Introduction: Dyslipidemia is a major health problem in Indonesia which leads to other diseases. Triglycerides (TG) is one of the increased lipid fractions which is related to dyslipidemia. Simvastatin is the main therapy in Indonesia to lower TG levels. The use of virgin coconut oil (VCO) becomes popular as a blood cholesterol-lowering agent even though the effects on TG level remains elusive. Since there is no empirical evidence VCO as a complementary therapy of simvastatin on TG levels, this experimental trial is conducted to determine the effect of simvastatin and VCO combination therapy on blood TG levels in dyslipidemic male white rats.Methods: 42 white rats (Rattus norvegicus) divided into 5 treatment groups and preliminary test of dyslipidemia. The blood was collected intracardially and TG level was measured by using spectrophotometric methods. Independent variables are simvastatin and VCO and combination therapy while the dependent variable is TG levels of white rats. The data was afterwards analyzed using Mann-Whitney U test.Results: The control group’s triglyceride plasma level, receiving standard diet, is 59±13,4 mg/dl. The VCO-only group shows 72±35,4 mg/dl while simvastatin-only group shows 61,9±22,6 mg/dl triglyceride level. Combination therapy of simvastatin and VCO shows 62,3±31,3 mg/dl (p>0,05).Conclusion: There were no significant differences between groups with control. However,the use of VCO with particular dose and duration of therapy, will significantly decrease triglyceride level in the blood as effective as simvastatin. VCO has no efficacy while used as combination therapy of simvastatin.

This research aimed to annalyse the effects of virgin coconut oil and olive oil on the length, diameter and weight of the femur bone in white male rat (Rattus norvegicus). This research used 25 white male rats and consists of 5 treatments with 5 replications. This research used the treatment P0: as a control, P1: feed with an addition of 5 ml virgin coconut oil / kg feed, P2: feed with an addition of 10 ml virgin coconut oil / kg of feed, P3: feed with an addition of 5 ml olive oil / kg feed and P4: feed with  an addition of 10 ml olive oil / kg feed. This research used a method called Completely Randomized Design (CRD). The variables measured in this research are the weight, length and diameter of white male rats. The data was analyzed using Analysis of Variance (ANOVA) at 95% level. The results showed that the growth of femur bone in male rat does not have different response after giving virgin coconut oil and olive oil. Keywords : VCO, olive oil, femur bone, rat

Dyslipidemia is an abnormal lipid metabolism which may cause fat degeneration on hepatocytes cells and elevated triglyceride serum level. Dyslipidemia can be prevented by the consumption of high antioxidant food. Noni(Morinda citrifolia) contains many antioxidant such as flavanoid, kuersetin, tannin, and saponin, which are able to prohibit the elevation of ROS.This research is aimed to find out the effect of noni(Morinda citrifolia) extract in reducing the number of hepatocyte’s cells with fat degeneration and decreasing the triglyceride level which is elevated due to high cholesterol diet induction.In this study white rats divided randomly into 4 groups, control group (K-), high cholesterol diet induced group (K+), high cholesterol diet induced with 100 mg/Kg BW noni extract group (P1), high cholesterol diet induced with 200 mg/Kg BW noni extract group (P2). The result shows that noni  extract with dose 100 mg/Kg BW and  200 mg/Kg BWcan reduce the number of hepatocytes cells with fat degeneration (p= 0,026 and p=0,027) and decrease the level of triglyceride serum (p=0,036 and p=0,010).The conclusion is noni extract with dose 100 mg/KgBW reduces effectively  the number of hepatocyte’s cells with fat degeneration and decreases the level of triglyceride serum which increase because of high cholesterol diet.

