In this study, we investigated the beneficial effects of milk biofortified with antioxidants on the health of children. Two experiments were conducted: experiment one evaluated the milk of 24 Jersey dairy cows (450 ± 25 kg of body weight (BW); 60 ± 30 days in milk dry matter intake (DIM)) given different diet treatments (CON = control diet; COANT = diet with vitamin E and selenium as antioxidants; OIL = diet with sunflower oil; and OANT = diet with sunflower oil containing more vitamin E and selenium as antioxidants), and experiment two evaluated the effect of the milk produced in the first experiment on the health of children (CON = control diet; COANT = diet with vitamin E and selenium as antioxidants; OIL = diet with sunflower oil; OANT = diet with sunflower oil containing more vitamin E and selenium as antioxidants; and SM = skim milk). One hundred children (8 to 10 years old) were evaluated in the second experiment. Blood samples were collected at 0 days of milk intake and 28 and 84 days after the start of milk intake. The cows fed the COANT and OANT diets showed greater selenium and vitamin E concentrations in their milk (p = 0.001), and the children who consumed the milk from those cows had higher concentrations of selenium and vitamin E in their blood (p = 0.001). The platelet (p = 0.001) and lymphocyte (p = 0.001) concentrations were increased in the blood of the children that consumed milk from cows fed the OANT diet compared to those in the children that consumed SM (p = 0.001). The children who consumed milk from cows fed the OIL diet treatment had increased concentrations of low density lipoprotein (LDL) and total cholesterol in their blood at the end of the supplementation period compared to children who consumed SM. The results of this study demonstrate that the consumption of biofortified milk increases the blood concentrations of selenium and vitamin E in children, which may be beneficial to their health.

Objective: To investigate the effect of antibiotics and/or probiotics on broiler performance, some serum metabolites, cecum microflora composition, and ileum histomorphology under the Egyptian conditions. Design: Randomized controlled experimental study. Animals: Two hundred forty 1-day-old Ross (308) chicks were reared till 35 days of age. Procedures: The birds were randomly allocated into four main groups: a control diet without additives (CON); probiotic (Lactobacillus acidophilus) supplemented diet (PRO); antibiotic (Avilamycin) supplemented diet (ANT) and a mix group (AP) that received antibiotic in the diet form 1 to 4 days of age and treated during the rest of the experimental period with probiotics. Results: Chickens fed on probiotic or antibiotic diets had linear improvement in live body weight (LBW) and feed conversion ratio (FCR) compared with the control group, while the best LBW and FCR were in the AP group. An improvement in the nutrient digestibility was observed in the probiotic added groups (PRO and AP). Serum cholesterol and low-density lipoprotein cholesterol contents decreased when antimicrobial (probiotic or antibiotic) supplementations were used, while there was an increase in high-density lipoprotein cholesterol contents, serum total protein, and albumin levels. Among all groups, cecum Clostridium perfringens and Escherichia coli counts decreased; however, there was an increase in Lactobacillus count compared to the control group. In probiotic supplemented groups (PRO and AP), a significant (P<0.05) improvement in ilea architecture. Conclusion and clinical relevance: Using probiotic after initial treatment with an antibiotic in broiler diets had a positive effect on broiler growth performance, gut health (improved cecum microbial populations and ileum histomorphology), and nutrient digestibility.

