Effects of 1% and 3% Mobe Leaf Extract Gel on Socket Wound Healing after Tooth Extraction

Tooth extraction is a process of removing teeth from the alveolar bone. In wound healing, fibroblast are very important cells. The main purpose of this study was to determine the effect of mobe leaf 1% and 3% extract gel (Artocarpus lakoocha) on fibroblast proliferation in post extraction tooth socket wound healing. This research used 16 samples of wistar rats, divided into 4 groups, a positive control group, a negative control group and a 1% and 3% mobe leaf extract gel group. The left mandibular incisors were extracted, then 1% and 3% gels of mobe leaf extract were applied on day 1 to day 7. Data analysis was calculated using the Kruskal-Wallis test on clinical data and one way ANOVA test for microscopic. The result of the socket wound healing activity test for a good concentration of mobe leaf extract gel was 3%. This research shows significant resultith p-value of 0.018 (< 0.05) on the closure of the socket wound clinically which means the closure of the wound accelerates because of the mobe leaf 3% extract gel treatment. The distance of fibroblast on microscopically shows significant resultith a p-value of 0.002 (< 0.05), which means that there was an enlargement of the distance fibroblast at the socket wound closure with application of mobe leaf 3% extract gel. From the results of the study it can be concluded that mobe leaf 3% extract gel has the best ability to show acceleration the closure of the socket wound either clinically or microscopically.

Pendahuluan: Pencabutan gigi di dunia kedokteran gigi adalah tindakan yang sering dilakukan. Pasca terjadinya pencabutan gigi, maka tulang pada soket akan rusak. Penyembuhan tulang ditandai dengan terbentuknya sel-sel osteoblas baru. Daun binahong merupakan daun yang mengandung flavonoid, alkaloid, terpenoid, asam askorbat, saponin, tanin dan asam oleanolik. Kandungan dari daun binahong dapat meningkatkan jumlah osteoblas pasca pencabutan gigi. Tujuan dari penelitian adalah menganalisis pengaruh pemberian gel ekstrak daun binahong (Anredera cordifolia) konsentrasi 5% terhadap peningkatan osteoblas pada proses penyembuhan luka pasca pencabutan gigi. Metode: Jenis penelitian ini merupakan penelitian eksperimental laboratorium murni dengan rancangan post test only control group design. Subyek penelitian berupa 45 ekor tikus dibagi menjadi 3 kelompok, yaitu kelompok gel ekstrak daun binahong, kontrol negatif dan kontrol positif. Gigi insisivus sentral kiri rahang bawah dilakukan pencabutan gigi kemudian diaplikasikan gel ekstrak daun binahong 5%, CMC-Na 1% dan iod gliserin. Tikus diterminasi pada hari ke 3,5,7,14 dan 21 pasca dilakukan pencabutan gigi untuk selanjutnya dibuat preparat histologis dengan pengecatan hematoksilin eosin. Jumlah sel osteoblas dihitung dengan optilab dan software image raster. Hasil: Hasil uji parametrik one way anova menunjukan terdapat perbedaan jumlah osteoblas yang bermakna antar kelompok (p<0,05) pada masing-masing hari. Hasil LSD (Least Significance Different) menunjukkan terdapat perbedaan yang bermakna (p<0,05) pada hari ke 5,7, 14 dan 21 antara kelompok perlakuan dengan kontrol negatif dan terdapat perbedaan yang bermakna pada hari ke 7 dan 14 antara kelompok perlakuan dan kontrol positif. Simpulan: Gel ekstrak daun binahong konsentrasi 5% berpengaruh dalam meningkatkan jumlah sel osteoblas pasca dilakukan pencabutan gigi.Kata kunci: Pencabutan gigi, gel ekstrak daun binahong, osteoblas. ABSTRACTIntroduction: Tooth extraction is the most common treatment in dentistry. After tooth extraction, the bone in the socket will be damaged. The formation of new osteoblast characterises bone healing. Binahong leaves contain flavonoids, alkaloids, terpenoids, ascorbic acid, saponins, tannins and oleanolic acids. This research was aimed to investigate the effectivity of binahong (Anredera cordifolia) leaf extract gel with a concentration of 5% in increasing the number of osteoblasts at the tooth extraction wound healing process. Methods: This research was an experimental laboratory with a post-test only control group design. Forty-five rats were divided into 3 groups; binahong leaf gel group, negative control group, and positive control group. Mandibular left central incisive was extracted, then binahong leaf gel, 1% CMC-Na, and glycerin were applied on the wound socket in different groups. All rats then terminated at day 3, 5, 7, 14, and 21 post-extraction and processed for the histological examination then coloured by haematoxylin-eosin. The number of osteoblasts was calculated by Optilab and raster image software. Results: The ANOVA test result showed significant differences in the number of osteoblasts among groups (p<0.05) on each day. The LSD (Least Significance Different) test result showed significant differences (p<0.05) at day 5, 7, 14, and 21 post-extraction, between binahong leaf gel group compared to the negative control group, and also at day 7 and 14 post-extraction between binahong leaf gel group compared to the positive control group. Conclusion: Binahong leaf extract gel with a concentration of 5% is effective in increasing the number of osteoblast in the wound healing process after tooth extraction.Keywords: Tooth extraction, Binahong leaf extract gel, osteoblast.