ABSTRACT This study aims to determine the effect of infusion of conjoined pumpkin leaves (Sechium edule) on reducing total blood cholesterol of male white rats (Rattus norvegicus). The subjects of this study were 15 male white rats with an average body weight of 200 grams which were divided into 5 groups, each group consisted of 3 mice. The method used is a laboratory experiment with a completely randomized design. The results were obtained from 2 measurements of blood cholesterol levels, namely measurements before and after treatment. The treatment begins with the provision of high-fat foods for 48 days. On the 49th day a blood cholesterol level was measured before treatment. Furthermore, treatment was given to each group, namely aquades in the negative control group, simvastatin in the positive control group, and squash leaves infusion with their respective doses in the dose group I (40%), the dose group II (20%), and the dose group III (10%). Measurement of cholesterol levels after treatment was carried out on day 54. Data were analyzed by Paired t-test and One Way ANOVA. The analysis showed that there were no significant differences between treatment groups. Judging from the change in average and percentage, 40% infusion dose of siamese pumpkin leaves gives the best reduction in cholesterol levels. Keywords: Cholesterol, pumpkin leaves, male white mouse infusion. ABSTRAK Penelitian ini bertujuan untuk mengetahui pengaruh pemberian infusa daun labu siam (Sechium edule ) terhadap penuruan kolesterol darah total tikus putih jantan (Rattus norvegicus). Subjek penelitian ini berupa tikus putih jantan berjumlah 15 ekor dengan berat badan rata-rata 200 gram yang dibagi dalam 5 kelompok, masing-masing kelompok sebanyak 3 ekor. Metode yang digunakan yaitu eksperimen laboratorium dengan rancangan acak lengkap. Hasil penelitian diperoleh dari 2 kali pengukuran kadar koleterol darah yaitu pengukuran sebelum dan sesudah perlakuan. Perlakuan dimulai dengan pemberian makanan tinggi lemak selama 48 hari. Pada hari ke49 dilakukan pengukuran kadar kolesterol darah sebelum perlakuan. Selanjutnya diberikan perlakuan pada tiap kelompok yaitu aquades pada kelompok kontrol negatif, simvastatin pada kelompok kontrol positif, dan infusa daun labu siam dengan dosis masing-masing pada kelompok dosis I (40 %), kelompok dosis II (20 %), dan kelompok dosis III (10%). Pengukuran kadar kolesterol sesudah perlakuan dilakukan pada hari 54. Data diananlisis dengan Paired t-test dan One Way ANOVA. Hasil analisa menunjukan tidak terdapat perbedaan yang bermakna antar kelompok perlakuan. Dilihat dari perubahan rerataan dan presentase, dosis infusa  40% daun labu siam memberikan penurunan kadar koleterol terbaik. Kata kunci : Infusa daun Labu siam, kolesterol, tikus putih jantan.

AbstrakPenyakit jantung koroner (PJK) merupakan penyebab utama kematian di berbagai belahan dunia termasuk di Indonesia. Salah satu pemicu terjadinya PJK adalah tingginya kadar kolesterol dalam darah. Buah mahkota dewa telah lama digunakan oleh masarakat untuk menurunkan kadar kolesterol dan mengobati berbagai penyakit kardiovaskuler.Telah dilakukan penelitian untuk melihat efek infus buah mahkota dewa terhadap pencegahan peningkatan kadar kolesterol pada tikus putih jantan yang diberi diet lemak tinggi (DLT). Penelitian ini menggunakan 30 ekor Rattus novergicus jantan yang berumur 2,5 – 3 bulan dengan berat badan 250 - 350 gram. Hewan coba dibagi secara acak atas lima kelompok dan diperlakukan sebagai berikut:Kelompok I diberi diet standar 555 global feed dan 1 ml aquades, kelompok II diberi diet lamak tinggi(DLT) yang terdiri dari 2% Kolesterol, 10% kuning telur itik, 18% lemak sapi dan 78% diet standar dan 1 ml aquades, kelompok III diberi DLT dan 1 ml infus yang mengandung 97 mg mahkota dewa., kelompok IV diberi DLT dan 1 ml infus yang mengandung 194 mg mahkota dewa, kelompok V diberi DLT dan 1 ml infus yang mengandung 388 mg mahkota dewa. Infus diberikan per oral setiap hari selama 56 hari. Diet dan air minum diberikan ad libitum.Pada hari ke 0, 14, 28, 42 dan 56 darah diambil melalui vena centralis ekor untuk ditentukan kadar kolesterol totalnya secara enzimatik menggunakan KIT kolesterol CHOD-PAP merk Diasys. Absorbans diukur dengan spektrofotometer Genesis 20 pada panjang gelombang 546 nm. Data dianalisis dengan GLM pengukuran berulang dan one-way anova.Hasil penelitian menunjukan : pemberian infus yang mengandung 