Alkaloids of the Lycopodiaceae family are of great interest to researchers due to their numerous properties and wide applications in medicine. They play a very important role mainly due to their potent antioxidant, antidepressant effects and a reversible ability to inhibit acetylcholinesterase (AChE) enzyme activity. This property is of immense importance due to the growing problem of an increasing number of patients with neurodegenerative diseases in developed countries and a lack of effective and efficient treatment for them. Numerous studies have shown that Lycopodiaceae alkaloids are a rich source of AChE inhibitors. In the obtaining of new therapeutic phytochemicals from plant material, the extraction process and its efficiency is crucial. Therefore, the aim of this work was to optimize the conditions of modern PLE to obtain bioactive alkaloids from two Lycopodium species: L. clavatum L. and L. annotinum L. Five different solvents of different polarity were used for prepared plant extracts in order to compare the alkaloid content in and thereby effectiveness of the entire extraction. PLE parameters were used based on multiple studies conducted that gave the highest alkaloids recovery. Crude extracts were purified using solid-phase extraction (SPE) on Oasis HLB cartridge and examined by HPLC/ESI-QTOF–MS of the highly abundant alkaloids. To the best of our knowledge, this is the first time such high recoveries have been obtained for known Lycopodiaceae alkaloids. The best extraction results of alkaloid-lycopodine were detected in the dichloromethane extract from L. clavatum, where the yield exceeded 45%. The high recovery of annotinine above 40% presented in L. annotinum was noticed in dichloromethane and ethyl acetate extracts. Moreover, chromatograms were obtained with all isolated alkaloids and the best separation and quality of the bands in methanolic extracts. Interestingly, no alkaloid amounts were detected in cyclohexane extracts belonging to the non-polar solvent. These results could be helpful for understanding and optimizing the best conditions for isolating potent AChE inhibitors.

Guayule (Parthenium argentatum) is one of the most promising alternatives to produce natural rubber. As the guayule leaves represent a waste byproduct in the rubber extraction process, their exploitation might contribute to the valorization of the whole plant biomass. The specific aim of our study was to determine the antioxidant capacity and the content of phenolic compounds in leaves and twigs of different guayule lines cultivated in a Mediterranean environment. The antioxidant capacity and the contents of phenolic were affected significantly by guayule accession and harvest time. Overall means in twigs represented about 60% of the corresponding leaf values for antioxidant capacity as DPPH (1,1-diphenyl-2-picrylhydrazyl) and not tannic phenolic and about 55% as ABTS (2,2′-azinobis (3-ethylbenzothiazoline-6-sulphonic acid) diammonium salt), total phenolic, tannic phenolic and total flavonoid contents. Five individual phenolic compounds were identified in methanolic extracts of leaves. Neochlorogenic acid (62.5–174.8 mg g−1 DW) and chlorogenic acid (28.6–41.1 mg g−1 DW) were the most abundant phenolic acids. In addition to the compounds quantified in leaves, cynarin was identified only in twigs and for the first time in guayule biomass. Our results indicate that guayule leaves and twigs represent a rich source of antioxidants for potential applications in fodder, nutraceutical and pharmacological sectors.

Even though the methodology used for the determination ofβ-blockers andβ-agonists in environmental samples is based mainly on solid-phase extraction (SPE) and gas chromatography or liquid chromatography with mass spectrometric detection, the available literature data on the applied SPE procedures is rather sparse. In this paper such comparison is presented. Moreover, the usefulness of the eight SPE cartridges for the determination of fiveβ-blockers (acebutolol, atenolol, metoprolol, nadolol, and propranolol) and twoβ-agonists (salbutamol and terbutaline) in environmental aqueous samples using GC techniques is tested. Among them, three (the trifunction sorbent Strata Screen C, the copolymers LiChrolut EN, and the functionalized copolymer Isolute ENV+) were used for the first time for this purpose. It was confirmed that polystyrene-divinylbenzene-N-vinylpyrrolidone copolymers (PS-DVB-VP, Strata-X, and Oasis HLB cartridges) have a better potential than a cation-exchange sorbent for the extraction of the target drugs from environmental water samples. However, it should be stressed out that the direct application of the tested SPE conditions for the analysis of real environmental water samples is not possible, and such parameters, like volume of loading sample, appropriate solvents for washing and elution steps, and so forth, must be optimized again in order to achieve satisfactory recovery values for the target compounds.

