The role of phenolic compounds on antihypercholeserol activity of Virgin Coconut Oil (VCO) has been investigated. The in vivo studies ware carried out by treatment of two groups of Wistar white mouse (Ratus norvegicus) using high phenolic VCO and low phenolic VCO respectively, followed by analysis of lipid profile in blood and liver serum of the mouse. In addition a group of hypercholesterol mouse was treated with low phenolic VCO and the blood serum lipid profile was compared with untreated hypercholesterol mouse. The results show that phenolic compound play an important role on antihypercholesterol of VCO. Group of mouse treated with high phenolic VCO have better lipid profile (blood serum: total cholesterol: 70 mg/dL, triglyceride: 76 mg/dL, HDL: 20 mg/dL, LDL: 35 mg/dL; liver serum: total cholesterol:7 mg/dL, triglyceride: 19 mg/dL) compared with the group treated with low phenolic VCO (blood serum: total cholesterol: 82 mg/dL, triglyceride: 100 mg/dL, HDL: 21 mg/dL, LDL: 41 mg/dL; liver serum: total cholesterol: 9 mg/dL, triglyceride: 34 mg/dL). Hypercholesterol mouse tests shown that low phenolic VCO treatment result in decreasing of blood serum cholesterol level by 52.10% which was not significantly different compared to untreated mouses (decreasing of blood serum cholesterol level by 48.61%). Keywords: antihypercholesterol, phenolic compound, VCO, in vivo

ABSTRACT

Effect of Virgin Coconut Oil (VCO) Emulsion Diet onLipid Profile of White Rats (Rattus norvegicus)

Virgin Coconut Oil (VCO) is a functional food product that ispopular in the society. VCO-emulsion is a VCO emulsified with pineapplejuice. This study was conducted to find out the effect of VCO-emulsiondiet on lipid profile of mouse (Rattus norvegicus) strain wistar which wasalready treated by inducting hyperlipidemia using lard (180g/100g) andyolk (4ml/days). This research was conducted from January to December2009 at the Laboratory of Coconut and Other Palm Trees ResearchInstitute (BALITKA), the Laboratory of FMIPA Sam Ratulangi UniversityManado, the Laboratory of Experiment Animal Research Unit GadjahMada University Yogyakarta, and the Laboratory of PAU IPB Bogor.Eighteen mice were randomly divided into 3 groups. Group I was thecontrol group treated with aquadest only, group II was treated with VCO-emulsion diet, and group III was treated with pure VCO diet. The contentof total cholesterol, HDL, LDL, and triacylglicerol were measured at theend of the treatment using enzymatic method. In this study, there weresome methods used for measurements: CHOD-PAP method for level oftotal cholesterol, PVS method for level of LDL cholesterol, CHOD-PAPmethod for level of HDL cholesterol, and GPO-PAP method for measuringlevel of triacylglicerol. The study results showed that diet of 0.945ml/daysof VCO and 3.78ml/days of VCO emulsion for 7 days significantlydecreased the level of total cholesterol and the level of LDL cholesterol, and increased the level of HDL cholesterol in the blood of Wistar mouse(α=0.05). Whereas it was shown that only by diet of VCO emulsion(α=0.05) decreased the level of triacylglicerol. Thus, VCO emulsionsignificantly lowered cholesterol in the experimental mouse better thanpure VCO diet only.

Key words : Virgin coconut oil(VCO) , VCO emulsion, lipid profile

Objectives. Several experimental studies have reported antiobesity and lipid-improving effects of Citrus unshiu. However, clinical studies on its effects are lacking. This study was designed to evaluate the impact of Citrus unshiu peel pellet (CUPP) on obesity and lipid profile. Methods. For 118 patients with body mass index (BMI) > 23 who took Citrus unshiu peel pellet (CUPP) for 4 weeks in a Public Health Center, laboratory and biometric readings before and after CUPP administration were analyzed. Results. Mean age of these subjects was 53.8±10.6 years (range: 18-75 years). There were 88 (74.6%) females in the study sample (n = 118). A significant (p < 0.01) decrease in BMI from 27.47±2.24 to 27.27±2.22 was observed in all subjects after CUPP treatment and 65.3% (N = 77) of them lost 1.03±0.83 kg of weight after 4 weeks of treatment. Total cholesterol level was significantly (p < 0.01) decreased from 204.0±37.4 mg/dL to 193.5±36.5 mg/dL. Significant (p < 0.05) decreases in levels of low-density lipoprotein, cholesterol, and triglyceride were also observed. Conclusions. These results suggest that CUPP in practice could help weight control and improve total cholesterol level. Findings of this study provide clinical foundation for future large-scale trials to establish clinical benefits of CUPP.

Relevance. Virgin Coconut Oil (VCO) contains a lot of medium chain fatty acids, combined with coffee grounds (Coffea Arabica Linn.) and activated carbon (Activated carbon coconut Nucifera L) into a preparation that can moisturize and brighten the skin. Objective. The purpose of this study was to make cosmetic cleansing preparations containing three natural ingredients. Methods. This study evaluates the organoleptic body scrub preparations, homogeneity, dispensability, and pH. The ingredients were tested using in-vivo and clinical irritation along with the effectiveness of the preparation (moisture and brightness). Results. Organoleptic testing and homogeneity were confirmed to have dark black color, the distinctive smell of coffee, homogeneity, spreadability in the range of 4 cm, and the pH was at a safe pH for the skin so that irritation results also did not show edema and erythema both in-vivo divided into 3 groups (n = 3) and clinically (n = 30). In addition, this body scrub provides moisture and brightness to the skin for 2 months of use, p <0.05. Conclusions. VCO body scrub preparations, coffee grounds, and activated carbon are preparations that have the potential to be cosmetic cleansers.

Rats were fed diets containing 20% oils and 9% fatty acids and the effect of these compounds on cholesterol metabolism was studied. Unsaturated oils and fatty acids increased total cholesterol in the liver and stimulated the incorporation of C14-acetate into cholesterol both in vivo and in liver homogenates. Saturated material such as coconut oil and lauric acid had the opposite effect with respect to amount of liver cholesterol and to in vivo incorporation. The saturated material had no significant effect on synthesis in homogenates. The effect of oils in the diet was rapid, the stimulating effect of rapeseed oils being observed after the rats had been placed on the diet for as short a period as 3 days.
