The aim of present study was to formulate and evaluate herbal gel containing Solanum nigrum extract for prevention of wound healing. Topical gel formulation was designed by using Solanum nigrum extract as API and HPMC E5 as a gelling agent. The herbaceous plant Solanum nigrum (kamuni) reported for healing action and newer research studies and methodologies are being carried to find active chemical constituents which not only promise fast healing but also will reduce the complication and cost. Extracts of this medicinal plant are useful in the treatment of several health problems such as bacterial infections, ulcer, cancer, tuberculosis, arthritis and inflammatory. The plant Solanum nigrum (kamuni) leaves is the best traditional medicine for the study of wound healing activity. The concept of wound healing is changing from day to day. Ayurveda is the richest source of plant drugs for management of wounds and Solanum nigrum L. is one such. The plant is used as haemostatic and wound healing agent from ethno pharmacological point of view. The prepared gel was characterized for their physicochemical parameters i.e., preliminary phytochemical analysis, quantitative analysis, appearance, spreadability, pH, viscosity, in-vitro diffusion study and stability study.

Mometasone furoate (MF) is a glucocorticoid prodrug that faces the problem of poor aqueous solubility. Nanoemulsion-based topical gel of MF was formulated to enhance its solubility and potential of treating skin conditions. The selection of oil, surfactant and co-surfactant was done based on their solubility with the drug. The nanoemulsion was prepared using rose oil as the oil phase. Tween 80 and Transcutol P were used as surfactant and co-surfactant and they were blended in different ratios (1:0, 1:1, 2:1 and 3:1 w/w). The pseudo ternary diagrams were developed using these excipients and formulations exhibiting considerable nanoemulsion region were selected. The formulations were optimized by using Design Expert software for the globule size and cumulative percent release. The nanoemulsion formulations were characterized for in vitro release and stability study. The optimized nanoemulsions consisting of 2 % w/w oil, 30 % w/w Smix (Surfactant: Co-surfactant) and 67.9 % w/w water were consolidated into Carbopol 940 gelling agent to prepare three nanoemulsion-based gel formulations or nanoemulgels (NEG1-NEG3). Nanoemulgels were evaluated for their stability and ex vivo permeation of MF. The outcomes suggested that skin permeation of MF from all the nanoemulgel formulations was significantly enhanced as compared to the marketed mometasone furoate topical formulation.

Herbal medicines widely improved for primary health care because of better cultural acceptability and better compatibility with human body and lesser side effects. The aim of present study was to formulate and evaluate of herbal gel containing Allium cepa extract. Topical gel formulation was designed by using Allium cepa extract and carbopol 934 as a gelling agent and different excipients. The herbaceous plant Allium cepa (onion). It has great health significance and is consumed for nutritional and health benefits for last centuries. It contains flavonoids compound. Plant extract used for prevention of hair loss. Raw onion eaten in salad form this is play very important role in health benefits. Gel was prepared by incorporating extract of Allium cepa in gel at a particular step in order to prepare non greasy formulation. Gel Formulation F3 Batch was optimized in the concentration 1.5% of Carbopol 934. Different gel formulations were evaluated for their physical appearance; pH, Spreadability, Homogeneity in-vitro diffusion ,Viscosity and Drug Excipients comparability study are carried out.

Objective: Most of the drugs are relevant to BSC class II and class IV having solubility problems. Cocrystallization of drug with conformer is an immense approach used to explore the physicochemical properties of drug. The objective of the present work was to design formulate and evaluate the drug cocrystals of poorly soluble drug apremilast (APR) with saccharin. Methods: Cocrystals of APR were prepared using the solvent evaporation technique. The saturated solubility study and in vitro dissolution study of cocrystals were carried out. The prepared cocrystals were characterized by differential scanning calorimetry (DSC), X-ray diffraction (XRD), and Fourier-transform infrared (FTIR) spectroscopy. The topical gel of APR cocrystals was formulated optimized and evaluated using three-level factorial design. Results: The cocrystals of APR were prepared in 1:1 molar ratio with saccharin. APR cocrystals showed the improvement in solubility and dissolution as compared to pure APR. The formation of cocrystals was confirmed from change in endothermic peak of DSC and from shifting of FTIR spectra of cocrystals. Crystallinity of cocrystal was confirmed from XRD pattern and noteworthy change in 2θ values of the intense peak. The topical gel of APR cocrystals was formulated and optimized using three-level factorial design using Carbapol-940 and hydroxypropyl methylcellulose (HPMC) as a gelling agent. Conclusion: The cocrystals with altered physicochemical properties of APR were prepared with saccharin and formulated as a topical gel to overcome the problems related to oral administration.

