AbstractOrthosiphon aristatus (Blume) Miq. of the Lamiaceae family, called as kumis kucing in Indonesia, is a valuable medicinal plant for their pharmacological properties. The present study comprised of fifteen genotypes of O. aristatus was undertaken to evaluate the genotypes based on phytochemical content and pharmacological activities of leaves ethanol extract. Chemometric analysis (correlation and principal component analysis) was also used to investigate the genetic variability based on phytochemical content and pharmacological activities of O. aristatus genotypes. Results of phytochemical characterization showed that total phenolic ranged from 1.48 to 36.08 (maximum in A15) mg GAE/g DW, total flavonoid ranged from 0.10 to 3.07 (maximum in A15) mg QE/g DW, sinensetin ranged from 0.36 to 4.02 (maximum in A11) mg/g DW, and rosmarinic acid ranged 0.06 to 7.25 (maximum in A7) mg/g DW. Antioxidant activity was tested using DPPH and FRAP assay. Antioxidant results showed that DPPH ranged from 1.68 to 15.55 (maximum in A15) μmol TE/g DW and FRAP ranged from 0.07 to 1.60 (maximum in A1 and A7) μmol TE/g DW. The genotype A8 showed the highest cytotoxic activities against HeLa (66.25%) and MCF-7 (61.79%) cell lines. Maximum α-glucosidase inhibitory activity was recorded in genotype A2 with the value of 62.84%. The genotypes A1, A2, A7, A11, and A15 were identified as superior based on their phytochemicals content and pharmacological activities coupled with chemometric analysis. This finding is important for breeding studies and also the pharmaceutical perspective of O. aristatus.

Durio kutejensis has known potential as herbal medicine in Kalimantan. Many things can be explored from D. kutejensis related pharmacological activity in every part of the plant included root bark. This study investigated the phytochemical content with a qualitative test and tested cytotoxic activities of D. kutejensis root bark on MCF-7 cell lines. Cytotoxic activity tested on MCF-7 cells with MTT assay method. The result showed that D. kutejensis root bark contains flavonoid, tannin, terpenoid, phenol, and saponin. IC50 value for ethanol extracts of root bark, n-hexane fractions, ethyl acetate fractions, and doxorubicin on MCF-7 cells are 761.29; 280.5; 207.08; and 0.25 μg/mL, respectively. In conclusion, D. kutejensis root bark has some secondary metabolites but no cytotoxic activity on MCF-7 cells. Further research on other compounds to be investigated and test to other pharmacological activity.

Crab apple (Malus baccata (L.) Borkh.) was mainly distributed in Europe as an ornamental plant, but the nutritional properties of its edible fruits were not fully revealed. The aim of the current study was to characterize the phytochemical composition of ripen carb apple fruits and to evaluate their nutritional and antioxidant potentials. The fruits were assayed for moisture and ash content, proteins, lipids, carbohydrates, titratable acidity (TA), pH, total phenolic compounds and natural pigments. Among the analyzed carbohydrates cellulose was found in the highest content (6% dw), followed by sugars (sucrose, glucose and fructose) and 1.8 % dw uronic acids. The total chlorophylls and carotenoids contents in their fruits were 6.51 and 4.80 μg/g fw, respectively. Total monomeric anthocyanins were not detected. The highest content of total phenolic compounds (2.67 mg GAE/g fw) was found in 95 % ethanol extract from fruits, while the total flavonoids were relatively low – 0.1 mg QE/g fw. DPPH assay (17.27 mM TE/g fw) and FRAP assay (14.34 mM TE/g fw) demonstrated in vitro antioxidant activities of crabapple. Malus baccata fruits were evaluated as a rich source of dietary fibers and phenolic compounds with significant antioxidant potential that could be used in human nutrition.

Locally grown crops with phytochemical antioxidant content i.e. goroho plantain and yellow pumpkin were used to develop composite flour. Mixture experiment with simplex lattice design was used for formulation to study the effect of blending goroho plantain flour (GF) with yellow pumpkin flour (PF) on phytochemical content and antioxidant capacity of developed composite flours. Responses measured including phytochemical content (total phenolic and carotenoid), antioxidant capacity (DPPH assay, FRAP assay, phosphomolybdenum assay, and reducing power assay), colour values of flours, proximate components, and sensory quality of biscuits made from composite flours. The results showed that increase in proportion of PF improved the phytochemical content, antioxidant capacity, and proximate components with the exception of carbohydrates. However, substitution of GF with PF reduced sensory ratings for all the sensory attributes of biscuits developed from composite flours. Among biscuits made from composite flours, sensory ratings for aroma, colour, texture, and taste were not statistically different (p>0.05). PF can be used as fortification material to improve the phytochemical antioxidant content in composite flours prepared from GF and PF, or flours from other locally grown plants.