Wound refers to partly damage or loss of body tissue. One way that has been used by the people in helping the process of wound healing is the use of medicinal plants, such as gotu kola (Centella asiatica). Gotu kola (Centella asiatica)  is kind of plant that found almost in all territories of Indonesia which functions as a cell revitalizing substance which quickens wound healing those are burnt, skin ulcer and to prevent keloid. This study is aimed to know the effect of giving gotu kola leaf extract (Centella Asiatica) against to the healing of the wound incision on male white rats (Rattus Norvegicus) wistar strain. This study is a pure experimental research using the randomized post test only control group design with the subjects are 30 male white rats (Rattus norvegicus) wistar strain that were divided into 3 groups, namely K1, K2, and K3. The incision wound were made on the back of all white rat in each group with a length of ± 2 cm and a depth of ± 2 mm. The wound in the negative control group (K1) was not given any substance; in the positive control group (K2) was given povidone iodine 10%; and in the treatment group (K3) was given gotu kola leaf extract at a dose of 37,5 mg/150 grBB. The study was done for 10 days. Furthermore, wound observation and monitoring is held. Then the results were processed by using One Way ANOVA. The result study of wound incision healing on male white rats (Rattus norvegicus) wistar strain shows that there is significant difference among the negative control group (K1), the positive control group (K2), and the treatment group (K3) on One Way ANOVA test which significant p-value are 0,027 (p < 0.05). The result of Post-Hoc LSD test also shows a significant differences among the K1 and K3 groups, and K2 and K3 groups which significant p-values are 0,011 and 0,038 (p < 0,05). From these results, it can be concluded that gotu kola leaf extract (Centella asiatica) effective on quickens wound healing of the wound incision on male white rats (Rattus norvegicus) wistar strain.

Background: The process of wound healing is influenced by various factors such as age, hormones, and wound care. Wound care is done to accelerate wound healing which can be done by various methods, one of them is traditional care. Traditional wound care can use medicinal plants. Rhodomyrtus tomentosa is a medicinal plant that has an antioxidant, anti-inflammatory, antitumor and antibacterial content. Thus this study aims to evaluate the effectiveness of the antiseptic solution of the Rodhomyrtus tomentosa leaf extract on wound healing in male Wistar rats. Method: this research is pure experimental research with post test only control group design. Thirty male white rats were divided into five groups, namely negative control, positive control, Rhodomyrtus tomentosa leaf extract 15%, 30%, and 60%. Rhodomyrtus tomentosa leaf extraction was carried out by maceration method with 70% ethano solvent. The extraction results are divided into 3 concentrations (15%, 30% and 60%). The wound healing process was evaluated by measuring the length of the wound manually from 0 to 10 days in each group. Meanwhile, the number of fibroblast cells was calculated through hematoxylin eosin (HE) staining and observed using an Olympus CX41 microscope with a 10x magnification and objective lens magnification in 3 fields. Result: There was a significant difference in the reduction in wound length (p =< 0,000) between the five experimental groups (Rhodomyrtus tomentosa leaf extract solution 15%, 30% and 60%, negative control and positive control. Solution of rhodomyrtus tomentosa leaf extract accelerated the increase in the number of fibroblasts compared to the negative control group (p = 0.003), but did not make a difference (p = 0.403) with the positive control group. Rhodomyrtus tomentosa leaf extraction solution had the same microscopic effect on the number of fibroblasts with a positive control group given 0.9% NaCl solution. Conclusion: There was a significant difference in the number of fibroblasts between all groups, but no difference in wound healing length.

