Astaxanthin is a potent antioxidant compared with vitamins and other antioxidants. However, astaxanthin extract from shrimp processing waste has not yet been used in cosmetic products. This study aimed to explore the natural astaxanthin from shrimp shells for antioxidant and antityrosinase activities as well as potential toxicity. The antioxidant activities were performed with 2,2-diphenyl-1-picrylhydrazyl (DPPH), 2,2’-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical scavenging, β-carotene bleaching, and singlet oxygen quenching assays. The results revealed that astaxanthin extract demonstrated potent antioxidant activities against DPPH and ABTS radicals, and prevented the bleaching of β-carotene and quenching of singlet oxygen (EC50 17.5 ± 3.6, 7.7 ± 0.6, 15.1 ± 1.9 and 9.2 ± 0.5 μg/mL, respectively). Furthermore, the astaxanthin extract could inhibit tyrosinase activity (IC50 12.2 ± 1.5 μg/mL) and had no toxic effects on human dermal fibroblast cells. These results suggested that shrimp astaxanthin would be a promising dietary supplement for skin health applications.

This review summarizes studies of protection against singlet oxygen and radical damage by carotenoids. The main focus is on how substitutions of the carotenoid molecules determine high antioxidant activities such as singlet oxygen quenching and radical scavenging. Applied assays were carried out either in vitro in solvents or with liposomes, and in a few cases with living organisms. In the latter, protection by carotenoids especially of photosynthesis against light- and UV-stress is of major importance, but also heterotrophic organisms suffer from high light and UV exposure which can be alleviated by carotenoids. Carotenoids to be compared include C30, C40 and C50 molecules either acyclic, monocyclic or bicyclic with different substitutions including sugar and fatty acid moieties. Although some studies are difficult to compare, there is a tendency towards mono and bicyclic carotenoids with keto groups at C-4/C-4’ and the longest possible polyene structure functions to act best in singlet oxygen quenching and radical scavenging. Size of the carotenoid and lipophilic substituents such as fatty acids seem to be of minor importance for their activity but hydroxyl groups at an acyclic end and especially glycosylation of these hydroxyl groups enhance carotenoid activity.

Antioxidant activities of acetone and ethyl acetate extracts from Metaplexis japonica Makino, one of famous medicine plants in the eastnorth region of China, named luomo in Chinese, were examined by a DPPH (1,1-Diphenyl-2-picrylhydrazyl) radical-scavenging assay and a β-carotene-linoleic acid test. In DPPH, the antioxidant activity of the acetone extracts, ethyl acetate extracts and derivative were IC50 were 313.21, 266.92 and 118.78μg/mL, respectively. In the β-carotene-linoleic acid test, IC50 were 285.09, 351.57 and 123.89μg/mL. It was concluded that Metaplexis japonica Makino and its derivatives might be a potential natural source of antioxidants .

Antioxidants are helpful in prevention of several diseases related with oxidative stress including neurodegenerative disorders. In recent studies, carbazoles were given proof of promising antioxidant activities. In this article, 9-ethyl-9H-carbazole hydrazone derivatives were synthesized, characterized and their in vitro antioxidant activity and possible cytotoxic effects were investigated. Furthermore, protective effect of the synthesized derivatives against amyloid β-induced damage in PC12 neuronal cells was examined by using MTT assay. The newly synthesized carbazoles were found to have radical scavenging activity with a varying potency both in cell-free and cell-based in vitro assays. Several compounds, especially such as 3d and 3e, 3m and 3n bearing two halogen groups on the phenyl ring, were found to have cytotoxic activity. However, their cytotoxic activities were not higher than that of melatonin. Several compounds also significantly protected neuronal PC12 cells against amyloid β-induced damage, which can be defined as neuroprotective agents. (4-(2-((9-Ethyl-9H-carbazol-3-yl)methylene)hydrazinyl)benzonitrile) 3r was found as the most active compound with both radical scavenging activity and neuroprotective effects against amyloid β-induced damage. These findings might provide an alternative strategy for developing novel carbazole derivatives for management of neurodegenerative diseases, such as Alzheimer's disease.

