Plants and plant-based products have been used for a long time for medicinal purposes. This study aimed to determine the antioxidant and anti-α-glucosidase activities of eight selected underutilized plants in Malaysia: Leucaena leucocephala, Muntingia calabura, Spondias dulcis, Annona squamosa, Ardisia elliptica, Cynometra cauliflora, Ficus auriculata, and Averrhoa bilimbi. This study showed that the 70% ethanolic extract of all plants exhibited total phenolic content (TPC) ranging from 51 to 344 mg gallic acid equivalent (GAE)/g dry weight. A. elliptica showed strong 2,2-diphenyl-1-picrylhydrazyl (DPPH) and nitric oxide (NO) scavenging activities, with half maximal inhibitory concentration (IC50) values of 2.17 and 49.43 μg/mL, respectively. Most of the tested plant extracts showed higher inhibition of α-glucosidase enzyme activity than the standard, quercetin, particularly A. elliptica, F. auriculata, and M. calabura extracts with IC50 values of 0.29, 0.36, and 0.51 μg/mL, respectively. A total of 62 metabolites including flavonoids, triterpenoids, benzoquinones, and fatty acids were tentatively identified in the most active plant, i.e., A. elliptica leaf extract, by using ultra-high-performance liquid chromatography (UHPLC)–electrospray ionization (ESI) Orbitrap MS. This study suggests a potential natural source of antioxidant and α-glucosidase inhibitors from A. elliptica.

Objective: The present investigation focuses on the use of Cardiospermum halicacabum L. in their phytochemical and biological activities. Methods: In this study, in vivo stem and in vitro callus ethanolic extracts of C. halicacabum were tested for their phytochemical attributes by qualitative method, Fourier transform infrared (FTIR), antioxidant, antibacterial, and bioactive compound properties. The bactericidal activity of the in vivo stem and in vitro callus extract has been evaluated in both Gram+ve and Gram-ve microorganisms using the disk diffusion method. Results: The highest frequency (78%) of well developed, dark green organogenic callus was induced from stem explant on Murashige and Skoog (MS) medium supplemented with 0.7 mg/l 2,4-Dichlorophenoxyacetic acid (2, 4-D) and 0.5 mg/l benzyl adenine (BA). The results of FTIR spectra confirmed the presence of functional groups in wild stem and in vitro callus extract of C. halicacabum with various peaks. The total phenolic content in ethanolic extract of in vivo plant and in vitro callus was 80.46 mg gallic acid equivalent (GAE)/g dry weight and 76.4 mg GAE/g dry weight, respectively. The highest percentage of tannins was measured at 78.03 in wild stem ethanol extracts followed by 75.22 in callus extract. The antioxidant activity of 2,2-diphenyl-2- picrylhydrazyl (DPPH) ethanol extract was found to be 206.54 μg/ml. IC50 values of the stem extracts of C. halicacabum are 306 μg/ml and 286 μg/ml in callus extract, respectively. Antibacterial activity of the ethanol extract was higher for Staphylococcus aureus (S. aureus) with a 17 mm zone of inhibition. Conclusion: The present investigation recommended that the callus ethanolic extract function as a good source of biologically active compounds and natural antioxidants.

Chemical compositions (fatty acids, total phenolic compounds, flavonoids) and the in-vitro biological activities (antioxidant and antimicrobial activity; and growth induction of edible fungal strains) were compared for four varieties of mango seeds. Hexanic extract (HE) showed a fatty acid profile with a higher proportion of oleic acid. In the ethanolic extract were found variable total phenolic contents (103 to 125 mg GAE/g dry weight) and flavonoid contents (0.72 to 0.8 mg QE/g dry weight). This study reports for the first time the presence of procyanidin B1 in ethanolic extracts. The antioxidant activity showed IC50 values ranging from 3.09 to 3.42 μg/mL for ABTS•+ and 12.17 to 13.93 μg/mL for DPPH•. The ethanolic extract from the seed of the Yulima variety showed the highest percentages of inhibition against Staphylococcus aureus. Residues removed from ethanolic extraction of the seed kernel (EKR) induced the growth of edible fungal strains: Lentinus crinitus and Pleurotus tubarius. The data obtained show the potential of the seeds from these mango varieties, which could lead to alternative uses in various industry sectors and the use of this agricultural byproduct.

