Telah dilakukan penelitian untuk menganalisis kandungan metabolit sekunder dan efektivitas antioksidan dari ektrak etanol bunga kasumba turate (Carthamus tinctorius L.), serta potensinya sebagai antihiperkolesterolemia. Bunga kasumba turate yang telah dikeringanginkan, dihaluskan dan dimaserasi dengan pelarut etanol 70%, lalu dievaporasi pelarutnya. Ekstrak etanol (EE) yang diperoleh dipartisi berturut-turut dengan pelarut n-heksana, etil asetat dan air, sehingga diperoleh ekstrak fraksi n-heksana (FH), etil asetat (FEA) dan air (FA). Selanjutnya, EE, FH, FEA, dan FA diuji kandungan metabolit sekundernya (metode Harborne) dan efektivitas antioksidannya (metode DPPH). Hasil penelitian menunjukkan bahwa EE dan FEA mengandung alkaloid, fenolik, flavonoid, saponin, tanin dan triterpenoid. FA mengandung fenolik, flavonoid, saponin, tanin dan triterpenoid. FH mengandung alkaloid, flavonoid dan triterpenoid. Efektivitas antioksidan bunga kasumba turate (dinyatakan dalam IC50) yang tertinggi pada FEA, diikuti FH, FA dan EE, dengan nilai IC50 53,59, 75,45, 77,43, dan 89.,9 µg/mL. Hasil kajian menunjukkan bahwa bunga kasumba turate dapat menghambat oksidasi LDL dan menurunkan kadar kolesterol darah. The ethanol extract (EE) obtained was partitioned successively with n-hexane, ethyl acetate and water as solvents, so that the extracts of the n-hexane (FH), ethyl acetate (FEA) and water (FA) fractions were obtained. Furthermore, EE, FH, FEA, and FA were tested for their phytochemical content (Harbourne method) and antioxidant effectiveness (DPPH method). The results showed that EE and FEA contained alkaloids, phenolics, flavonoids, saponins, tannins and triterpenoids. FA contains phenolics, flavonoids, saponins, tannins and triterpenoids. FH contains alkaloids, flavonoids and triterpenoids. The antioxidant effectiveness of casumba turate flower (expressed in IC50) was highest in FEA, followed by FH, FA and EE, with IC50 values of 53,59, 75,45, 77,43, and 89,19 g/mL, respectively. The results of the literature review show that kasumba turate flowers can inhibit LDL oxidation and reduce blood cholesterol levels. This study concluded that kasumba turate flower extract has a strong antioxidant effectiveness and antihypercholesterolemic effect.Keywords: Antihypercholesterolemia; antioxidant; Carthamus tinctorius L.; ethanol

Characterization of antioxidant compounds from the seeds of basil (Ocimum basilicum L.) has been done. Extraction is done by maceration method using ethanol solvent, fractionation by TLC and column chromatography, antioxidants test using DPPH method, and characterization of antioxidant compound using GCMS. Ethanol extract and results of fractionation ethanol extract of basil seeds using n-hexane, ethyl acetate, n-butanol and methanol-water extracts show that n-butanol extract has the highest antioxidant activity with IC50 values ​​of 41.90 ppm. Results of column chromatography n-butanol extract using n-hexane : ethyl acetate (1:9) as mobile phase yielded 5 fractions with fraction 4 (F4 isolate) has dominant stain of active antioxidants after being sprayed DPPH reagent, it had IC50 values ​​of 39,70 ppm and total phenolic content of 0,003 mg/g. Isolate F4 suspected contains two active compounds as antioxidant which is terpenoid and phenolic compound group, namely squalene and 1,4-di-tert-buthyl-phenol identified by GCMS.DOI :http://dx.doi.org/10.15408/jkv.v0i0.3598

ABSTRACTKersen (Muntingia calabura L.) is a plant that has begun to be eliminated and was rarely used because it is often considered to have no economic value and lack of knowledge about its utilization, whereas kersen plants contain flavonoids, saponins, and tannins which were have high benefit for health. The content of metabolites is affected by soil nutrient elements and difference place of growth. This study aims to determine the potential of kersen leaves grown in North Minahasa based on phytochemical content, ability of antioxidant activity, and toxicity. Kersen leaves were extracted using sequential maceration method with n-hexane, ethyl acetate, and ethanol as solvents. Phytochemical Screening using several reagents which tailored to the type of phytochemical test. 1.1-diphenyl-2-picrylhydrazyl (DPPH) method is used to