Medicinal plants possess therapeutic potential and are used to treat various diseases around the world. Eclipta prostrate (L.) is a medicinal herb that has extensive application in the native medicinal system. In any therapeutic activity chemical constituents play an important role. Eclipta prostrata has been investigated in this study for its antioxidant, antimicrobial and antidiabetic activity in vitro. The well-known research protocol available in the literature established qualitative analysis of the different phytochemical constituents and quantitative analysis of total phenol and flavonoids. The hydroalcoholic extracts of the leaves and seeds of Eclipta prostrata exhibited significant and dose-dependent antioxidant activity including ability to donate electron. To analyze the antimicrobial activity, Leaves hydroalcoholic extracts and Eclipta prostrate seeds were tested against two selected strains using a well-diffusion method and showing significant inhibitory action against all the strain tested. In addition, the dose-dependent α-amylase inhibitory activity with an IC50 value of acarbose, leaves, and seed extract was found to be 364.89μg/ml and 438.43μg/ml, respectively, indicating that Eclipta prostrate is a promising source as an herbal medicine. Silver nanoparticles have attracted great interests widely in medicine due to its great characteristics of antibacterial activity. In this research, the antibacterial activity and biocompatibility of a topical gel synthesized from polyvinyl alcohol, chitosan, and silver nanoparticles were studied. Hydrogels with different concentrations of silver nanoparticles (15 ppm, 30 ppm, and 60 ppm) were evaluated to compare their antibacterial activity, nanoparticles’ sizes, and in vivo behaviors. The resulted silver nanoparticles in the hydrogel were characterized by TEM showing the nanoparticles’ sizes less than 22 nm. The in vitro results prove that the antibacterial effects of all of the samples are satisfied. However, the in vivo results demonstrate the significant difference among different hydrogels in wound healing, where hydrogel with 30 ppm shows the best healing rate.

Topical delivery of clobetasole propionate (CP) offers several formulation related problems due to poor water solubility and photo degradation property. In the present investigation, topical gel of CP was formulated using Acrylamide/ Sodium Acryloyldimethyl taurate copolymer (SEPINEO™ P 600) as a gelling agent and evaluated with respect to different physicochemical parameters such as pH, viscosity, bio-adhesivity, spreadability, in vitro drug release and photo stability. Permeation of CP gel was studied using freshly excised pig ear skin for 24 h. The cumulative permeation of drug through excised rat skin was 3.0 ± 1.2 mg cm-2 with the corresponding flux value of 0.24 ± 0.09 mg cm-2 h -1 . The in vitro release studies showed 101.43±1.12 % drug release over 10 h. The selected formulation was found to be effective with respect to percent drug content, permeation characteristics, pH, viscosity, and photostability. Therefore, CP gel could be very promising alternative for the topical drug delivery.

Butanol and hexane leaves extracts ofParkinsonia aculeataL. (Fabaceae) were assessed for its antioxidant potential byin vitromethods. Phytochemical analysis and antioxidant activity of plant extracts were studied using differentin vitroassays. UPLC analysis of extracts was carried out for the identification of chemical constituents. The total phenolic contents of the butanol and hexane leaf extract were 42 mgGAE/g and 34 mgGAE/g whereas flavonoid contents of these extracts were found to be 0.044 mgRE/g and 0.005 mgRE/g, respectively. Among both extracts, butanol extract shows maximum inhibition (%) of 93.88%, 80.02%, 52.06%, 94.68%, and 69.37% in DPPH, non-site-specific and site-specific, FTC, and TBA assays and absorbance of 0.852 and 0.522 in reducing power and CUPRAC assay at the highest concentration tested. The FRAP and TAC values of butanol extract were found to be 678 μM Fe(II)/g and 36 mgAAE/100 mg. UPLC analysis of extracts revealed the presence of various polyphenols. The tested plant extracts were found to possess potent antioxidant and free radical scavenging activity which may be due to the presence of flavonoids and polyphenols.

