The aim of this research was to know the effectiveness of guava filtrate supplementation in coconut water- egg yolk dilution on quality of liquid semen stored at 5oC of Bali cattle. Semen collected from a five year old Bali cattle using artificial vagina. Semen of good quality were kept in six tubes based on treatment then stored at 5oC. Treatments of the research were P0 : coconut water 80% + egg yolk 20% without guava filtrate; P1 : coconut water 80% + egg yolk 20% + 0.8% guava filtrate; P2 : coconut water 80% + egg yolk 20% + 0.9% guava filtrate; P3 : coconut water 80% + egg yolk 20% + 1.0 % guava filtrate; P4 : coconut water 80% + egg yolk 20% + 1.1 % guava filtrate and P5 : coconut water 80% + egg yolk 20% + 1.2 % guava filtrate. Each treatment was replicated 8 times making 48 experimental units. No. 36a/E/KPT/201621PENDAHULUAN Salah satu solusi yang dapat digunakan untuk pengembangan program Inseminasi buatan (IB) secara cepat dan mudah pada sapi bali adalah penggunaan semen cair. Penggunaan semen cair dapat meningkatkan kinerja IB pada sapi bali di Nusa Tenggara Timur (NTT). Keunggulan lain semen cair dapat diproduksi menggunakan bahan pengencer herbal berbasis bahan lokal dan peralatan yang sederhana serta mudah diperoleh dan tidak tergantung dengan persediaan nitrogen cair. Hasil akhir dari metabolisme spermatozoa adalah terbentuknya radikal bebas berupa derivat oksigen di antaranya adalah single1 oksigen (1O2), tripel1 oksigen (3O2), superokside anion (O2-), hidroksil radikal (OH) dan nitrit oxide (NO-) yang semuanya disebut radical oksigen species (ROS). Single1 oksigen dapat merusak ikatan rangkap pada asam lemak sehingga dapat menyebabkan kerusakan Deoxyribo Nuclead Acid (DNA) dan protein. Single1 oksigen bila bereaksi dengan asam amino histidin akan membentuk enzim yang dapat menyebabkan denaturasi protein. Kerusakan spermatozoa pada penyimpanan suhu 5%C akibat radikal bebas dan cold shock inilah merupakan penyebab utama disfungsi semen (Sharma et al., 2000). Oksidasi fosforilasi yang terganggu menyebabkan peningkatan radikal bebas dalam semen. Kadar radikal bebas yang terganggu menyebabkan peningkatan radikal bebas dalam semen. Kadar radikal bebas yang tinggi dalam sel dapat mengoksidasi lipid, protein dan DNA. Lipid membran plasma semen memiliki fosfolipid dengan kadar yang tinggi menyebabkan semen rentan terhadap radikal bebas (Sanoeka dan Kurpisz, 2004). Antioksidan bertindak mengikat asam lemak tak jenuh dan mencegah terjadinyareaksi berantai. Pada proses penyimpanan semen akan terjadi kerusakan membran plasma spermatozoa akibat terbentuknya perioksidasi lipid. Antioksidan-pemutus rantai seperti yang terkandung dalam jambu biji dapat menghambat perioksidasi lipid dalam membran melalui radical peroxyl (RO) dan alkoxyl (ROO) pengurai. Pengunaan jambu biji yang difilter dalam pengencer air kelapa kuning telur dapat menjaga kualitas spermatozoa (motilitas, keutuhan akrosom, viabilitas dan morfologi spermatozoa) semen cair sapi bali selama penyimpanan pada suhu 5%C. Dosis jambu biji yang difilter yang terbaik dalam pengencer air kelapa kuning telur, akan terbaik pula dalam mempertahankan kualitas spermatozoa sampai tujuan IB. Adapun tujuan penelitian ini adalah menguji berbagai level pemberian filtrat jambu biji (FJB) dalam pengencer air kelapa kuning telur terhadap motilitas, viabilitas, membran plasma utuh (MPU) dan tudung akrosom utuh (TAU) spermatozoa sapi bali yang disimpan pada suhu 5%C.METODE PENELITIAN Penelitian ini telah dilakukan di Laboratorium Reproduksi milik Yayasan Wiliams dan Laura yang berlokasi di Tilong, Desa Oelnasi, Kec. Kupang Tengah, Kab. Kupang, Nusa Tenggara Timur, dan berlangsung selama delapan bulan. Materi yang digunakan dalam penelitian ini adalah semen sapi bali yang ditampung dari satu ekor sapi bali jantan berumur lima tahun milik Yayasan Williams dan Laura yang telah dilatih, memiliki performans yang baik, dan organ reproduksi normal. Pakan yang diberikan adalah hijauan berupa rumput dan legum dan pemberian konsentrat secukupnya (dedak padi dan jagung giling).and P5 : 9%, 21.25%, 17.56% and 19.30%. Result of statistical analysis showed that there were a significant effect (P<0.05) between treatment on motility, viability, MPU and TAU of spermatozoa of Bali cattle till the third day of storage. It can be concluded that the supplementation of guava filtrate 0.9% in dilution of coconut water 80% - egg yolk 20% had been able to maintain motility, viability, MPU and TAU of spermatozoa of Bali cattle till the third day of storage at 5oC.

