Antimicrobial Activity of Calotropis gigantea against Staphylococcus aureus: Eco-Friendly Management

Calotropis gigantea (Madaar) Linn. is a potent medicinal herb that has active compounds in the form of alkaloids, glycosides, lactones, and steroids. All these active compounds have immune-modulatory and physiological roles of different types; the plant is also reported as effective in treating skin, digestive, respiratory, circulatory, and neurological disorders and was used to treat fevers, elephantiasis, nausea, vomiting, and diarrhea. In this study, the phytochemical and the zone of inhibition was measured in C. gigantea L. It proves that C. gigantea L. is more effective against Staphylococcus aureus. Aqueous extract of C. gigantea L. was extracted by the aqueous method. In the qualitative phytochemical analysis presence of various secondary metabolites were found as alkaloids, flavonoids, tannins, and saponin. In the quantitative analysis, carbohydrate was found in C. gigantea L. about concentration is found (470 μg/mL). Antimicrobial activity was also quite good against S. aureus. The study demonstrates that the C. gigantea L. contains the presence different of bioactive compounds indicated a potent antimicrobial activity of C. gigantea L. against S. aureus so that we called as eco-friendly management.

The present study was designed to find out phytochemical analysis and antimicrobial activity of leaves of Mulberry plant. The phytochemical analysis was performed on quantitative and qualitative basis. The qualitative basis indicated the presence of alkaloids, glycosides, flavonoids, steroids, tannins, saponins and Anthraquinone. The quantitative analysis showed alkaloids (40%), glycosides (20.05%), flavonoids (14%), steroids (3.5%), tannins (11.9%) and saponins (11.5%) and anthraquinone (0.5%) presence. Antimicrobial activity against bacteria has been observed in the leaves of Mulberry (Morus alba) and showed zone of inhibition, Escherichia coli (7±3.3) mm, Staphylococcus aurous (8±3.1), Bacillus subtilis (9±2.5), Pasturella multocida (8±3.1) mm.

The medicinal value of the plants is due to their chemical components that bring a definite physiological action on the human body to prevent the diseases. In this work, we investigated the antimicrobial activity of leaves’ extracts of Quercus robur L., collected from the Algerian upper highlands, on ten bacterial strains and one fungal strain known to be pathogenic. First, we performed a qualitative phytochemical analysis, and second, antimicrobial activity tests performed by agar diffusion method (disc and well) with the determination of MIC by broth macro-dilution method. Given the results, it appears that obtained macerates of Quercus robur L. were rich in bioactive phytoconstituents such as alkaloids, anthraquinones, saponins, tannins, and other components. The yield of aqueous and methanolic macerates of leaves was 8.5 ± 1.41 and 22.4 ± 4.36%, respectively. The bacterial resistance was relatively important to several antibiotics, namely, ampicillin, amoxicillin + clavulanic acid for strains of Escherichia coli and Salmonella sp. However, Staphylococcus aureus strains were resistant to fusidic acid, penicillin, and oxacillin; while Enterococcus faecalis was resistant to fusidic acid, penicillin, oxacillin, and ticarcillin. The antibacterial activity of the macerates toward tested microbial strains showed that the aqueous and methanolic macerates of the leaves were proportional to the tested concentration and active not only against Gram-positive and Gram-negative bacteria but also on the fungal species Candida albicans. The estimated MIC for Escherichia coli, Enterococcus faecalis, and Staphylococcus aureus was in the order of 10 mg/mL, which seems more effective than toward Salmonella sp., Klebsiella pneumoniae, Pseudomonas aeruginosa, and Candida albicans which were in the order of 30 mg/mL. These preliminary results confirm that the part of the studied plant had a very good antimicrobial activity that was proportional to the serial concentrations of the tested extracts.

The presented work represents phytochemical analysis, Antioxidant assay and Antimicrobial activity of Bhut jolokia pepper (Capsicum chinense Jacq) extracted by ethanol as solvent. The ethanolic extract of Bhut jolokia pepper showed the presence of terpenoids, steroids, saponins and flavonoids. Antimicrobial assay was done with varying concentration (250-1000µg/ml) of pepper extract using tetracycline as control by well diffusion method, the extract at 750µg/ml shown best inhibition zone and Staphylococcus aureus showed the highest zone of inhibition at all concentration compared to other bacteria species with maximum zone of inhibition of 27mm. The DPPH scavenging assay for antioxidant activity at 517nm showed positive activity for scavenging, scavenging increased with the concentration of extract. Thus, Bhut jolokia could be effective in treating diseases caused by Staphylococcus aureus from antimicrobial assay result.

