The paper presents review of authors? own results as well as results of other researchers on the effects of nutritional factors in overcoming of problems in transition period, 30 days prior and 70 days post calving, and to ensure satisfactory productive and reproductive performances. The significance of proper diet balance for total and non degradable protein, sufficient amount of energy, addition of some minerals and vitamins as well as additives in order to achieve good health, high milk production and satisfactory reproductive performances are pointed out. In general, all mentioned factors have great influence in increasing economical effects in cattle breeding.

Dairy cows need sufficient physically effective fibre (peNDF) in their diet to induce chewing with the latter stimulating salivation and maintaining rumen health. Thus, monitoring of chewing activity can be a non-invasive tool to assess fibre adequacy, and thus helping in the optimization of the diet. The objective of this study was to investigate and compare chewing activities of cows during transition period and in the course of lactation. Simmental dairy cows, in four different production groups such as dry period (from 8 to 6 weeks ante-calving), calving (24 h before and after calving), early-lactation (7–60 days in milk), and mid-lactation (60–120 days in milk) were used in the study. Cows were fed partial mixed rations supplemented with different amounts of concentrates. The chewing and drinking activity were recorded using rumination-halters (RumiWatch System, Itin+Hoch GmbH, Liestal, Switzerland). Feed data analysis showed that the peNDF content of the partial mixed ration (PMR) was highest during dry period, decreased around parturition, reaching the nadir in the lactation, in all cases, however, exceeding the peNDF requirements. Chewing data analysis showed that rumination time decreased (p < 0.05) in the time around parturition (from 460 min/d during dry period to 363 min/d 24 h before calving) and increased again in early-lactation (505 min/d), reaching a maximum in mid-lactation (515 min/d). Eating time was lowest for cows during early-lactation (342 min/d) and the highest for those in mid-lactation (462 min/d). Moreover, early-lactation cows spent less time (p < 0.05) drinking (8 min/d) compared to other groups (e.g., 24 min/d the day before calving and 20 min/d postpartum). Monitoring of chewing activity might be a useful tool to assess rumen disorder risks and welfare of the cows during the transition period. It further shows promising results to be used as a tool to identify cows that are shortly before calving.

Background: Dairy goats are highly susceptible to subclinical hyperketonemia (SCHK) during the transition period. This study aimed to compare the variation in metabolic parameters and surrogate indexes of insulin resistance (sIR) between goats with SCHK and clinically healthy (HEAL) goats during the transition period. Methods: Twenty Guanzhong dairy goats were assorted to HEAL (n = 10) and SCHK (n = 10) groups according to the blood β-hydroxybutyrate (BHBA) concentrations. The blood samples were taken from the jugular vein of each goat at −3, −2, −1, 0 (partum), +1, +2, and +3 weeks relative to kidding to analyses GLU and INS. The sIR was calculated from blood metabolic parameters. Results: Compared with the HEAL goats, the insulin concentrations were significantly higher in SCHK goats during the first three weeks postpartum. The QUICKI, revised QUICKI (RQUICKI), and RQUICKIBHBA were significantly lower in goats with SCHK at 1 week postpartum, while the homeostasis model assessment-IR (HOMA-IR) was significantly higher. Conclusion: Goats with SCHK made more efforts through elevated insulin levels at early lactation than HEAL goats, thereby maintaining the normal glucose concentrations.

AbstractIn dairy cows, the period from the end of lactation through the dry period and into the transition period, requires vast physiological and immunological changes critical to mammary health. The dry period is important to the success of the next lactation and intramammary infections during the dry period will adversely alter mammary function, health and milk production for the subsequent lactation. MicroRNAs (miRNAs) are small non-coding RNAs that can post transcriptionally regulate gene expression. We sought to characterize the miRNA profile in dry secretions from the last day of lactation to 3, 10, and 21 days post dry-off. We identified 816 known and 80 novel miRNAs. We found 46 miRNAs whose expression significantly changed (q-value < 0.05) over the first three weeks of dry-off. Additionally, we examined the slopes of random regression models of log transformed normalized counts and cross analyzed the 46 significantly upregulated and downregulated miRNAs. These miRNAs were found to be associated with important components of pregnancy, lactation, as well as inflammation and disease. Detailing the miRNA profile of dry secretions through the dry-off period provides insight into the biology at work, possible means of regulation, components of resistance and/or susceptibility, and outlets for targeted therapy development.

