Prebiotic oligosaccharides are widely used as human and animal feed additives for their beneficial effects on the gut microbiota. However, there are limited data to assess the direct effect of such functional foods on the transcriptome of intestinal epithelial cells. The purpose of this study is to describe the differential transcriptomes and cellular pathways of colonic cells directly exposed to galacto-oligosaccharides (GOS) and fructo-oligosaccharides (FOS). We have examined the differential gene expression of polarized Caco-2 cells treated with GOS or FOS and their respective mock-treated cells using mRNA sequencing (RNA-seq). A total of 89 significant differentially expressed genes were identified between GOS and mock-treated groups. For FOS treatment, a reduced number of 12 significant genes were observed to be differentially expressed relative to the control group. KEGG and Gene Ontology functional analysis revealed that genes up-regulated in the presence of GOS were involved in digestion and absorption processes, fatty acids and steroids metabolism, potential antimicrobial proteins, energy-dependent and -independent transmembrane trafficking of solutes and amino acids. Using our data, we have established complementary non-prebiotic modes of action for these frequently used dietary fibers.

Garlic (Allium sativum) is an essential vegetable that has been widely utilized as seasoning, flavoring, culinary and in herbal remedies. Garlic contains several characteristic organosulfur compounds, such as diallyl sulfide, allicin (diallyl thiosulphate), γ-glutamylcysteine, and S-allyl cysteine (alliin) and ajoene, which garlic has beneficial effects on inflammation, oxidative stress markers, hypertension, hyperlipidaemia and endothelial function in vitro or in animal model. These bioactive molecules are also playing pivotal role in livestock and fisheries production apart from its application in humans. Supplementation of animal feed with garlic and its related products is consistent with the modern agricultural concept of organic animal husbandry. This review compiles the information describing the effects of feeding garlic and its extracts on selected performance parameters in animals (chicken, rabbits, ruminants, pigs and fish). This review may provide reference for scientists and entrepreneurs to investigate the applications of feeds added with garlic and allicin by-products for the improvement of animal husbandry and aquatic production.

We evaluated whether olive leaves (OLs) are effective as feed additives and supplements for ruminants and the potential methane reduction effects during in vitro fermentation. Two Hanwoo cows (460 ± 20 kg) equipped with cannula were fed Timothy hay and corn-based feed 3% of the body weight at a ratio of 6:4 (8:30 a.m. and 5:00 p.m.). Ruminal fluid from the cows was collected and mixed before morning feeding. In vitro batch fermentation was monitored after 12 and 24 h of incubation at 39 °C, and OLs were used as supplements to achieve the concentration of 5% in the basal diet. At 12 h of fermentation, methane production decreased in the 5% OLs group compared to that in the control group, but not at 24 h. The proportion of cellulose-degrading bacteria, Fibrobacter succinogenes, Ruminococcus albus, and Ruminococcus flavefaciens, tended to increase in the 5% OLs group at 12 h. The amount of ammonia produced was the same as the polymerase chain reaction result for Prevotella ruminicola. At 12 h, the proportion of Prevotella ruminicola was significantly higher in the 5% OLs group. OLs may be used incorporated with protein byproducts or other methane-reducing agents in animal feed.

Изучена целесообразность использования в рационах телят черно-пестрой породы комплексной минерально-витаминной кормовой добавки (КМВКД). Установлено пониженное содержание в кормах таких элементов, как кальций, фосфор, магний, сера, марганец, медь, цинк и кобальт. Соотношение питательных и минеральных веществ, степень их доступности для усвоения организмом не обеспечивают их потребность. Использование КМВКД способствовало улучшению всех гематологических показателей. Все испытываемые рецепты, имеющие в своем составе разные варианты макро-, микроэлементов и витаминов положительно влияют на обменные про- цессы в организме животных. У молодняка опытной группы, получавшей подкормку по рецепту 1, количество эритроцитов в сравнении с животными контрольной группы увеличилось на 6,6, гемоглобина на 6,93 соответственно, по рецепту 2 11,05 и 11,39 по рецепту 3 16,75 и 15,27, то есть соотношение макро- и микроэлементов, имеющихся в составе КМВКД и взаимодействие между собой меди, железа и кобальта сыграли особую роль на концентрацию форменных элементов. У молодняка, получавшего подкормку по рецепту 1 количество общего белка в сравнении с животными контрольной группы увеличилось на 1,99, кальция на 16,02, каротина на 10,92 соответственно, по рецепту 2 4,91, 25,96 и 19,12 по рецепту 3 на 3,58, 27,07 и 20,21. Значительное повышение содержания белка, кальция, фосфора и каротина по сравнению с животными контрольной группы объясняется тем, что недостаток их в рационах восполнялся за счет используемой подкормки, в результате их содержание увеличилось в сыворотке, но не выходило за пределы нормативных данных и соответствовало физиологическим нормам.Practicability of using a complex mineral and vitamin feed additive (CMVFA) in the ration of black-and-white calf breed was studied. Decreased content of such elements as calcium, phosphorus, magnesium, sulfur, manganese, copper, zinc, and cobalt was registered. The correlation of nutritive and mineral substances, and the level of their availability for digestion did not provide for their need. CMVFA use facilitated the increase of all hematologic parameters. Tested recipes which had different content of sapropel and zeolite, as well as various types of macronutrients, microelements, and vitamins, positively influenced the digestive processes in animal bodies. Young stock from the test group that was receiving food additives according to recipe No. 1 showed 6.6 increase in the quantity of red blood cells compared to control group animals, and 6.93 increase in hemoglobin quantity the ones who were fed according to recipe No. 2 demonstrated the increase by 11.05 and 11.39 recipe No. 3 showed 16.75 and 15.27 increase, which means that the ratio of macronutrients and microelements present in CMVFA and the mutual interaction of copper, iron, and cobalt played a special role in the concentration of formed elements due to the fact that they are integral elements of blood. Total protein of young stock that was receiving food additives according to recipe No. 1 was 1.99 higher than in test group animals calcium 16.02 higher carotene 10.92 as for recipe No. 2 the values were increased by 4.91, 25.96, and 19.12 recipe No. 3 3.58, 27.07, and 20.21. Substantial increase in the content of protein, calcium, phosphorus, and carotene compared to control group animals can be explained by the fact that their insufficiency in animal feed was replenished due to the use of feed additives, and as a result their content increased in the serum but did not go beyond the norm and corresponded to physiological norms.