evaluate antioxidant activity, and Brine Shrimp Lethality Test (BSLT) method is used to evaluate toxicity. The result of this study indicate that the n-hexane extract contains phenols, flavonoids, and tannins, with IC50 value 12.54 μg/mL, and LC50 value 881 μg/mL. Ethyl acetate extract contains phenols, flavonoids, tannins, and saponins, with IC50 value 61.3 μg/mL, and LC50 value 1758 μg/mL. Penelitian ini bertujuan untuk mengetahui potensi dari daun kersen yang tumbuh di Minahasa Utara berdasarkan kandungan fitokimia, kemampuan aktivitas antioksidan, dan toksisitasnya. Ekstrak daun kersen diekstraksi dengan metode maserasi sekuensial menggunakan pelarut n-heksan, etil asetat, dan etanol.  Skrining fitokimia menggunakan beberapa reagen yang disesuaikan dengan jenis uji fitokimia. Metode 1.1-diphenyl-2-picrylhydrazyl (DPPH) digunakan untuk mengevaluasi aktivitas antioksidan, dan metode Brine Shrimp Lethality Test (BSLT) digunakan untuk mengevaluasi toksisitas. ABSTRACTDayak onion plant (Eleutherine americana Merr.) or commonly called forest onion is a typical plant of Central Kalimantan that has been used by local peoples for cure a various types of diseases. This study aims to determine the activity of antioxidant compounds from Dayak bulb ethanol extracts from Kotamobagu City. This research method is an experimental laboratory. Initially the Dayak bulb sample was extracted maceration with 96% ethanol solvent then evaporated until a thick extract was obtained. As a parameter, the DPPH method (1,1-diphenyl-2-picrylhydrazyl) was used to test the antioxidant activity of the samples at concentrations of 50, 60, 70, 80 and 90 mg / L and as a comparison standard used Vitamin C with concentrations of 2, 4, 6, 8 and 10 mg / L. The results obtained showed antioxidant activity in Dayak bulb ethanol extracts with IC50 values of 41.46 mg / L and on vitamin C IC50 values of 1.04 mg / L. Based on these results it can be concluded that the onion bulb ethanol extract has very strong antioxidant activity. Keywords: Dayak Onion bulbs, Activity, Antioxidant, DPPH  ABSTRAK Tanaman bawang Dayak (Eleutherine americana Merr.) atau biasa disebut bawang hutan merupakan tanaman khas Kalimantan Tengah yang sudah dipergunakan masyarakat lokal sebagai obat berbagai jenis penyakit. Penelitian ini bertujuan untuk mengetahui aktivitas senyawa antioksidan dari ekstrak etanol bulbus bawang Dayak yang berasal dari Kota Kotamobagu. Metode penelitian ini adalah eksperimental laboratorium. Awalnya sampel bulbus bawang Dayak diekstraksi maserasi dengan pelarut etanol 96% kemudian dievaporasi hingga didapatkan ekstrak kental. Sebagai parameter, digunakan metode DPPH (1,1-diphenyl-2-picrylhydrazyl) untuk pengujian aktivitas antioksidan sampel pada konsentrasi 50, 60, 70, 80 dan 90 mg/L dan sebagai standar pembanding digunakan Vitamin C dengan konsentrasi 2, 4, 6, 8 dan 10 mg/L. Hasil penelitian yang didapat menunjukkan aktivitas antioksidan pada ekstrak etanol bulbus bawang Dayak dengan nilai IC50 sebesar 41,46 mg/L dan pada vitamin C nilai IC50 sebesar 1,04 mg/L. Berdasarkan hasil tersebut dapat disimpulkan bahwa ekstrak etanol bulbus bawang Dayak memiliki aktivitas antioksidan yang sangat kuat. Kata kunci: Bulbus Bawang Dayak, Aktivitas, Antioksidan, DPPH

Antioxidants are substances that can slow down the oxidation process of free radicals. Limpasu plant (Baccaurea lanceolata (Miq) Muell. Arg), an indigenous plant of Borneo, is a natural antioxidant source. The purpose of this study was to determine the antioxidant activity of the limpasu pericarpium extract. The extraction of the limpasu pericarpium was done by maceration method using solvents with increasing polarity ranging from n-hexane, ethyl acetate, and methanol. Antioxidant activities of the three extracts were measured by the DPPH and FRAP methods. The IC50 values of n-hexane, ethyl acetate, and methanol extracts, as well as quercetin using the DPPH method were 517,45 µg/mL, 530,64 µg/mL, 10,63 µg/mL and 6,83 µg/mL, respectively. Meanwhile, the IC50 values obtained from FRAP method were 198,96 µg/mL, 190,07 µg/mL, 661,36 µg/mL, and 7,09 µg/mL, respectively. The results revealed that the methanol extract is more potent than other extracts tested for antioxidant activity.