ABSTRAKAir kelapa muda varietas viridisdapat dijadikan pengencer aletrnatif semen cair bagi program IB di daerah minim sarana semen beku. Tujuan penelitian ini untuk menguji pengaruh penggunaan air kelapa muda viridissebagai bahan pengencer terhadap kualitas semen cair kambing Boer setelah didinginkan. Dilaksanakanselama 3 bulan di Laboratorium Fakultas Peternakan UBUnit SumberSekar,Malang. Metodenya yaitu eksperimen. Semen dari  3 pejantan Boer umur 3-5 tahun, dikoleksi seminggu sekali dengan VB. Air kelapa mudaviridis umur 5-7 bulan serta tris aminomethane sebagai kontrol. Didesain menggunakan Rancangan Acak Kelompok (RAK) dengan 2 perlakuan yaitu P0 (tris aminomethane + 10% KT) dan  P1 (air kelapa muda viridis + 10% KT) masing-masing diulang 10 kali. Data dianalisis dengan analisis Ragam (Anova) dengan software Genstat 18. Variabelnya yaitu motilitas individu, viabilitas dan abnormalitas. Hasil penelitian yaitu motilitas individu pada P1bertahan sampai 4 hari (40,5± 24,3%), viabilitas terbaik sampai hari ke-5 (42±24,6%), abnormalitas terendah di hari ke-7(1,31± 0,6). Kesimpulannya, Pengencer air kelapa muda viridis dapat mempertahankan kualitas semen cair kambing Boer selama 4 hari untuk motilitas dan 5 hari untuk viabilitas.Kata Kunci:pengencer, air kelapa, varietas viridisABSTRACTYoung viridis coconut water could be used as an alternative to liquid semen diluent for artificial insemination program in the area with limited facility for frozen semen production. This study evaluated the use of young coconut water as a diluent on liquid semen quality of Boer goat after cold storage. This study was carried out for 3 months at Sumber Sekar Laboratory, Faculty of Animal Husbandry, University of Brawijaya, Malang. The semen was collected from 3 Boer bucks aged at 3 to 5 years old. The semen collection was done once a week with the aid of artificial vagina. The diluents used were young Viridis coconut (5 to 7 months old) and tris aminomethane. The method used was an experiment in a randomized block design with 2 treatments and 10 replicates. The treatments used were T0: tris aminomethane + 10% egg yolk (control) and T1:  young Viridis coconut water + 10% egg yolk. Data were analyzed by analysis of variance using Genstat 18 software. The variables measured were sperm individual motility, viability, and abnormality. The results showed that the sperm individual motility in T1 survived up to 4 days (40.5± 24.3%), the best viability at 5 days (42.0±24.6%),  while the lowest abnormality at 7 days (1.31±0.6). It could be concluded that: 1. Tris aminomethane diluent has higher quality with the storage length up to 9 days, 2. Young Viridis coconut water diluent could preserve liquid semen quality of Boer goat up to 4 days for sperm motility and 5 days for sperm viability.Keywords: diluents, coconut water, viridis variety