Sarcopoterium spinosum (L.) is a medicinal plant traditionally used for the treatment of various diseases including cancer in the Near- and Middle East. The fractions and constituents of the ethanol extract of S. spinosum were screened for in vitro cytotoxic activities on Jurkat (acute T-lymphoblastic leukemia), HeLa (cervical adenocarcinoma), MCF-7 (mammary gland adenocarcinoma), Caco-2 (human colorectal adenocarcinoma), and MDA-MB-231 (mammary gland adenocarcinoma) cell lines using the MTT (3-(dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) assay. The ethanol extract was subsequently re-extracted with ethyl acetate and in its sub-fraction obtained by column chromatography three compounds (stachydrine, benzalkonium chloride and rutine) were the first time identified by nuclear magnetic resonance (NMR) analyses. The most active subfraction showed cytotoxic activity against HeLa, MCF-7, and Caco-2 cell lines. The three compounds mentioned, as standards of high-performance liquid chromatography (HPLC) quality, were studied individually and in combination. Cytotoxic activity observed might be due to the presence of benzalkonium chloride and rutin. Benzalkonium chloride showed the strongest growth suppression effect against HeLa cells (IC50 8.10−7 M) and MCF-7 cells (IC50 5.10−6 M). The mixture of stachydrine and benzalkonium chloride allowed a synergistic cytotoxic effect against all tested cancer and normal cells to be obtained. Anti-cancer activity of the plant extract of S. spinosum remains under-investigated, so this research describes how the three major compounds identified in the ethyl acetate extract can exert a significant dose dependent in vitro cytotoxicity.

Plant natural products have a long history of being a rich source of neuroactive drugs.The total phenolic and flavonoid content, and antioxidant activity of ethanol, chloroform and aqueous extracts of AlgerianMedicagoSativaL, leaves were measured to discover new potential sources of Algerian natural antioxidants and neuroactives. All extracts were non-neurotoxic up to 1 mg/ ml for 24 h exposure. The total phenolcontent of M.Sativa extracts ranged between 38 and 69 mg GAE per g extracts. All of the extracts displayed antioxidant capacityassessed by two methods, but at different levels depending on the concentration and the extraction solvent used. The overall antioxidant potency was in the order water > chloroform> ethanol extracts, and the phenolic content exhibited a positive correlation with the antioxidant activity of M. Sativa extracts.Despite a weaker antioxidant activity, the ethanol extract elicited a calcium spike and suppressed the KCl-induced depolarisation response in neurons, indicating effect on ligand-and/or voltage gated calcium channels. In contrast, the chloroform and water extractsdisplayed no apparent direct functional effects. In conclusion, the antioxidant and functional properties related to phenolic content of Algerian Medicago Sativa L. in water and ethanol extracts, respectively.

Dates seeds are one of the byproducts of the date palm juice industry which has the potential for further processing. Dates seeds contain active compounds that are known to have ability to inhibit αglucosidase. This study aims to measure the activity of 70% ethanol extract and the fraction of date palm seeds with a mechanism in inhibiting αglucosidase. The measurement of inhibitory activity was carried out by extracting date palm seeds using 70% water and ethanol. The extraction product is then fractionated with n-hexane, ethyl acetate, n-butanol, and water. Then the total phenolic and flavonoid calculations were calculated for each extract and fraction. Measurement of α-glucosidase inhibition activity was measured based on reaction of substrate to enzyme by the addition of inhibitors. The results showed that the ethanolextract had the highest total phenolic and flavonoid, which were 76.86 mg GAE / g and 21.19 mg QE / g, respectively. n-hexane fraction have the best inhibitory activity seen from the IC50 value, which is 12.69 mg/L. The kinetics of inhibition of 70% ethanol extract of date palm seeds are mixed inhibition. Keywords: α-glucosidase, date seeds, flavonoids, kinetic inhibition, phenolics

The crude ethanol extracts of four Indonesian medicinal plants namely Curcuma xanthorrhiza Roxb.,Phyllanthus niruri Linn., Andrographis paniculata Ness., and Curcuma aeruginosa Roxb. wereexamined for their antioxidant (radical scavenging) activity using 2, 2-diphenyl-2-picrylhydrazyl(DPPH) free radical and cytotoxicity using brine shrimp lethality test (BSLT). The total phenoliccontent was used the Folin-Ciocalteu method. IC50 values for DPPH radical scavenging activityranged from 14.5 to 178.5 μg/ml, with P. niruri having the lowest value and therefore the mostpotent, and C. aeruginosa having the highest value. LC50 values for BSLT ranged from 210.3 to593.2 μg/ml, with C. xanthorrhiza and A. paniculata having the lowest and highest values,respectively. The total phenolic content of the Indonesian plants ranged from 133.0 ±3.7 to863.3±54.7 mg tannic acid equivalent per 1 g extract, with C. aeruginosa and P. niruri having thelowest and highest values, respectively. A positive correlation between free radical scavengingactivity and the content of phenolic compounds was found in the four of Indonesian medicinal plants.

