Betaine aldehyde dehydrogenase 1 (BADH1), a paralog of the fragrance gene BADH2, is known to be associated with salt stress through the accumulation of synthesized glycine betaine (GB), which is involved in the response to abiotic stresses. Despite the unclear association between BADH1 and salt stress, we observed the responses of eight phenotypic characteristics (germination percentage (GP), germination energy (GE), germination index (GI), mean germination time (MGT), germination rate (GR), shoot length (SL), root length (RL), and total dry weight (TDW)) to salt stress during the germination stage of 475 rice accessions to investigate their association with BADH1 haplotypes. We found a total of 116 SNPs and 77 InDels in the whole BADH1 gene region, representing 39 haplotypes. Twenty-nine haplotypes representing 27 mutated alleles (two InDels and 25 SNPs) were highly (p < 0.05) associated with salt stress, including the five SNPs that have been previously reported to be associated with salt tolerance. We observed three predominant haplotypes associated with salt tolerance, Hap_2, Hap_18, and Hap_23, which were Indica specific, indicating a comparatively high number of rice accessions among the associated haplotypes. Eight plant parameters (phenotypes) also showed clear responses to salt stress, and except for MGT (mean germination time), all were positively correlated with each other. Different signatures of domestication for BADH1 were detected in cultivated rice by identifying the highest and lowest Tajima’s D values of two major cultivated ecotypes (Temperate Japonica and Indica). Our findings on these significant associations and BADH1 evolution to plant traits can be useful for future research development related to its gene expression.

Sorghum [Sorghum bicolor (L.) Moench] seed germination is sensitive to salinity, and seed priming is an effective method for alleviating the negative effects of salt stress on seed germination. However, few studies have compared the effects of different priming agents on sorghum germination under salt stress. In this study, we quantified the effects of priming with distilled water (HP), sodium chloride (NaCl), potassium chloride (KCl), calcium chloride (CaCl2), and polyethylene glycol (PEG) on sorghum seed germination under 150 mM NaCl stress. The germination potential, germination rate, germination index, vigor index, root length, shoot length, root fresh weight, shoot fresh weight, root dry weight, and shoot dry weight were significantly reduced by salt stress. Different priming treatments alleviated the germination inhibition caused by salt stress to varying degrees, and 50 mM CaCl2 was the most effective treatment. In addition, the mitigation effect of priming was stronger on root traits than on shoot traits. Mitigation efficacy was closely related to both the type of agent and the concentration of the solution. Principal component analysis showed that all concentrations of CaCl2 had higher scores and were clearly distinguished from other treatments based on their positive effects on all germination traits. The effects of the other agents varied with concentration. The priming treatments were divided into three categories based on their priming efficacy, and the 50, 100, and 150 mM CaCl2 treatments were placed in the first category. The 150 mM KCl, 10% PEG, HP, 150 mM NaCl, 30% PEG, and 50 mM KCl treatments were placed in the second category, and the 100 mM NaCl, 100 mM KCl, 20% PEG, and 50 mM NaCl treatments were least effective and were placed in the third category. Choosing appropriate priming agents and methods for future research and applications can ensure that crop seeds germinate healthily under saline conditions.

Abstract A laboratory experiment regarding germination and seedling growth test was conducted with three black gram genotypes tested under three salinity levels (0, 75 and 150 mM), for 10 days, in sand culture within small plastic pot, to investigate the germination and seedling growth characteristics. Different germination traits of all black gram genotypes, like germination percentage (GP), germination rate (GR), coefficient of velocity of germination (CVG) greatly reduced, as well as mean germination time (MGT) increased with increasing salt stress. At high salt stress, BARI Mash-3 provided the highest GP reduction (28.58%), while the lowest was recorded (15.79% to control) in BARI Mash-1. Salinity have the negative impact on shoot and root lengths, fresh and dry weights. The highest (50.32% to control) and lowest reduction (36.39%) of shoot length were recorded in BARI Mash-2 and BARI Mash-1, respectively, under 150 mM NaCl saline conditions. There were significant reduction of root lengths, root fresh and dry weight, shoot length, shoot fresh and dry weight in all genotypes under saline condition. The genotypes were arranged as BARI Mash-1 > BARI Mash-3 > BARI Mash-2, with respect to salinity tolerance.

