ABSTRAK Sinar ultraviolet dapat memberikan dampak yang merugikan terhadap kulit. Meskipun sinar ultraviolet juga memberikan manfaat seperti memediasi sintesis vitamin D, namun paparan berlebih sinar ultraviolet dapat menyebabkan kelainan kulit dan dapat menimbulkan resiko kesehatan, seperti atropi, perubahan pigmen, keriput, dan malignancy (keganasan). Reaksi tersebut timbul karena pembentukan radikal bebas dan untuk mengatasinya diperlukan antioksidan. Ekstrak daun Aquilaria microcarpa diketahui mengandung flavanoid, yang dapat berfungsi sebagai antioksidan. Pemanfaatan ekstrak daun Aquilaria microcarpa dalam bentuk sediaan merupakan langkah untuk meningkatkan kenyamanan pemakaian dan memaksimalkan manfaat ekstrak. Penelitian ini bertujuan untuk menentukan kemampuan sediaan ekstrak daun Aquilaria microcarpa dalam memperbaiki kulit yang terpapar sinar matahari. Kulit yang digunakan dalam penelitian adalah kulit mencit dewasa galur Balb/C yang telah diberi perlakuan selama 8 hari.  Pengamatan dilakukan terhadap lapisan epidermis dan dermis terhadap masing-masing ketebalan dan jumlah melanosit. Hasil penelitian menunjukkan sediaan ekstrak daun Aquilaria microcarpa memiliki potensi memperbaiki kulit yang terpapar sinar ultraviolet. Kata Kunci: paparan sinar ultraviolet, Aquilaria microcarpa,   ABSTRACT Ultraviolet rays could give negative effect to skin. Although ultraviolet rays also give benefit, for example, catalyzing vitamin D synthesis, but excessive exposure of ultraviolet rays may cause skin disorders, such as atropy, pigment changes, wrinkles, and malignancy. Those reactions caused by free radicals formation and need antioxidant to reduce them. Aquilaria microcarpa leaves extract were revealed to have flavanoid, which can be used as antioxidant. Its usage as dosage form was a step to increase acceptability and to maximize benefit of the extract. This research was aimed to determined Aquilaria microcarpa leaves extract ability to recover skin-previously exposed to ultraviolet rays. Skin used in this research were full grown mice Balb/C whom treated after 8 days. Epidermis thickness and melanocyte cell numbers were counted and compared with UV group and control group. The result showed Aquilaria microcarpa leaves extract as dosage form has potency to recover skin which exposure to ultraviolet rays. Keywords: ultraviolet rays exposure, Aquilaria microcarpa

Background: Tuberculosis (TB) is a global health problem, Indonesia had the second rank of the case in the world after India which is the fourth cause of death. Giving vitamin D together with anti-tuberculosis drugs can increase healing proses because vitamin D (anti-microbial immunomodulators) kills Mycobacterium tuberculosis.Objectives: This study aims to measure the effect of giving anti-tuberculosis drugs with vitamin D on changes in AFB in patients with pulmonary tuberculosis.Methods: This study was used a quasi-experiment design, pre-test and post-test control group was doing at 25th March – 25th July 2019. This research has been conducted in Pidie Regency. The intervention group (17 samples with tuberculosis drugs) was given Softgels vitamin D 5000 IU for 4 months, while the control group was only anti-tuberculosis drug therapy. The data were analyzed statistically using the Wilcoxon and Mann Whitney tests at a significance level of 95%.Results: Before the intervention, the BTA status of both the intervention and control group was similar (p= 0.061). After the intervention, there was a decrease in AFB interactions (p= 0.000). There was a decrease in the contribution of AFB in the control group before with after intervention (p= 0.000). There are those who support (p= 0.033) giving vitamin D the acceleration of pulmonary tuberculosis treatment. Conclusion: It is better to administer vitamin D to the successful treatment of pulmonary TB in patients undergoing anti-tuberculosis drug therapy.

