<p>Usaha pengendalian hama ulat grayak (Spodoptera litura) di tingkat petani masih mengandalkan pestisida sintetik. Tujuan penelitian adalah mengeksplorasi bakteri potensial pengendali hama ulat grayak (S. litura) dan menguji ketahanan bakteri potensial pada bahan pembawa kompos dan zeolit. Isolat tanah diisolasi dari tiga jenis sumber, yaitu sampel tanah daerah rhizosfer (padi, kelapa sawit, terung, jagung), sampel buah busuk (kakao, kelapa sawit, jambu air), dan sampel bangkai serangga (ulat api, belalang, kumbang tahi, kupu-kupu) yang diambil dari kawasan Dramaga dengan metode purposive sampling. Penelitian di laboratorium meliputi isolasi bakteri, uji patogenitas, pewarnaan gram, pengamatan morfologi koloni, uji toksisitas, uji biokimia, dan uji bahan pembawa. Berdasarkan penelitian ini didapatkan dua strain yang berpotensi sebagai agens biokontrol dengan kemampuan membunuh hama yang tinggi pada pengujian toksisitas tahap kedua, yaitu IRJ 10 (tingkat kematian 90%) dan ISU 4 (tingkat kematian 100%). Kedua isolat ini merupakan anggota genus Bacillus. Pada uji bahan pembawa kompos dan zeolit, penurunan jumlah sel bakteri pengendali hama paling tinggi adalah pada bahan pembawa zeolit dibandingkan dengan menggunakan bahan pembawa kompos. Jumlah sel bakteri pengendali hama pada masa penyimpanan 3 minggu masih di atas 108 CFU/g.</p><p><strong>Keywords</strong></p><p>Bakteri; Bahan pembawa; Ekplorasi; Agens pengendali hama; Ulat grayak (Spodoptera litura)</p><p><strong>Abstract</strong></p><p>The effort to control the Spodoptera litura at the farm level still used synthetic pesticides. This research aimed to explore potential bacteria as biological control of S. litura and do viability test of potential bacteria on compost and zeolite carrier. Soil potential bacteria had been isolated from three sources, including rhizosphere soil samples (rice, oil palm, eggplant, corn), rotten fruit samples (cocoa, oil palm, water), and insect samples (fireworms, locusts, dung beetles, butterflies) taken from Dramaga area with the purposive sampling method. Stages of laboratory study include isolation of bacterial isolates, pathogenicity tests, gram staining, colony morphology observation, toxicity test, biochemical test, and viability test. Two strains that have potential as biocontrol agents with a high ability to kill pests in the second stage of toxicity testing are IRJ 10 (90% mortality rate) and ISU 4 (100% mortality rate). Both of these isolates are members of the genus Bacillus. The highest number of viability was found in zeolite carriers. The number of bacterial cells in the three-week storage period is still above 108 CFU/g.</p>

Fresh fruit bunches (FFB) consist of fruit be composed grade in few spikelet. Fruit at a spikelet can be distinguished into performed fruit namely internal fruit, middle fruit and outer fruit as soon as each section contain parthenocarpy fruits. This research was conducted for determine composition and content fatty acid of oil at internal fruit, middle, outer and parthenocarpy fruit from oil palm fruit. Samples of fruit came from 3 – 5 spikelet the central of FFB. Result showed that oil content of outer fruit (46.9 + 9.9)% trend higher be compared middle fruit (42.8 + 10.3)% and internal fruit (39.1 + 9.5)%. Parthenocarpy fruits have a low oil content (14.2 + 16.2)% except yellowish fruit trend high relatively oil content. The main components of fatty acid at outer fruit, middle and internal are palmitic acid, oleic, linoleic and stearic with mean value respectively (44.8 – 45.8)%, (37.6 – 38.0)%, (9.9 – 10.9)% and (4.6 – 4.8)%. Oil content at parthenocarpy fruit have amount main component of fatty acid with performed fruit but composition of palmitic acid (40.0 + 5.9)% and oleic (34.6 + 8.4)% lower while linoleic acid (16.9 + 8.5)% and linolenic (1.6 + 1.8)% higher be compared to performed fruit. Simalungun variety has the highest oil content in the part of fruit, with that PPKS 540 and La Mé respectively. PPKS 540 variety has the highest oleic acid content while PPKS 718 has the highest linoleic content.