This study investigated the effect of clofibrate treatment on expression of target genes of peroxisome proliferator-activated receptor (PPAR)-α and various genes of the lipid metabolism in liver and adipose tissue of pigs. An experiment with 18 pigs was performed in which pigs were fed either a control diet or the same diet supplemented with 5 g clofibrate/kg for 28 days. Pigs treated with clofibrate had heavier livers, moderately increased mRNA concentrations of various PPAR-α target genes in liver and adipose tissue, a higher concentration of 3-hydroxybutyrate, and markedly lower concentrations of triglycerides and cholesterol in plasma and lipoproteins than control pigs ( P < 0.05). mRNA concentrations of sterol regulatory element-binding proteins (SREBP)-1 and -2, insulin-induced genes ( Insig) -1 and Insig-2, and the SREBP target genes acetyl-CoA carboxylase, 3-methyl-3-hydroxyglutaryl-CoA reductase, and low-density lipoprotein receptor in liver and adipose tissue and mRNA concentrations of apolipoproteins A-I, A-II, and C-III in the liver were not different between both groups of pigs. In conclusion, this study shows that clofibrate treatment activates PPAR-α in liver and adipose tissue and has a strong hypotriglyceridemic and hypocholesterolemic effect in pigs. The finding that mRNA concentrations of some proteins responsible for the hypolipidemic action of fibrates in humans were not altered suggests that there were certain differences in the mode of action compared with humans. It is also shown that PPAR-α activation by clofibrate does not affect hepatic expression of SREBP target genes involved in synthesis of triglycerides and cholesterol homeostasis in liver and adipose tissue of pigs.

Quercetin is a typical flavonoid present mostly as glycosides in plant foods; it has attracted much attention for its potential beneficial effects in disease prevention. In this study, we examined human volunteers after the short-term ingestion of onion, a vegetable rich in quercetin glucosides. The subjects were served diets containing onion slices (quercetin equivalent: 67.6–93.6 mg/day) with meals for 1 wk. Quercetin was only found in glucuronidase-sulfatase-treated plasma, and its concentration after 10 h of fasting increased from 0.04 ± 0.04 μM before the trial to 0.63 ± 0.72 μM after the 1-wk trial. The quercetin content in low-density lipoprotein (LDL) after glucuronidase-sulfatase treatment corresponded to <1% of the α-tocopherol content. Human LDL isolated from the plasma after the trial showed little improvement of its resistance to copper ion-induced oxidation. It is therefore concluded that conjugated metabolites of quercetin accumulate exclusively in human blood plasma in the concentration range of 10−7 ∼ 10−6 M after the short-term ingestion of vegetables rich in quercetin glucosides, although these metabolites are hardly incorporated into plasma LDL.

Penyakit jantung koroner (PJK) terutama disebabkan oleh aterosklerosis karena hiperkolesterolimia. Oksidasi low density lipoprotein (LDL) oleh reactive oxygen species (ROS) sebagai penyebab utama proses aterogenik dapat dicegah dengan kehadiran antioksidan seperti vitamin E. Buah dan sayuran banyak mengandung vitamin. Salah satu buah yang mengandung vitamin E adalah buah merah (Pandanus conoideus Lam.). Penelitian ini bertujuan untuk mengetahui pengaruh pemberian ekstrak buah merah terhadap kadar LDL darah pada tikus putih (Rattus norvegicus L.). Jenis penelitian ini adalah penelitian eksperimental laboratorium yang didesain mengikuti Rancangan Acak Lengkap (RAL) dengan 6 perlakuan dan 4 kali ulangan dengan parameter penelitian adalah kadar LDL darah tikus putih. Data dianalisis menggunakan one way anova dilanjutkan dengan uji Duncan 95%. Berdasarkan hasil penelitian, minyak buah merah memiliki kemampuan untuk menurunkan kadar LDL darah tikus putih.