 At the beginning of pandemic of SARS-CoV-2 children were thought to go through COVID 19 in  a definitely milder way than adults, in most cases even asymptomatically. However, even though the period of infection in pediatric patients is benign, SARS-CoV-2 found a way to take its toll on children’s health. Here comes PIMS-TS:  Paediatric inflammatory multisystem syndrome - temporally associated with SARS-CoV-2 (PIMS-TS) [ICD-10: U 10.9]. Since the global pandemic of COVID-19 is still on the go, for over a year now, SARS-CoV-2 tends to constantly mutate and affect children at increasingly young age either by oligosymptomatic respiratory viral infection, or more distant SARS-CoV-2-related medical condition- PIMS-TS. Paediatric inflammatory multisystem syndrome - temporally associated with SARS-CoV-2 (PIMS-TS), or Multisystem inflammatory syndrome in children (MIS-C) as it’s also called, is a rare systemic disease manifested by persistent fever and extreme inflammatory reaction following exposure to SARS-CoV-2, 2-4 weeks before the symptoms occur. It’s a brand new medical condition, described for the first time in May 2020. PIMS-TS affects 1 out of 1000 children infected with SARS-CoV-2 and can lead to very dangerous cardiological complications such as acute pancarditis, shock or aneurysms of coronary arteries. Efficient treatment performed in the accurate time and hospitalization in ICU allow to overcome PIMS-TS in the prevalent number of patients and get better in a few days. Mortality of PIMS-TS ranges from 1,5% to 2% of all cases.

AbstractThis work focuses on identifying the thermal conductivity of composites loaded with phase-change materials (PCMs). Three configurations are studied: (1) the PCMs are divided into identical spherical inclusions arranged in one plane, (2) the PCMs are inserted into the matrix as a plate on the level of the same plane of arrangement, and (3) the PCMs are divided into identical spherical inclusions arranged periodically in the whole matrix. The percentage PCM/matrix is fixed for all cases. A comparison among the various situations is made for the first time, thus providing a new idea on how to insert PCMs into composite matrices. The results show that the composite conductivity is the most important consideration in the first case, precisely when the arrangement plane is parallel with the flux and diagonal to the entry face. In the present work, we are interested in exploring the solid-solid PCMs. The PCM polyurethane and a wood matrix are particularly studied.

he existence of oil-water transition layer brings a great trouble to the dehydration of oil gathering and transportation system. It leads to raising the electric current of dehydrator and becoming worse of the deoiling and dehydrating properties of the treatment equipment, resulting in the serious influences on oil recovery. For the efficient treatment of the transition layer, it is necessary clearly to understand the structure and composition of the layer. In this paper, the physical structure and chemical composition of the layer were systematically, layer by layer and phase by phase, analyzed by modern instrumental methods The results show that (1)the layer is an emulsion which is composed of oil, water and suspended solids. The water phase has characteristics of weak alkaline,high salinity and viscous polymer. The oil phase contains many natural emulsifiers such as colloid, asphaltene and so on. The solid phase mainly concludes FeS particle which plays a decisive role in suspended solids; (2) the typical transition layer is composed of water and oil which accounts for above 90%, the content of solid impurity, which controls the emulsion of the layer, is less than 10%. Compared with oil phase, the water content of typical transition layer is larger with the density of 0.9~1.0 g/L and high freezing point. The analytical data can be adopted for the treatment of oil-water transition layer and smoothly run operations for oil gathering and transportation.

This study was conducted to develop a selective and sensitive method for the determination of bisphenol A (BPA) levels in milk and dairy products. A method based on solvent extraction with acetonitrile and solid-phase extraction (SPE) was developed for the analysis of BPA in milk, yogurt, cream, butter, pudding, condensed milk, and flavored milk, and a method using two SPE cartridges (OASIS HLB and Florisil cartridge) for skim milk was also developed. The developed methods showed good recovery levels (77 to 102%) together with low detection limits (1 μg/liter for milk, yogurt, pudding, condensed milk, flavored milk, and skim milk and 3 μg/liter for cream and butter). These methods are simple, sensitive, and suitable for the analysis of BPA in milk and dairy products. When 40 milk and dairy products were analyzed by the proposed methods, BPA was not identified in noncanned products, but its levels ranged from 21 to 43 μg/kg in canned products, levels that were 60- to 140-fold lower than the migration limits in the European Union and Japan.