97, 194 dan 388 mg mahkota dewa dapat mencegah peningkatan kadar kolesterol dibanding kontrol (-). Terdapat perbedaan yang signifikan (p<0,05) antara kelompok 194 dan 388 mg dengan kelompk 97 mg, dimana kelompok 194 dan 388 mg mencegah peningkatan lebih besar. Tidak terdapat perbedaan (p>0,05) antara kelompok yang mendapat 194 dengan kelompok yang mendapat 388 mg mahkota dewa.ARTIKEL PENELITIAN52Disarankan untuk meneliti efek pencegahan peningkatan pada profil lipid yang lain dan melihat efek penurunan infus mahkota dewa pada hewan coba yang hiperkolesterolemik.Kata kunci : Mahkota dewa, kolesterolAbstractCoronary heart disease (CHD) is the leading cause of death in many parts of the world including in Indonesia. One of the triggers of CHD are high blood cholesterol levels. The Crown of God (Phaleria macrocarpa) has long been used by people to lower cholesterol and treat various cardiovascular diseases. Has done research to observe the effect of infusion Phaleria to prevent an increase in cholesterol levels in male white rats fed a high fat diet (HFD). This study used 30 male Rattus novergicus old from 2.5 to 3 months with body weight 250-350 grams. Experimental animals were randomized over five groups and treated as follows:Group I was given a standard diet 555 of global feed and 1 ml aquades, group II were given a diet high fat (HFD) consisting of 2% cholesterol, 10% of duck egg yolk, beef 18% fat and 78% standard diet and 1 ml aquades, group III was given the DLT and 1 ml infusion containing 97 mg of crown gods., group IV were given DLT and 1 ml infusion containing 194 mg crown of god, group V were given HFD and 1 ml infusion containing 388 mg the crown of god. Infusion given orally every day for 56 days. Diet and drinking water provided ad libitum.On days 0, 14, 28, 42 and 56 blood was collected via the tail vein centralis for total cholesterol is determined enzymatically using cholesterol Chod-PAP KIT Diasys. Absorbance was measured with a spectrophotometer Genesis 20 at 546 nm wavelength. Data were analyzed by GLM repeated measurements and one-way ANOVA.The results showed: the infusion containing 97, 194 and 388 mg crown of god can prevent an increase in cholesterol levels compared to controls (-).There are significant differences (p <0.05) between groups 194 and 388 mg with 97 mg faction, which groups 194 and 388 mg prevent the increase is greater. No difference (p> 0.05) between the group receiving 194 with the group receiving 388 mg crown of gods.It is advisable to examine the effect of preventing an increase in other lipid profiles and observe the effect of decreasing infusion crown of gods on hypercholesterolemic animal.Key word : Phaleria macrocarpa, cholseterol

Abstract: Intraperitoneal adhesion is an attachment between intraperitoneal tissue and organ in the form of fibrosis. The occurences of intraperitoneal adhesion are 67-93% after laparotomy and 97% after gynecological operation. Adhesion between wound and omentum occurs in 80% of patients; 50% involve the intestines. Prevention and inhibition of adhesion formation can be done by decreasing post traumatic inflammation. There are several substances that can be used for that purpose, inter alia: anti-inflammation, anti-histamine, anti-coagulant (heparin), anti-oxidant, proteolytic enzymes, and tissue plasminogen activator. It is already known that virgin coconut oil (VCO) has several important roles, such as: anti-inflammation, antithrombosis, mechanical barrier, and antioxidant due to its tocopherol content. This study aimed to obtain the influence of several doses of intraperitoneal VCO on intraperitoneal adhesion macroscopically and microscopically after laparotomy. This was an analytical interventional study using control. Samples were male Wistar rats (Rattus norvegicus) with an average body weight of 175-200 g. The results of Kruskal-Wallis chi-square tests were 16.381, df = 4, and Asymp sig = 0.003 (P < 0.05) for macroscopical grading (Zühlke) meanwhile 15.160, df = 4, and Asymp sig = 0.004 (P < 0.01) for microscopical grading (Yilmaz). Conclusion: There was a statistically significant difference in macroscopical grades according to Zühlke and in microscopical grades according to Yalmiz among the five groups concerning the occurence of intraperitoneal adhesion in Rattus norvegicus.Keywords: intraperitoneal adhesion, virgin coconut oil (VCO).Abstract: Adhesi intraperitonial adalah timbulnya