H2S papers as presumptive tests for Salmonella contamination in tropical drinking water

Coliform bacteria may not be adequate as sole indicators of recent faecal contamination in tropical waters. Salmonella spp. in particular have been found in tropical waters in the absence of traditional indicator bacteria. Remote areas without access to a laboratory have no opportunity for salmonellae analysis as portable tests are currently not available. H2S papers indicate the presence of hydrogen sulphide producing bacteria, a characteristic shared by the majority of Salmonella spp.. The potential of H2S papers to act as a presumptive test for salmonellae in the absence of coliform bacteria was assessed. Salmonella spp. grew in the H2S medium, with an optimum incubation temperature of 37°C and a recommended length of incubation for a negative result of 48 hours. The presence of high numbers of the type of noncoliform bacteria commonly found in drinking water did not affect the performance of the H2S strips. H2S papers are recommended for use in conjunction with a coliform test as a presumptive test for the presence of Salmonella spp. in drinking water.

Epidemics arising from waterborne diseases are a global health problem. Faecal contamination of drinking water is the main cause of these outbreaks. According to WHO (1996) for drinking water to be safe, a 100 ml sample should not contain any coliform bacteria. The standard methods currently used for routine testing have many limitations especially when applied in remote areas. The H2S method has been developed as an on-site, inexpensive and easy to use method to test drinking water for remote and rural areas. The present work analyses the reliability of the H2S method for detecting faecal contamination in drinking water. The minimum level of faecal coliforms that could be detected and the incubation period required at various levels of contamination were studied. The range of temperatures at which the method was effective and the incubation period required at various temperatures were also determined. The H2S method was found to be able to detect contamination down to a level of 1 CFU/100 ml of coliform bacteria. Although the H2S method could be used at a temperature range of 20 to 44°C, temperatures between 28 to 37°C gave faster results. An incubation period of only 24 hours was required at 37°C, which was found to be the most suitable incubation temperature. The incubation period increased with a decrease or increase in temperature.

Enteropathogenic and indicator bacteria become injured in drinking water with exposure to sublethal levels of various biological, chemical and physical factors. One manifestation of this injury is the inability to grow and form colonies on selective media containing surfactants. The resulting underestimation of indicator bacteria can lead to a false estimation of water potability. m-T7 medium was developed specifically for the recovery of injured coliforms (both “total” and fecal) in drinking water. The m-T7 method was used to survey operating drinking water treatment and distribution systems for the presence of injured coliforms that were undetected with currently used media. The mean recovery with m-Endo LES medium was less than 1/100 ml while it ranged between 6 and 68/100ml with m-T7 agar. The majority of samples giving positive results with m-T7 medium yielded no detectable coliforms with m-Endo LES agar. Over 95% of the coliform bacteria in these samples were injured. Laboratory experiments were also done to describe the virulence of injured waterborne pathogens. Enteropathogens including Salmonella tvphimurium, Yersinia enterocolitica and Shigella spp. required up to 20 times the chlorine levels to produce the same injury in enterotoxigenic Escherichia coli (ETEC) and nonpathogenic coliforms. Similar results were seen with Y. enterocolitica exposed to copper. The recovery of ETEC was followed by delayed enterotoxin production, both invitro and in the gut of experimental animals. This indicates that injured waterborne enteropathogenic bacteria can be virulent.

According to the latest SDG recommendations, guidelines on access to safe drinking water have enabled the construction of boreholes in remote areas of developing countries. In Togo,  particularly in the savannah region where access to drinking water remains a major problem for the population, many boreholes have been built for this purpose. The objective of this study is to evaluate the bacteriological quality of the waters of some boreholes built in four (04) prefectures of the savannah region in Togo. A total of 68 samples of drilling water intended for human consumption were collected between January and February 2019 for bacteriological analysis.  These analyses were  carried out according to the standardized routine methods of the French Association for Standardization (AFNOR). The parameters sought or counted in these  samples are those retained by  the 2007 European Union criteria for water intended for human consumption. The analysis reveal that the samples of borehole water are at 50% and 90%   of unsatisfactory hygienic quality respectively compared to the Total Coliforms (CT) and the Total Aerobic Mesophilic Flora (FAMT) which are indicative germs of hygiene failure. The correlation of the germs sought made it possible to distinguish two groups of indicators of contamination: those  responsible for  hygiene  failures and old fecal contamination (ASR) Conclusion: Since the majority of the borehole water analyzed is contaminated by germs indicating  hygiene deficiencies, adequate  treatment of these waters and monitoring of their quality  are necessary in order to protect the population of the savannah region against probable diseases linked to faecal contamination germs.

