<p class="BodyAbstract">Avocado leaves (Persea americana Mill.) have many benefits in our health because the leaves contain many secondary metabolites. One of the secondary metabolites in avocado leaves is quercetin. Quercetin is the largest compound of the flavonols group in which percentage of quercetin and its glycosides is 60-75% of total flavonoids. The purpose of this study was to isolate quercetin compounds from ethyl acetate fraction of ethanol extract of avocado leaves and to determine the presence of quercetin compounds in the avocado leaves. The leaves were extracted by maceration using 95% ethanol and fractionated with ethyl acetate. Identification of quercetin compounds using liquid chromatography-mass spectrometry (LC-MS). The results of this study indicated that avocado leaf extract contained quercetin compounds with a retention time of 5.83 minutes with molecular weight m/z = 300.50-301.50 [M-H]<sup>+</sup>.</p><p class="BodyAbstract"> </p><p class="BodyAbstract">Keywords: Avocado leaves, quercetin, LC-MS <strong></strong></p><p class="BodyAbstract"><strong> </strong></p><p class="BodyAbstract">Daun alpukat (Persea americana Mill.) banyak memiliki kegunaan dalam kesehatan karena mengandung banyak metabolit sekunder. Salah satu metabolit sekunder yang terkandung dalam daun alpukat adalah kuersetin. Kuersetin adalah senyawa kelompok flavonol terbesar yang mana kuersetin dan glikosidanya berada dalam jumlah sekitar 60-75% dari flavonoid. Tujuan dari penelitian ini adalah untuk mengisolasi senyawa kuersetin dari fraksi etil asetat dari ekstrak etanol daun alpukat dan untuk mengetahui adanya senyawa kuersetin dalam daun alpukat. Daun alpukat diekstraksi dengan metode maserasi menggunakan pelarut etanol 95% dan difraksinasi dengan etil asetat. Identifikasi senyawa kuersetin dengan menggunakan kromatografi cair-spektrometri massa (KC-SM). Hasil dari penelitian ini menunjukkan bahwa dalam ekstrak daun alpukat mengandung senyawa kuersetin dengan waktu retensi 5,83 menit dengan berat molekul m/z= 300,50-301,50 [M-H]<sup>+</sup>.</p><p> </p><p>Kata kunci<strong>: </strong>Daun alpukat, kuersetin, KC-SM</p>

 Objective: The objective of this study is to evaluate the antioxidant activity of Tarenna polycarpa (Miq.) koord. Ex Valenton. Leaf extract and fractions.Methods: Antioxidant activity was examined by DPPH method.Results: Ethanol extract, n-hexane fraction, and ethyl acetate fraction with DPPH assay measured as half maximal inhibitory concentration were 55.21, 109.73, and 42.04 μg/mL, respectively.Conclusions: The results reveal that T. polycarpa extract and fractions have strong antioxidant potential. Our further study is to isolate compounds responsible for antioxidant components.

Objective: The objective of the study was to evaluate brine shrimp lethality of Blighia sapida stem-bark extract and its fractions and identify the bioactive constituents in the ethyl acetate fraction (EAF) using gas chromatography-mass spectrometry (GC-MS) technique. Methods: The ethanol extract (EE) and its fractions were subjected to lethality assay, and GC-MS analysis of EAF was carried out. Results: The lethality test showed a concentration-dependent mortality rate in the brine shrimp nauplii for the EE and its fractions. GC-MS analysis of EAF of the extract revealed the existence of 13 peaks of the GC-MS chromatogram with only one prominent compound, n-hexadecanoic acid (peak area of 10.13%). Conclusion: The result revealed the presence of 13 bioactive components in the EAF of the extract, the majority of which have been reported for different biological activities, hence, justifies the use of the plant in the treatment and management of different diseases ethnomedicinally.