perlengketan berupa fibrosis antara jaringan dan organ di dalam rongga abdomen. Kejadian adhesi intraperitoneal sekitar 67-93% setelah operasi laparotomi bedah dan mencapai 97% pada operasi ginekologi. Adhesi antara luka dan omentum terjadi pada 80% pasien dan sekitar 50% melibatkan usus. Untuk mencegah atau mengurangi pembentukan adhesi dapat dilakukan dengan menurunkan inflamasi pasca trauma melalui bahan atau obat anti-inflamasi, anti-histamin, anti-koagulan (heparin), anti-oksidan, enzim proteolitik, dan tissue plasminogen activator. Virgin Coconut Oil (VCO) telah diketahui berperan sebagai antiinflamasi, antitrombotik, barier mekanik, dan antioksidan dengan bahan aktif utama tokoferol. Penelitian ini bertujuan untuk mengetahui pengaruh VCO secara makroskopik dan mikroskopik terhadap adhesi intraperitoneal dengan menganalisis perbandingan dosis VCO yang diberikan intraperitoneal pada hewan coba tikus Wistar. Jenis penelitian ini analitik intervensional dengan kontrol. Hewan coba ialah tikus Wistar (Rattus norvegicus) yang diseragamkan berat badan 175-200 g, jenis kelamin jantan, dan variannya. Hasil uji Kruskal-Wallis chi-square ialah 16,381 dengan df = 4 dan Asymp sig = 0,003 (P < 0,05) untuk penilaian makroskopik (Zühlke) sedangkan nilai 15,160, dengan df = 4 dan Asymp sig = 0,004 (P < 0,01) untuk penilaian mikroskopik (Yilmaz). Simpulan: Terdapat perbedaan bermakna dalam derajat makroskopik menurut Zühlke dan derajat mikroskopik menurut Yilmaz pada ke-5 perlakuan dalam kejadian adhesi intraperitoneal pada Rattus norvegicus.Kata kunci: adhesi intraperitoneal, VCO

Latar belakang: Adhesi peritoneal sering terjadi setelah operasi abdomen, dimana adhesi kemudian menjadi sumber morbiditas. Adanya perlekatan menimbulkan keluhan seperti nyeri perut, mempersulit pembedahan berikutnya, sampai terjadinya obstruksi usus. Pemakaian obat-obatan farmakologik sebagai ajuvan telah banyak dilakukan untuk pencegahan adhesi paska operasi. Pada penelitian ini diteliti perbandingan efektivitas antara penggunaan Virgin Coconut Oil (VCO) dan Triamcinolone Acetonide (TCA) dalam mencegah terbentuknya adhesi intraperitoneal paska laparatomi Metode: Penelitian eksperimen dengan pengukuran perbandingan derajat adhesi secara makroskopik dan mikroskopik pada 30 tikus jantan Rattus norvegicus strain Wistar. Subjek dibagi dalam tiga kelompok. Satu kelompok diberikan VCO pada caecum yang telah diabrasi, satu kelompok diberikan TCA, dan kelompok yang lain menjadi kontrol. Hasil: Tampak bahwa sebagian hewan-hewan tersebut (n = 14, atau 47%) tidak menunjukkan adanya adhesi secara makroskopik dan mikroskopik. Pada 16 tikus dengan fibrosis pun, semuanya berkategori ringan dan tipis (tingkat 1) menurut klasifikasi Yilmaz. Dalam pembagian gambaran makroskopik Zuhlke, tiga perempat (n = 12) dari tikus-tikus dengan adhesi tergolong tingkat 1 ataupun 2. Diskusi: Penggunaan preparat TCA dan VCO terbukti membantu mengurangi angka kejadian adhesi intraperitoneal sesuai kepustakaan dan penelitian sebelumnya, yang mengungkapkan bahwa TCA dan VCO mengandung zat-zat yang secara langsung berfungsi mencegah adhesi baik secara lokal maupun sistemik karena kinerjanya dalam proses inflamasi. Simpulan: Penggunaan TCA dan VCO memiliki efektivitas yang bermakna dalam pencegahan adhesi intraperitoneal dibandingkan dengan kelompok kontrol, namun dalam perbandingan antara kedua preparat yaitu TCA dan VCO, tidak ditemukan perbedaan efektivitas yang bermakna.

Background: The aim of this study was to evaluate the nutritional effects of supplementation with virgin coconut oil (VCO) in Wistar rats over a sub-chronic period (6 weeks).Methods: Twelve Wistar rats were used and randomly assigned to receive (n = 6): control - lab chow; coconut oil (CO) - lab chow with added virgin coconut oil (20%). Food and caloric intake, weight gain, food efficiency, body mass index, femur and tibia length, bone mineral composition and blood biochemistry were evaluated.Result: The CO group showed an energy intake closed to control group. Also, the supplementation with VCO generated a decrease in total blood cholesterol as compared to the control group. However, the CO group showed accumulation of fat mass, shorter femur length and anxiogenesis in relation to the control group. These results indicate few beneficial effects from the sub-chronic use of VCO and indicate that its consumption in large quantities for long periods should be questioned.