ABSTRACTIn broiler chickens, feed additives, including prebiotics, are widely used to improve gut health and to stimulate performance. Xylo-oligosaccharides (XOS) are hydrolytic degradation products of arabinoxylans that can be fermented by the gut microbiota. In the current study, we aimed to analyze the prebiotic properties of XOS when added to the broiler diet. Administration of XOS to chickens, in addition to a wheat-rye-based diet, significantly improved the feed conversion ratio. XOS significantly increased villus length in the ileum. It also significantly increased numbers of lactobacilli in the colon andClostridiumcluster XIVa in the ceca. Moreover, the number of gene copies encoding the key bacterial enzyme for butyrate production, butyryl-coenzyme A (butyryl-CoA):acetate CoA transferase, was significantly increased in the ceca of chickens administered XOS. In this group of chickens, at the species level,Lactobacillus crispatusandAnaerostipes butyraticuswere significantly increased in abundance in the colon and cecum, respectively.In vitrofermentation of XOS revealed cross-feeding betweenL. crispatusandA. butyraticus. Lactate, produced byL. crispatusduring XOS fermentation, was utilized by the butyrate-producingAnaerostipesspecies. These data show the beneficial effects of XOS on broiler performance when added to the feed, which potentially can be explained by stimulation of butyrate-producing bacteria through cross-feeding of lactate and subsequent effects of butyrate on gastrointestinal function.

Yogurt is a traditional fermented dairy product, prepared with starter cultures containing Streptococcus thermophilus and Lactobacillus bulgaricus that has gained widespread consumer acceptance as a healthy food. It is widely accepted that yogurt cultures have been recognized as probiotics, due to their beneficial effects on human health. In this study, we have characterized technological and health-promoting properties of autochthonous strains S. thermophilus BGKMJ1-36 and L. bulgaricus BGVLJ1-21 isolated from artisanal sour milk and yogurt, respectively, in order to be used as functional yogurt starter cultures. Both BGKMJ1-36 and BGVLJ1-21 strains have the ability to form curd after five hours at 42 °C, hydrolyze αs1-, β-, and κ- casein, and to show antimicrobial activity toward Listeria monocytogenes. The strain BGKMJ1-36 produces exopolysaccharides important for rheological properties of the yogurt. The colonies of BGKMJ1-36 and BGVLJ1-21 strains that successfully survived transit of the yogurt through simulated gastrointestinal tract conditions have been tested for adhesion to intestinal epithelial Caco-2 cells. The results reveal that both strains adhere to Caco-2 cells and significantly upregulate the expression of autophagy-, tight junction proteins-, and anti-microbial peptides-related genes. Hence, both strains may be interesting for use as a novel functional starter culture for production of added-value yogurt with health-promoting properties.