Objective: The aim of this study is to investigate antioxidant activity and phytochemical screening of ethanol extract, fractions of water, ethyl acetate, and n-hexane from mistletoe tea (Scurrula atropurpurea Bl. Dans).Methods: Simplicia extracted using soxhlet equipment with 96% ethanol. Fractionation was conducted using liquid-liquid extraction using a solvent of water, ethyl acetate and n-hexane. Screening of phytochemical and antioxidant activity was performed against these fractions. Antioxidant activity was determined by 2,2-diphenyl-1-picrylhydrazyl method using ultraviolet-visible spectrophotometry with ascorbic acid as standard. Phytochemical screening was conducted based on the method of Farnsworth.Results: The IC50 values of ethanol extract, water fraction, fraction of ethyl acetate, and n-hexane fraction were 21.92 ppm, 89.57 ppm, 14.08 ppm, and 162.09 ppm, respectively, whereas for ascorbic acid was 4.41 ppm. The ethanol extract and ethyl acetate fraction contained compounds were the same group, polyphenolic, tannins, flavonoids, monoterpenoid, steroids, triterpenoids, and quinones. Fraction of water contained compounds such as polyphenolic group, flavonoids, monoterpenoids, sesquiterpenoids, steroids, and triterpenoids. n-hexane fraction compounds contained steroids and triterpenoids.Conclusion: The ethanol extract, water fraction, ethyl acetate fraction, and n-hexane fraction showed antioxidant activities. Antibacterial activity of aqueous and solvent extracts of E. cotinifolia leaves were tested against some human pathogenic bacteria viz. Escherichia coli, Klebsiella pneumonia, Bacillus subtilis, Bacillus cereus, Salmonella typhi, Enterobacter aerogenes and Staphylococcus aureus by agar cup diffusion and broth microdilution methods. Antioxidant properties were evaluated for different solvent extracts by diphenyl picryl hydrazyl (DPPH), nitric oxide (NO) and hydrogen peroxide methods and IC50 values were calculated and compared with the standard ascorbic acid and butylated hydroxyanisole. Among the aqueous and organic solvent extracts, methanol and ethyl acetate, showed significant activity against B. subtilis and E. aerogenes which recorded a maximum inhibition zone of 17.25 mm. Minimum inhibitory concentration of methanol and ethyl acetate extracts for different bacteria ranged from 0.3- 1.25 mg/mL. In DPPH method, IC50 values of chloroform, petroleum ether, ethyl acetate and methanol were found to be 15, 17, 18 and 19 mg/mL, respectively, lesser than the standard, ascorbic acid (25 mg/mL). Phytochemical analysis of aqueous, ethyl acetate and methanol extract showed the presence of flavonoids, terpenoids, tannins and steroids. Further work is in progress to isolate the active compound(s).