The objective of this study to examine phytochemical analysis of sonchus oleraceus l, (asteraceae) (moliata) extracts which were extracted by four solvents, petroleum ether, chloroform, methanol and distill water and to assess their potential antibacterial against four standard bacteria gram-positive and gramnegative (bacillus subtilis staphylococcus aureu, salmonella typhi, and pscudomona aeruginosa) and antioxidant activities. (moliata) extracts using cold maceration method to extract sonchus oleraceus l, (asteraceae) locally known as (moliata) extracts, where dpph assay and paper disc diffusion assay were employed to evaluate the antioxidant and antibacterial activities respectively. The results showed that tannins, triterpenes, a sterols, flavonoids and cardiac glycosides compounds were present in the sonchus oleraceus l, (asteraceae) lives (moliata) extracts. The methanol extract of leaves showed high antioxidant potential (77± 0.03 % ) and chloroform showed low antioxidant potential activity (4±0.09) compare with propyl gallate as standard (93± 0.010 %) against dpph. Chloroform extract was effective and give moderate zone of inhibition against staphylococcus aureus. Methanol extract was effective and showed moderate zone of inhibition against staphylococcus aureus, bacillus subtilis, salmonella typhi and pseudomonas aeruginosa.

Comparative study of plant extracts crude and aqueous, methanolic and ethanolic with antibiotics, provide evidence that calotropis procera extracts has the similar antibacterial activity as these antibiotics against test pathogens i.e. Salmonella typhi and E.coli. The analysis of antimicrobial activity of aqueous, methanolic and ethanolic extract of leaves and flower of Calotropis procera was carried out in disc method and also determined MIC value at 600nm through optical density using spectrophotometer. The zone of inhibition produced by extracts was examined and compares it with zone produced by antibiotics. The effect exhibited by ethanolic extract of leaves and flower was significantly greater than the aqueous and methanolic extract of leaves and flower. Crude extracts i.e. latex, leaves, fruit and flower crude extracts. Among them, flower crude extracts shows similar zone of inhibition to test anitibiotics.while in MIC value, we made different concentration of extracts and antibiotics.i.e for crude we made 25%, 50%.75% and 100% concentration of crude juice and for the aqueous, methanolic and ethanolic we made 0.5mg/mL,1mg/mL,2mg/mL,3mg/mL,4mg/mL, 5mg/mL and 6mg/mL in Dimethyl salfoxide, and same antibiotics concentration. Aqueous leaves extracts show MIC at 0.5mg/ml against E.coli while against Salmonella it shows MIC at 1mg/ml... We also determine phytochemical analysis for presences of different compounds in crude extracts. The obtained results provide a support for the use of Calotropis procera, in traditional medicine and suggest its further advance investigation.

The aim of this study was to investigate the antibacterial effect and identify the phytochemical constituents of Moringa oleifera leaves and seeds extract on Staphylococcus aureus (S. aureus) clinical isolates using agar well diffusion method. The samples were collected from the premises of Usmanu Danfodiyo University, Sokoto. The Seeds and Leaves were collected fresh. They were extracted using methanol and ethyl acetate. Various concentrations from 100mg/ml to 500mg/ml were prepared. The test bacteria used is Staphylococcus aureus obtained from Microbiology laboratory of the Usmanu Danfodiyo University. The bacteria were re-identified using biochemical tests. The bacterial inoculums were standardized to McFarland scale 0.5. Zone of inhibition were read after 24 hours of incubation at 370C.The results of the antibacterial study revealed that the methanolic leaves extracts at 500 mg/ml had effect on S. aureus with zone of inhibition of 20mm. The methanolic seed extract have effect on S. aureus with zone of inhibition of 19.5mm. The MIC for the leave and seed extracts for Staphylococcus aureus was 250mg/ml. The MBC was 500mg/ml. The results of the phytochemical analysis revealed the presence of flavonoid, tannins, saponins, cardiac glycosides, alkaloids, volatile oil, saponin glycosides, and glycosides but anthraquinone and steroids were absent in the extracts. The zones of inhibition showed that both the methanolic and ethyl acetate extracts at 500mg/ml were active to all the tested bacteria. ANOVA and Duncan Multiple Mean Range test was used to analyze the data. Based on Duncan’s grouping, there is significant difference between the solvents and the concentrations used.