Two new abietane diterpenoids (1,2), along with five known diterpenoids (3–7), were first isolated and purified from the stems of Clerodendrum bracteatum. The structures of the new compounds were established by extensive analysis of mass spectrometric and 1-D, 2-D NMR spectroscopic data. Their antioxidant activities were determined on DPPH radical scavenging and ABTS. The in vitro cytotoxic activities of the compounds were evaluated against the HL-60 and A549 cell lines by the MTT method.

The aim of this study is to evaluate in vitro antioxidant activities of Ruta montana L.  extracts. This activity was evaluated by three methods : DPPH (2, 2'-diphenyl- 1- picrylhydrazy), bleaching of β-carotene and chelation of ferrous iron. Results showed that ethyl acetate extract (EAE) represents the highest amount of total polyphenols, tannins and flavonoids with 257,1 ± 0,703µg gallic acid equivalent/mg of extract,  251 ± 1.41 µg tannic acid equivalent /mg of extract,117,4 ± 3,451 µg quercetin equivalents/mg of extract, 139,5 ± 4,107 µg rutin equivalents/mg of extract, respectively. In the DPPH assay, ethyl acetate extract showed the higher scavenging capacity (IC50 = 0.044 ± 0.001 mg/ml) followed by methanol, aqueous and chloroform extract. Whereas, AqE showed the best chelating effect and the best inhibitory capacity of the coupled oxidation of linoleic acid/ β-carotene. The present study aimed to estimate and compare, using in vitro assays, the antioxidant activities of alcoholic extracts of saffron stigma and spath as by-products of the saffron crop. The antioxidant activity was evaluated using in vitro assays such as total antioxidant capacity, DPPH free radical, Scavenging of H2O2, FRAP and β-carotene bleaching assay. The results allowed us to evidence the presence of products with antioxidant and bioactive capacities in spaths and stigmas of Crocus sativus. The response of each extract depends on the type of the test. Extract from spaths protect β-carotene from bleaching more than the extract of stigmas and extract from stigmas revealed a significant reducing power activity than that of spaths. As a result, we can consider these extracts especially that of spaths like a new source of antioxidant compounds.

Gedi’s leave is one of the raw material in tinutuan porridge, a traditional food from Manado Indonesia. The leaves was extracted by various solvent to dissolve all of the compound. The aim of this study was to compare the profile and antioxidant activities of the extract in various sequence of solvent. Hexane, aseton, and methanol were used to dissolve a non polar, semi polar and polar component, respectively. All of the extract were analysed for its antioxidant activities. The profile of extract were indicated by total phenol and total flavonoid meanwhile antioxidant activities wasmeasured by radical scavenging activity DPPH, metal chelating and singlet oxygen quenching. The result indicated the sequence of hexane – aseton – methanol contain the highest of total phenol and flavonoid compare to the others with 10.67±0.49 mg GAE/g extract and 2.33±0.026 mg quersetin/g extract. The sequence also showed the highest of antioxidant activities at 150 μg/mL extract with 67.47%; 48.07% and 38.66% for percentage of inhibition DPPH, value of metal chelating and singlet oxygen, respectively.Keywords: Gedi’s leave, sequencial extraction, phenolic, flavonoid and antioxidant ABSTRAKDaun Gedi (Abelmoschus manihot L) merupakan salah satu bahan utama tinutuan, makanan tradisional Manado.Penelitian tentang profil dan aktivitas antioksidan dari daun Gedi telah dilakukan. Tujuan penelitian ini adalah mengevaluasi dan membandingkan profil dan aktivitas antioksidan dari daun gedi yang diekstraksi secara sekuensial dengan pelarut heksana, aseton dan metanol. Ekstrak daun gedi selanjutnya dianalisis kandungan total fenolik dan flavonoid, sedangkan aktivitas antioksidannya dilakukan secara in vitro meliputi penangkal radikal bebas DPPH, pengkelat logam dan penstabil oksigen singlet. Hasil penelitian menunjukkan bahwa ekstrak sekuensial heksanaaseton-metanol(ESHAM) memiliki total fenol dan total flavonoid yang tertinggi dibandingkan dengan ekstrak lainnya,masing-masing sebesar 10,67±0,49 mg GAE/g ekstrak dan 2,33±0,026mg kuersetin/g ekstrak. ESHAMjuga memiliki aktivitas antioksidan yang paling tinggi, dengan persentase penghambatan DPPH sebesar 67,47%; persen pegkelat logam sebesar48,07% dan persen penghambatan oksigen singlet sebesar 38,66% pada konsentrasi 150μg/mL esktrak. Kesimpulan senyawa fenolik pada daun gedi bersifat polar sehingga menghasilkan aktivitas antioksidan tertinggi pada pelarut polar.Kata kunci: Daun gedi, ekstraksi sekuensial, fenolik, flavonoid dan antioksidan