A biocontrol rhizobacterial strain of Bacillus subtilis AF 1 grown for 6 h was coinoculated with Aspergillus niger at different time intervals and microscopic observations revealed adherence of bacterial cells to the fungal mycelium. Bacterial cells multiplied in situ and colonized the mycelial surface. Growth of AF 1 resulted in damage to the cell wall, followed by lysis. AF 1 inoculation into media containing A. niger at 0, 6, and 12 h suppressed >90% fungal growth, while in 18- and 24-h cultures fungal growth inhibition was 70 and 56%, respectively, in terms of dry weight. In dual culture the fungal growth was not accompanied by formation of spores. The mycelial preparation of A. niger as principal carbon source supported the growth of B. subtilis, as much as chitin. Extracellular protein precipitate from B. subtilis culture filtrate had a significant growth-retarding effect on A. niger. Groundnut seeds bacterized with B. subtilis showed a reduced incidence of crown rot in A. niger infested soil, suggesting a possible role of B. subtilis in biological control of A. niger.Key words: mycolytic bacteria, Bacillus subtilis, Aspergillus niger, biological control.

ABSTRACT This study aims To find out the constraints faced by extension workers in providing agricultural extension in Sukasari Village Mesuji Raya District Ogan Komering Ilir Regency and To know the response of farmers to the implementation of agricultural extension in the Sukasari Village Mesuji Raya District Ogan Komering Ilir Regency. This research was carried out in Sukasari Village Mesuji Raya District Ogan Komering Ilir Regency in March to May 2017. The research method used is qualitative methods. While the sampling method used is simple random Purposive sampling will 28 samples of palm oil farmer. So the sample studied from members of the population as a whole that is as much as 28 oil palm farmers. Data collection methods used are interviews and observation methods. The data collected consists of primary data and secondary data. Method of processing and analysis of data used is using descriptive analysis method with qualitative approach. The results showed that agricultural extension facilities used extension workers are motorcycles, aids and props. Infrastructure that is not yet supported is not yet all extension workers get a service house near the location of counseling, Distance taken by agricultural extension workers in carrying out their duties is about 3 km for the location of the activities of the farmer's house and about 5 km for the location of agricultural gardens. Farmer's response to the organik fertilizer extension activiti b using cattle manure for oil palm plantation with total score or 47,50 which means getting high score.  ABSTRAK Penelitian ini bertujuan Untuk mengetahui kendala yang dihadapi penyuluh dalam memberikan penyuluhan pertanian di Sukasari Kecamatan Mesuji Raya Kabupaten Ogan Komering Ilir dan Untuk mengetahui tanggapan petani terhadap pelaksanaan penyuluhan pertanian di Desa Sukasari Kecamatan Mesuji Raya Kabupaten Ogan Komering Ilir. Penelitian ini dilaksanakan di Desa Sukasari Kecamatan Mesuji Raya Kabupaten Ogan Komering Ilir pada bulan Maret sampai dengan bulan Mei 2017. Metode penelitian yang digunakan adalah metode penelitian kualitatif. Sedangkan metode penarikan contoh yang digunakan adalah acak sederhana Purposive sampling dengan anggota populasi sebanyak 28. Maka sampel yang diteliti dari anggota populasi secara keseluruhan yaitu sebanyak 28 petani kelapa sawit. Metode pengumpulan data yang digunakan adalah wawancara dan observasi. Data yang dikumpulkan terdiri dari data primer dan data sekunder. Metode pengolahan dan analisis data yang digunakan yaitu menggunakan metode analisis diskriptif dengan pendekatan kualitatif. Hasil penelitian menunjukan bahwa Sarana penyuluhan pertanian yang digunakan penyuluh adalah sepeda motor, alat bantu dan alat peraga. Prasarana yang yang belum mendukung adalah belum semua penyuluh mendapatkan rumah dinas di dekat lokasi penyuluhan, Jarak yang ditempuh penyuluh pertanian dalam melaksanakan tugasnya adalah sekitar 3 km untuk kelokasi kegiatan dari rumah petani dan sekitar 5 km untuk lokasi kebun pertanian. Tanggapan petani terhadap kegitan penyuluhan pertanian pupuk organik dengan menggunakan kotoran ternak untuk tanaman kelapa sawit dengan jumlah keseluruhan skornya 47.5 yang artinya memperoleh nilai tinggi.

The study included manufacturing of fermented dairy products by using full cream milk of four kinds of mammalian: (Buffalos, Cows, Sheep's and Goats); with the use of Lactobacillus casei as a starter for the production of fermented dairy products which had the ability to lower cholesterol percentage in the above mentioned products by (71.4, 70, 74.8 and 67.7)% respectively. The viability of Lb. casei had not been affected significantly during storage shelf life of 21days " The product shelf life " , keeping their therapeutic properties unaltered with high viable number of bacteria at time of consumption. The viable counts of the bacteria after storage period for manufactured products were (1.06× 109, 8.1× 108, 7.5× 108and 8× 108) CFU/ml respectively. These numbers represent a decrease equal to one logarithmic cycle for each of manufactured products of Cows, Sheep's and Goats milk, and the decrease of bacteria's viability of manufactured products of Buffalos milk was less than one logarithmic cycle. Results of statistical analysis showed that there was highly significant differences (P<0.05) in the viable bacterial cells counts between manufactured products. By sensory comparison of the manufactured fermented products together, the results shows that the manufactured products from Buffalos milk was the best then the manufactured products of Cows milk then Sheep's milk then goats milk