All over the world, hundreds of plants have been identified based on researchers and experimental evidence as good sources of medicinal agents. The bioactive components (phytochemicals) of both the seeds and pulp of Cola milleni were extracted using ethanol as solvent. The bioactive components detected were alkaloids, tanins, saponins, cardiac glycosides, carbohydrates, sterols, resins and terpenes while Flavonoids, anthraquinones, anthracyanides and phenol were not detected for both the seed and pulps. Antimicrobial activity of the ethanol extract (Seed and pulp) against Staphylococcus aureus, Escherichia coli and Penicillium notatum was carried out using standard techniques. Staphylococcus aureus had the highest zone of inhibition for pulp having a range of 9.7mm±0.58mm - 19.7mm±2.52mm while Penicllium notatum had the least with 0.00mm. S.aureus also had the highest zone of inhibition range of 14.3mm±2.08mm - 21.3mm±1.53mm for the seed extract while penicillium had the least inhibition range of 5.0mm±1.00mm - 5.7mm±0.58. E.coli showed the highest minimum inhibitory concentration with ethanol extract of the pulp (160mg/ml) while penicillium notatum was not reactive. The minimum inhibitory concentration of seed against penillium notatum was the highest (160mg/ml) while staphylococcus aureus showed the lowest of 40mg/ml. The antimicrobial activity is as a result of the presence of phytochemicals detected, which suggest the use of the plant for the treatment of diseases caused by these organisms. Parkia biglobosa (Jacq) is a wild leguminous plant found in North-Central zone of Nigeria with high calorific value, essential proteins, fatty acids, and vitamins. The study investigated the antimicrobial activity of crude extracts of fermented and unfermented P. biglobosa seeds on selected clinical microbial isolates namely, Candida albicans, Staphylococcus aureus, Pseudomonas aeruginosa and Escherichia coli. P. biglobosa seeds were obtained from Oja-Oba market in Ilorin, Kwara State, Nigeria. The samples were pre-treated and pulverized into powder. The extraction was achieved with acetone and water and qualitative phytochemical analysis was performed following standard procedures. The antimicrobial activity of the extracts against the isolates was determined by agar well diffusion method. Qualitative phytochemical screening of the crude extracts showed the presence of tannins, alkaloid, flavonoid, saponin and glycosides. P. aeruginosa was sensitive to the aqueous extract of fermented seeds having a zone of inhibition of 14.00±1.00mm while for unfermented seeds it was 10.00±2.00 mm at 100 mg/ml. The acetone extracts of both fermented and unfermented seeds revealed antibacterial activity against P. aeruginosa with zone of inhibition of 17.00±3.00 mm and 18.00±0.00 mm respectively. In conclusion, the crude extracts of the fermented and unfermented P. biglobosa seeds at a concentration of 75 and 100 mg/ml respectively have antimicrobial effect on the clinical isolates.

Crinum defixum Ker - Gawler is a bulbous herb which has widely used in indigenous medicine for many diseases; mainly leaf and bulb extracts are of medicinal importance. Leaf extract is used as a treatment for pimples, body-ache, dropsy, carbuncle, paronychia, leprosy, fever and diarrhoea. The bulbs are crushed and applied on to piles and itching. The present study to determine the possible phytochemicals and antimicrobial activity of various leaf extracts of Crinum defixum Ker - Gawler plant. The phytochemical screening demonstrated the presence different types compound like flavonoids, tannins, phenolic compounds, saponins and glycosides. The ethanol, methanol and crude extracts of Crinum defixum Ker - Gawler showed significant antibacterial activity against Escherichia coli, Staphylococcus aureus, pseudomonas aeruginosa, proteus vulgaris and antifungal activity against Candida albicans, Aspergillus flavus when compare to different concentrations (μg/mL). The antimicrobial potential of test compounds was determined on the basis of mean diameter of zone of inhibition around the disk in millimetres. Thus, the present findings revealed the medicinal potential of Crinum defixum Ker - Gawler to develop a drug against various human ailments.

ABSTRACTSponge is a multi-cell marine biota whose tissue and organ functions are very simple. Sponges have considerable potentially in producing active compounds that can be used in the pharmaceutical world. This study aimed to determine the presence of antimicrobial activity from extracts and fractions of the Stylissa carteri Sponge on against the microbes of Staphylococcus aureus, Escherichia coli, and Candida albicans. The extraction process was carried out by maceration using ethanol solvent, and fractionation was carried out using methanol, n-hexane and chloroform solvents. Antimicrobial activity was carried out by disk diffusion agar method. The results showed that the crude ethanol extracts and methanol fractions of the Stylissa carteri sponge actively inhibited the growth of Staphylococcus aureus, Escherichia coli, and Candida albicans microbes. Keywords: Stylissa carteri, antimicrobial, Staphylococcus aureus, Escherichia coli, Candida albicans.  ABSTRAKSpons merupakan biota laut multi sel yang fungsi jaringan dan organnya sangat sederhana. Penelitian ini bertujuan untuk menentukan adanya Aktivitas Antimikroba dari ekstrak dan fraksi Spons Stylissa carteri terhadap mikroba Staphylococcus aureus, Escherichia coli, dan Candida albicans. Dilakukan proses ekstraksi dengan cara maserasi terhadap sampel menggunakan pelarut etanol, dan dilakukan fraksinasi menggunakan pelarut metanol, n-heksan dan kloroform. Aktivitas antimikroba dilakukan dengan metode difusi agar. Hasil penelitian menunjukkan bahwa ekstrak kasar dan fraksi metanol dari spons Stylissa carteri aktif menghambat pertumbuhan mikroba Staphylococcus aureus, Escherichia coli, dan Candida albicans.  Kata Kunci : Stylissa carteri, antimikroba, Staphylococcus aureus, Escherichia coli, Candida albicans.
