Formulation of Composite Flour with Antioxidant from Goroho Plantain Flour (Musa Acuminafe, sp) and Yellow Pumpkin Flour (Cucurbita moschata) and Its Application on Biscuit Making

Locally grown crops with phytochemical antioxidant content i.e. goroho plantain and yellow pumpkin were used to develop composite flour. Mixture experiment with simplex lattice design was used for formulation to study the effect of blending goroho plantain flour (GF) with yellow pumpkin flour (PF) on phytochemical content and antioxidant capacity of developed composite flours. Responses measured including phytochemical content (total phenolic and carotenoid), antioxidant capacity (DPPH assay, FRAP assay, phosphomolybdenum assay, and reducing power assay), colour values of flours, proximate components, and sensory quality of biscuits made from composite flours. The results showed that increase in proportion of PF improved the phytochemical content, antioxidant capacity, and proximate components with the exception of carbohydrates. However, substitution of GF with PF reduced sensory ratings for all the sensory attributes of biscuits developed from composite flours. Among biscuits made from composite flours, sensory ratings for aroma, colour, texture, and taste were not statistically different (p>0.05). PF can be used as fortification material to improve the phytochemical antioxidant content in composite flours prepared from GF and PF, or flours from other locally grown plants.

ABSTRAK Tanaman-tanaman lokal yang mengandung antioksidan, yaitu pisang goroho dan labu kuning, digunakan untuk mengembangkan tepung komposit.  Penelitian dilakukan untuk mengevaluasi efek pencampuran tepung pisang goroho dengan tepung labu kuning terhadap kandungan fitokimia antioksidan (total fenolik dan beta karoten) dan kapasitas antioksidan (kemampuan menangkal radikal DPPH dan daya reduksi) dari tepung komposit yang dihasilkan dengan menggunakan bantuan ultrasonik pada tahapan ekstraksi. Formulasi tepung komposit yang digunakan (tepung pisang goroho:tepung labu kuning) adalah 75:25, 50:50, dan 25:75. Kandungan total fenolik tepung komposit berkisar antara 98,2- 78,8 mg/kg GAE, dan kandungan beta karoten tepung komposit berkisar antara 186,1-497,8 µg/g. Hasil penelitian menunjukkaan peningkatan  kandungan fitokimia antioksidan dan kapasitas antioksidan pada tepung komposit terjadi dengan meningkatnya proporsi tepung labu kuning. ABSTRACT Locally grown crops with antioxidant content i.e. goroho plantain dan yellow pumpkin were used to develop composite flours. Flour formulations (goroho plantain flour: yellow pumpkin flour) used were 75:25, 50:50, and 25:75.  Total phenolic content of composite flours ranged from 98.2-178.8 mg/kg GAE, and beta carotene content of composite flours ranged from 186,1-497,8 µg/g. The study showed that increased in phytochemical antioxidant content and antioxidant capacity of composite flours were due to increasing in yellow pumpkin flour proportion in composite flour.

ABSTRACT The objective of this work was to evaluate the physicochemical and microbiological characteristics, antioxidant capacity and phenolic compounds of organic cherry tomatoes grown under fertigation with organic dairy cattle wastewater (DCW) with different nitrogen rates. Tomato plants, grown in an agroecological farm in Seropédica, State of Rio de Janeiro, Brazil, were subjected to four different nitrogen rates (T1=0, T2=50, T3=100 and T4=150% of N). The moisture, lipids, ashes, protein and total fiber contents, soluble solids (ºBrix), reducing and total sugars (%), pH and total titratable acidity (mg NaOH per 100 g) were evaluated. The total phenolic content (TPC) and the antioxidant capacity was determined by the DPPH and FRAP methods. The different nitrogen rates (%N) affected the pH, protein and soluble solids contents. The increase in %N increased the antioxidant capacities, according to the DPPH assay, and TPC. On the other hand, the tomatoes under fertigation with the highest %N presented lower antioxidant capacities according to the FRAP assay. The fertigation did not affect the microbiological characteristics of the tomatoes, which presented fecal coliforms count <3 NMP g-1 and absence of Salmonella in 25 g.