Plants such as wheat and barley that are strategically important crops need to be considered to develop a comprehensive toxicity profile for nanoparticles (NPs). The present study was aimed to investigate the effects of chitosan and SiO2 NPs on wheat and barley plants. Two factorial experiments (seeds priming and direct exposure) were performed based on a completely randomized design in four replications. Results showed that the seeds priming with the NPs had not significant effect on germination parameters such as Germination Percentage (GP), Germination Rate (GR), Germination Value (GV), Mean Germination Time (MGT), Pick Value (PV) and Mean Daily Germination (MDG). In contrast, exposure of the seeds to the NPs had significant effects on these parameters. In both experiments, treatments had significant effects on shoot, seedling, root length, fresh and dry weight, as well as vigor indexes as compared to the control. In most traits, the best concentration of NPs was 30 ppm, whereas applications of the NPs with 90 ppm displayed adverse effects on majority of the studied traits. According to these results, selectivity in applications of NPs with suitable concentration and method is essential for different plant species.

Salt stress inhibits normal plant growth and development by disrupting cellular water absorption and metabolism. Therefore, understanding plant salt tolerance mechanisms should provide a theoretical basis for developing salt-resistant varieties. Here, we cloned ThTrx5 from Tamarix hispida, a salt-resistant woody shrub, and generated ThTrx5-overexpressing transgenic Arabidopsis thaliana lines. Under NaCl stress, the germination rate of overexpressing ThTrx5 lines was significantly increased relative to that of the nontransgenic line; under salt stress, superoxide dismutase (SOD), peroxidase (POD), catalase (CAT), and glutathione levels and root length and fresh weight values of transgenic ThTrx5 plants were significantly greater than corresponding values for wild-type plants. Moreover, with regard to the transcriptome, comparison of differential gene expression of transgenic versus nontransgenic lines at 0 h and 3 h of salt stress exposure revealed 500 and 194 differentially expressed genes (DEGs), respectively, that were mainly functionally linked to catalytic activity and binding process. Pull-down experiments showed that ThTrx bound 2-Cys peroxiredoxin BAS1-like protein that influences stress response-associated redox, hormone signal transduction, and transcription factor functions. Therefore, this work provides important insights into ThTrx5 mechanisms that promote salt tolerance in plants.

Tartary buckwheat (Fagopyrum tataricum (L.) Gaertn.) is a nutritional crop, which has high flavonoid content. However, buckwheat is a salt sensitive glycophyte cereal crop and the growth and grain yield of buckwheat are significantly affected by soil salinity. In this study, we performed a comprehensive analysis of the transcriptome and metabolome of salt treated-buckwheat to understand the effects of salinity on buckwheat. A total of 50,681,938 clean reads were acquired from all samples. We acquired 94,950 unigenes with a mean length of 1133 bp and N50 length of 1900 bp assembly. Of these, 63,305 unigenes (66.7%) were matched in public databases. Comparison of the transcriptome expression patterns between control and salt treated groups showed that 4098 unigenes were up-regulated and 3292 unigenes were down-regulated significantly. Further, we found that genes involved with amino acid, lipid and nucleotide metabolism were most responsive to salt stress. Additionally, many genes involved in secondary metabolite biosynthesis changed significantly following treatment. Those affected included phenylpropanoid biosynthesis and flavonoid biosynthesis. Chromatographic analysis was used to examine the differences in concentration of flavonoids, carotenoids, amino acids and organic acids in the samples following treatment. There was a significant increase in rutin (12.115 mg/g dry weight), following salt stress; whereas, six carotenoids (lutein, zeaxanthin, 13Z-β-carotene, α-carotene, E-β-carotene and 9Z-β-carotene) did not significantly respond to salt stress. Ultimately, our data acts as a valuable resource for future research on buckwheat and can be used as the basis for future analysis focused on gene-to-metabolite networks in buckwheat.

Teff is an annual grass originated in Ethiopia, grown for both human food and animal feed. Salinity and moisture stress are the main limiting factors of agricultural development on arid and semi-arid regions. The aim of this study was to investigate the effect of salinity due to NaCl on germination of teff. A set of experiments was conducted under a completely randomized design with four replications of 200 seeds at two different suboptimal germination temperatures (15 and 25 °C) in the dark for 10 days. In order to create salt stress conditions during the germination process, seven solutions were used (0 as control, 80, 160, 240 and 320, 400 and 480 mM NaCl). The germination performance was evaluated by final germination percentage and mean germination time. The presence of NaCl reduced germination, especially above 240 mM for 15 °C and 400 mM for 25 °C. The mean germination time increased with higher NaCl levels.