The fruits of Canarium tramdenum are commonly used as foods and cooking ingredients in Vietnam, Laos, and the southeast region of China, whilst the leaves are traditionally used for treating diarrhea and rheumatism. This study was conducted to investigate the potential use of this plant bark as antioxidants, and α-amylase and α-glucosidase inhibitors. Five different extracts of C. tramdenum bark (TDB) consisting of the extract (TDBS) and factional extracts hexane (TDBH), ethyl acetate (TDBE), butanol (TDBB), and water (TDBW) were evaluated. The TDBS extract contained the highest amount of total phenolic (112.14 mg gallic acid equivalent per g dry weight), while the TDBB extract had the most effective antioxidant capacity compared to other extracts. Its IC50 values were 12.33, 47.87, 33.25, and 103.74 µg/mL in 2,2-diphenyl-1-picrylhydrazyl (DPPH), 2,2′-azino-bis (ABTS), reducing power (RP), and nitric oxide (NO) assays, respectively. Meanwhile, the lipid peroxidation inhibition of the four above extracts was proximate to that of butylated hydroxytoluene (BHT) as a standard antioxidant. The result of porcine pancreatic α-amylase inhibition showed that TDB extracts have promising effects which are in line with the commercial diabetic inhibitor acarbose. Interestingly, the inhibitory ability on α-glucosidase of all the extracts was higher than that of acarbose. Among the extracts, the TDBB extract expressed the strongest activity on the enzymatic reaction (IC50 = 18.93 µg/mL) followed by the TDBW extract (IC50 = 25.27 µg/mL), TDBS (IC50 = 28.17 µg/mL), and TDBE extract (IC50 = 141.37 µg/mL). The phytochemical constituents of the TDB extract were identified by gas chromatography–mass spectrometry (GC-MS). The principal constituents included nine phenolics, eight terpenoids, two steroids, and five compounds belonging to other chemical classes, which were the first reported in this plant. Among them, the presence of α- and β-amyrins were identified by GC-MS and appeared as the most dominant constituents in TDB extracts (1.52 mg/g). The results of this study revealed that C. tramdenum bark possessed rich phenolics and terpenoids, which might confer on reducing risks from diabetes. A high quantity of α- and β-amyrins highlighted the potentials of anti-inflammatory, anti-ulcer, anti-hyperlipidemic, anti-tumor, and hepatoprotective properties of C. tramdenum bark.

Crocus sativus L. (saffron) has been traditionally used as a food coloring or flavoring agent, but recent research has shown its potent pharmacological activity to tackle several health-related conditions. Crocus sp. leaves, and petals are the by-products of saffron production and are not usually used in the medicine or food industries. The present study was designed to determine the chemical composition of the water and ethanolic extracts of C. sativus leaves and test their cytotoxic activity against melanoma (IGR39) and triple-negative breast cancer (MDA-MB-231) cell lines by MTT assay. We also determined their anti-allergic, anti-inflammatory, and anti-viral activities. HPLC fingerprint analysis showed the presence of 16 compounds, including hydroxycinnamic acids, xanthones, flavonoids, and isoflavonoids, which could contribute to the extracts’ biological activities. For the first time, compounds such as tectoridin, iristectorigenin B, nigricin, and irigenin were identified in Crocus leaf extracts. The results showed that mangiferin (up to 2 mg/g dry weight) and isoorientin (8.5 mg/g dry weight) were the major active ingredients in the leaf extracts. The ethanolic extract reduced the viability of IGR39 and MDA-MB-231 cancer cells with EC50 = 410 ± 100 and 330 ± 40 µg/mL, respectively. It was more active than the aqueous extract. Kaempferol and quercetin were identified as the most active compounds. Our results showed that Crocus leaves contain secondary metabolites with potent cytotoxic and antioxidant activities.

