Aeromonas salmonicida are specific bacteria that can cause infections and death to the cultivation of carp (Cyprinus carpio) during larval stage. Death in carp can be prevented by a vaccine, but the vaccine can only be given on the seed over the age of 3 weeks. Maternal vaccination needs to be done to improve the immune system of the larvae by means of inactivated whole cell vaccine A. salmonicida on broodstock ready to spawn. Aims to determine the effectiveness of vaccines on breeders carp to the parent antibody titer test and larvae, as well Survival Rate (SR) and the Relative Percent Survival (RPS) larvae. This research was conducted with a completely randomized design, 4 treatments A (control); B (0.3 ml/kg); C (0.4 ml/kg); D (0.5 ml/kg) and 3 repetitions. The results show that the antibody titer of 0.3 ml dose capable of providing agglutination reaction to pitting 7th (64x dilution) in broodstock, and vaccine doses 0,4ml on broodstock able to give agglutination reaction to the larvae until all 6 wells (32x dilution). A dose of 0.4 ml/kg resulted the highest SR and RPS with 96.11% and 81.25% respectively. Clinical symptoms of redness in control larvae was spread throughout the body whereas on the vaccine treatment was only in certain body parts.Keywords: A. salmonicida, vaccines, maternal immunity, larva, specific immune respon

Common carp (Cyprinus carpio) cultivation is often hampered by a disease attack, one of them is the attack of Edwarsiella tarda. Lactic acid bacteria (LAB) can be used as an alternative to prevent diseases in fish by increasing the body's resistance. This research aimed to determine the most effective isolates of LAB that increase of the resistance of carp fingerlings to the attack of E. tarda bacteria and see which isolates can produce the highest survival. The LAB isolates used were the result of isolation from the gut of carp. This study used a Completely Randomized Design (CRD) consisting of four treatments with three replications. The fish were immersed with different LAB isolates, CcB7, CcB8, and CcB15 in the same density of 108 cells / mm3. Immersion was carried out for 30 minutes with a frequency of seven days. While during the research, two immersions were carried out before the challenge test against E. tarda bacteria. The parameters observed were the number of leukocytes, hematocrit, erythrocyte, differential leukocytes, survival rate, and clinical symptoms that appeared. The results showed that all LAB isolates used in this study could increase the body resistance of carp against the attacks of E. tarda bacteria. The LAB CcB7 isolate was the most effective for enhancing the body resistance of carp fish withthe highest increase level of  leukocyte, erythrocyte, and hematocrit were 18 ± 0.057,7 ± 0,077, and 0.26± 7.31% respectively. After being challenged with E. tarda bacteria producing mild clinical symptoms, the highest increase is in monocyte and neutrophil cells was 20 and 62% respectively, the highest reduction in lymphocytes was – 9%  and the highest survival rate was 80%.

Penelitian ini bertujuan untuk mendeterminasi keberadaan Koi Herpes Virus (KHV) pada ikan koi (Cyprinus carpio koi). Sampel ikan diambil dari Kabupaten Kepulauan Sangihe. Penelitian di lakukan di Laboratorium Biologi Fakultas Matematika dan Ilmu Pengetahuan Alam Universitas Sam Ratulangi dengan menggunakan metode Polymerase Chain Reaction (PCR). Hasil pemeriksaan PCR terhadap sampel ikan koi mengindikasikan bahwa sampel ikan koi telah terinfeksi oleh virus KHV, yang ditandai dengan munculnya pita DNA pada hasil visualisasi elektroforesis agarosa. Hasil pemeriksaan pada morfologi ternyata sampel ikan yang digunakan menunjukkan gejala-gejala klinis terserang KHV seperti mata pucat, insang berwarna pucat serta produksi lendir yang berlebihan. Beberapa sampel lainnya walaupun secara morfologi belum menunjukan gejala-gejala klinis tetapi melalui pemeriksaan PCR telah terindikasikan terinfeksi KHV. Kata kunci: KHV, PCR, ikan koi   The purpose of this study was to determine the presence of Koi Herpes Virus (KHV) in Koi (Cyprinus carpio koi). Fish samples were taken from the Sangihe Archipelago Regency. Laboratory work conducted at the Laboratory of Biology Faculty of Mathematics and Natural Sciences University of Sam Ratulangi using Polymerase Chain Reaction (PCR) method. The samples indicated that the koi fish samples were infected with KHV. It is shown by the appearance of DNA bands on agarose electrophore­sis visualization. The morphological examination of the body indicated clinical symptoms of KHV infection, such as pale eyes, pale gills and excessive mucus production. Several other samples, although without clinical symptoms, have shown to be infected as indicated by PCR test. Keywords: KHV, PCR, Koi.

Sangkuriang catfish (Clarias gariepinus) is a leading commodity of Indonesian freshwater fishery. Production continues to increase in 2016 reached 22%. cultivators experience constraints on seeding and enlargement due to ectoparasites Trichodina sp. this parasite attacks the outer part of the body marked by the spots on the body. Prevention and treatment of many usses of chemicals that can adversely affect the human body and environtment, so it takes another alternative to replace these chemicals that can adversely affect the human body and environment, so it takes another alternative to replace these chemicals. Cherry Leaf contain compounds Flavonoid, Tannin, Triterpenoid, Saponin and Polifenol as antioxidants, Cherry Leaf useful as an antibacterial. The method used in this research is the experimental method and using Completely Randomized Design (RAL). Concentrations of cherry leaf extract (Muntingia calabura) given include 1%, 2%, 3%, 4% and 5% there were 5 treatments and 4 replications. The data obtained were analyzed by ANAVA method (variance analysis) followed by LSD test. The results showed that the use of cherry leaf extract (Muntingia calabura) in treating Trichodina sp parasite has a very real effect with the best value on the presentation of Leukocrit is 2% concentration that produces Leukocrit percentage of 2.68% and percentage of Hematocrit 2% with Hematocrit yield of 11.83%. The clinical symptoms are white spots in all body, pale color, decreased appetite of the mucus that comes out, often rubbing the body to the wall. The result of water quality measurement shows temperature: 24 - 27,2oC, DO: 4,1 - 6,9 and pH: 6 - 8.KEY WORDS: Muntingia calabura, Trichodiniasis, Sangkuriang Catfish, Leukocrit, Hematocrit.