<p>ABSTRAK</p><p>Virgin Coconut Oil (VCO) merupakan salah satu produk panganfungsional yang populer di masyarakat. Cita rasa VCO dapat diperkayadalam bentuk emulsi mengandung sari buah nenas. Penelitian inidilakukan untuk mengetahui pengaruh diet emulsi VCO terhadap profillipid tikus (Rattus norvegicus) yang sebelumnya diinduksi hiperlipidemiadengan lemak babi 180g/100g ransum dan kuning telur bebek dengandosis 4 ml/hari. Penelitian dilakukan bulan Januari sampai Desember 2009di Laboratorium Balai Penelitian Tanaman Kelapa dan Palma Lain(BALITKA), Laboratorium FMIPA Universitas Sam Ratulangi Manado,Laboratorium UPHP (Unit Penelitian Hewan Percobaan) UniversitasGadjah Mada Yogyakarta serta Laboratorium PAU IPB Bogor. Delapanbelas tikus dibagi secara acak ke dalam 3 kelompok (6 tikus perkelompok). Kelompok I adalah kelompok kontrol yang diberi akuades,kelompok II diberi diet emulsi VCO, dan kelompok III diberi diet VCOmurni. Kandungan kolesterol total, HDL, LDL, dan triasilgliserol diukursetelah akhir perlakuan dengan metode enzimatik. Kadar kolesterol totaldiukur menggunakan metode CHOD-PAP, kadar kolesterol LDLmenggunakan metode PVS, kadar kolesterol HDL menggunakan metodeCHOD-PAP dan kadar triasil gliserol menggunakan metode GPO-PAP.Hasil penelitian menunjukkan bahwa konsumsi 0,945ml/hari VCO dan3,78ml/hari emulsi VCO selama 7 hari dapat menurunkan kadar kolesteroltotal, kadar kolesterol LDL, serta meningkatkan kadar kolesterol HDLdarah tikus Wistar secara signifikan (=0,05), sedangkan penurunan kadartriasilgliserol hanya ditunjukkan oleh konsumsi emulsi VCO (=0,05).Dengan demikian, emulsi VCO yang diperkaya dengan sari buah nenasberperan lebih baik dari VCO murni dalam menurunkan kolesterol padahewan uji tikus.</p><p>Kata kunci: Virgin coconut oil (VCO), emulsi VCO, profil lemak</p><p>ABSTRACT</p><p>Effect of Virgin Coconut Oil (VCO) Emulsion Diet onLipid Profile of White Rats (Rattus norvegicus)</p><p>Virgin Coconut Oil (VCO) is a functional food product that ispopular in the society. This research was conducted from January to December2009 at the Laboratory of Coconut and Other Palm Trees ResearchInstitute (BALITKA), the Laboratory of FMIPA Sam Ratulangi UniversityManado, the Laboratory of Experiment Animal Research Unit GadjahMada University Yogyakarta, and the Laboratory of PAU IPB Bogor.Eighteen mice were randomly divided into 3 groups. Group I was thecontrol group treated with aquadest only, group II was treated with VCO-emulsion diet, and group III was treated with pure VCO diet. The contentof total cholesterol, HDL, LDL, and triacylglicerol were measured at theend of the treatment using enzymatic method. In this study, there weresome methods used for measurements: CHOD-PAP method for level oftotal cholesterol, PVS method for level of LDL cholesterol, CHOD-PAPmethod for level of HDL cholesterol, and GPO-PAP method for measuringlevel of triacylglicerol. The study results showed that diet of 0.945ml/daysof VCO and 3.78ml/days of VCO emulsion for 7 days significantlydecreased the level of total cholesterol and the level of LDL cholesterol,and increased the level of HDL cholesterol in the blood of Wistar mouse(α=0.05). Whereas it was shown that only by diet of VCO emulsion(α=0.05) decreased the level of triacylglicerol. High levels of blood cholesterol or hypercholesterolemia is an important risk factor for a cardiovascular disease. In order to overcome this problem, giving statins drug such as simvastatin is an intervention that is often applied. Besides using this drug, it can also be accompanied by consuming herbal plants which is Ashitaba leaves (Angelica keiskei). It contains Chalcones which demonstrates anti tumour property and also antioxidant activity that can lower cholesterol levels, especially LDL in the blood. This study aimed to investigate the effect of Ashitaba leaves (Angelica keiskei) steeping administration in assisting simvastatin drugs to decrease the levels of Low Density Lipoprotein (LDL) in the blood serum of male white rats Strain wistar (Rattus Norvegicus) with hypercholesterolemia. The population for this study was male white rats Strain wistar (Rattus Norvegicus) with samples taken in this study were 33 white rats. Data analysis for this study is using One Way Anova test.The result showed that supplementation of simvastatin treatment using steeping Ashitaba leaves might slightly reduce LDL level among male white rats Strain wistar (Rattus Norvegicus) with hypercholesterolemia.