Modern civilization is facing more than hundreds of disorders associated with free radicals and natural antioxidants from non-edible plants are gaining importance to fight these disorders. The intention of this report is to evaluate a well known medicinal weed Ageratum conyzoides stems for its antioxidant and cytotoxic Effects. Antioxidant potentiality of the crude methanolic extract of the Ageratum conyzoides (AC) stems was investigated on DPPH scavenging activity, reducing ability, total antioxidant capacity as well as total phenolic contents. Cytotoxic study was done by brine shrimp lethality bioassay and vincristin sulphate was used as standard. The total phenols and total antioxidant capacity of AC was found to be 38.125 ± 2.01mg/g equivalent of gallic acid and 333.37 ± 4.22mg/gm equivalent of ascorbic acid, respectively. The percentage (%) scavenging of DPPH free radical of the extract was found to be concentration dependent with IC50 value 46.01 ± 2.23µg/ml while IC50 value of standard ascorbic acid was found to be 29.56 ± 0.11?g/ml. The reducing power of AC was found to be concentration dependent. The cytotoxicity exhibited by AC was found promising with LC50 value 1.32?g/ml, comparing with the LC50 (0.689?g/ml) values of vincristin sulphate. Zilla spinosa is commonly used in traditional medicine to treat gastrointestinal disorders and diabetes. In this study, aqueous ethanol (AE) and aqueous methanol (AM) extracts from aerial parts and roots of Z. spinosa were investigated. The total phenolic (TPC) and flavonoid (TFC) contents and antioxidant capacities in terms of 1,1-diphenyl-2-picrylhydrazyl (DPPH) scavenging, hydrogen peroxide (H2O2), and ferric reducing antioxidant power (FRAP) assays were determined, and the correlations among the results were assessed using Pearson’s correlation. The antimicrobial activity was assessed through agar disc diffusion and broth microdilution assays. Phytochemical screening showed that Z. spinosa extracts had alkaloids, glycosides, saponins, triterpenoids, phenols, and flavonoids in different abundances. The aerial part-AE extract contained low TPC (30.17 ± 4.24 mg GAE/g) and TFC (7.40 ± 1.02 mg QE/g) and displayed significant antioxidant capacity in the DPPH (IC50 = 52.17 ± 7.30 μg/mL), H2O2 (91.22 ± 2.60 μg/mL), and FRAP (EC50 = 98.70 ± 2.21 μg/mL) assays. By contrast, the root-AM extract contained high amounts of TPC (87.72 ± 7.75 mg GAE/g) and TFC (25.60 ± 1.57 mg QE/g). It showed significantly high antioxidant activity with IC50 values of 12.33 ± 1.88 μg/mL in the DPPH and 39.37 ± 2.59 μg/mL in the H2O2 assays, as well as reducing power capacity with an EC50 value of 20.82 ± 1.14 μg/mL in the FRAP assay. Both TPC and TFC were exhibited negative correlations ( p < 0.01 ) with the IC50 and EC50 values obtained in the applied antioxidant assays. The aerial part-AM extract showed the highest inhibitory activity against Staphylococcus aureus (26.5 ± 0.20 mm), followed by Shigella flexneri (19.4 ± 0.40 mm) and Proteus mirabilis (17.7 ± 0.49 mm). S. aureus was the most affected microorganism with a minimum inhibitory concentration of 128 μg/mL against the aerial part-AM extract. Interestingly, all evaluated extracts showed potent antifungal activity against Candida albicans. However, aerial part-AM was the most effective, with an inhibition zone of 12.6 ± 0.17 mm. The results concluded that Z. spinosa possesses different phytochemicals displaying significant antioxidant and antimicrobial activities, thus lending credence to its use in traditional medicine.