The present work evaluated the reversible meiosis inhibition effect on the development of swine embryos produced by in vitro fertilization (IVF) or parthenogenetic activation (PA). The efficiency of PZM3 and NCSU23 embryo culture media was also evaluated. Oocytes from ovaries collected at a slaughterhouse were subjected to IVM in two different groups: CHX (cycloheximide 5 µM for 10 h) and control, both with TCM-199 + 3.05 mM glucose + 0.91 mM sodium pyruvate + 10% porcine follicular fluid (pFF) + 0.57 mM cystein + 10 ng epidermal growth factor (EGF)/mL + 10 IU eCG/mL + 10 IU hCG/mL for the initial 22 h. In the remaining period (20 h for CHX and 22 h for control), medium without hormones was utilized. After IVM, oocytes were denuded and fertilized for 6 h (IFV) or the matured oocytes were submitted to activation by electric pulses (PA) (2 DC of 1.5 kV/cm for 30 µs), incubated for 1 h in culture medium with 10 μM of CHX, and again submitted to the same electric pulses for 60 µs. Embryo development was evaluated by cleavage rate on Day 3 and blastocyst rate and blastocyst cell number on Day 7 of culture. Cleavage and blastocyst rates were analyzed by the equality-of-two-ratios test and cell number by the Kruskal-Wallis and Mann-Whitney tests (P < 0.05). In relation to IVF, the PZM3 medium was more efficient than NCSU23 for cleavage rate in the CHX group (PZM3: 68.4%, NCSU23: 44.4%) and had a better blastocyst rate in the control group (PZM3: 13.4%, NCSU23: 5.6%). With reference to PA, NCSU23 presented better cleavage and blastocyst rates than PZM3 in the CHX group (NCSU23: 89.5%, PZM3: 78.5% and NCSU23: 20.4%, PZM3: 13.0%, respectively). In the control group, only the NCSU23 blastocyst rate was higher than that for PZM3 (NCSU23: 22.5%, PZM3: 10.8%). No culture medium effect on cell number mean of IVF and PA blastocysts was observed. Maturation block improved cleavage rates in IVF groups cultured with PZM3 (68.4% and 50.6%, respectively, for CHX and control) and in PA groups cultured with NCSU23 (89.5% and 80.3%, respectively, for CHX and control), but no improvement of blastocyst rates in both groups (IVF and PA) was verified. Table 1 below shows that maturation block decreased the IVF and increased the PA blastocyst cell numbers. As older oocytes are more effectively activated, oocytes blocked with CHX achieved the maturation stage faster than the control group, therefore resulting in high-quality PA blastocysts. In conclusion, PZM3 was more efficient for IVF embryo production in contrast to NCSU23, whereas NCSU23 can be indicated for PA embryo production. Moreover, maturation blockage with CHX influenced blastocyst cell number, decreasing in IVF embryos and increasing in PA embryos. Table 1. BackgroundVitamin D has an immunomodulatory role but the effect of therapeutic vitamin D supplementation in SARS-CoV-2 infection is not known.AimEffect of high dose, oral cholecalciferol supplementation on SARS-CoV-2 viral clearance.DesignRandomised, placebo-controlled.ParticipantsAsymptomatic or mildly symptomatic SARS-CoV-2 RNA positive vitamin D deficient (25(OH)D<20 ng/ml) individuals.InterventionParticipants were randomised to receive daily 60 000 IU of cholecalciferol (oral nano-liquid droplets) for 7 days with therapeutic target 25(OH)D>50 ng/ml (intervention group) or placebo (control group). Patients requiring invasive ventilation or with significant comorbidities were excluded. 25(OH)D levels were assessed at day 7, and cholecalciferol supplementation was continued for those with 25(OH)D <50 ng/ml in the intervention arm. SARS-CoV-2 RNA and inflammatory markers fibrinogen, D-dimer, procalcitonin and (CRP), ferritin were measured periodically.Outcome measureProportion of patients with SARS-CoV-2 RNA negative before day-21 and change in inflammatory markers.ResultsForty SARS-CoV-2 RNA positive individuals were randomised to intervention (n=16) or control (n=24) group. Baseline serum 25(OH)D was 8.6 (7.1 to 13.1) and 9.54 (8.1 to 12.5) ng/ml (p=0.730), in the intervention and control group, respectively. 10 out of 16 patients could achieve 25(OH)D>50 ng/ml by day-7 and another two by day-14 [day-14 25(OH)D levels 51.7 (48.9 to 59.5) ng/ml and 15.2 (12.7 to 19.5) ng/ml (p<0.001) in intervention and control group, respectively]. 10 (62.5%) participants in the intervention group and 5 (20.8%) participants in the control arm (p<0.018) became SARS-CoV-2 RNA negative. Fibrinogen levels significantly decreased with cholecalciferol supplementation (intergroup difference 0.70 ng/ml; P=0.007) unlike other inflammatory biomarkers.ConclusionGreater proportion of vitamin D-deficient individuals with SARS-CoV-2 infection turned SARS-CoV-2 RNA negative with a significant decrease in fibrinogen on high-dose cholecalciferol supplementation.Trial register numberNCT04459247.