Abstract. Andayani S, Dadiono MS, Elwira WT, Setyawan FH. 2020. Potency of aloe extract as immunostimulant for carp (Cyprinus carpio) against Aeromonas salmonicida. Biodiversitas 21: 860-864. The objective of the study was to be able to know the cellular and humoral non-specific immune response to Aeromonas salmonicida after aloe extract administration. This study employed Complete Randomized Design with 3 treatments and 3 replications, control treatment (K), A =2 ppm of aloe extract application in the media, B = 16 ppm of aloe extract in the media, and C = 30 ppm of aloe extract in the media. The extract was given through one-hour soaking at day 0 and day 7, and then challenge-tested to 107 cells.ml-1 of A. salmonicida into the media for one day. Results showed that administration of aloe extract could raise plasma protein levels from 443.0 to 662.0 mg.Kg-1 antibody from 1.2576 to 3.3974, and phagocytic activity from 30.61% to 45.94%. Similarly, the differential value of leucocyte (lymphocyte from 58.6% to 73.70%, neutrophils from 6.09 to 8%, and monocyte from 5.70% to 7.72%). As conclusion, aloe extract at the dose of 2 ppm could raise the cellular and humoral non-specific immune. Therefore, the use of aloe extract as immunostimulant at the dose of 2 ppm applied through immersion could also apply other methods, such as oral and injection.

One of the factors that greatly influence the successful growth of cultivated fish is feed. The aim of this study was to examine the effect of iodine addition on the growth and survival rate of carp (Cyprinus carpio) seeds. This study used a completely randomized design (CRD) method with 4 treatments and 3 replications. 10 fish per unit each, with treatment P0 (feed without adding iodine), P1 (feed + iodine at a dose of 10 mg / kg), P2 (feed + iodine at a dose of 20 mg / kg), P3 (feed + iodine at a dose of 30 mg / kg). The results showed that there was no significant effect (p> 0.05) on the growth rate, feed conversion and survival rate of goldfish. So it can be concluded that the addition of iodine in feed supplementation up to a dose of 30 mg / kg does not have a significant effect on the growth rate of length and absolute weight, feed conversion, survival rate and iodine content in the body of goldfish (Cyprinus carpio). The best recommended dose of iodine for growth is 20 mg / kg of feed, although the results were not significantly different for growth. However, it can have an effect on the iodine content in the body of goldfish.

Aeromonas salmonicida merupakan bakteri penyebab furunculosis pada ikan yang mengakibatkan kerugian ekonomi di dalam budidaya ikan air tawar. Penelitian bertujuan mengetahui gambaran darah ikan Mas (Cyprinus carpio) yang diinfeksi oleh A. salmonicida, juga dilakukan pemeriksaan histopatologi. Sebanyak empat isolat atipikal A. salmonicida telah diisolasi dari ikan di empat daerah di Indonesia yaitu Pontianak, Semarang, Yogyakarta, Jambi dan satu isolat atipikal A. salmonicida subjenis smithia dari ATCC sebagai kontrol. Sebanyak 45 ekor ikan mas berukuran 1215 cm dibagi menjadi lima kelompok. Kelompok 14 diinfeksi dengan A. salmonicida 0,1ml x 10 4 sel/ml secara intraperitoneal dari empat isolat berbeda. Kelompok ikan 5/kontrol tidak dilakukan infeksi bakteri. Isolat A. salmonicida yang berasal dari Pontianak menunjukkan jumlah leukosit total dan kadar hemoglobin meningkat jelas pada hari ke-7 sesudah infeksi, disertai peningkatan jumlah rata-rata heterofil, limfosit dan monosit pada hari ke-7 sesudah infeksi. Berdasarkan hasil pemeriksaan darah menunjukkan bahwa isolat A. salmonicida dari Pontianak merupakan isolat patogen yang menyebabkan reaksi akut jika dibandingkan dengan isolat dari daerah lain. Isolat A. salmonicida dari Yogyakarta menyebabkan perubahan patologi paling ringan. Isolat dari Jambi dan Pontianak menyebabkan lesi kulit hingga lapisan otot, epicarditis mulai hari ke-3 sesudah infeksi.

AbstractTo describle how respiratory tract infections (RTIs) that occurred in children with allergic asthma (AA) on allergen immunotherapy (AIT) during an influenza season. Data including clinical symptoms and treatment history of children (those with AA on AIT and their siblings under 14 years old), who suffered from RTIs during an influenza season (Dec 1st, 2019–Dec 31st, 2019), were collected (by face to face interview and medical records) and analyzed. Children on AIT were divided into 2 groups: stage 1 (dose increasing stage) and stage 2 (dose maintenance stage). Their siblings were enrolled as control. During the study period, 49 children with AA on AIT (33 patients in stage 1 and 16 patients in stage 2) as well as 49 children without AA ( their siblings ) were included. There were no significant differences in occurrences of RTIs among the three groups (p > 0.05). Compared with children in the other two groups, patients with RTIs in stage 2 had less duration of coughing and needed less medicine. Children on AIT with maintenance doses had fewer symptoms and recovered quickly when they were attacked by RTIs, which suggested that AIT with dose maintenance may enhance disease resistance of the body.