The objective of this study was to examine the effect of seminal plasma on viability of Boer goat spermatozoa and effectiveness of sugar palm juice as an alternative extender during preservation at 5ºC. Semen of two Boer goats were collected using an artificial vagina. Fresh semen were evaluated and divided in equal volume into four tubes. Semen in the first and second tubes diluted with 80% sugar palm juice + 20% egg yolk (P1) and Andromed (P2), respectively. Semen in the third and fourth tubes were centrifuged with 3,000 RPM for 20 minutes, and the supernatant removed. diluted with 80% sugar palm juice + 20% egg yolk (P3) and Andromed (P4), respectively. Diluted-semen were preserved in refrigerator at 5oC, and quality of the spermatozoa including motile spermatozoa (MS), live spermatozoa (LS), and intact plasma membrane (IPM) were evaluated every day for four days. Results of this study showed that at day-2 preservation, mean percentages of MS, LS, and IPM for P2 (72, 83.4, and 83.4%), P3 (72, 82.6, and 82.2%), and P4 (72, 83, 83.8%) were significantly (P<0.05) higher than P1 (3, 24.8, and 25.2%). At day-3 preservation, mean percentages of MS, LS, and IPM for P2 (57, 65.6, and 69.6%) was significantly (P<0.05) higher than P3 (21, 34.8, and 31.8%), P4 (22, 33.6, and 31.2%), and P1 (0, 0, and 0%). In conclusion, semen of Boer goat to be preserved with extender containing egg yolk should be removed seminal plasma. Sugar palm juice containing egg yolk could be used as an extender for Boer goat semen, but should be applied in the AI program immediately after the semen is diluted.

Dimethyl Sulfoxide (DMSO) is one type of cryoprotectant which has a low molecular weight so that it is easier to enter cells when cryopreservation. The purpose of this study was to explore the optimal concentration of DMSO in modified coconut water (mCW) extender that were able to maintain frozen sperm quality of bali bulls. Semen was collected from two four-year old bali bulls by artificial vagina. Good quality semen diluted with mCW (young coconut water + 20% egg yolk + 7.5 % moringa leaf extract) and supplemented by 3, 5, or 7% DMSO. Semen was filled into 0.25 ml ministraw, and was incubated in a refrigerator at 5°C for four hours, frozen on the surface of liquid nitrogen for 10 minutes and then dipped into liquid nitrogen. The quality of post thawing sperm was measured 24 hours later by placing the ministraw of frozen semen into water at 37oC for 30 seconds. Data were analyzed by analysis of variance and continued with Duncan test. Postthawing observations showed that bali bulls sperm cryopreserved at 3% DMSO yielded higher motility and viability (p<0.05) i.e. 36 and 44.15%, than DMSO 5% i.e. 18 and 23.65%, and DMSO 7% i.e. 7 and 12.62%. The recovery rate of sperm cryopreserved at 3% DMSO was also higher (p<0.05) than DMSO 5 and 7%, successively 45.65, 23.06, and 8.86%. The results of this study concluded that the optimal concentration of DMSO in mCW diluent to maintain frozen sperm quality of bali bulls was 3%.

This study aims to determine the quality of Bali cattle spermatozoa diluted using sugarcane juice-egg yolk. This research was carried out in the stables and Laboratory of the Faculty of Agriculture, University of Timor in August-September 2020. Semen was collected from an adult male bali cattle, aged ± 4.5 years in healthy condition. The method used in this study is an experimental method using a completely randomized design consisting of 4 treatments and 5 replications. Each treatment was T1: 0.075 ml semen + 0.3 ml of egg yolk sugarcane juice diluent and stored for 24 hours, T2: semen 0.075 ml + 0.3 ml of diluent for egg yolk cane juice and stored for 48 hours, T3: semen 0.075 ml + 0.3 ml of diluent for egg yolk sugarcane juice and stored for 72 hours, T4: semen 0.075 ml + 0 ,3 ml of egg yolk sugarcane juice diluent and stored for 96 hours. The variables measured were individual motility, viability, spermatozoa abnormalities, and semen pH. The results showed that the T4 treatment on a shelf life of 96 hours still showed a good average value with individual spermatozoa motility values of 45%, spermatozoa viability of 77.3%, spermatozoa abnormalities of 12.5% and semen pH 6.6. It can be concluded that the use of cane juice-egg yolk diluent has a positive impact because it is able to maintain the viability of spermatozoa for 96 hours.