This study was aimed at isolating and identifying secondary metabolites contained in the ethyl acetate fraction of parasitic leaves (Dendrophthoe falcata (L.f)) Ettingsh which attaches to the stem of Mindi plants (Melia azedarach). The subjects in this study were leaves of parasitic plants on Mindi plants. While the objects in this study were secondary metabolites from the ethyl acetate fraction of the leaves of parasite plants attached to the mindi plant. The research method was carried out by extracting macerated leaves of D. falcata with ethanol solvents. The ethanol extract obtained was partitioned using n-hexane, chloroform, and ethyl acetate. Ethyl acetate fraction was separated by gravity column chromatography (CCG). The relatively non-polar fraction that has shown a single stain is tested for purity using 3 different types of solvents and identified using UV-Vis, IR, and GC-MS. The results showed that the compounds that could be isolated and identified from the relatively non-polar ethyl acetate fraction of D. falcata leaves were stigmast-5-en-3β-ol with an abundance of 6.49%, having a molecular weight of m / z 414 with base peak 55. Subjek dalam penelitian ini adalah daun tumbuhan benalu pada tanaman mindi. Sedangkan objek dalam penelitian ini adalah senyawa metabolit sekunder dari fraksi etil asetat daun tumbuhan benalu yang menempel pada tanaman mindi. Metode penelitian dilakukan dengan ekstraksi maserasi daun D. falcata dengan pelarut etanol. Ekstrak etanol yang diperoleh dipartisi menggunakan n-heksana, kloroform, dan etil asetat. Fraksi etil asetat dipisahkan secara kromatografi kolom gravitasi (KKG). Fraksi relatif non polar yang sudah menunjukkan noda tunggal diuji kemurniannya menggunakan 3 jenis pelarut yang berbeda dan diidentifikasi menggunakan UV-Vis, IR, dan GC-MS. Hasil penelitian menunjukkan senyawa yang dapat diisolasi dan diidentifikasi dari fraksi etil asetat relatif nonpolar daun D. falcata adalah stigmast-5-en-3β-ol dengan kelimpahan 6,49 %, memiliki berat molekul m/z 414 dengan base peak 55. Senyawa sterol merupakan senyawa yag lazim ditemukan dalam tumbuhan benalu.

One chemichal component that is often reported to have pharmacological activity against treatment or prevention of disease is flavonoids which can be found in gedi hijau leaves. Staphylococcus aureus is the main cause of infection in humans which can cause various severe infections. This study aims to determine the total flavonoids content, minimum inhibitory concentration (MIC). To determine the correlation between total flavonoids and antibacterial activity of extract and fractions (Abelmoschus manihot L.), The method of this study was done by obtaining the results of total flavonoid content, testing the antibacterial activity by dilution using a positive control of ciprofloxacin. The results showed the highest total flavonoid content in ethyl acetate fraction, and the best antibacterial activity in ethyl acetate fraction. Correlation between total flavonoid content from extract and fraction (Abelmoschus manihot L.) with antibacterial activity against Staphylococcus aureus bacteria on ethyl acetate fraction, n-hexane fraction, and ethanol extract at a concentration of 1000 µg / mL had good values with kneeling significance values according to 0.012 (p <0.05), 0.034 (p <0.05), and 0.023 (p <0.05) with total flavonoid content affecting how much antibacterial activity is 40% for ethyl acetate fraction, 35% fraction n -heksane and 33% ethanol extract. Keywords         : Antibacterial, Total Flavonoids, Staphylococcus aureus, Abelmoschus manihot L. ABSTRAK Salah satu komponen kimia yang sering dilaporkan memiliki aktivitas farmakologi terhadap pengobatan atau pencegahan penyakit adalah flavonoid yang dapat ditemukan pada daun gedi hijau. Staphylococcus aureus merupakan kuman penyebab utama infeksi pada manusia yang dapat menyebabkan berbagai infeksi berat. Penelitian ini bertujuan menentukan kandungan flavonoid total, aktivitas antibakteri dan korelasi antara flavonoid total dengan aktivitas antibakteri dari ekstrak dan fraksi (Abelmoschus manihot L.). Metode penelitian ini dilakukan dengan mendapatkan hasil kandungan flavonoid total, uji aktivitas antibakteri dengan cara dilusi menggunakan kontrol positif ciprofloxacin. Hasil penelitian didapatkan kandungan flavonoid total tertinggi pada fraksi etil asetat, dan aktivitas antibakteri paling bagus pada fraksi etil asetat. Korelasi antara kandungan flavonoid total dari ekstrak dan fraksi (Abelmoschus manihot L.) dengan aktivitas antibakteri terhadap bakteri Staphylococcus aureus pada fraksi etil asetat, fraksi n-heksane, dan ekstrak etanol pada konsentrasi 1000 µg/mL memiliki nilai yang baik baik dengan nilai signifikansi bertutut-turut 0,012 (p <0,05), 0,034  (p <0,05), dan 0,023 (p <0,05) dengan kandungan flavonoid total mempengaruhi seberapa besar aktivitas antibakteri yaitu 40% untuk fraksi etil asetat, 35% fraksi n-heksane dan 33% ekstrak etanol.Kata kunci          : Antibakteri, Flavonoid Total, Staphylococcus aureus, Abelmoschus manihot L.