Polyphenolic enriched extracts from two species of Cyperus, Cyperus glomeratus and Cyperus thunbergii, possess mammalian arginase inhibitory capacities, with the percentage inhibition ranging from 80% to 95% at 100 µg/mL and 40% to 64% at 10 µg/mL. Phytochemical investigation of these species led to the isolation and identification of two new natural stilbene oligomers named thunbergin A-B (1–2), together with three other stilbenes, trans-resveratrol (3), trans-scirpusin A (4), trans-cyperusphenol A (6), and two flavonoids, aureusidin (5) and luteolin (7), which were isolated for the first time from C.thunbergii and C. glomeratus. Structures were established on the basis of the spectroscopic data from MS and NMR experiments. The arginase inhibitory activity of compounds 1–7 was evaluated through an in vitro arginase inhibitory assay using purified liver bovine arginase. As a result, five compounds (1, 4–7) showed significant inhibition of arginase, with IC50 values between 17.6 and 60.6 µM, in the range of those of the natural arginase inhibitor piceatannol (12.6 µM). In addition, methanolic extract from Cyperus thunbergii exhibited an endothelium and NO-dependent vasorelaxant effect on thoracic aortic rings from rats and improved endothelial dysfunction in an adjuvant-induced arthritis rat model.

The article draws attention to the sociocultural aspect of the Siberian Railway Committee's activities in the early twentieth century. Historiographic analysis showed a research interest in the status of the Committee in the context of the organization of management of the Russian Empire's Eastern outskirts. Taking into account the broad powers of the Siberian Railway Committee, the author notes isolated studies of the social aspect in its activities. The aim of this publication is an attempt to create a holistic view of the activities of the Committee for the implementation of social tasks aimed at improving the life of railway employees at the TransBaikal section of the railway in the early twentieth century. The study was conducted on the basis of the documentation of the Siberian Railway Committee. A number of documents are introduced into scholarly discourse for the first time. Based on the structural and functional approach, using a set of historical research methods, it has been revealed that, after the commissioning of the Trans-Baikal section of the Siberian Railway, considerable attention was paid to solve sociocultural problems aimed at improving the life of railway employees. The preparatory commission chaired by A.N. Kulomzina and the Main School Committee implemented social programs. The author has defined the procedure for the formation of the committee, its structure, and principles of its activity. For the first time, personal data of the school committee's members elected on the Trans-Baikal Railway are introduced into scholarly discourse. The information of the committee's activities of the opening and maintenance of primary schools at railway stations has been summarized. The obtained statistics convincingly prove the effectiveness of the committee in the field of school education. The author notes that the Siberian Railway Committee achieved a very successful development of the school network by applying administrative and financial efforts. The author, for the first time, provides data on the organization of libraries and public convocations for the employees on the Trans-Baikal Railway. She draws attention to the organization of medical care for the employees; establishes the organization order and types of medical institutions; generalizes information about the staff of hospitals and obstetric centers, and the number of patients. The author concludes that the Siberian Railway Committee had an organized and balanced approach to solving sociocultural problems that occupied an important place in its activities. The Siberian Railway Committee's social programs in a number of areas were ahead of those of other government departments.

There has been increasing interest in the antidepressant effects of the muscarinic cholinergic receptor antagonist scopolamine. Here we assess, for the first time, whether a transdermal scopolamine patch is sufficient to induce changes in cognition that are consistent with the reported cognitive and antidepressant effects of scopolamine. A scopolamine or placebo patch was administered to healthy volunteers ( n=33) for 17 h in a double-blind, between-subject procedure. There was no clear effect of scopolamine patch on emotional cognition, verbal or working memory, suggesting that the effective dose of scopolamine available through the patch is too low to represent a viable antidepressant mechanism.

Azithromycin is an effective and well-known antimicrobial agent. In the present study, a simple, sensitive and specific LC/MS/MS method has been developed and validated for the quantification of Azithromycin in  human serum samples using Clarithromycin as internal standard. Azithromycin was extracted from biological matrix  by using solid phase extraction process. The chromatographic separation was performed on Luna C18 (3 ?, 2x150   mm) column with a mobile phase consisting of 35 mM ammonium acetate buffer (mobile phase-A) and acetonitrile  and methanol in ratio of 90:10 ( as mobile phase-B) at a flow rate of 0.25 mL/min. The method was validated over a  linear concentration range of 0.5?50.0 ng/mL and limit of quantification (LOQ) was 0.5 ng/mL with a coefficient of  correlation (r2) = 0.9998. The intra-day and inter-day precision expressed as relative standard deviation were 1.64% – 8.43% and 2.32% – 9.92%, respectively. The average recovery of azithromycin from serum was 98.11%. The method  was successfully applied to a pharmacokinetic study after oral administration of Azithromycin 200 mg/5 ml suspension in healthy Bangladeshi volunteers.