With the understanding of the role of antioxidants in preventing degenerative and age-related diseases caused by oxidative stress, and taking into account the multitude of pharmacological applications such as antidiabetic, antifungal, cardioprotection, immunostimulant, interest in plants rich in this respect has increased. Ocimum basilicum L. (purple) is a one-year, fragrant spice herb with its own aroma. In this study, chemical characterization of Ocimum basilicum L. plant was performed and inhibition effects on urease and a-amylase were investigated. Total phenolic content of Ocimum basilicum L. leafs ethanolic and water extracts were 320.08±2.03, 388.15±1.05 mg GAE/100g; total flavonoids were 282.57±1.12, 307.75±0.89 mg QE/100g; antioxidant capacity of samples were 0.46±0.01 and 0.52±0.02  mM Fe+2/mg extract, 0.46±0.01; IC50 values of urease were 18.77±0.22, 20.19±0.15  % and IC50 values of α-amylase were 0.47±0.01, 0.42±0.01 µg/mL, respectively. It is determined that ethanolic extract of leaves is rich in linalool, linolenin, phytol and α-humulene. The datas show that the leaves of the plant may be effective on two important diseases such as Diabetus mellitus and H. pylori.

Lepidium sativum L. is a rich source of polyphenols that have huge medicinal and pharmaceutical applications. In the current study, an effective abiotic elicitation strategy was designed for enhanced biosynthesis of polyphenols in callus culture of L. sativum. Callus was exposed to UV-C radiations for different time intervals and various concentrations of melatonin. Secondary metabolites were quantified by using high-performance liquid chromatography (HPLC). Results indicated the total secondary metabolite accumulation of nine quantified compounds was almost three fold higher (36.36 mg/g dry weight (DW)) in melatonin (20 μM) treated cultures, whereas, in response to UV-C (60 min), a 2.5 fold increase (32.33 mg/g DW) was recorded compared to control (13.94 mg/g DW). Metabolic profiling revealed the presence of three major phytochemicals, i.e., chlorogenic acid, kaemferol, and quercetin, in callus culture of L. sativum. Furthermore, antioxidant, antidiabetic, and enzymatic activities of callus cultures were significantly enhanced. Maximum antidiabetic activities (α-glucosidase: 57.84%; α-amylase: 62.66%) were recorded in melatonin (20 μM) treated callus cultures. Overall, melatonin proved to be an effect elicitor compared to UV-C and a positive correlation in these biological activities and phytochemical accumulation was observed. The present study provides a better comparison of both elicitors and their role in the initiation of physiological pathways for enhanced metabolites biosynthesis in vitro callus culture of L. sativum.

We previously reported the inhibitory potentials of momilactones A (MA) and B (MB) against key enzymes related to type 2 diabetes and obesity. In this study, antioxidant and anti-skin-aging activities of MA and MB were investigated and compared with tricin, a well-known antioxidant and antiaging flavonoid in rice. MA, MB, and tricin were purified from rice husk by column chromatography and their biological activities were subsequently assayed by in vitro trials. The contents of MA, MB, and tricin of different commercial rice cultivars in Japan were quantified and confirmed by ultra-performance liquid chromatography-electrospray ionization-mass spectrometry (UPLC-ESI-MS) and high-performance liquid chromatography (HPLC) analyses. The antioxidant assays revealed a synergistic activity of the mixture MA and MB (MAB, 1:1, v/v). In addition, in 2,2’-azino-bis (ABTS) assay, IC50 values of MAB (0.3 mg/mL) and tricin (0.3 mg/mL) was 4-fold and 9-fold greater than that of individual MB (1.3 mg/mL) or MA (2.8 mg/mL), respectively. The in vitro enzymatic assays on pancreatic elastase and tyrosinase indicated that MA and MB were potential to relief skin wrinkles and freckles. In detail, MA exerted higher inhibition on both enzymatic activities (30.9 and 37.6% for elastase and tyrosinase inhibition, respectively) than MB (18.5 and 12.6%) and MAB (32.0 and 19.7%) at a concentration of 2.0 mg/mL. Notably, MA and the mixture MAB exhibited stronger inhibitions on elastase and tyrosinase in comparison with tricin and vanillin. MA, MB, and tricin in rice are potential to develop cosmetics as well as supplements for skin aging treatments.