ABSTRACTAn experiment was performed with two groups of nine British Friesian cows to compare the effect of calving in fat or thin condition on (1) the mobilization and functional activity of subcutaneous adipose tissue, (2) the mobilization of skeletal muscle, (3) the development and resolution of fatty infiltration of the liver and (4) the chemistry and haematology of blood. Sampling was performed at various times during the dry period and subsequent lactation. There were no differences between groups in the amount of adipose tissue mobilized between 4 weeks before and 26 weeks after calving. The lipogenic and lipolytic capacities of isolated adipocytes were also not different between groups at any time although major changes occurred in both over the calving period and during early lactation. Acetate oxidation to carbon dioxide was higher in adipocytes isolated from thin cows particularly after calving. More muscle fibre area was lost in the fat cows compared with the thin cows between 4 weeks before and 4 weeks after calving and the fat cows had greater infiltration of fat in the liver at 1 and 4 weeks after calving than the thin cows. The mean white-cell count was lower and the packed-cell volume was higher in the fat cows than in the thin cows at 1 week after calving. The major differences between groups in blood composition were increased concentrations of copper, non-esterified fatty acids, bilirubin and enzymes such as ornithine carbamyl transferase in the fat cows after calving. These results suggest that fat and thin cows respond differently to the metabolic demands of early lactation and that some of these differences render fat cows more susceptible to disease.

Early lactating goats show insulin resistance with respect to extramammary glucose utilization. However, much less is known about the two major factors, insulin and plasma amino acid concentration, that regulate protein metabolism in lactating goats. To examine this question, the in vivo effect of acute insulin was studied in goats during early lactation (12-31 days postpartum), midlactation (98-143 days postpartum), and the dry period (approximately 1 yr postpartum). Insulin was infused (at 0.36 or 1.79 nmol/min) under euglycemic and eukaliemic clamps. In addition, appropriate amino acid infusion was used to blunt insulin-induced hypoaminoacidemia or to create hyperaminoacidemia and maintain this condition under insulin treatment. Leucine kinetics were assessed using a primed continuous infusion of L-[1-14C]-leucine, which started 2.5 h before insulin. In all animals the insulin treatments failed to stimulate the nonoxidative leucine disposal (an estimate of whole body protein synthesis) under both euaminoacidemic and hyperaminoacidemic conditions. Thus, in goat as well as humans, infusion of insulin fails to stimulate protein synthesis even when combined with a substantially increased provision of amino acids. In contrast, insulin treatments caused a dose-dependent inhibition of the endogenous leucine appearance (an estimate of whole body protein degradation). Under euaminoacidemia the initial slope from the plot of the endogenous leucine appearance as a function of plasma insulin (an insulin sensitivity index) was steeper during early lactation than when compared with the dry period. A similar trend occurred during midlactation but not to any significant degree. These differences were abolished under hyperaminoacidemia. It was concluded that the ability of physiological insulin to inhibit protein degradation was improved during lactation, demonstrating a clear-cut dissociation between the effects of insulin on protein and glucose metabolism. This adaptation no doubt may provide a mechanism to save body protein.

The work reported in this Research Communication describes the modification in epithelial cell populations during the first and the last month of milking in Holstein Friesian cows that have undergone different management during the dry period, and we report the differential expression of CD49f+ and cytokeratin18+ cell subpopulations. Twenty six cows were randomly divided into 2 balanced groups that were housed at stocking density of either 11 m2 (CTR) or 5 m2 from 21 ± 3 d before the expected calving until calving. Cells collected from milk samples taken in early lactation and late lactation were directly analysed for CD45, CD49f, cytokeratin 14, cytokeratin 18 and cell viability. We observed a differential expression with a significant reduction in CD49f+ (P < 0·01) and cytokeratin 18+ (P < 0·05) cells in early lactation. Differences were still evident in late lactation but were not significant. These observations suggest that mammary epithelial cell immunophenotypes could be associated with different animal management in the dry period and we hypothesise they may have a role as biomarkers for mammary gland function in dairy cows.