Plasma calcium, phosphorus, alkaline phosphatase, total protein, and albumin were measured during pregnancy in a group of Asian women living in the south of England and in a control group. The Asian women had slightly lower mean plasma calcium concentrations than the control group. Exposure of Asian women to sunshine appeared to be low, and it was inferred that they probably had little endogenous synthesis of cholecalciferol as a result. Both Asian and control subjects ingested similarly low amounts of vitamin D. The Asian women consumed greater amounts of phytate and vegetable fibres than the controls, and it is suggested that these substances may reduce absorption of insoluble lipids (including cholecalciferol) and calcium in a setting where the dietary intake and endogenous synthesis of vitamin D are already borderline for the subjects' requirements.

Background Vitiligo is a well-recognized pigmentary disorder of the skin and /or mucous membrane characterized by circumscribed ivory or chalky white macules devoid of identifiable melanocytes. The pathogenesis of vitiligo is complex and still not well understood. According to autocytotoxic hypothesis, oxidative stress has been suggested to be the initial pathogenic event in melanocyte degeneration. The role of free radicals and oxidative damage in the pathophysiology of vitiligo has been documented in recent studies.Objective To evaluate the role of oxidative stress in patients with vitiligo and of healthy controls by measuring levels of the oxidant malondialdehyde (MDA) and antioxidants vitamin C and vitamin E in serum and catalase (CAT) in erythrocytes.Method A total of 80 clinically diagnosed cases of vitiligo and 80 control subjects were included in the study to assess the activity of MDA, vitamin C and vitamin E in serum and CAT in erythrocytes of patients and controls by using the spectrophotometric assay.Result There was statistically significant increase in the levels of MDA in patients with vitiligo compared to the control group (p<0.001). No significant difference was found in the levels of vitamin C (p=0.411) and vitamin E (p=0.771) between the patients with vitiligo and control group. The levels of CAT in the vitiligo patients were found to be significantly lower than those of controls (p<0.001).Conclusion Increased oxidative stress and decreased catalase have been observed in vitiligo patients and the data suggesting that the free radicals may be involved in the destruction of melanocytes or dysregulation of melanogenesis.Kathmandu University Medical Journal Vol.12(2) 2014: 132-136

Background: Most people believe that electric cigarettes have no negative effect on health,  which causes them to use it more. However, exposure to the smoke from these cigarettes is bad for the health and causes cell damage. Antioxidants play an important role in preventing cell damage, and they can be obtained through the oral administration of selenomethionine.Design and methods: This study used an experimental method and a post-test control group design. Male Wistar rats, which were exposed to cigarette smoke were given selenomethionine orally and then tested for the presence of free radicals. The measurement of lung tissue damage was conducted by assessing the level of malondialdehyde in the blood and immunohistochemistry (IHC) of the lung tissue.Result: The study showed that differences in the time of administration of selenomethionine affect the levels of malondialdehyde in the blood and expression of malondialdehyde in the lung tissue (p<0.05). Consequently, the two groups showed a strong (r=0.861) and significant (p=0.000) relationship with each other.Conclusion: The early application of selenomethionine can prevent increased levels of malondialdehyde in the blood and lung tissue due to exposure to e-cigarette smoke.

Abstract. The aim of this study was to investigate the effects of age on pecking behaviour and plumage quality of free-range laying hens during first access to range. Six hundred 16-week-old layer pullets obtained from a commercial company were housed in a experimental free-range house. The experimental house consisted of a slat-and-litter floor with range area and was divided into three similar pens. The pullets were randomly assigned to a control and two experimental groups according to age at first access to range area. The birds were allowed into the range area for the first time at 18, 20 and 22 weeks of age in the groups of treatment I, treatment II and control, respectively. Pecking behaviour and plumage quality of the birds were measured for 8-week intervals from 24 to 48 weeks of age. Total feather damage significantly increased with age in all groups (P < 0.040, P < 0.006, P < 0.001). Compared to the control group, birds allowed into the range area earlier had less feather damage. The feather score of the tail body region in all groups was significantly greater (more damage) than in the others (P < 0.001). There were no significant differences for the total pecking behaviour of the birds in the groups at 24, 32, 40 and 48 weeks of age. Incidents of gentle feather pecking in every period were more frequent, while aggressive pecking was less frequent (P < 0.026, P < 0.007, P < 0.001). It can be said that access to range area as early as possible at the beginning of the laying period may be useful to reduce the negative effect of feather pecking in free-range layer chickens.