The therapeutic failures and the increasingly high costs of treating resistant bacterial infections calls for alternative means of care. The present study was initiated to evaluate the antifungal properties of the Harungana madagascariensis methanol barks extract and potent interactions with some usual drugs. The extract was prepared by maceration of the dry stem bark powder in methanol. Phytochemical analysis was carried out by colorimetric assays. Antifungal activity as well as interactions between extract, Nystatin or Fluconazole was evaluated by broth microdilution method. The therapeutic efficacies of H. madagascariensis extract and Fluconazole – extract mixture as compared to Fluconazole extract was studied in experimental models of oral and vulvovaginal candidiasis in rats by oral route. Results showed that methanol extract of H. madagascariensis stem barks had antifungal activity ranging from 128 to 1024 µg/ml. This extract also had a fungicidal activity on all of the tested yeasts. Moreover, the extract presented in vitro synergetic and additive interactions with Nystatin and Fluconazole. In both oral and vaginal infection model, all the treatment significantly reduced (P less than 0.05) the number of colony formant unit (UFC) of C. albicans compared with untreated control. Moreover, significant decrease in the percentage of animals showing positive cultures was observed in rats treated with Fluconazole-extract mixture. In both therapeutic efficacy studies, the histological findings confirmed the microbiological results. The results of this study constitute a base for the usage of H. madagascariensis stem barks in association with Fluconazole to overcome yeast infection.

Rhizomes of Reynoutria japonica Houtt. are a traditional Chinese medicinal herb (Polygoni cuspidati rhizoma, hu zhang) used for treatment of numerous diseases including wound healing support. The aim of this study was to provide evidence for the value of this herbal drug’s traditional use as a gingival healing treatment as well as to obtain the most active extract. In vitro studies were performed using primary human gingival fibroblasts (HGFs) with determination of viability (MTT assay), cell proliferation (the confocal laser scanning microscope (CLSM) was used to visualize histone 3 expression), cell migration (wound healing assay), and evaluation of the expression of collagen type III (immunocytochemical staining) after incubation with extracts from R. japonica rhizomes (25% or 40% ethanol or 60% acetone). In addition to these extracts, commercial dental rinse (containing chlorhexidine digluconate 0.2%) was tested as the gold standard of choice for gum healing in dental practice. The studied extracts were qualitatively and quantitatively characterized using the validated HPLC/DAD/ESI-HR-QTOF-MS method. Total phenols and tannins content were determined using the Folin–Ciocalteu assay. Low concentration of all extracts after 24 h incubation caused significant increase in HGF viability. This effect was most pronounced at a concentration of 50 µg/mL, which was selected for further experiments. All extracts (at 50 µg/mL) stimulated HGF to proliferate, migrate, and increase collagen III synthesis, but with different strength. The highest stimulated proliferation and migration activity was observed after incubation with 25% EtOH, which according to phytochemical analysis may be related to the highest content of resveratrol and an appropriate composition of procyanidins. The 25% EtOH extract from R. japonica rhizomes appears to be a promising gingival wound healing agent worthy of animal and clinical trials.

AbstractSerious infection caused by drug-resistant gram-negative bacteria and their secreted toxins (e.g., lipopolysaccharide) is a serious threat to human health. Thus, treatment strategies that efficiently kill bacteria and reducing the impact of their toxins simultaneously are urgently required. Herein, a novel antibacterial platform composed of a mesoporous copper silicate microsphere (CSO) core and a platelet membrane (PM) shell was prepared (CSO@PM). CSO@PM specifically targets bacteria owing to formyl peptide receptors on the PM and, combined with photothermal therapy (PTT), exhibits highly effective bacter icidal activity. Importantly, CSO@PM can adsorb lipopolysaccharide secreted by gram-negative bacteria, resulting in inflammation reduction. Thus, CSO@PM stimulates re-epithelialization and granulation-tissue formation, promoting wound healing. Moreover, this antibacterial platform exhibits no obvious toxicity at all the test concentrations in vitro and in vivo. Thus, CSO@PM exhibits a robust antibacterial effect and a strong toxin-adsorption capacity, facilitating the clinical treatment of many bacterial infections and the development of next-generation antibacterial nanoagents.