This study aims to find the treatment of diabetes using natural materials by exploring plants in the province of East Nusa Tenggara. his research was conducted out by extracting the Strychnos nitida G.Don stem using a method of maceration by ethanol 70%. Ethanol extract was then fractionated using n-hexane and ethyl acetate. Simplicia from maceration and fractionation results were then tested for antioxidant activity, α-glucosidase inhibition activity and identification of active compounds. The results showed that ethyl acetate fraction had the lowest IC50 value of 86.83 μg / ml. Results of the α-glucosidase activity test showed that ethyl acetate fraction and n-heksan fraction at 900 ppm had the highest percentage of inhibition of 34.23% and 33.89%. Identification using LCMS/MS method showed that ethyl acetate fraction consist of Benzenemethamine, N, N-dioctyl- as an antioxidantcompound and compound 24-methyl-5-cholestone-hexol as an antidiabetic compound. From the results of this study, we concluded that the extract of kayu ular Strychnos nitida G.Don stem has inhibition activity toward α-glucosidase enzyme.

This study is aimed at determining antibacterial activity from ethanol extracts and the most active fraction of cassava leaves against clinical isolates of Staphylococcus epidermidis and Propionibacterium acnes. Research carried out by the experimental method involved determination of plants, extraction with maceration method, fractionation with liquid-liquid extraction, antibacterial activity testing of extracts and fractions by agar diffusion method, determination of most active fraction from the extract, and minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) testing of most active fraction by microdilution method. The results showed that ethanol extracts of cassava leaves had antibacterial activity against both bacteria with the most active fraction indicated by ethyl acetate. MIC values of ethyl acetate fraction against S. epidermidis were in the concentration range of 2.5%–5.0% (w/v) and against P. acnes were in the concentration range of 1.25%–2.5% (w/v). The MBC value of ethyl acetate fraction against S. epidermidis was at a concentration of 5% (w/v), while P. acnes was at a concentration of 2.5% (w/v). From the results of this study, it can be concluded that the ethanol extract of cassava leaves (Manihot esculenta Crantz) has antibacterial activity against clinical isolates of Staphylococcus epidermidis as well as on Propionibacterium acnes. The fraction with the best activity from the ethanol extract of cassava leaves to the two test bacteria was shown by ethyl acetate fraction. It is suggested that cassava leaves are possible to be developed into standardized antiacne herbal.

