Even today, the leading cause of visual impairment is age related cataract.  Among the risk factors for cataract, tobacco is one of the modifiable risk factors. Association between smoking and increased oxidative stress among cataract patients has been shown by some studies. But studies related to smokeless tobacco use are not done. The aim of the study was to estimate oxidative stress by markers like enzymatic antioxidant Superoxide Dismutase (SOD) and lipid peroxiation product Malondialdehyde (MDA) in tobacco user age related cataract (ARC) subjects and compare them with tobacco nonuser age related cataract (ARC) subjects. This cross sectional study was performed in 120 subjects divided into 2 groups – 60 tobacco nonuser age related cataract subjects and 60 tobacco user age related cataract subjects. 60 tobacco user cataract subjects divided into 5 groups - tobacco smokers, tobacco chewers, tobacco mishri users, dual tobacco chewers with mishri users and dual smokers with smokeless tobacco users. Oxidative stress was assessed by estimation of erythrocytic SOD and serum MDA. The study showed that in tobacco user group age of cataract patients was significantly less than tobacco nonuser cataract patients (P<0.05).  Serum MDA levels were significantly high and erythrocytic SOD levels were significantly low in tobacco user cataract patients than tobacco nonuser cataract patients (P<0.001).  Nuclear cataract was significantly more in tobacco user group (P<0.05).  On comparison of subtypes of cataract in tobacco user subgroup, no significant difference was found. These results suggested that age related Cataract occurs at an earlier age in tobacco users due to increased oxidative stress. Also nuclear cataract was significantly associated with tobacco use.

Background: In Pakistan age related vision disturbances are mainly due to cataract. Various studies have reported relationship of ocular lesion with senile changes and diabetes mellitus resulting in reduced quality of life due to vision. Oxidative stress is an important factor in the process of cataractogenesis. The pathogenesis of the cataract may involve decreased activity of antioxidant scavenging system which includes non-enzymatic natural antioxidants as biomolecules such as carotenoids and vitamins. So, it is planned to investigate the level of serum antioxidant vitamins in diabetic cataract patients and in non-diabetic cataract patients.Methods: The study was conducted at Biochemistry department, Al-Tibri Medical College Karachi from October 2016 to October 2017. Ninety pre diagnosed cataract patients were selected from Al-Ibrahim Eye Hospital Karachi 40 normal control subjects were selected from the same population with same socioeconomic group. The demographic data was analyzed. The random blood sugar, antioxidant vitamins (C, A and E) and malondialdehyde were analyzed in the blood sample of control and cataract patients. The data was analyzed by SPSS version 20.Results: There was no significant difference in the level of vitamin C, A, E and MDA between diabetic and non-diabetic cataract patients, but the blood levels of vitamins of control are higher as compared to the cataract patients. The level of MDA is significantly high in cataract patients as compared to control. Antioxidant vitamin E was negatively correlated with serum malondialdehyde in cataract patients.Conclusions: It is concluded that in diabetic and non-diabetic cataract low level of serum antioxidant vitamins may be a contributory factor for cataractogenesis.

Oxidative stress is a major factor in the pathogenesis of age-related macular degeneration (AMD). Iron may catalyze the Fenton reaction resulting in overproduction of reactive oxygen species. Transferrin receptor 2 plays a critical role in iron homeostasis and variability in its gene may influence oxidative stress and AMD occurrence. To verify this hypothesis we assessed the association between polymorphisms of theTFR2gene and AMD. A total of 493 AMD patients and 171 matched controls were genotyped for the two polymorphisms of theTFR2gene: c.1892C>T (rs2075674) and c.−258+123T>C (rs4434553). We also assessed the modulation of some AMD risk factors by these polymorphisms. The CC and TT genotypes of the c.1892C>T were associated with AMD occurrence but the latter only in obese patients. The other polymorphism was not associated with AMD occurrence, but the CC genotype was correlated with an increasing AMD frequency in subjects withBMI<26. The TT genotype and the T allele of this polymorphism decreased AMD occurrence in subjects above 72 years, whereas the TC genotype and the C allele increased occurrence of AMD in this group. The c.1892C>T and c.−258+123T>C polymorphisms of theTRF2gene may be associated with AMD occurrence, either directly or by modulation of risk factors.

Objective. Neonates have a high risk of oxidative stress during anesthetic procedures. The predictive role of oxidative stress biomarkers on the occurrence of brain injury in the perioperative period has not been reported before. Methods. A prospective cohort study of patients requiring major surgery in the neonatal period was conducted. Biomarker levels of nonprotein-bound iron (NPBI) in plasma and F2-isoprostane in plasma and urine before and after surgical intervention were determined. Brain injury was assessed using postoperative MRI. Results. In total, 61 neonates were included, median gestational age at 39 weeks (range 31–42) and weight at 3000 grams (1400–4400). Mild to moderate brain lesions were found in 66%. Logistic regression analysis showed a significant difference between plasma NPBI in patients with nonparenchymal injury versus no brain injury: 1.34 umol/L was identified as correlation threshold for nonparenchymal injury (sensitivity 67%, specificity 91%). In the multivariable analysis, correcting for GA, no other significant relation was found with the oxidative stress biomarkers and risk factors. Conclusion. Oxidative stress seems to occur during anaesthesia in this cohort of neonates. Plasma nonprotein-bound iron showed to be associated with nonparenchymal injury after surgery, with values of 1.34 umol/L or higher. Risk factors should be elucidated in a more homogeneous patient group.

