iodine was administered to the hens of the 1st control group. Starch-based iodine product was administered to the birds of the 2nd, 3rd and 4th trial groups into the lower third of the neck, under the wing and into the rump area in a dose of 1.5 mg per head. The egg production intensity in the trial groups was higher by 21.3-28.8%. Egg weight in the 1st control group was 65.6 ± 1.33g which was lower than that in the trial groups by 4.4% (2nd group); 3.7% (3rd group) and 1.5% 4th group). All eggs belonged to “select” grade. The 3rd and 4th trial groups had less egg white by 3.8 and 2.7%; the egg-yolk content increases in the eggs of the 2nd, 3rd and 4th trial groups by 1.2%, 9.4% and 2.4%, respectively, and the eggshell weight by 1.2-9.4% as com-pared to the control. The ratio of egg white and egg-yolk weight in the eggs of the trial groups was lower than in the control group by 9.38-19.29%, but the ratio of egg-yolk to egg white was by 10.4-23.9% higher. The following indices were higher in the trial groups as compared to the control: egg white index -by 1.4-17.1%; egg-yolk -by 3.2-6.4%; Haugh unit -by 7.2-11.1%; eggshell thickness -by 3.9-8.3%. The best results were obtained with the administra-tion of iodine to the hens in the region of the lower third of the neck. The enrichment of the body of laying hens of the Rodonit cross with iodine makes it possible to obtain the profitability of edible egg production at the level of 28.6-41.0%.

In this study, it was investigated the effect of clove oil supplementation at increasing levels into laying hens’ diets on performance, egg quality traits, some blood parameters and yolk TBARS (Thiobarbituric Acid Reagent) values. For this purpose 96 Lohman white laying hens, 28 weeks of age, were divided into four treatment groups. Control group was fed with basal diet (C) and treatment groups were fed with diets formed by addition of clove oil at 50 ppm (CO1), 100 ppm (CO2) and 150 ppm (CO3), respectively. During the trial, feed and water were given as ad-libitum, and poultry house was illuminated for 17 hours. Experiment lasted for 13 weeks. Addition clove oil at increasing rates into diet did not affect the live weight. The data analysed as polynomial showed that supplementation of clove oil into layer diet linearly improved feed conversion ratio and increased the egg production. But, daily feed consumption, egg weight, damaged egg ratio and egg quality parameters were not affected by treatments. Serum parameters such as triglyceride, glucose, aspartate aminotransferase, alanine aminotransferase and calcium were not affected by the clove oil supplementation. TBARS values in C, CO1, and CO2 were found higher than the CO3 group fed with diet including 150 ppm clove oil. In conclusion, clove oil at 150 ppm level in diets of laying hens could be used due to extend the egg shelf life and to decline serum cholesterol content.

The objective of this study was to compare the effects of feed supplementation of laying hens with manganese from its inorganic and organic sources on performance and some parameters of egg quality. Ninety-six hens at 20 weeks of age were randomly allocated to 4 dietary treatments, each consisting of 6 replicates (4 birds per replicate). The control group was fed unsupplemented basal diet (BD) with only natural background Mn level of 46.4 mg/kg feed. For the three experimental treatments, the BD was supplemented with 120 mg Mn/kg either from Mn-sulphate or Mn-chelate of protein hydrolysate (Mn-Pro) or Mn-chelate of glycine hydrate (Mn-Gly). After 8 weeks of dietary treatments the egg production, egg weight, feed intake, and feed efficiency were not affected by dietary treatments. Regardless of the sources, Mn supplementation to feed resulted in significantly decreased percentages of cracked eggs compared to the unsupplemented control group. The thickness, weight, proportion, and index of eggshell were significantly elevated in all groups supplemented with Mn. The intake of Mn-Gly resulted in considerably increased Mn deposition in egg yolk compared to the control eggs. In the control and Mn-sulphate groups yolk malondialdehyde (MDA) started to increase after 20 and 30 days of egg storage respectively, whereas in eggs from hens given organic Mn-sources this parameter was not affected up to 40 days. Although there were no significant differences in MDA values between the treatments until 20 days of storage, the Mn-sulphate group showed significantly higher MDA concentration in yolks compared to the control group after 30 days of storage. These results demonstrate that supplementation of hens&rsquo; diet with Mn has positive effects on eggshell quality. Feed supplementation with Mn from organic sources appears to be more effective in preventing yolk lipid oxidation during cold storage of eggs than that from Mn-sulphate. &nbsp;