Background: Peperomia pellucida L'HBK or known as messengers in the Indonesian plant is a herbaceous plant that belongs to the family Piperaceae. This study aimed to determine the effect of the stem and leaf extract cream messengers to the healing process of burns in rats (Rattus norvegicus. Methods: This was an experimental study using a completely randomized design. Test animals were divided into three groups, each - each group consisted of 3 rats. The first group is the negative control group (distilled water), the second group is a positive control group (Bioplacenton®), the third group is the group treated stem and leaf extract cream errand. The diameter of the wound and fibroblasts observed histopathology and is used as an indicator of the healing process of burns. The burns were treated and observed the healing effect for 20 days. Data were analyzed statistically wound diameter using ANOVA followed by LSD test. Results: The results showed the cream extracts of stems and leaves telling effect on the healing process of burns on rats. Conclusion: The results also showed that the treatment group and the leaf stem extract cream messengers and control groups positively influence the healing process of burns significantly when compared to the negative control group. Meanwhile, the treatment group stem and leaf extract cream messengers have no preformance difference influence the healing process of burns a significant positive control group

The aim of this study was to determine the effect of neem leaf extract (Azadirachta indica) on parasitemia of rats infected with Trypanosoma evansi (T. evansi) Aceh local isolate. A total of 24 male rats aged three months were used in this study and randomly divided into six treatment groups equally. The negative control group (K0) without T. evansi infection and neem leaf extract, the positive control group (K1) was infected with T. evansi but no neem leaf extract given, group K2, K3, K4, and K5 were infected with 5x104 T. evansi and were given neem leaf extract after patent infection with dose of 50, 100, 400, and 800 mg/kg BW respectively. The extract was given orally for three consecutive days. On the fourth day, rat blood was drawn for parasitemia examination. The results showed that no T. evansi detected in rats in negative control group (K0), while parasitemia in group K1; K2; K3; K4; and K5 was 12,295 x106/mL; 10,495 x106/mL; 9,360 x106/mL; 5,080x106/mL; and 2,398x106/mL of blood, respectively. Percentage of inhibition of parasitemia in K2, K3, K4, and K5 reached 14.64, 23.78, 58.68, and 80.50%, respectively. Based on the result of the study, neem leaf extract of 800 mg/kg BW gave the highest reduction of parasitemia in rats infected with T. evansi.