Background: The prevalence of vitamin D deficiency is increasing due to changes in lifestyle and dietary habits. The aim of this study was to compare the serum levels of 25-hydroxy vitamin D between patients with bronchial asthma and the healthy control group. Patients and Methods: In this case-control study, 45 patients with asthma and 45 healthy subjects were enrolled and the level of serum 25 (OH) vitamin D was measured in both groups. In addition, a welltrained observer assessed airway reversibility, peak flowmetry and spirometry in the participants. The data were statistically analyzed using t test, one-way analysis of variance (ANOVA), and chi-square test with Stata software (version 11). Results: The mean age (±SD) of participants were 49.06 ±16.43 and 46.13 ±16.10 years in case and control groups, respectively (P=.394). The prevalence of vitamin D deficiency was high in both groups (69% in case and 65.5% in control groups). The mean (±SD) serum 25 (OH) vitamin D was 16.24 (±14.98) ng/ml in case group and 17.70 (±16.07) ng/mL in control group (P=.657). We found a positive correlation between the levels of vitamin D and the amount of FEV1 (r=0.2). Conclusions: According to the present study, the mean serum levels of vitamin D differences were not statistically significant between asthmatic patients and control group. However, the results of this study showed a positive relationship between forced expiratory volumes in first second (FEV1) and vitamin D levels

Purpose: To determine the levels of Vitamin D in children with myopia and to compare them with age matched controls. Study Design:  Case control study. Place and Duration of Study:  The study was conducted in Naseer Memorial Hospital, Dadhyal Azad Kashmir from March 2016 to March 2017. Material and Methods:  Two hundred patients were selected using convenient sampling technique and were divided two groups (group I Myopic and group II control). Myopia was labeled if after subjective refraction a Spherical Equivalent (SE) of −0.50 diopters (D) or more was found. Vitamin D levels were measured by radioimmunoassay technique with Diasorin SR® kit following the user’s manual. Vitamin D levels less than 20 ng/ml were considered Vitamin D deficient following the standards of American academy of pediatrics. The collected data was entered in the statistical package for social sciences (SPSS) version 21 for analysis. Independent t–test was used to determine the significant difference of means between controls and patients. P-values less than 0.05 were considered significant. Results:  Mean age of controls and myopes were 10.65 ± 3.9 and 10.20 ± 2.5 years respectively. Vitamin D levels in myopic children were found to be 14.95 ± 3.75 ng/ml and there was no significant difference in mean values of Vitamin D levels in myopic and control group. Conclusion:  We found no difference in Vitamin D levels of myopic and non myopic children and concluded that Vitamin D has no role in development or progression of myopia.

Purpose: To investigate the effect of combination of vitamin D and anti-tuberculosis drugs on serum interleukin-1β (IL-1β), interferon-γ (IFN-γ) and helper T 17 (Th17) cell-associated cytokine levels for the treatment of spinal tuberculosis (TB). Methods: Ninety-two spinal TB patients were assigned without bias to two groups (46/group): study group (vitamin D combined with anti-TB drug group) and control group (anti-TB drug group). After treatment for 8 weeks, clinical effectiveness, adverse reactions, visual analog scale (VAS) score, spinal cord injury grade, and serum levels of IL-1β, IFN-γ, Th17, IL-10, TGF-β1, IL-17 and IL-23 were assayed with ELISA, and compared between groups. Results: Study group total effectiveness was significantly higher than that in the control group (95.65 % vs 80.43 %, p < 0.05). Before drug administration, VAS score, degree of spinal cord injury and serum levels of IL-1β, IFN-γ, IL-10, TGF-β1, IL-17 and IL-23 were comparable in the study and control patients (p > 0.05). However, post-treatment, these parameters significantly decreased in both groups (p < 0.05), but were markedly lower in study group patients, relative to controls (p < 0.05). Conclusion: The use of combined treatment of vitamin D and anti-TB drugs is an effective and safe way to alleviate inflammatory response and improve the immunity of spinal TB patients via the regulation of the levels of Th17 cell-related factors.
