Asymptomatic urinary tract infections were recovered from the urine of 40 of 100 rabbits, and identical bacteria were isolated when the rabbits were retested. Urine samples which yielded significant growths of bacteria also had pus cells. Some specimens yielded more than 2 different isolates, and staphylococci were the most frequently isolated bacteria. Oxalate and uric acid crystals were seen in 6% of the samples, particularly those with significant growths of Pseudomonas aeruginosa. No ova, parasites or fungal elements were seen in 'wet-mount' preparations and no fungi were seen when urine samples were cultured. Female rabbits showed a higher number of bacterial isolates than males. Animals with significant isolates were treated with suitable antibiotics and repeat samples yielded no growth.

Tube cystostomy is a surgical method used for managing obstructive urolithiasis and involves placement of a Foley catheter into the urinary bladder. We identified and evaluated the antibiotic resistance patterns of bacteria isolated from indwelling Foley catheters following tube cystostomy in goats with obstructive urolithiasis. Urine samples collected over a 10-y period from catheter tips at the time of removal were submitted for bacteriologic culture and antibiotic susceptibility testing. Resistance patterns to antibiotics, trends in the resistance patterns over the study period, and the probability of a bacterial isolate being resistant as a function of the identity of the isolate and antibiotic tested were determined. A total of 103 urine samples from 103 male goats with obstructive urolithiasis managed surgically with tube cystostomy were included in the study. Aerococcus (36.9%) and Enterococcus (30.1%) were isolated most frequently. The susceptibility patterns of all bacteria isolated did not change over the study period ( p > 0.05). Proportions of isolates resistant to 1, 2, and ≥3 antibiotics were 36.9%, 18.5%, and 23.3%, respectively. Thus, 41.8% of bacterial isolates were resistant to 2 or more antibiotics tested. The probability of Aerococcus spp., Escherichia coli, and Pseudomonas aeruginosa isolates to be resistant to ampicillin, ceftiofur, erythromycin, penicillin, or tetracycline ranged from 0.59 to 0.76.

Urinary tract infection (UTI) is one of the most common diseases encountered worldwide and is a major public health problem in terms of morbidity and financial costs. A cross-sectional study was conducted in Alka Hospital, Lalitpur, Nepal from February to July, 2014. A total of 353 midstream urine samples were collected from postmenopausal women visiting Alka Hospital, Lalitpur, Nepal. The samples were examined by microscopically and culture methods. The isolated organisms were identified by conventional microbiological methods. Antibiotic susceptibility test was performed by modified Kirby-Bauer disc diffusion method according to CLSI (2011) guidelines. Among 353 urine samples processed, 32 % (113) showed significant bacteriuria. Out of 113 bacterial isolates, the prevalence of gram negative bacteria was 97.3% (110) while that of gram positive was 2.7% (3). The most predominating organisms causing UTI were Escherichia coli (84.9%) followed by Klebsiella pneumoniae (5.3%), Enterococcus faecalis (1.7%), Providencia spp. (1.7%), Klebsiella oxytoca (1.7%), Proteus mirabilis (1.7%), Proteus vulgaris (0.9%), Citrobacter freundii (0.9%) and Staphylococcus aureus (0.9%). Antibiotic susceptibility tests of the isolates showed that, most of the gram negative bacterial isolates were sensitive to Piperacillin+Tazobactam followed by Amikacin, Imipenem and Nitrofurantoin, while they were resistant to Amoxicillin. All gram positive isolates were sensitive to Amoxicillin and resistant to Gentamycin. This study showed that higher aged postmenopausal women (>80 years) were at higher risk for UTI than those of lower aged ones (<80 years).

Objectives: The present study is aimed to determine frequency of bacteria in urine and pus sample along with the antibiotic resistance profile of isolated bacteria. Study Design: Cross-Sectional Study. Setting: Real-Time PCR Laboratory at Dabgari Garden, Peshawar with the Collaboration of Health Biotechnology Laboratory, Department of Biotechnology, Abdul Wali Khan University, Mardan. Period: June 2018 to July 2019. Material & Methods: The urine and pus samples were collected from suspected patients and were cultured on appropriate culture media. The biochemical tests were also performed after growing on culture plates for identification and confirmation of bacterial isolates. The disc diffusion technique was used to evaluate the antibiotic pattern of retrieved isolates. Result: A total of 525 samples of pus and urine samples were collected from different regions of Peshawar. Out of the total, 237 (45.1%) samples were found positive for bacterial growth whereas the remaining (54.9%) were observed negative. Among total positive isolates, 220 (92.8%) were from urine samples, and 17 (7.2%) were from pus samples. The predominant isolate was Escherichia coli (E. coli) (90.3%) retrieved from positive samples, followed by Pseudomonas aeruginosa (7.2%), Klebsiella pneumonia (1.3%), and Staphylococcus aureus (1.3%). The most effective antibiotic was Fosfomycin against bacteria whereas Nalidixic acid, Nitrofurantoin, and Amoxicillin/clavulanic acid were found less effective against bacterial isolates. Conclusion: Most frequent bacteria isolated was E. coli and the most efficient drug was Fosmomycin and the least was Nitrofurantoin, and Amoxicillin.