This study was designed to investigate the prophylactic effect of aqueousextract of black currant concentrate on some biological markers related withheart disease in male rat treated H2O2. It also aimed at comparing theprophylactic effect of black currant concentrate to that of vit E.Forty adult male rats were divided randomly into equal groups (tenrat/group) & were treated as follows for 42 days. Rats in the first group (G І)were received normal water with oral intubation of sun flower oil 1ml /rat andconsider as control group. Animals of the second group (G Π) were received0.5% H2O2 in drinking water, while rats of the third group (G Ш) were received0.5% H2O2 in drinking water with oral intubation of vit E 400 I.U/Kg.B.Wdiluting in sun flower oil for each rat daily .While animals in the fourth group (GІV) were intubated daily 60mg/kg body weight of aqueous extract of blackcurrant concentrate plus 0.5% H2O2 in drinking water.Fasting blood samples were collected at 0, 21, and 42 days of experimentto study the following parameters:A - Platelet count (PC) and prothrombin time (PT).B- Serum concentration of total cholesterol TC, triacylglycerol TAG, Highdensity lipoprotein-cholesterol HDL-C, Low density lipoprotein-cholesterolLDL-C and Very low density lipoprotein- cholesterol VLDL-C of each groupwere measured. These parameters were regarded as biomarkers ofatherosclerosis and coronary heart disease (CHD). Abstract The components of biological variation in serum vitamin E in relation to serum cholesterol, triglycerides, high- and low-density lipoprotein cholesterol (HDL-C, LDL-C), apolipoprotein A-I (apo A-I), and apo B were examined in 26 healthy volunteers who had monthly blood samplings during one calendar year. The estimated CVs for vitamin E were: interindividual, 19.9%, and intraindividual, 11.9%; the index of individuality (I-index) was 0.59. The I-indices for all lipid variables were &lt; 0.51. Serum concentrations of vitamin E, cholesterol, triglycerides, HDL-C, LDL-C, and apo B were lower in spring than in the other seasons. The peak-trough differences in the yearly variations, expressed as a percentage of the mean, were for vitamin E 14.5%, cholesterol 16.2%, triglycerides 14.5%, and LDL-C 24.3%. A significant common annual rhythm was expressed in vitamin E or lipid variables and in the changes in ambient temperature the weeks before blood sampling (inverse relations). There were highly significant positive time relations between serum vitamin E and cholesterol, triglycerides, and apo B. Subjects with higher homeostatic setpoints of cholesterol showed higher homeostatic setpoints of vitamin E, triglycerides, LDL-C, and apo B.

Genetic, epidemiological and pharmacological data have led to the conclusion that antagonizing or inhibiting Proprotein convertase subtilisin/kexin type 9 (PCSK9) reduces cardiovascular events. This clinical outcome is mainly related to the pivotal role of PCSK9 in controlling low-density lipoprotein (LDL) cholesterol levels. The absence of oral and affordable anti-PCSK9 medications has limited the beneficial effects of this new therapeutic option. A possible breakthrough in this field may come from the discovery of new naturally occurring PCSK9 inhibitors as a starting point for the development of oral, small molecules, to be used in combination with statins in order to increase the percentage of patients reaching their LDL-cholesterol target levels. In the present review, we have summarized the current knowledge on natural compounds or extracts that have shown an inhibitory effect on PCSK9, either in experimental or clinical settings. When available, the pharmacodynamic and pharmacokinetic profiles of the listed compounds are described.

Atherosclerosis is associated with acute cardiovascular conditions, such as ischemic heart disease, myocardial infarction, and stroke, and is the leading cause of morbidity and mortality worldwide. Our understanding of atherosclerosis and the processes triggering its initiation is constantly improving, and, during the last few decades, many pathological processes related to this disease have been investigated in detail. For example, atherosclerosis has been considered to be a chronic inflammation triggered by the injury of the arterial wall. However, recent works showed that atherogenesis is a more complex process involving not only the immune system, but also resident cells of the vessel wall, genetic factors, altered hemodynamics, and changes in lipid metabolism. In this review, we focus on foam cells that are crucial for atherosclerosis lesion formation. It has been demonstrated that the formation of foam cells is induced by modified low-density lipoprotein (LDL). The beneficial effects of the majority of therapeutic strategies with generalized action, such as the use of anti-inflammatory drugs or antioxidants, were not confirmed by clinical studies. However, the experimental therapies targeting certain stages of atherosclerosis, among which are lipid accumulation, were shown to be more effective. This emphasizes the relevance of future detailed investigation of atherogenesis and the importance of new therapies development.