ABSTRACT Background: Recent studies showed that Interleukin-8 (IL-8), activated cytokine immune response which plays an important role in the development of acute and chronic gastritis. Harmless anti-inflammatory therapeutic alternatives have been proposed, for example, the consumption of Sechium Edule Jacq. Swartz (chayote). Antioxidant (flavonoid) and cell regeneration (alkaloid) agents were found in chayote. This study aimed to determine the effect of chayote Sechium Edule Jacq. Swartz extracts on the level of IL-8 in Wistar rats with aspirin- induced gastritis. Subjects and Method: This was a randomized controlled trial (RCT) conducted at the laboratory of Mathematics and Natural Science, Universitas Sumatra Utara from January to February 2020. A total of 35 male Wistar rats was selected for this study and randomly allocated into 7 groups: (1) Negative control; (2) Positive control; (3) 100 mg/ kg BW chayote ethanol extract ; (4) 200 mg/kg BW chayote ethanol extract; (5) 100 mg/ kg BW chayote ethyl acetate fraction; (6) 200 mg/kg BW chayote ethyl acetate fraction; and (7) 20 mg omeprazole. The rats in positive control and treatment groups were induced with aspirin (200mg/ kg BW). The negative control group received no intervention. The dependent variable was level of IL-8 measured by ELISA. The independent variables were treatment status. The data were analyzed by One Way Anova and post hoc test. Results: The mean differences of IL-8 level were not statistically significant between study groups (p= 0.327). Mean of IL-8 level was higher in positive control group (Mean= 160.80; SD= 6.90) than in negative control group (Mean= 141.20; SD= 10.98). The lowest IL-8 level was in 100mg/ kg BW chayote ethanol extract group (Mean= 149.94; SD= 40.4), followed by 200mg/ kg BW (Mean= 152.4; SD= 30.73) and 100mg/ kg BW (Mean= 164.60; SD= 25.04) chayote ethyl acetate fraction groups, 20 mg omeprazole group (Mean= 170.60; SD= 21.58), and 200 mg/ kg BW chayote ethanol extract group (Mean= 176.80; SD= 10.98). Conclusion: The low dose (100mg/ kg BW) chayote ethanol extract has the most potential antiinflammation effect on in vitro gastritis with the lowest IL-8 level of all doses of chayote ethanol extract, chayote ethyl acetate fraction, and omeprazole. Kasturi (Mangifera casturi) is a typical plant of South Kalimantan which has only been used for consumption because of its distinctive taste and aroma. Mangifera indica, which is in the same genus as Mangifera casturi, shows antioxidant, antibacterial, antifungal, and antidiarrheal activity, so it is suspected that this plant has the same activity. Crude methanol extract from the meat of Casturi mango was fractionated using n-hexane, ethyl acetate, and methanol-water. Testing for antibacterial activity and minimum inhibitory concentration (MIC) were carried out using the disk diffusion method (Kirby-Bauer). The positive control used was Chloramphenicol because it has an antibacterial range that is rather broad and the negative control used was 10% DMSO. The fraction that had the greatest antibacterial activity was the ethyl acetate fraction; with the formation of a clear zone of 7.98mm against Pseudomonas aeruginosa and 7.52mm against Bacillus cereus, with a minimum inhibitory concentration of 2mg/mL. The next test was a cell leakage analysis which revealed an increase in absorbance at MIC 1 and MIC 2. The Gas Chromatography – Mass Spectrometry (GC-MS) results from the ethyl acetate fraction revealed the presence of phenolic, flavonoid, and glycoide content, with the highest compound concentration of 5-Hydroxymethylfurfural. Mangifera indica yang masih satu genus dengan Mangifera casturi menunjukkan aktivitas antioksidan, antibakteri, antijamur, dan antidiare sehingga diduga tanaman ini memiliki aktivitas yang sama. Ekstrak kasar metanol daging buah mangga kasturi difraksinasi dengan menggunakan n-heksan, etil asetat, dan metanol-air. Pengujian aktivitas antibakteri dan kadar hambat minimum dilakukan dengan menggunakan metode difusi cakram (Kirby- Bauer). Kontrol positif yang digunakan adalah Chloramfenikol karena memiliki spektrum antibakteri yang cukup luas dan kontrol negatif yang digunakan adalah DMSO 10%. Fraksi yang memiliki aktivitas antibakteri terbesar pada penelitian ini adalah fraksi etil asetat dengan terbentuknya zona bening sebesar 7,98mm terhadap Pseudomonas aeruginosa dan 7,52mm terhadap Bacillus cereus dengan kadar hambat minimum 2mg/mL. Uji selanjutnya adalah uji kebocoran sel yang menyatakan adanya peningkatan absorbansi pada KHM 1 dan KHM 2. Hasil pemeriksaan Gas Chromatography – Mass Spectrometry (GCMS) dari fraksi etil asetat menyatakan adanya kandungan fenolik, flavonoid, dan glikosida dengan senyawa tertinggi yaitu 5-Hydroxymethylfurfural. Disease cancer caused by abnormal growth of tissue where there has been an error, fast and out of control. Judging