Objective: Evaluation of beneficial effects of the seed of Linum usitatissimum in congestive heart failure.Methods: Methanolic extract of seeds of Linumusitatissimum (MELU) was prepared using soxhlet apparatus and oil of seed of Linumusitatissimum (OLU) was isolated using Clevenger apparatus. The positive inotropic action of methanolic extract of seeds of Linum usitatissimum was evaluated using Langendorff’s assembly (in vitro study). Beneficial effects of methanolic extract and oil of seeds of Linum usitatissimum were carried out by doxorubicin (1 mg/kg, i. p. within 3 w) to induce congestive heart failure (in vivo study). Parameters like electrocardiogram (ECG) recording and cytosolic Ca2+level and histopathology of the heart were carried out. In same study diuretic action was evaluated using Lipschitz model.Results: Methanolic extract of seeds of Linum usitatissimum showed significantly increased in positive inotropic effect (force of contraction 48.8±1.53 mm) as compared to control group (force of contraction 17.5±0.76 mm) on Langendorff”s study (in vitro study). In doxorubicin-induced congestive heart failure both MELU and OLU showed significant decreased QT (Note: In cardiology, the QT interval is a measure of the time between the start of the Q wave and the end of the T wave in the heart's electrical cycle. There is no full form for this medical word) interval. The histopathologic study indicated the least damage to the architecture of myocardial membrane. MELU and OLU increased urine output (5.66±0.16 ml and 6.58±0.15 ml respectively) significantly in Lipschitz model as compared to disease control group (4.58±0.15 ml).Conclusion: Present research work emphasizes that the seeds of Linum usitatissimum is beneficial in the management of congestive heart failure because of having positive inotropic effect, diuretic activity and control of rhythmicity of heart.

Background: Lacidipine, a third-generation calcium channel blocker, exerts beneficial effects on the endothelium of hypertensive patients in addition to blood pressure lowering. However, the detailed mechanism underlying Lacidipine-related endothelial protection is still elusive.Methods: Sixteen spontaneous hypertensive rats (SHRs) were randomly divided into two groups: Lacidipine-treated SHR group and saline-treated control group. Tail systolic blood pressure was monitored for four consecutive weeks. Endothelial cells (ECs) were pretreated with Lacidipine prior to being stimulated with H2O2, bleomycin, or Lipopolysaccharides (LPS) in vitro. Then, cell activity, migration, and senescence were measured by Cell Counting Kit-8 assay, transwell assay, and β-galactosidase staining, respectively. The fluorescent probe 2′, 7′-dichlorofluorescein diacetate (DCFH-DA) was used to assess the intracellular reactive oxygen species (ROS). Related protein expression was detected by Western blotting and immunofluorescence.Results: Our data showed that Lacidipine treatment lowered the blood pressure of SHRs accompanied by the elevation of CXCR7 expression and suppression of P38 and CCAAT/enhancer-binding protein beta (C/EBP-β) compared with the control group. In vitro experiments further demonstrated that Lacidipine increased the cell viability and function of ECs under oxidative stress, cell senescence, and inflammatory activation via the CXCR7/P38/signaling pathway.Conclusions: Our results suggested that Lacidipine plays a protective role in EC senescence, oxidative stress, and inflammatory injury through the regulation of CXCR7/P38/C/EBP-β signaling pathway.

Humic substances are well known human nutritional supplement materials and play important performance-enhancing roles as animal feed additives, too. For decades, ingredients of humic substances have also been proven to carry potent antiviral effects against different viruses. Here, the antiviral activity of a humic substance containing ascorbic acid, Se- and Zn2+ ions intended as a nutritional supplement material was investigated against SARS-CoV-2 virus B1.1.7 Variant of Concern (Alpha Variant) in a VeroE6 cell line. Results show that this combination has a significant in vitro antiviral effect at a very low concentration range of its intended active ingredients. Even picomolar concentration ranges of humic substances, vitamin C and Zn/Se ions in the given composition were enough to achieve fifty percent viral replication inhibition in the applied SARS-CoV-2 virus inhibition test. The Study was conducted to evaluate the additive effect of selected feed additives (Glycerol, a mixture of exogenous fibrolytic enzymes (EFE), a mixture of probiotics and rumen protected methionine (RPM) under both in vitro and in vivo conditions. The optimum dose of pre-mixture of feed additives (PFA) was evaluated by in vitro gas production technique in two stages. The marginal increase in Y-max and in vitro neutral detergent fiber digestibility (IVNDFD) for each incremental increase in PFA was compared graphically and it was found to be 1.7 %. In in vivo trial, 12 crossbred HF cows were equally divided into 3 groups based on milk yield and average days in lactation. The optimized dose of PFA was added to the concentrate supplements without (T1) or with buffer (T2), control group (CG) was fed with the basal diet (roughage: concentrate ratio of 60:40). Results indicated that PFA with or without buffer had no significant effect on nutrient intake and digestibility. Fat corrected milk yield (FCM), milk fat, total solids (TS) and milk protein were significantly (P Lass Than 0.01) increased in TGs than that of CG. From the results, it may be concluded that optimized PFA had an additive effect on milk production efficiency but not on nutrient intake and digestibility. Further, different groups of feed additives can be evaluated for additive effect, which may pave the way for commercial product preparation.