There is little evidence in the veterinary literature to determine if antihistamines are as effective as glucocorticoids in the management of atopic dermatitis in dogs. Two randomised control trials directly compared an antihistamine, fexofenadine, to methylprednisolone although one was a small pilot study and only used four dogs in each treatment group. The third study was a cross-over placebo-controlled trial testing a variety of antihistamines and prednisone with limited reporting of statistical analysis of the data. None provide sufficient quality of evidence to recommend the use of antihistamines over glucocorticoids and further comparative study is needed.

Objective: The cleaning and shaping of root canals is undeniably a step of great importance for the success of endodontic treatment. With the aim of optimizing the time and quality of this step, numerous instruments of different materials and shapes have been developed over the years. One of these instruments was nickel-titanium files, which have greater flexibility and resistance, allowing a better prognosis for endodontic treatment. In this study, two endodontic files, Reciproc Blue and V-File, were compared with the aim of evaluating their performance after the third use. Materials and methods: 60 acrylic blocks wrapped with laminated paper and filled with Nankin ink were used. Instrumentation was performed with an endodontic engine, by an experienced operator. After instrumentation, the blocks were photographed so that the photographs were superimposed. Results: After statistical analysis, it was found that there was no significant difference in the performance of the two groups after the third use. Conclusion: There was no significant difference between the two studied groups, and both are safe for the preparation of curved canals, but it is necessary that the manufacturer's recommendation is followed, regarding the number of canals instrumented with the same file.

The objective of this research was to examine the effectiveness of coconut water with various egg yolk concentrations in maintaining the viability of epididymal spermatozoa of crossbreed cattle preserved at 5oC. Five testis with epididymides of crossbreed cattle were obtained from slaughterhouse. Epididymal spermatozoa was collected by the combination of slicing, flushing and tissues pressure methods of cauda epididymides with saline solution (0.9% NaCl). Collected-spermatozoa was equally divided in volume into four tubes and diluted with lactose extender containing 20% egg yolk (control), 90% coconut water + 10% egg yolk (CWEY10), 85% coconut water + 15% egg yolk (CWEY15), and 80% coconut water + 20% egg yolk (CWEY20), repectively. Diluted-spermatozoa was stored in refrigerator at 5oC. Quality of dilutedspermatozoa including percentages of motile spermatozoa (MS), live spermatozoa (LS), spermatozoa with intact plasma membrane (IPM) were evaluated every day during four days of storage. Data were analyzed by using completely randomized design with four treatments and five replicates. Means were compared with significant difference test at 0.05 significant level. Results of this study showed that mean of spermatozoa concentration, percentage of MS, percentage of LS, percentage of spermatozoa abnormal, and percentage of IPM of crossbreed cattle fresh epididymal spermatozoa were 1,414 million cell/ml, 72%, 85%, 9%, and 90%, respectively. At day-4 of the storage, percentages of MS, LS, and IPM of control (43, 52.2, 59.2%) and CWEY20 (42, 52, 59%) were significantly (P<0.05) higher than CWEY10 (33, 45.4, 52.8%) and CWEY15 (37, 50, 54.6%). In conclusion, lactose and CWEY20 extenders could be maintaining the quality of epidydimal spermatozoa of crossbreed cattle for three days preservation at 5oC and two days for CWEY10 and CWEY15.

The aim of this research was to evaluate the Bali cattle semen quality during cryopreservation with different types of extenders in term of live, total motility, progressive motility, and abnormality of post-thawed bull sperms. The treatments were AndroMed® (T0), Tris-based egg yolk diluent (T1), Tris-based coconut water diluent (T2), Coconut water egg yolk diluent (T3). Bulls’ semen was collected from two adult Bali cattle maintained at the semen production facility at Bali Artificial Insemination Center, Tabanan Bali. The age of the bulls were 6 years old. Sperm live, total motility, progressive motility, and abnormality were analyzed with computer assisted sperm analysis (CASA) post-dilution, before and after thawed. The study was replicated five times, and data were analyzed using analysis of variance (ANOVA). The results showed that AndroMed ® and Tris-based egg yolk had significantly higher sperm live, total motility, progressive motility and abnormalities of spermatozoa for post-dilution, after equilibration and post thawed than Tris egg yolk coconut water and Coconut water egg yolk diluent. It was concluded that AndroMed® and Tris-based egg yolk can be considered as the best suitable extender for Bali cattle sperm cryopreservation. Coconut water had a deleterious effect when supplemented with 20% in tris and egg yolk.