AbstractOrthosiphon aristatus (Blume) Miq. of the Lamiaceae family, called as kumis kucing in Indonesia, is a valuable medicinal plant for their pharmacological properties. The present study comprised of fifteen genotypes of O. aristatus was undertaken to evaluate the genotypes based on phytochemical content and pharmacological activities of leaves ethanol extract. Chemometric analysis (correlation and principal component analysis) was also used to investigate the genetic variability based on phytochemical content and pharmacological activities of O. aristatus genotypes. Results of phytochemical characterization showed that total phenolic ranged from 1.48 to 36.08 (maximum in A15) mg GAE/g DW, total flavonoid ranged from 0.10 to 3.07 (maximum in A15) mg QE/g DW, sinensetin ranged from 0.36 to 4.02 (maximum in A11) mg/g DW, and rosmarinic acid ranged 0.06 to 7.25 (maximum in A7) mg/g DW. Antioxidant activity was tested using DPPH and FRAP assay. Antioxidant results showed that DPPH ranged from 1.68 to 15.55 (maximum in A15) μmol TE/g DW and FRAP ranged from 0.07 to 1.60 (maximum in A1 and A7) μmol TE/g DW. The genotype A8 showed the highest cytotoxic activities against HeLa (66.25%) and MCF-7 (61.79%) cell lines. Maximum α-glucosidase inhibitory activity was recorded in genotype A2 with the value of 62.84%. The genotypes A1, A2, A7, A11, and A15 were identified as superior based on their phytochemicals content and pharmacological activities coupled with chemometric analysis. This finding is important for breeding studies and also the pharmaceutical perspective of O. aristatus.

Many plants with high antioxidant activity are great of significant in the pharmaceutical and cosmetic industries. Oxidative stress plays a major part in the development of many diseases including cancer, which is known imbalance free radicals and antioxidants. Herein, new natural antioxidant compounds have great interest in the scientific research. The genus Silene is a major group in the Caryophyllaceae family. In Turkey, Silene species have been used for several medicinal purposes such as skin softening, asthma, bronchitis. In our study, the antioxidant capacity of three Silene species (S. conoidea, S. dichotoma and S. italica) were evaluated by different in vitro assays, including free radical scavenging, reducing power, metal chelating, and phosphomolybdenum. In addition, total phenolic and flavonoid contents were analyzed spectrophotometrically. The water extracts contained higher total phenolic content than ethyl acetate extracts. All extracts showed antioxidant capacity. This data indicated that Silene species could potentially be used as antioxidant sources in pharmaceutical and cosmetic areas.

Abstract Heracleum lasiopetalum Boiss, Kelussia odoratissima Mozaff., and Echinophora platyloba DC. belong the Apiaceae family. They are Iranian endemic plants. These three herbs have been used as food additives in traditional preparations such as pickles. Antioxidant activity (AA) of methanol extracts (ME) of the plants was evaluated by three assays, including DPPH, FRAP, and TEAC. From all three assays, comparing all the MEs for their IC50 and EC1 values, E. platyloba had the highest AA. Total phenolic content (TPC) of the extracts ranged from 74 to 120 mg TAE/g. The extract of H. lasiopetalum exhibited the highest TPC. The flavonoids content (FC) of the extracts ranged from 7.63 to 14.52 mg RE/g, from which the extract of E. platyloba had the highest flavonoids concentration. A positive correlation between the FC and AA in DPPH assay was found. A significant correlation was also found between the TPC and AA in FRAP assay. These results suggested that the level of AA in these plants varied in a great extent. Our results indicated that extract of E. platyloba could be an important dietary source of flavonoids compounds with high antioxidant capacity. In addition, E. platyloba can be used as an alternative preservative and natural flavor instead of synthetic ones in food industry (especially pickles)