Research on the antioxidant activity of the local onion roots of Palu (Allium cepa Var Aggregatum L.) with various polarity solvents has been carried out. This study aims to determine the potential of Palu local shallot root extract as an antioxidant and determine the IC50 value of the Palu local shallot root antioxidant activity. The method applied in this research is maceration using n-hexane, ethyl acetate, and ethanol. The extracts from each solvent were tested for their antioxidant activity by the DPPH method. The results of the study showed that the IC50 value in n-hexane extract, ethyl acetate extract, and ethanol extract were 579.98 ppm, 518.28 ppm 315.83 ppm and as a comparison used ascorbic acid obtained IC50 of 53.69 ppm. Based on the results obtained, the best antioxidant activity is in polar solvents (ethanol extract) compared to other solvents. This study aimed to investigate phytochemical screening and antioxidant activity of n–hexane, ethyl acetate and ethanol extract from lakoocha leaves. The powdered simplicia was macerated with n–hexane, ethyl acetate and ethanol 96% successively, filtered, then concentrated using rotary evaporator to obtain n–hexane extract, ethyl acetate extract and ethanol extract. Phytochemical screening and antioxidant activity was performed against these extracts. Antioxidant activity was determined by DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging method using ultraviolet-visible spectrophotometer at wavelength of 516 nm after incubated for 60 minutes in dark place. Quercetin was used as positive control. The result of phytochemical screening showed n-hexane extract contains steroid, ethyl acetate extract contain steroid, tannin, glycoside, flavonoid and saponin, whereas ethanol extract contain tannin, glycoside, flavonoid and saponin. The IC50 value of n–hexane, ethyl acetate and ethanol extract was 1062.03±1.42 ppm, 323.18±0.02 ppm and 99.23±0.07 ppm respectively, whereas for quercetin was 2.32±0.01 ppm. This study showed that ethanol extract had antioxidant activity with strong category whereas n-hexane extract and ethyl acetate extract had inactive antioxidant activity with very weak categories.       Keyword: Antioxidant Activity, DPPH, Lakoocha leaf

ABSTRAK Penelitian ini dilakukan untuk mengetahui senyawa aktif yang terkandung melalui pengujian fitokimia dan aktivitas antioksidan pada tumbuhan obat di pulau Tidore. Uji aktivitas antioksidan menggunakan metode DPPH dilanjutkan dengan perhitungan Inhibition Concentration 50% (IC50). Hasil yang diperoleh adalah biji buah mojoi terkandung senyawa alkaloid dan saponin, buah coro terkandung alkaloid, flavonoid, dan saponin, pada daun ofo terkandung  alkaloid, tanin, flavonoid, steroid, dan saponin dan pada rimpang kuso mafola terkandung alkaloid, tanin, flavonoid, steroid, dan saponin. Nilai IC50 sebagai berikut biji rimpang kuso mafola 37,30 ppm, buah coro 250,17 ppm, daun ofo 976,10 ppm  dan buah mojoi 1001, 07 ppm. Kata kunci :Tumbuhan  obat, DPPH, antioksidan dan uji fitokimia. In the test results to the phytochemical , that mojoi fruit seeds contained alkaloids and saponins, fruit coro (alkaloids, flavonoids, and saponins), ofo leaves (alkaloids, tannins, flavonoids, steroids, and saponins) and ethanol extract of rhizome kusomafola (alkaloids, tannins, flavonoids, steroids, and saponins. In  calculation of IC50 values ​​for rhizome kusomafola 37.30 ppm, 250.17 ppm coro fruit, 976.10 ppm ofo leaf extract, and fruit seed extract mojoi 1001.07 ppm. The study about antioxidant activity test of coffee parasite leaves (Loranthus ferrugineus Roxb.) by 1,1-diphenyl-2-pikrilhidrazyl (DPPH) method has been done. The aim of this study was to determine the content and activity of secondary metabolites, flavonoids and antioxidant, which found in the coffee parasite leaves. Dry powder of parasite coffee leaves (Loranthus ferrugineus Roxb.) as much as 0.5 kg were macerated by 2 L of ethanol solvent at room temperature for 4 x 24 hours, then mixed and filtered. Ethanol filtrate was evaporated at 30-40°C by using a rotary evaporator to obtain the crude extract of coffee parasite leaves. The results of the phytochemical screening showed positively that the extract was containing alkaloid, flavonoids, saponins, tannins and steroid compounds.The results of the antioxidant activity test by using DPPH method was obtained that the amount of antioxidant activity of the samples of ethanol extract had a very strong antioxidant activity against radical DPPH 0.05 mM, with IC50 values was obtained 6.063 ppm. Whereas, for comparison of ascorbic acid was about 3.127 ppm.