This experiment was conducted in (Zawita) area in Dohuk governorate / Iraq, and the morphological characteristics were studied by selecting four sites, four trees from each site, four sectors and, choosing twenty trees in each sector. Statistical analysis (RCBD) was done using the SPSS system. Through this study, it was found that Pinus brutia Ten. In Zawita, there is a clear effect on some morphological characteristics of the seedlings resulting from it. The first site was one of the best sites followed by the second site where he gave seedlings of good quality in terms of stem length with an average of 39.25 cm and the total weight of wet and dry seedlings with averages of 13.97 and 35.34 g respectively, and the third site was the lowest studied sites and gave seedlings of less quality than the rest of the sites where he scored Stem length averaged 23.06 cm while wet and dry weight was 7.68 and 22.17 g, respectively. As for the correlation between the variables, the relationship was positive and strong among all the variables except those between the length of the root and the rest of the variables except with the characteristic of the wet root weight. The highest correlation values were between total dry weight and total wet weight recording 0.960 and the lowest between the length of the root and the dry vegetative weight where the value was 0.070.

ABSTRAKPengencer konvensional berbasis kuning telur yang bersumber dari hewan membawa risiko kontaminasi mikroba dan menghalangi jarak pandang pemeriksaan spermatozoa di bawah mikroskop. Penelitian ini dilaksanakan` untuk menilai apakah substitusi kuning telur dengan lesitin kedelai di dalam pengencer berbasis-tris dapat mempertahankan kualitas spermatozoa kerbau lumpur pada penyimpanan 5°C selama 120 jam. Ejakulat dari 3 ekor kerbau umur 3-4 tahun dengan bobot badan 450-500 kg ditampung dengan menggunakan vagina buatan.Semen diencerkan dengan pengencer konvensional yang mengandung 20% kuning telur (kontrol) dan pengencer lesitin dengan konsentrasi1%, 2%, 3% dan 4%(perlakuan) dan disimpan pada 5°C selama 120 jam. Parameter spermatozoa dinilai pada interval 0, 72, dan 120 jam setelah semen yang diencerkan disimpan 5°C.Persentase motilitas dan spermatozoa normal tidak berbeda nyata (P<0,05) antara pengencer 1% lesitin dan kontrol. Namun pengencer 1% lesitin mampu mempertahankan kedua parameter tersebut setelah penyimpanan 5°C selama 120 jam. Persentase viabilitas, keutuhan membran dan tudung akrosom spermatozoa pada pengencer 1% lesitin dibandingkan pengencer kontrol lebih tinggi (P<0,05) dari pengencer perlakuan 2, 3, dan 4% lesitin. Disimpulkan bahwa pengencer 1% lesitin dapat menyubstitusi pengencer konvensional berbasis kuning telur untuk penyimpanan semen kerbau pada 5ºC selama 120 jam.Kata kunci: kuning telur, lesitin, pengencer, semen cair, semen kerbauABSTRACTConventional egg yolk-based diluent animal origin carries the risk of microbial contamination and interference with microscopic examination. This study was conducted to assess whether substitution of egg yolk with soy lecithin in tris-based diluents can maintain the quality of buffalo spermatozoa stored at 5°C for 120 h. Ejaculate from 3 buffaloes aged 3-4 years and body weight of 450-500 kg was collected using an artificial vagina. Semen was diluted with diluent containing 20% egg yolk (control) and diluent with concentrations of lecithin 1%, 2%, 3% and 4% (treatment). Spermatozoa parameters were assessed at intervals of 0, 72 and 120 h after the diluted semen was stored at 5°C for 120 h. The percentages of motility and normal spermatozoa did not differ significantly (P<0.05) between 1% lecithin and control diluents. However, 1% of lecithin was able to maintain both parameters after stored at 5°C for 120 h. The percentages of viability, membrane integrity and functional acrosome of spermatozoa in 1% lecithin were higher (P<0.05) than control and the other three treatment diluents. In conclusion, 1% of lecithin diluent could substitute conventional egg yolk-based diluent for the storage of buffalo semen at 5ºC for 120 h.Keywords: buffalo semen, egg yolk, lecithin, diluents, liquid semen