Solubility of phytoconstituents depends on the polarity of the extraction medium used, which might result in the different pharmacological responses of extracts. In line with this, ethnomedicinally important food plant (i.e., Caralluma tuberculata extracts) have been made in fourteen distinct solvent systems that were then analyzed phytochemically via total phenolic amount estimation, total flavonoid amount estimation, and HPLC detection and quantification of the selected polyphenols. Test extracts were then subjected to a battery of in vitro assays i.e., antioxidants (DDPH scavenging, antioxidant capacity, and reducing power estimation), antimicrobial (antibacterial, antifungal, and antileishmanial), cytotoxic (brine shrimps, THP-1 human leukemia cell lines and normal lymphocytes), and protein kinase inhibition assays. Maximum phenolic and flavonoid contents were computed in distilled water–acetone and acetone extracts (i.e., 16 ± 1 μg/mg extract and 8 ± 0.4/mg extract, respectively). HPLC-DAD quantified rutin (0.58 µg/mg extract) and gallic acid (0.4 µg/mg extract) in methanol–ethyl acetate and methanol extracts, respectively. Water–acetone extract exhibited the highest DPPH scavenging of 36 ± 1%. Total reducing potential of 76.0 ± 1 μg/mg extract was shown by ethanol chloroform while maximum total antioxidant capacity was depicted by the acetone extract (92.21 ± 0.70 μg/mg extract). Maximal antifungal effect against Mucor spp., antileishmanial, brine shrimp cytotoxicity, THP-1 cell line cytotoxicity, and protein kinase inhibitory activities were shown by ethyl acetate-methanol (MIC: 50 µg/disc), n-hexane (IC50: 120.8 ± 3.7 µg/mL), ethyl acetate (LD50: 29.94 ± 1.6 µg/mL), distilled water–acetone (IC50: 118 ± 3.4 µg/mL) and methanol–chloroform (ZOI: 19 ± 1 mm) extracts, respectively. Our findings show the dependency of phytochemicals and bioactivities on the polarity of the extraction solvent and our preliminary screening suggests the C. tuberculata extract formulations to be tested and used in different ailments, however, detailed studies remain necessary for corroboration with our results.

The present study was carried out to demonstrate the combined effects of different maize flour, ascorbic acid and sugar on the physical, textural and sensory properties of composite breads. The composite flour was prepared using 70% of wheat flour and 30% of flour obtained from grain of differently coloured maize - light blue, blue, red and yellow maize flour. Chemical characterization of composite flours made with four different types of maize was also assessed. Furthermore, the content of total phenolics, flavonoids, anthocyanins, phenolic acids and antioxidant capacity in composite flours was determined. The total of 12 breads that were prepared, four of which were control composite breads, four breads with ascorbic acid, and four were breads with ascorbic acid and sugar. The content of total phenolic compounds showed clear differences among all composite flours. The anthocyanins content determined in composite flours was in the following descending order: blue>red>light blue, while in the yellow maize composite flour anthocyanins were not detected. The results showed that the addition of AsA (0.025%) and sugar (5%) negatively affected the volume as well as the specific volume of composite wheat-maize breads. The texture analysis showed that the addition of AsA in amount of 0.025% had no impact on springiness, cohesiveness and resilience of bread crumb, while it increased crumb hardness. However, composite breads made with AsA and AsA and sugar showed a more compact structure, with a larger number of cells and smaller mean cell areas. Bread samples with AsA and sugar in the tested doses had the lowest springiness, which is indicative of brittleness and reflects the tendency of the bread to crumble when slicing. Results of the sensory evaluation revealed that the AsA and sugar addition had a generally positive effect on the investigated sensory attributes.

Abstract Carrot (Daucus carota L.) is one of the main root vegetables rich in bioactive compounds with appreciable health-promoting properties, largely consumed in Algeria. In the current study, the storage effect (at 4 °C throughout 12 days) on bioactive compound stability and the antioxidant activity of two Algerian orange carrot varieties (Supermuscade and Touchon) were investigated. Total phenolic content of samples was determined by the Folin–Ciocâlteu method. Antioxidant capacity was determined spectrophotometrically, based on the evaluation of Free Radical Scavenging Activity (FRSA) using DPPH radical and Ferric Reducing Power (FRP). The results showed that the Touchon variety is richer in phenolics, flavonoids, and carotenoids and presents higher antioxidant activity in comparison with the Supermuscade variety. At the end of storage, the bioactive compound content and antiradical activity increased significantly (p < 0.05). Also, an extremely significant correlation (p < 0.001) was observed between the antioxidant contents and the antioxidant capacities of aqueous carrot extracts.