Summary Individuals with hypertension and diabetes mellitus are at high risk of cerebrovascular and cardiovascular morbidity and mortality. Recent advances in the multifactorial pathophysiology of atherogenesis provide important information about the complex interrelations between traditional risk factors, inflammation and oxidative stress in mediating all stages of atherosclerosis. The objective of the study was to determine if some inflammatory and oxidative stress markers in patients with arterial hypertension and diabetes mellitus differ from those in healthy age-matched controls. Our results revealed a significant difference in blood pro/antioxidant activities in hypertensive diabetics and the controls. The investigation of inflammatory and oxidative stress markers along with traditional risk factors proves useful in complex assessment of vascular risk and primary prophylaxis of cerebrovascular and cardiovascular events.

Objective: The aim of this work is the study of risk factors for breast cancer in a population of women in the north-western regions of Algeria (Saida and El-Bayadh) Methodology: A case-control study was carried out on 40 women including 20 cancerous women and 20 control women, in order to determine the dietary patterns and general characteristics of the population studied and to compare the hematological and biochemical parameters, and markers of oxidative/antioxidant status between control women and breast cancer women. Results: The results obtained show that there is no significant difference in the average age however the body mass index was significantly increased in cancer women, revealing overweight (p <0.05), compared to control women. The marital status of the target population is predominantly a married woman status and the level of education shows a relatively low level in breast cancer cases compared to women controls. The illiteracy rate is exceptionally high in cancer women compared to controls (35% versus 20%). In addition, the levels of glucose, creatinine, HDL and ALAT are similar between the two groups of women. The contents of urea, cholesterol, triglyceride, LDL, ASAT, malondialdehyde and uric acid are significantly increased in women with breast cancer compared to women controls. But the vitamin C content is significantly reduced in cancer women compared to control women. Conclusion: We conclude from this study that there is an association between breast cancer and lipid alteration, oxidative stress, age, increase in BMI, socio-demographic data, professional situation, age, means of menstruation and menopause, and ultimately the lifestyle. All these factors are risk factors for breast cancer in the region of Saida and El-Bayadh. Keywords: Breast Cancer, Risk Factors, Dietary Patterns, Oxidative Stress.

Endothelial senescence is believed to constitute the initial pathogenesis of the atherosclerotic cardiovascular disease (ASCVD). MicroRNA-335-5p (miR-335-5p) expression is significantly upregulated in oxidative stress-induced endothelial cells (ECs). Sirtuin7 (SIRT7) is considered to prevent EC senescence, yet data on its response to ASCVD risk factors are limited. This study analyzed the elevated levels of miR-335-5p and the decreased levels of SIRT7 in human umbilical vein endothelial cells (HUVECs) , and found that high glucose, tumour necrosis factor-α (TNF-α), and H2O2 are the three contributing factors that induced cellular senescence. The current study also assessed premature endothelial senescence and decreased proliferation, adhesion, migration, and nitric oxide secretion in HUVECs with these risk factors together with SIRT7-siRNA transfection. It found that the miR-335-5p inhibitor attenuated the downregulation of SIRT7 expression induced by oxidative stress in HUVECs, and SIRT7 overexpression exerts a rescue effect against miR-335-5p induced endothelial dysfunction. Furthermore, the direct binding of miR-335-5p to SIRT7 was observed in HEK-293T. Therefore, it can be inferred that miR-335-5p downregulates the expression of SIRT7 in human cells. Current findings may provide deeper insights into the underlying mechanisms of endothelial senescence and potential therapeutic targets of ASCVD as well as other age-related diseases.

Abstract Background Phacoemulsification using phaco-chop technique has many challenging features in cataract patients with highly liquefied vitreous. This study aimed to compare the intraoperative parameters and safety between prechop technique and traditional phaco-chop in phacoemulsification for these patients. Methods A total of 54 eyes of 54 patients with high myopia-related or post-vitrectomy cataract that underwent phacoemulsification combined with intraocular lens implantation were included in this retrospective study. Of them, 25 eyes that received manual prechop were included in the prechop group, and 29 eyes with best match of age, axial length and nuclear opalescence (NO) that received standardized phaco-chop were included as the control group. The intraoperative complications and surgery parameters were compared between groups. Results No surgical complications were observed in the prechop group, while 2 eyes of posterior capsular rupture and 1 eye with a broken ciliary zonule (10.3%) were found in the control group. There was no significant difference in phaco time, average energy, and cumulative dissipated energy (CDE) between groups (all P > 0.05), but for hard nuclear cataract with NO grading ≥ 5, prechop group had less phaco time (P = 0.008) and CDE (P = 0.029). The correlations between phaco time vs. NO (r = 0.762 vs. 0.581, both P < 0.005) and CDE vs. NO (r = 0.717 vs. 0.668, both P < 0.001) seemed to be weaker in the prechop group as compared to the control group. Conclusions The prechop technique which seemed to have less intraoperative complications, reduced phaco time and CDE compared to standardized phaco-chop might be a good alternative for cataract patients with highly liquefied or vitrectomized vitreous, especially those with hard nuclear cataract.