The consumption of herbal teas has become popular in recent years due to their attractive flavors and outstanding antioxidant properties. The Five-Golden-Flowers tea is a herbal tea consisting of five famous edible flowers. The effects of microwave-assisted extraction parameters on the antioxidant activity of Five-Golden-Flowers tea were studied by single-factor experiments, and further investigated using response surface methodology. Under the optimal parameters (53.04 mL/g of solvent/material ratio, 65.52 °C, 30.89 min, and 500 W), the ferric-reducing antioxidant power, Trolox equivalent antioxidant capacity, and total phenolic content of the herbal tea were 862.90 ± 2.44 µmol Fe2+/g dry weight (DW), 474.37 ± 1.92 µmol Trolox/g DW, and 65.50 ± 1.26 mg gallic acid equivalent (GAE)/g DW, respectively. The in vivo antioxidant activity of the herbal tea was evaluated on alcohol-induced acute liver injury in mice. The herbal tea significantly decreased the levels of aspartate aminotransferase, total bilirubin, and malonaldehyde at different doses (200, 400, and 800 mg/kg); improved the levels of liver index, serum triacylglycerol, and catalase at dose of 800 mg/kg. These results indicated its role in alleviating hepatic oxidative injury. Besides, rutin, chlorogenic acid, epicatechin, gallic acid, and p-coumaric acid were identified and quantified by high performance liquid chromatography (HPLC), which could contribute to the antioxidant activity of the herbal tea.

Ethanolic extract and its n-hexane and chloroform soluble fractions of the leaves of Gardenia coronaria Buch. Ham. were screened for their bio-activities. Preliminary phytochemicals screening of crude extract revealed the presence of glycosides, alkaloids, tannins, saponins, reducing sugars and flavonoids in different extracts. In antibacterial test, maximal zone of inhibition obtained against B. cereus (15.3 mm) and in antifungal test maximal zone found against C. albicans (12.7 mm) by the chloroform extract. The crude extract of the plant exhibited notable anti-inflammatory activity. The antioxidant activity was evaluated by DPPH free radical scavenging method where the scavenging activity was concentration dependent with IC50 values of 5.15μg/ml, 8.75μg/ml and 12.71μg/ml for ethanol, nhexane and chloroform extract, respectively. In the castor oil-induced antidiarrheal assay, the crude extract of the plant significantly increased the latency and total count of defecation. Acetic acid-induced writhing reflex due to analgesia was inhibited by 31.18% (by ethanol), 23.67% (by n-hexane) and 24.73% (by chloroform) at 500 mg/kg body wt. in experimental mice. The extractives at doses from 31.25-500 μg/ml, produced inhibition of amylase activity in a dose dependent manner.Bangladesh Pharmaceutical Journal 20(2): 139-147, 2017

<p>Antioxidant based drug preparations are used in the prevention and management of complex diseases which include atherosclerosis, stroke, diabetes, alzheimer’s disease and cancer. Diabetes mellitus is a metabolic disorder of glucose metabolism. The management of blood glucose level is the hallmark in the treatment of this ailment, which may be achieved through the use of oral hypoglycemic drugs such as biguanides, insulin secretagogues, and 𝛼-amylase and glucosidase inhibitors. Although several biological activities had been reported for  <em>Momordica</em> <em>foetida </em>and <em>Momordica</em> <em>charantia</em>; it appears there is limited information on the  biological activity of <em>Momordica</em> <em>cissoides</em>. The purpose of this study is to compare the antioxidant, 𝛼-amylase and 𝛼-glucosidase inhibitory activities of <em>Momordica</em> <em>charantia </em>(M1), <em>Momordica</em> <em>foetida </em>(M2)<em>, </em>and <em>Momordica</em> <em>cissoides</em>  (M3) to establish a chemotaxonomic relationship between them. The antioxidant activities measured by DPPH scavenging properties, metal ion chelation, hydrogen peroxide scavenging and ABTS revealed that <em>M. foetida</em> had the highest inhibition potential, followed by <em>M. charantia</em> and the least being <em>M. cissoides</em>. In contrast, the antioxidant activities measured by FRAP,  the total phenolic content , flavonoids and tannins revealed that <em>M. cissoides</em> had the best antioxidant potential, while <em>M. foetida</em> had the least activity. The comparative α-amylase and glucosidase inhibitory studies performed demonstrated that the extracts of <em>M.cissoides </em>had the highest inhibitory potentials. Thus, the plant can be used in the management of diabetes.</p>