Waterborne diseases are still common in developing countries as drinking water sources are contaminated and feasible means to reliably treat and disinfect these waters are not available. Many of these developing countries are in the tropical regions of the world where sunlight is plentiful. The objective of this study was to evaluate the effectiveness of combining solar radiation and solar heating to disinfect contaminated water using a modified Family Sol*Saver System (FSP). The non-UV transmittable cover sheet of the former FSP system was replaced with an UV transmittable plastic cover sheet to enable more wavelengths of sunlight to treat the water. Disinfection efficiency of both systems was evaluated based on reduction of the natural populations of faecal coliform, E. coli, enterococci, C. perfringens, total heterotrophic bacteria, hydrogen sulphide producing bacteria and FRNA virus. The results showed that under sunny and partly sunny conditions, water was heated to critical temperature (60°C) in both the FSP systems inactivating more than 3 log (99.9%) of the concentrations of faecal coliform and E. coli to undetectable levels of <1 CFU/100mL within 2-5 h exposure to sunlight. However, under cloudy conditions, the two FSP systems did not reduce the concentrations of faecal indicator bacteria to levels of <1 CFU/100mL. Nonetheless, sufficient evidence was obtained to show that UV radiation of sunlight plus heat worked synergistically to enhance the inactivation of faecal indicator bacteria. The relative log removal of indicator microorganism in the FSP treated water was total heterotrophic bacteria < C. perfringens < F RNA virus < enterococci < E. coli < faecal coliform. In summary, time of exposure to heat and radiation effects of sunlight were important in disinfecting water by solar units. The data indicated that direct radiation of sunlight worked synergistically with solar heating of the water to disinfect the water. Thus, effective disinfection was observed even when the water temperature did not reach 60°C. Finally, the hydrogen sulphide test is a simple and reliable test that householders can use to determine whether their water had been sufficiently disinfected.

Water is the main source for life and also the most severe substance caused by pollution. The mandatory parameters for determining microbiological quality of drinking water are total non-fecal Coliform bacteria and Coliform fecal (Escherichia coli). Coliform bacteria are a group of microorganisms commonly used as indicators, where these bacteria can be a signal to determine whether a water source has been contaminated by bacteria or not, while fecal Coliform bacteria are indicator bacteria polluting pathogenic bacteria originating from human feces and warm-blooded animals (mammals) . The water inspection method in this study uses the MPN (Most Probable Number) method which consists of 3 tests, namely, the presumption test, the affirmation test, and the reinforcement test. The results showed that of 15 drinking water samples 8 samples were tested positive for Coliform bacteria with the highest total bacterial value of sample number 1, 15 (210/100 ml), while 7 other samples were negative. From 8 positive Coliform samples only 1 sample was stated to be negative fecal Coliform bacteria and 7 other samples were positive for Coliform fecal bacteria with the highest total bacterial value of sample number 1 (210/100 ml).