Abstract: High blood cholesterol is a major risk factor for atherosclerosis. Gedi plants contain polyphenolic compounds that can lowering blood cholesterol levels. This study aimed to find out the effect of gedi (Abelmoschus manihot L) leaves extract on histopatologic features of aorta of those wistar rats, which had been administered an atherogenic diet. This was an experimental study on animal using 20 Wistar rats as samples. The rats were divided into 4 different groups, 5 rats for each group. Negative control group was fed only pelet, positive control group was fed pork fat as much as 2 ml/day, group C was fed on pork fat and gedi leaf extract, and group D was fed on pork fat then continued with gedi leaf extract. The results indicated that administration of 30 mg gedi leaf extract showed a histopathologic feature which is decreasing of foam cells. Conclusion: Microscopically, aorta of those rats fed on pork lard diet showed a number of foam cells in tunica intima and media whereas the rats that were fed on pork lard along with gedi leaf extract, generally there were no foam cells, but there aresome parts which contain few foam cells. On the group of Wistar rats which were fed on pork lard, continued with gedi leaf extract showed less foam cells compared to the positive control group.Keywords: Gedi leaf extract, pork lard, foam cell.Abstrak: Kadar kolesterol darah yang tinggi merupakan faktor risiko utama terjadinya aterosklerosis. Tumbuhan gedi mengandung senyawa polifenol yang dapat menurunkan kadar kolesterol darah. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian ekstrak daun gedi terhadap gambaran histopatologi aorta tikus wistar dengan diet aterogenik. Hewan uji yang digunakan berupa 20 ekor tikus Wistar yang dibagi ke dalam 4 kelompok, masing-masing kelompok terdiri dari 5 ekor tikus. Kelompok kontrol negatif hanya diberikan pelet biasa, kelompok kontrol positif diberikan diet lemak babi 2ml/hari, kelompok C diberikan diet lemak babi bersamaan dengan ekstrak daun gedi, dan kelompok D diberikan lemak babi kemudian dilanjutkan dengan pemberian ekstrak daun gedi. Hasil penelitian memperlihatkan bahwa pemberian ekstrak daun gedi 30 mg mampu memberikan gambaran histopatologi aorta berupa berkurangnya jumlah sel busa. Simpulan: Secara mikroskopik, aorta tikus wistar yang diberi lemak babi menunjukkan adanya penumpukan sel-sel busa pada tunika intima dan media, sedangkan pada aorta tikus wistar yang diberi lemak babi bersamaan dengan ekstrak daun gedi maupun yang dilanjutkan dengan ekstrak daun gedi ditemukan berkurangnya jumlah sel-sel busa.Kata Kunci : Ekstrak daun gedi, Lemak babi, Sel busa.

Multidrug-resistant tuberculosis (MDR-TB) is a tuberculosis infection that is resistant to the treatment at least two of the most powerful anti-tuberculosis drugs, such as Isoniazid and Rifampisin. Increased cases of MDR-TB in morbidity and mortality become obstacles in the control of tuberculosis (TB), thus requiring supportive treatment of natural ingredients that can contribute in the treatment of TB, such a noni fruit. The main objective of this study was extract of noni fruits to inhibition the growth of strain MDR-TB bacteria, and compered it with the anti-TB drugs. The Morinda c. Linn (Noni) fruits was extraced by ethanol (96%). The extract was filtered through whatman No.1 filter paper, evaporated to dryness on a water bath until the solvent evaporated completely and yield of the crude extract. The experiment were divided into 3 groups, i.e.: negative control: group I; positive control: group II; crude extracts noni fruit: group III: combinations of crude extracts noni fruit and anti-TB drugs (K, AK, and OF). Each group was divided into three groups’ doses of 30 mg/ml, 40 mg/ml and 50 mg/ml. The Anti-tuberculosis activities of extracts noni fruit and K, AK, and OF against TB-MDR bacteria were tested by susceptibility test using proportion method in Lowenstein-Jensen (LJ) media.The anti-tubercular activity of noni fruits was determined by the minimum inhibitory concentration (MIC) of the bacterial growth at various doses 30 mg/ml, 40 mg/ml, and 50 mg/ml. The research design used post-test only contol group, and analyzed using analysis of variance and post hoct test. The extracted of Morinda c.Linn (noni) fruits have antitubercular activity to inhibiton of growth MDR-TB bacteria at various doses (p value=0,000). At a dose 30 mg/mlthe mean rate of the growth colonies of MDR TB-bacteria whit the mean rate 59,00 ± 27,81, and at a dose 40 mg/ml was1,50 ± 2,81. While at a dose of 50 mg/ml the bacterial colonies of MDR-TB did not grow in media. The combination of Morinda c.Linn (noni) fruit with anti-tuberculosis drugs, was the smallest groups to inhibit and eliminate MDR-TB bacteria at a dose 30 mg/ml(0,00 ± 00.00). The experimental results confirmed the extracted of Morinda c.Linn (noni) fruits have antitubercular activity as well as anti-TB drugs, and the combination of the extracted of Morinda c.Linn (noni) fruits and anti-TB drugs was the best groups to inhibiton of growth MDR-TB bacteria.   Keywords: TB-MDR Bacteria, Morinda c. Diabetes is a metabolic disease that can affect the male reproductive system. The condition of hyperglycemia will increase the production of ROS, causing oxidative stress. Oxidative stress causes microangiopathy, triggers cell apoptosis, interferes with spermatogenesis, thereby affecting the diameter of the seminiferous tubules. Kebar grass contains of flavonoid and vitamin E which can be used to reduce ROS reactions and inhibit oxidative stress. This research was to prove that kebar grass extract can increase diameter of the seminiferous tubules of diabetic mice. Total of 30 samples were divided into 5 groups: negative control group (K-) was given CMC-Na, positive control group (K+) was given metformin 2mg/kg, groups P1, P2 and P3 received 2mg/kg metformin and kebar grass extract with different dosages (67.5mg/kg, 135mg/kg and 270mg/kg). The treatment lasted for 35 days. On the 36th day the mice were sacrificed and the testes were taken, then histological observations were made with HE staining to measure the diameter of tubules. The result showed that there was a significant difference in the diameter of seminiferous tubules with p value=0,000. Kebar grass extract can increase the diameter of seminiferous tubules of diabetic mice.