Screening of chickpea lines for salt tolerance through seed germination and early seedling growth is crucial for their evaluation. Seeds of 30 chickpea genotypes were germinated on a sand bed irrigated with saline (3, 6, 9, 12 dS/m) and control solutions upto 30 days. At the early seedling stage (25-30 days), germination percentage, chlorophyll content, proline, root length, shoot length and seedling dry weight were found to be affected due to salinity. Salt tolerance index (STI) for plant biomass maintained a significant correlation with chlorophyll, proline, shoot length, and root length, which indicated that these parameters could be used as selection criteria for screening chickpea genotypes against salt stress. Significant differences in shoot length, root length, and seedling dry weight in 30-day-old seedlings were observed among selected chickpea genotypes as well. From the overall observation of germination characterstics and early seedling growth, it is concluded that the chickpea genotypes, HC-1, HC-5, ICC 867, ICC 5003, H-10-41 showed better salt tolerance as compared to the available salt tolerant check variety.

Salt tolerances of 3 cultivars, Menemen puccinellia (Puccinellia ciliata Bor), Tyrrell and Dundas [tall wheatgrass, Thinopyrum ponticum (Podp.) Z. W. Liu and R. R. C. Wang], were compared with respect to their seed germination, adaptive responses to salt and waterlogging, seedling emergence, plant growth, shoot osmolality and mineral contents in a series of salt-stress experiments. An inverse normal distribution provided good fits for the time to seed germination. Under NaCl stress, 50% of the control (distilled water) seed germination rates of Menemen, Tyrrell and Dundas were achieved in 178.8, 300.9 and 296.8 mmol/L NaCl, respectively. Fifty percent of the control seedling emergence rates of these 3 cultivars were in 92.7, 107.2 and 113.5 mmol/L NaCl, respectively. The seed germination rates of these 3 cultivars under both salt and waterlogging stress were far lower than those germinated only under salt stress at the same salt level. Seed pretreatment by soaking seed in NaCl solutions greatly increased the seed germination rate under salt stress for Menemen and under both salt stress and waterlogging for Dundas. Tyrrell and Dundas were very similar in their tolerance to salt stress, and were significantly (P<0.05) more salt tolerant than Menemen in terms of seed germination and seedling emergence rate. Both shoot height and dry matter of these 3 cultivars were not statistically different among all salt stress levels during the seedling elongation period, indicating that the established plants of these 3 cultivars were very salt tolerant. The salt tolerance mechanisms of these 3 cultivars are possibly related to their abilities to maintain high osmolality in shoots by regulating high sodium and potassium contents, and reducing calcium deficiency under salt stress.

ABSTRACT: Soil salinity limits agricultural production and is a major obstacle for increasing crop yield. Common wheat is one of the most important crops with allohexaploid characteristic and a highly complex genome. QTL mapping is a useful way to identify genes for quantitative traits such as salinity tolerance in hexaploid wheat. In the present study, a hydroponic trial was carried out to identify quantitative trait loci (QTLs) associated with salinity tolerance of wheat under 150mM NaCl concentration using a recombinant inbred line population (Xiaoyan 54×Jing 411). Values of wheat seedling traits including maximum root length (MRL), root dry weight (RDW), shoot dry weight (SDW), total dry weight (TDW) and the ratio of TDW of wheat plants between salt stress and control (TDWR) were evaluated or calculated. A total of 19QTLs for five traits were detected through composite interval mapping method by using QTL Cartographer version 2.5 under normal and salt stress conditions. These QTLs distributed on 12 chromosomes explained the percentage of phenotypic variation by individual QTL varying from 7.9% to 19.0%. Among them, 11 and six QTLs were detected under normal and salt stress conditions, respectively and two QTLs were detected for TDWR. Some salt tolerance related loci may be pleiotropic. Chromosome 1A, 3A and 7A may harbor crucial candidate genes associated with wheat salt tolerance. Our results would be helpful for the marker assisted selection to breed wheat varieties with improved salt tolerance.