Alzheimer's disease is a neurodegenerative disorder characterized by memory loss and impairment of language. Alzheimer's disease is strongly associated with oxidative stress and impairment in the cholinergic pathway, which results in decreased levels of acetylcholine in certain areas of the brain. Hence, inhibition of acetylcholinesterase activity has been recognized as an acceptable treatment against Alzheimer's disease. Nature provides an array of bioactive compounds, which may protect against free radical damage and inhibit acetylcholinesterase activity. This study compares the in vitro antioxidant and anticholinesterase activities of hydroalcoholic extracts of five cultivars of Rosa Damascena Mill. petals (R. damascena 'Bulgarica', R. damascena 'Faik', R. damascena 'Iranica', R. damascena 'Complex-635' and R. damascena 'Complex-637') from Isparta, Turkey. The antioxidant activities of the hydroalcoholic extracts were tested for ferric ion reduction and DPPH radical scavenging activities. The anti-acetylcholinesterase activity was also evaluated. All rose cultivars showed a high potency for scavenging free radical and inhibiting acetylcholinesterase activity. There was a significant correlation between antioxidant and acetylcholinesterase inhibitory activity. Among cultivars, Complex-635 showed the highest inhibitory effect with an IC50 value of 3.92 µg/mL. Our results suggest that all these extracts may have the potential to treat Alzheimer's disease with Complex-635 showing more promise.

The investigation aimed to study the in vitro and in silico antioxidant properties of Melissa officinalis subsp. officinalis essential oil (MOEO). The chemical composition of MOEO was determined using GC–MS analysis. Among 36 compounds identified in MOEO, the main were beta-cubebene (27.66%), beta-caryophyllene (27.41%), alpha-cadinene (4.72%), caryophyllene oxide (4.09%), and alpha-cadinol (4.07%), respectively. In vitro antioxidant properties of MOEO have been studied in 2,2’-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) and 2,2-diphenyl-1-picrylhydrazyl (DPPH) free-radical scavenging, and inhibition of β-carotene bleaching assays. The half-maximal inhibitory concentration (IC50) for the radical scavenging abilities of ABTS and DPPH were 1.225 ± 0.011 μg/mL and 14.015 ± 0.027 μg/mL, respectively, demonstrating good antioxidant activity. Moreover, MOEO exhibited a strong inhibitory effect (94.031 ± 0.082%) in the β-carotene bleaching assay by neutralizing hydroperoxides, responsible for the oxidation of highly unsaturated β-carotene. Furthermore, molecular docking showed that the MOEO components could exert an in vitro antioxidant activity through xanthine oxidoreductase inhibition. The most active structures are minor MOEO components (approximately 6%), among which the highest affinity for the target protein belongs to carvacrol.

AbstractPectin is a kind of natural and complex carbohydrates which is extensively used in food, chemical, cosmetic, and pharmaceutical industries. Fresh sunflower (Helianthus annuus L.) heads were utilized as a novel source of pectin extracted by ammonium oxalate. The conditions of the extraction process were optimized implementing the response surface methodology. Under optimal extraction parameters (extraction time 1.34 h, liquid–solid ratio 15:1 mL/g, ammonium oxalate concentration 0.76% (w/v)), the maximum experimental yield was 7.36%. The effect of spray-drying and freeze-drying on the physiochemical properties, structural characteristics, and antioxidant activities was investigated by FT-IR spectroscopy, high performance size exclusion chromatography, and X-ray diffraction. The results showed freeze-drying lead to decrease in galacturonic acid (GalA) content (76.2%), molecular weight (Mw 316 kDa), and crystallinity. The antioxidant activities of pectin were investigated utilizing the in-vitro DPPH and ABTS radical-scavenging systems. This study provided a novel and efficient extraction method of sunflower pectin, and confirmed that different drying processes had an effect on the structure and properties of pectin.