from the fact of gender, more than 270,000 women die every year caused by cervical cancer. To inhibit the growth of cancer cells, a compound is needed that causes the cell cycle to stop so that the ability of cell proliferation decreases. Alkaloid compounds can inhibit proliferation through oxidative inhibition processes that can cause cancer. Mangrove plants have potential as anticancer, antimicrobial, and antioxidant. The content of chemical compounds found in mangroves are flavonoids, steroids, alkaloids, phenolites, saponins and tannins. These compounds show high antioxidant activity and are shown to have a real relationship with the properties of the material's bioactivity against cancer cells. One of the mangrove species is Rhizophora apiculata. The purpose of this study was to determine the IC50 value produced by Rhizophora apiculata mangrove leaf extract on HeLa cell viability and to see the effect of Rhizophora apiculata mangrove leaf extract dosage on HeLa cell viability. The method used in this research is the experimental method. The research parameters included yield, proximate test, phytochemical test, toxicity test, total phenol test, cytotoxicity test and LC-MS test. The experimental design used was a simple and complex completely randomized design (CRD) with the Tukey test.The results of this study showed that the highest yield was in the ethanol extract of 5.91%, while the n-hexane and ethyl acetate extracts respectively had yields of 1.18% and 1.31%. The results of the proximate test on the water content of leaves and powder were 64.53% and 13.86%, respectively, the results of the ash content in the leaves and powder of Rhizophora apiculata were 3.94% and 8.41%, respectively. while the water content in the extract obtained the highest yield in the ethanol extract of 21.42%, while the n-hexane extract and ethyl acetate extract were 11.08% and 15.42%, respectively. For phytochemical results, it was found that n-hexane extract only contained alkaloids, flavonoids and steroids. Ethyl acetate extract contains steroid compounds. Meanwhile, the ethanol extract contains the most bioactive compounds, namely saponins, flavonoids, tannins and triterpenoids. The toxicity test using the Brine Shrimp Lethality Test (BSLT) method resulted in the lowest IC50 of ethanol extract at 49.45 ppm while the n-hexane and ethyl acetate extracts were 251.63 ppm and 920.45 ppm respectively. In the total phenol test, the n-hexane extract was 66.79 mg GAE / 100 gr, 222.97 mg GAE / 100 gr ethyl acetate extract and 929.04 mg GAE / 100 gr ethanol extract. HeLa cell cytotoxicity testing using the MTT method (3- (4,5-dimethiltiazol-2-yl) -2,5-dipheniltetra zolium bromide) assay resulted in the highest cell viability value at a dose of 125 ppm of 46.97%. As for the doses of 250 ppm, 500 ppm 1000 ppm, and 2000 ppm resulted in a percentage of viability of 42.95% 37.70% 35.82% and 32.12%, respectively. The IC50 value of Rhizophora apiculata leaf extract was 64.42 ppm. This value indicates that the Rhizophora apiculata extract is toxic to HeLa cells.

Background: Binjai (Mangifera caesia) is a herb derived from South Kalimantan possessing antioxidant properties which promote wound healing inhibiting oxidation radicals. The natural antioxidants present in binjai leaves can be extracted by fractionation. Purpose: This study aimed to analyze the antioxidant activity of ethyl acetate fraction in 96% ethanol extract of binjai leaf. Methods: The study constituted a pure experimental study incorporating a post-test design with only random sampling technique consisting of two groups, namely; an ethyl acetate fraction as the treatment group and ascorbic acid as the positive control group. The leaves were treated in accordance with the soxhlet method and subsequently fractionated to extract ethyl acetate fraction. This was used to measure antioxidant activity with DPPH radical damping method using a UV-Vis spectrophotometer. A linear regression calculation was performed with a standard curve to quantify the IC50 value, before the ethyl acetate fraction underwent a qualitative test of secondary metabolite. Results: An independent t-test indicated significant differences between groups, an average value of IC50 in ascorbic acid of 13.812 ppm with 0.996 linearity and a fraction of ethyl acetate 38.526 ppm with a linearity of 0.999. In contrast, at this linearity value ascorbic acid and ethyl fraction acetate demonstrate a very high linear connection between concentration and inhibition. A secondary metabolite test conducted on the ethyl acetate fraction produced positive results for flavonoid, tannins, and phenol. Conclusion: Based on the IC50 parameters, the fraction of ethyl acetate in 96% ethanol extract of binjai leaf produces very strong antioxidant activity in the content of the compounds in the fraction, namely: flavonoid, tannins and phenol.