It was postulated that a phytobiotic preparation containing cinnamon oil and citric acid added to drinking water for chickens in a suitable amount and for a suitable time would beneficially modify the microbiota composition and morphology of the small intestine, thereby improving immunity and growth performance without inducing metabolic disorders. The aim of the study was to establish the dosage and time of administration of such a phytobiotic that would have the most beneficial effect on the intestinal histology and microbiota, production results, and immune and metabolic status of broiler chickens. The experiment was carried out on 980 one-day-old male chickens until the age of 42 days. The chickens were assigned to seven experimental groups of 140 birds each (seven replications of 20 individuals each). The control group (G-C) did not receive the phytobiotic. Groups CT-0.05, CT-0.1, and CT-0.25 received the phytobiotic in their drinking water in the amount of 0.05, 0.1, and 0.2 mL/L, respectively, at days 1–42 of life (continuous application, CT). The birds in groups PT-0.05, PT-0.5, and PT-0.25 received the phytobiotic in the same amounts, but only at days 1–7, 15–21, and 29–35 of life (periodic application, PT). Selected antioxidant and biochemical parameters were determined in the blood of the chickens, as well as parameters of immune status and redox status. The morphology of the intestinal epithelium, composition of the microbiome, and production parameters of chickens receiving the phytobiotic in their drinking water were determined as well. The addition of a phytobiotic containing cinnamon oil and citric acid to the drinking water of broiler chickens at a suitable dosage and for a suitable time can beneficially modify the microbiome composition and morphometry of the small intestine (total number of fungi p < 0.001, total number of aerobic bacteria p < 0.001; and total number of coliform bacteria p < 0.001 was decreased) improving the immunity and growth performance of the chickens (there occurred a villi lengthening p = 0.002 and crypts deepening p = 0.003). Among the three tested dosages (0.05, 0.1, and 0.25 mL/L of water) of the preparation containing cinnamon oil, the dosage of 0.25 mL/L of water administered for 42 days proved to be most beneficial. Chickens receiving the phytobiotic in the amount of 0.25 mL/L had better growth performance, which was linked to the beneficial effect of the preparation on the microbiome of the small intestine, metabolism (the HDL level p = 0.017 was increased; and a decreased level of total cholesterol (TC) p = 0.018 and nonesterified fatty acids (NEFA) p = 0.007, LDL p = 0.041, as well as triacylglycerols (TAG) p = 0.014), and immune (the level of lysozyme p = 0.041 was increased, as well as the percentage of phagocytic cells p = 0.034, phagocytosis index p = 0.038, and Ig-A level p = 0.031) and antioxidant system (the level of LOOH p < 0.001, MDA p = 0.002, and the activity of Catalase (CAT) p < 0.001 were decreased, but the level of ferric reducing ability of plasma (FRAP) p = 0.029, glutathione p = 0.045 and vitamin C p = 0.021 were increased).

Poor drinking water quality can affect pigs’ health and performance. The disinfection of water may enhance microbial water quality. In this study, bacteria and endotoxins in sodium hypochlorite-treated and -untreated water from one pig nursery were analyzed. Water samples were taken from incoming water and from compartments with treated and untreated water at the beginning and end of pipes and from nipples. The farm was visited 14 times to measure total bacteria counts and concentrations of Pseudomonas spp. and endotoxins. Additionally, the occurrence of coliform bacteria was analyzed. A mixed model analysis revealed significant reductions in total bacteria counts and Pseudomonas spp. in treated water at the beginning of pipes and at nipple drinkers. The differences between bacteria concentrations at the end of pipes had no clear trend. Endotoxin concentrations were approximately equal at the beginning of pipes and at nipple drinkers but were found to have differences at the end of pipes. The occurrence of coliform bacteria was significantly reduced in treated water. The application of sodium hypochlorite can significantly reduce bacteria in water pipes. Endotoxin concentrations were mostly unaffected by water treatment. Disinfection of the dead-end pipe sections failed, and thus these parts should be regarded as potential contamination sources.

Paired water samples were collected and analysed for thermotolerant coliforms (TTC) from 20 sources (17 developed or rehabilitated by Oxfam and 3 others) and from the stored household water supplies of 100 households (5 from each source) in 13 towns and villages in the Kailahun District of Sierra Leone. In addition, the female head of the 85 households drawing water from Oxfam improved sources was interviewed and information recorded on demographics, hygiene instruction and practices, sanitation facilities and water collection and storage practices. At the non-improved sources, the arithmetic mean TTC load was 407/100 ml at the point of distribution, rising to a mean count of 882/100 ml at the household level. Water from the improved sources met WHO guidelines, with no faecal contamination. At the household level, however, even this safe water was subject to frequent and extensive faecal contamination; 92.9% of stored household samples contained some level of TTC, 76.5% contained more than the 10 TTC per 100 ml threshold set by the Sphere Project for emergency conditions. The arithmetic mean TTC count for all samples from the sampled households was 244 TTC per 100 ml (geometric mean was 77). These results are consistent with other studies that demonstrate substantial levels of faecal contamination of even safe water during collection, storage and access in the home. They point to the need to extend drinking water quality beyond the point of distribution to the point of consumption. The options for such extended protection, including improved collection and storage methods and household-based water treatment, are discussed.