Wound is considered as a serious health problem, because it can interfere with quality of life due to pain, weakness, and decreased mobility and quality of life. This study aims to determine the effect ofj jathropha multifida, linn to the number of PMN and selt mast in the process of wound healing in rats with acute injury models. This study is an experimental research laboratory using the design of post test control group design Wistar rats were used as research subjects. Based on Tukey HSD test results, the average number of PMN leukocytes third day the negative control group had no significant difference in the positive control group (0.037) and treatment group (0004). On the seventh day the number of PMN leukocytes negative control group had no significant difference in the positive control group (0.069) and had a significant difference to the treatment group (0.00). Meanwhile, on the fourteenth day the number of PMN leukocytes negative control group had no significant difference in the positive control group (0.000) and had a significant difference to the treatment group (0.00). Based on the results of data analysis Annova one direction on the third day obtained a significant level of 0:50 means there is no significant difference in the number of mast cells in each group on the seventh day. While on day three significant values ​​of 0:08 was also no significant difference, as well as on the fourteenth day found no significant difference between the group with a value of 0.68. Conclusions there was a decrease in the number of Leukocyte PMN in rat wounds given a 10% stem extract ointment, and there was a decrease in the number of mast cells in the wounds of rodents given a 10% stem extract ointment. Suggestions need to be investigated regarding the use of distance stem extract in various concentrations to obtain effective results for wound healing and the need to examine other factors related to wound healing.

Background: Orthodontic tooth movement is a continual and balanced process between bone deposition and bone resorption in pressure and tension sites. Stichopus hermanii is one of the best fishery commodities in Indonesia. It is natural and contains various active ingredients such as hyaluronic acid, chondroitin sulphate, cell growth factor, eicosa pentaenoic acid (EPA) docosa hexaenoic acid (DHA) and flavonoid that potentially play a role in orthodontic tooth movement. Purpose: The aim of this study was to investigate the active ingredients of nanopowder Stichopus hermanii promoting bone resorption in tension area orthodontic tooth movement. Methods: A quantitative test for active ingredients of stichopus hermanii was conducted. Thirty two male Cavia cobaya were divisibled became four groups. K (–) groups as a negative control group (without treatment), K (+) groups as a positive control group which were provided with a separator rubber for orthodontic tooth movement, and P1, P2 groups, which were treated with 3% and 3.5% stichopus hermanii for orthodontic tooth movement. After treatment the cavia cobaya were sacrificed. TRAP-6 expression as a osteoclast marker was examined by means of an immunohistochemistry method. Results: A one-way Anova test confirmed that TRAP-6 expression was significantly increased with p = 0.00 (p≤0,05) in P2 compared to K (+). P2 to K (–), P2 to P1 and P1 to K (+) had no significant differences Conclusion: Nanopowder Stichopus hermanii 3.5% has an active ingredient that could increase osteoclast activity to resorb periodontal ligament and alveolar bone in tension areas of orthodontic tooth movement.