Sickle cell anaemia (SCA) is an inherited disease resulting from mutations in the β-globin chain of adult haemoglobin that results in the formation of homozygous sickle haemoglobin. It is associated with several complications including an altered blood picture and damage in multiple organs, including the kidneys. Kidney disease is seen in most patients with SCA and may affect glomerular and/or tubular function, thereby putting these patients at risk of urinary tract infections. However, there is a paucity of data on the prevalence of urinary tract infections (UTIs) among SCA patients in Zambia. This study aimed to determine the prevalence of UTIs and haematological and kidney function profiles among SCA patients at the University Teaching Hospitals, Lusaka, Zambia. This was a cross-sectional study conducted between April and July 2019 involving 78 SCA patients who presented at the UTH. Blood and midstream urine samples were collected from each participant using the standard specimen collection procedures. Full blood counts and kidney function tests were determined using Sysmex XT-4000i haematology analyser and the Pentra C200 by Horiba, respectively. Bacterial profiles of the urine samples were determined using conventional microbiological methods. We found that all the measured patients’ haemoglobin (Hb) levels fell below the WHO-recommended reference range with a minimum of 5 g/dl, a maximum of 10.5 g/dl, and a mean of 8 ± 1 g/dl. Fifty percent of the participants had moderate anaemia, while the other 50% had severe anaemia. The minimum WBC count of the participants was 0.02 × 109/L with a maximum of 23.36 × 109/L and a mean of 13.48 ± 3.87 × 109/L. Using the one-way analysis of variance test, we found no significant difference in mean WBC count and Hb concentration across various age-group categories that we defined. Bacteriuria was found in 25% of participants. The most common bacterial isolates were Staphylococcus aureus (32%) and coagulase-negative Staphylococci (32%). Klebsiella pneumoniae was 16%. We found no significant association between bacterial isolates and white blood cell count, age groups, sex, and anaemia severity p=0.41. None of the participants were diagnosed with kidney disease. There was a high prevalence of asymptomatic UTIs among SCA patients at UTH, which, when coupled with the marked leukocytosis and anaemia, may negatively impact the clinical outcome of the patients. Therefore, we recommend close monitoring of sickle cell patients in Zambia for such conditions to improve patients’ outcomes.

Objective: This study aimed to investigate the correlation between ureR and ureC genes with the production of urease by Proteus mirabilis.Methods: A total of 450 mid-stream urine samples have been collected from patients with urinary tract infection whom admitted to the hospitals in Annajaf Al-Ashraf province for consultancy during the period from October 2015 to February 2016.Out of 150 bacterial isolates, only 29 isolates were belong to P. mirabilis according to conventional methods (depending on microscopic and culturing examination as well as biochemical test) and molecular technique using 16SrRNA gene.Results: The results of phenotypic and genotypic detection of urease in P. mirabilis showed that all isolates were able to produce urease and possess ureR and ureC that encodes to urease by appearing of amplicon with molecular weight 359 and 533 bp, respectively, when electrophoresed on 1% agarose gel.Conclusion: A correlation has been found between ureR and ureC genes with the production of urease by P. mirabilis.

Objectives: The study was aimed to determine the prevalence of Extended Spectrum Beta Lactamase (ESBL) producing Gram negative pathogens from urine samples along with their antimicrobial resistance. Methods: This cross-sectional study was conducted from December 2015 to May 2016 at Everest Hospital, Kathmandu. Mid-stream urine samples were collected and processed for culture by standard loop streak method. Identified bacterial isolates were tested for Antibiotic Susceptibility by modified Kirby Bauer disc diffusion method and, were subjected to ESBL screening by using 30µg cefotaxime and ceftazidime. ESBL production was confirmed by combination disc method. Results: Of the three hundred urine samples, 22.7% (67/300) showed significant growth. Four different bacterial species were identified. Among the isolates, E. coli was the most common pathogen (71.64%) followed by Klebsiella pneumoniae (14.92%), Pseudomonas spp (8.95%) and Acinetobacter spp (4.48%). Altogether 92.54% (n=62) isolates were sensitive to gentamicin, 89.55% (n=60) to amikacin, and 79.10% (n=53) to nitrofurantoin. 70.10% (n=47) isolates were resistant to antibiotic ampicillin while 62.68% (n=42) were found as Multi-Drug Resistant (MDR) and 29.8% (n=20) were ESBL producers. Conclusions: The overall prevalence of MDR and ESBL among uropathogens is low in comparison to other studies though it is essential to have a regular monitoring of ESBL producing clinical isolates in laboratory practice.