В условиях научно-производственного опыта исследована эффективность введения кормовой добавки «Абиотоник» в рацион высокопродуктивных голштинизированных симментальских коров (в транзитный период и на раздое). «Абиотоник» представляет собой усовершенствованный аналог известного импортного препарата «Чиктоник», отличаясь от иностранного аналога многократно увеличенной концентрацией пептона и наличием высокодефицитных микроэлементов (селена и элементоорганического соединения йода). Наши исследования показали, что скармливание препарата «Абиотоник» в дозе 150 мл/гол. способствовало росту продуктивности коров на 5,84%, улучшило физико-химические свойства молока и повысило его пищевую ценность. Применение кормовой добавки способствовало также профилактике желудочно-кишечных заболеваний у коров, повышению их резистентности, о чём свидетельствовали показатели крови и бактериологического исследования кала. Заболеваемость коров в опытной группе не наблюдалась. Использование препарата «Абиотоник» коровам в заключительной стадии сухостойного периода и в начале лактации привело к сокращению времени отделения плаценты у коров после отёла на 6,54%, или на 0,4 часа, повышению живой массы новорождённого молодняка на 1,6% и сокращению сервис-периода у опытных коров на 8 дней по сравнению с контрольными. Использование кормовой добавки «Абиотоник» способствовало получению дополнительного дохода от реализации молока в размере 1152 руб./гол. Полученные экспериментальные данные позволяют рекомендовать использование отечественной импортозамещающей кормовой добавки «Абиотоник» в рационах коров транзитного периода и на раздое в дозе 150 мл/гол. ежедневно в последней фазе сухостоя и через день в течение 40 суток после отёла. Under the conditions of scientific and farm scale trial the efficiency of introduction of the feed additive "Abiotonic" into the diet of highly productive Holsteinized Simmental cows (during the transition period and for days in milk) was studied. "Abiotonic" is an improved analogue of the known imported preparation "Chiktonik" differing from the foreign analogue by its manifoldly increased peptone concentration and the presence of highly deficient microelements (selenium and organo-element compounds of iodine). Our researches have shown that feeding the preparation "Abiotonic" at a dose of 150 ml / head contributed to an increase in cow productivity by 5.84%, improved the physicochemical properties of milk and increased its nutritional value. The use of a feed additive also contributed to the prevention of gastrointestinal diseases in cows, an increase in their resistance as evidenced by haematological parameters and bacteriological feces analysis. The incidence of cows in the experimental group was not observed. The use of the preparation "Abiotonic" for cows in the final stage of the dry period and at the beginning of lactation reduced the time for separation of the placenta in cows after calving by 6.54% or 0.4 hours, increased the live weight of newborn young stock by 1.6% and reduced the service period of experienced cows by 8 days compared with the control ones. The use of the "Abiotonic" feed additive contributed to the receipt of additional income from the sale of milk in the amount of 1152 rubles / head. The obtained experimental data allow us to recommend the use of domestic import-substituting feed additive "Abiotonic" in the diets of cows in the transition period and for days in milk at a dose of 150 ml / head daily in the last phase of dry period and every other day for 40 days after calving.

Introduction: Hormonal and metabolic changes, as well as energy imbalance, can affect health, production and reproductive performance of dairy cows. In the present study, we evaluated phagocytosis and respiratory burst neutrophil activity during the transition period and early lactation and compared it with biochemical and hematological parameters in dairy cows. Methodology: Simmental cows (n = 21) were enrolled in the study. Whole blood samples were collected weekly from 3 weeks pre- calving until 6 weeks post calving. Basic metabolic and blood parameters were assessed by routine laboratory analyses, while neutrophil functions were analyzed by commercial test kits. Results: Optimal neutrophil response was observed pre and post calving. The highest value was recorded in the 6th week after calving (89.54 ± 7.61%) and being significantly higher (p < 0.01) as compared to values recorded at two and one week before and one week after calving. The percentage of activated neutrophils was high during the entire study period: from 70.80 ± 5.22% at the beginning of the study to 89.54 ± 7.61% at the end of the study. During the study period, production of Reactive Oxidative Species by neutrophils was positively correlated with β-hydroxybutyrat and non-esterified fatty acids values (0.454** and 0.423**, respectively) and calcium levels (0.164* and 0.212**, respectively). Conclusions: The most prominent changes in all parameters had no influence on phagocytic and respiratory burst activity of neutrophils. Neutrophil function is preserved at the optimal level during the transition period and early lactation in Simmental cows.

AbstractA partially balanced changeover design experiment was undertaken to evaluate the effects of level of fish oil inclusion in the diet on milk fat concentration and composition when offered to 50 lactating dairy cows in early lactation, which were offered either 5 or 10 kg concentrates/cow/day. Concentrates were formulated to contain similar concentrations of crude protein (CP), effective rumen degradable protein (ERDP) digestible undegradable protein and starch and to provide 0 (T0), 150 (T150), 300 (T300) or 450 (T450) g fish oil/cow/day or 300 g (T300B) fish oil/cow/day from a commercial fish oil premix. All animals were offered 5 kg treatment concentrate in two equal feeds through the in-parlour feeder at each milking. Additionally, cows offered the higher level of concentrates received a further 5 kg of T0 concentrate in two equal feeds per day through out-of-parlour feeders. Increasing the level of fish oil increased milk yield (P<0.01) and decreased the concentrations of fat (P<0.001) and protein (P<0.001). Increasing the level of fish oil decreased the concentrations of C6:0, C8:0, C10:0, C16:0, C14:0 and C18:0 and increased the concentrations of C18:1t, C20:0, C20:1, C20:4w6, C18:2c, C23:0, C20:5w3 and C18:2t. Relative to T300, T300B significantly decreased the concentrations of C20:4w6, but did not alter any of the other fatty acids. The transfer from feed to milk of EPA and DHA averaged 0.61 and 0.19 respectively across the three levels of fish oil supplementation. For T300 the efficiency of transfer of EPA was higher while the efficiency of transfer of DHA was lower to that for T300B. It is concluded that increasing fish oil supplementation decreased milk fat concentration by up to 15 g/kg. Also feeding fish oil is an efficient method of increasing EPA in the human diet through transfer into milk.