Honey maturity is an important factor in evaluating the quality of honey. We established a method for the identification of natural mature acacia honey with eighteen physicochemical parameters combined with chemometric analysis. The analysis of variance showed significant differences between mature and immature acacia honey in physicochemical parameters. The principal component analysis explained 82.64% of the variance among samples, and indicated that total phenolic content, total protein content, and total sugar (glucose, fructose, sucrose) were the major variables. The cluster analysis and orthogonal partial least squares-discriminant analysis demonstrated that samples were grouped in relation to the maturity coinciding with the results of the principal component analysis. Meanwhile, the 35 test samples were classified with 100% accuracy with the method of multi-physicochemical parameters combined with chemometric analysis. All the results presented above proved the possibility of identifying mature acacia honey and immature acacia honey according to the chemometric analysis based on the multi-physicochemical parameters.

Belimbing wuluh merupakan tanaman jenis buah dan obat tradisional. Tanaman belimbing wuluh sudah sering dimanfaatkan masyarakat salah satunya untuk mengobati penyakit seperti batuk dan radang rektum. Tujuan penelitian ini adalah menguji senyawa fitokimia, menganalisis kandungan total fenolik dan flavonoid, serta aktivitas antioksidan dan antiinflamasi secara in vitro pada ekstrak etanol daun belimbing wuluh. Metode yang dilakukan pada penelitian ini adalah ekstraksi daun belimbing wuluh, skrining fitokimia, perhitungan total fenol dan flavonoid, uji aktivitas antioksidan, dan uji aktivitas antiinflamasi. Senyawa fitokimia yang terkandung pada ekstrak etanol daun belimbing wuluh adalah saponin, tanin, steroid, flavonoid, dan alkaloid. Kandungan total fenolik dan flavonoid ekstrak etanol daun belimbing wuluh secara berturut-turut sebesar 39,03 dan 97,28 µg QE/mg. Ekstrak etanol daun belimbing wuluh memiliki aktivitas antioksidan yang tergolong sangat kuat, sementara aktivitas antiinflamasinya terutama ditunjukkan pada konsentrasi ekstrak 200 µg/ ml, memiliki nilai persen inhibisi hemolisis yang paling tinggi. Kesimpulannya, ekstrak etanol daun belimbing wuluh dapat menjadi salah satu sumber antioksidan dan antiinflamasi alami.Ethanol Extracts of Averrhoa Bilimbi Leaf Demonstrated Antioxidative and Anti-inflammatory ActivityAbstractAverrhoa bilimbi, a fruit-bearing plant, has been traditionally used for medicinal purposes such as treatment of cough and rectal inflammation. In this current work, ethanol extract of Averrhoa bilimbi leaf was analyzed, with regard to phytochemical composition, i.e. total phenolic, and flavonoid, antioxidant activity, and in vitro antiinflammation activity. Extraction of carambola leaves, phytochemical screening, total phenolic and flavonoid contens, assay of antioxidant activity, and assay of anti-inflammatory activity were done in this research. As a result, the leaf extract positively contained some phytochemical compounds, i.e. saponin, tannin, steroid, flavonoid, and alkaloid. Furthermore, total phenolic and flavonoid of the leaf extract was found at 39.03 and 97.28 µg QE/mg extract, respectively. Additionally, antioxidant activity of the leaf extract was classified as very strong, while its anti-inflammatory feature at extract concentration of 200 µg/ml exhibited the highest inhibition of hemolysis. In summary, the ability of Averrhoa bilimbi leaf ethanol extracts to act as antioxidative and anti-inflammatory agents was determined and this may open the use for natural antioxidant and anti-inflammatory agents.

Abstract. Nurcholis W, Khumaida N, Bintang M, Syukur M. 2021. GC-MS analysis of rhizome ethanol extracts from Curcuma aeruginosa accessions and their efficiency activities as anticancer agent. Biodiversitas 22: 1179-1186. This work aimed to evaluate the bioactive compounds and anticancer activity in rhizome extract of ten Curcuma aeruginosa accessions to explore their pharmacological values. The GC-MS analysis was used to identify bioactive compounds. The cytotoxic activity was performed against MCF-7 (Human breastadeno carcinoma) and Vero cell lines using MTT assay. The GC-MS analysis revealed 71 of the compounds as sesquiterpenes (36), monoterpenes (20), phenolics (5), diterpenes (4), phenanthrene (1), tetrapeptides (1), oxazole (1), triazine (1), piperidine (1), and oxygenated hydrocarbons (1). The isocurcumenol was the most dominant metabolite in ethanol extract of C. aeruginosa rhizome, with the highest produced by KP accession (22.01%) followed MD accession (21.12%). However, camphor and β-elemene were the metabolites produced by all accessions studied. In the Vero cell line as a normal cell, the cytotoxic activity varied from 13.28% (MD) to 45.17% (PW). Furthermore, the cytotoxic activity ranged from 1.16% (LC) to 49.70% (MD) against the MCF-7 cell line. The highest anticancer activity was produced in MD accessions; thus, it can be used as a source of quality raw materials for the pharmaceutical and food industry. Besides that, it can also be further developed to obtain superior varieties through plant breeding programs.