This research was carried out to determine the effect of Jerusalem artichoke (JA) with or without 5, 10% vetch (V) supplementation on performance, egg quality characteristics and egg cholesterol content. In the study, seventy-five 25 weeks-old commercial white laying hens were randomly divided into one control and 4 treatment groups each containing 15 hens. Control group was fed basal diet without JA or V. Treatment group 1, 2, 3 and 4 were fed diets containing 5% V, 5% JA, 5% JA + 5% V and 10% JA + 10% V, respectively. The feeding period lasted 16 weeks. In the study, it was determined that dried-ground JA contained dry matter, crude protein, ether extract, crude fibre, crude ash, inulin and metabolizable energy (ME) at the level of 93.30, 10.02, 0.36, 5.64, 9.05, 15.80% and 3 060 kcal/kg, respectively. At the end of the study, live weight was lower by 4.36&ndash;10.09% in the treatment group 10% JA + 10% V compared with the other groups, but feed efficiency was improved in this group. There were statistically significant differences between the groups in egg quality characteristics (P &lt; 0.05, P &lt; 0.001). Egg production was not affected by supplementation of 5, 10% JA with or without 5, 10% V supplementation. Egg yolk cholesterol and total cholesterol content were not different in the groups (P &gt; 0.05). As a result, the addition of JA with or without V has no adverse effect on performance and egg quality in hens. &nbsp;

The present study was conducted to find the effect of different levels of organic minerals on egg production and egg quality of laying hens. A total of 45 laying hens were selected and divided into three groups with 3 replicates of 5 birds each. The control group was provided with commercial ration having inorganic minerals and the other two groups were supplemented with organic minerals at the rate of 7.5 (OM-50) and 15 mg/kg (OM-100). The experiment lasted for 5 weeks including 1 week of adaptation period. Overall feed intake was significantly higher (P<0.05) in the control. Significantly lower feed intake was recorded in group OM-100. Numerically lower (better) FCR was recorded in OM-100 at all recorded stages. Significantly (P<0.05) higher egg weight, egg shell weight and egg shell thickness was found in OM-100 compared to the control. Similarly, albumin height, Haugh unit and yolk weight were significantly (P<0.05) higher in M-100. It was concluded from the present results that organic minerals at the level of 50 and 100 mg/kg improved the performance and egg quality of laying hens. However, better results were found in OM-100.

The study was conducted to determine the effect of antiprolactin drug (Bromergon®) and Peppermint (Mentha piperita L.) on broodiness, laying performance and egg quality in indigenous hens. The effect of modulation of prolactin concentration on egg production, sequence length and inter sequence pauses were studied by analyzing the oviposition records. Total sixty indigenous laying hens (30-40 weeks of age), were assigned for treatment with peppermint solution and Bromergon®. Sixty laying hens were randomly divided into 4 groups (A, B, C and D) and each group remained 15 hens. Group A was kept for control, Group B was treated with bromocriptine (Bromergon® SANDOZ) orally @ 640 ?g per bird per day, Group C with 50% peppermint solution @ 10 g per bird orally and Group D was with peppermint and Bromergon® combined at previous dose. Over the course of trial, incremental dietary peppermint (Group C) significantly (p?0.05) increased egg production, body weight of treated indigenous hens than other groups. Egg shell percentage, thickness and haugh unit of hens fed diets supplemented with peppermint were greater than that of hens fed the control diet. However, peppermint supplementation did not influence other egg quality characteristics like albumen and yolk percentages and albumen height. The treated birds had comparatively longer sequences and fewer pauses. It is concluded that the physiological pauses occur during ovulatory sequences can be disrupted effectively using Bromergon® and peppermint. Prolactin levels modulated which may interfere with follicular recruitment and subsequent oviposition thereby improves egg laying potential of the indigenous hens.Asian J. Med. Biol. Res.