Postharvest fungal disease is one of the significant factors that limits the storage period and marketing life of peaches, and even result in serious economic losses worldwide. Biological control using microbial antagonists has been explored as an alternative approach for the management of postharvest disease of fruits. However, there is little information available regarding to the identification the fungal pathogen species that cause the postharvest peach diseases and the potential and mechanisms of using the Bacillus subtilis JK-14 to control postharvest peach diseases. In the present study, a total of six fungal isolates were isolated from peach fruits, and the isolates of Alternaria tenuis and Botrytis cinerea exhibited the highest pathogenicity and virulence on the host of mature peaches. In the culture plates, the strain of B. subtilis JK-14 showed the significant antagonistic activity against the growth of A. tenuis and B. cinerea with the inhibitory rates of 81.32% and 83.45% at 5 days after incubation, respectively. Peach fruits treated with different formulations of B. subtilis JK-14 significantly reduced the mean disease incidences and lesion diameters of A. tenuis and B. cinerea. The greatest mean percent reduction of the disease incidences (81.99% and 71.34%) and lesion diameters (82.80% and 73.57%) of A. tenuis and B. cinerea were obtained at the concentration of 1 × 107 CFU mL−1 (colony forming unit, CFU). Treatment with the strain of B. subtilis JK-14 effectively enhanced the activity of the antioxidant enzymes-superoxide dismutase (SOD), peroxidase (POD) and catalase (CAT) in A. tenuis and B. cinerea inoculated peach fruits. As such, the average activities of SOD, POD and CAT were increased by 36.56%, 17.63% and 20.35%, respectively, compared to the sterile water treatment. Our results indicate that the isolates of A. tenuis and B. cinerea are the main pathogens that cause the postharvest peach diseases, and the strain of B. subtilis JK-14 can be considered as an environmentally-safe biological control agent for the management of postharvest fruits diseases. We propose the possible mechanisms of the strain of B. subtilis JK-14 in controlling of postharvest peach diseases.

Starvation is defined as a failure to ingest and digest food under some extrinsic limitation. Starvation could induce the behavioral consequences, such as searching, movement, parasitism efficiency of arthropods. In this study, we try to evaluate the hunger tolerance and movement rates of P. zhonghuajia on corn and sorghum, and observed the time of P. zhonghuajia biting S. frugiperda and the lethal efficiency of P. zhonghuajia on S. frugiperda under different starvation periods (6 h, 12 h, 24 h, 48 h, 72 h and 96 h). The results showed that without host the survival rates of P. zhonghuajia was significantly reduced over the 96 h. Starvation significantly increased the movement rates of P. zhonghuajia with increasing starvation from 6 to 24 h on the sorghum and corn leaves. When starved for 12 h, starvation significantly decreased the time P. zhonghuajia spent biting S. frugiperda. Proper hunger in P. zhonghuajia caused a 100% mortality rate in S. frugiperda within 2 hours, while a 96-h starved P. zhonghuajia parasited S. frugiperda for 16 hours to achieve a 100% mortality rate. In conclusion, a shorter period of starvation (24 h) could improve the movement rates and lethal efficiency of P. zhonghuajia against S. frugiperda, and ultimately enhance the biological control of P. zhonghuajia.

Three strains of Escherichia coli O157:H7 were grown in tryptic soy broth (TSB) or in a sterile cattle manure extract at 35°C for 18 ± 2 h. Aliquots from both inocula containing 106 CFU/ml were used to inoculate 1-cm3 cubes of beef muscle or adipose tissue by immersion for 20 min at 21°C. After removal from the inoculum, one-half of the samples were analyzed for bacterial cell numbers and pH, and the other half were stored at 4°C for 2 or 3 h before analysis. Samples were analyzed by enumerating bacteria present in liquid droplets deposited on the tissue and bacteria loosely or strongly attached to the tissue in order to determine attachment strength. Total numbers of cells on beef muscle tissue (bacteria in liquid droplets, as well as those loosely and strongly attached) were 5.65 ± 0.14 and 5.76 ± 0.26 log CFU/cm2 for E. coli O157:H7 inocula grown in TSB and manure extract, respectively. The differences in attachment strength between inocula from the two media were not significant (P &gt; 0.05). A 2-h storage period after exposure of muscle tissue to an E. coli O157:H7 inoculum did not influence attachment strength. Numbers of bacteria attached to adipose tissue and muscle (5.31 ± 0.08 and 5.48 ± 0.09 log CFU/cm2, respectively) were not significantly different (P &gt; 0.05). After 3 h at 4°C, the attachment strength of E. coli O157:H7 cells on muscle or adipose tissue had not changed. Overall, the culture medium and type of beef tissue did not affect the numbers of E. coli O157:H7 cells attached, nor the strength of their attachment, to muscle or adipose tissue.
