The objective of research was to isolate the probiotic bacteria from intestine of carp.  Isolation was performed by weighing 1 g of intestine, crushed and inserted in a reaction tube containing 9 mL of NaCl then centrifuged. Supernatant was taken and spread over the agar medium by scatter method. The bacteria-stocked medium was then incubated in an incubator at 28°C for 24-48 hours. The growing colonies were re-isolated on the agar medium to obtain pure colonies. The bacteria were then identified by morphological observation, resulting pigment and gram staining. Five nile tilapia (sized 8 – 10 cm) were used in pathogenicity test and placed in the aquarium. Fish were injected with 0.2 ml of Lactobacillus solution containing 1x107 CFU/mL via intraperitoneal injection. The results showed that the bacteria Lactobacillus sp had rod-shaped characteristics, white pigment and gram positive and  not pathogenic, which means it can be used as probiotic for aquaculture.Keywords : Lactobacillus sp, carp, probiotic, pathogenicity test

The objective of this research was to obtain probiotic bacteria isolated from the digestive tract of catfish Clarias batracus, to evaluate its effect on growth performances and survival rate of carp. After purified, the bacteria was identified through biochemical tests and selection was carried out through proteolytic, amylolytic, cellulolytic, tests as well as pathogenicity test. Probiotic candidate was administered orally to fish at different concentrations. The fish were fed for 30 days respectively as much as 3% of body weight per day with feeding frequency of two times a day. The results showed that addition of probiotics into feed significantly increased growth performance but not survival rate.Keywords: Probiotic, Clarias batracus,  growth performance, aquaculture. ABSTRAKTujuan dari penelitian ini adalah untuk mendapatkan bakteri probiotik yang diisolasi dari saluran pencernaan ikan lele Clarias batracus, dan mengevaluasi pengaruhnya terhadap kinerja pertumbuhan dan kelangsungan hidup benih ikan mas. Bakteri probiotik diidentifikasi melalui uji biokimia dan seleksi dilakukan melalui proteolitik, amilolitik, tes selulolitik dan patogenisitas. Kandidat probiotik diberikan pada ikan secara oral dengan konsentrasi berbeda Pemberian pakan berprobiotik berlangsung selama 30 hari sebanyak  3% dari berat badan per hari dengan frekuensi pemberian dua kali sehari. Hasil penelitian menunjukkan bahwa penambahan probiotik ke dalam pakan secara signifikan meningkatkan kinerja pertumbuhan namun tidak memengaruhi kelangsungan hidup benih ikan mas.Kata kunci : Probiotik, Clarias batracus, kinerja pertumbuhan, budidaya

The study aimed to isolate probiotic bacteria from the digestive organs of goldfish taken from Fish Hatchery, Tateli.  As much as 1 g of the intestine was crushed, put in 9 mL NaCl and centrifuged twice at 1000 rpm for 10 minutes. The supernatant was removed and diluted at a concentration of 10-2 - 10-3, then spread on the surface of the MRS media. The bacteria were incubated at 280C for 24-48 hours in the Incubator. The Gram Stain test found that the isolated bacterium was gram positive, biochemical tests found that bacterium isolated from the gut of carp was Lactobacillus sp. while pathogenicity test found that there was no mortality of fish until the 14th day after the challenge test.  So that it could be concluded that the bacteria obtained could be used as probiotics.Keywords:  probiotic, Lactobacillus sp., Gram Stain, pathogenesity

The study aims to isolate, characterize, and examine probiotic bacteria's inhibitory ability against Vibrio harveyi bacteria, both in-vitro and in vivo. Methods used in the study consist of 1) An Isolation of Candidate Probiotic Bacteria, 2) An Antagonistic Test of Candidate Probiotic Bacteria in vitro, 3) An Identification of Bacteria, 4) A Pathogenicity Test of Candidate Probiotic Bacteria, 5) An Antagonistic Test of Candidate Probiotic Bacteria against V. harveyi in vivo. According to the isolation of candidate probiotic bacteria, there are 18 isolated candidate probiotic. After being tested for its inhibitory ability in vitro, there are 8 isolates with zone of inhibition as follows: isolate MM 7 from intestine (22 mm), isolate MM 6 from intestine (12 mm), isolate MM 10 from sea water (10 mm), isolate MM 5 from intestine (9 mm), isolate MM 4 from intestine (8 mm), isolate MM 3 from intestine (7 mm), isolate MM 2.2 from intestine (7 mm), isolate MM 2.1 from intestine (7 mm). Eight genera of the candidate probiotic bacteria is derived from Portunid crab, they are Staphylococcus, Streptococcus, bacillus, vibrio, Alcaligenes, Lactobacillus, micrococcus. Before proceeding the V. harveyi bacterial challenge test in vivo, three potential isolates consisting of MM6, MM7 and MM10 as the probiotic bacteria are pathogenicity-tested against V. harveyi. The survival rate of Portunid crab on pathogenicity test using MM6, MM7 and MM10 generates 91.11-100%, while the control generates 100% survival rate. Variance analysis result through post-hoc Tukey's Honest Significant Difference (HSD) test at 95% confidence interval indicates that isolate MM7 and MM10 are significantly able to increase hatchling Portunid crab's survival rate.

The purpose of this study was to detect the presence of bacteria Aeromonas sp in tilapia (Oreochromis niloticus). Nine fish with an average body weight of 150-200 g were used in this study as samples. Sampling was conducted at three different locations which were representative of fish culture sites located on Lake Tondano. The number of sample gathered from each location was and 3 fish which were taken randomly. Fish samples were taken by using a scoop, then the samples were taken alive by placing in an oxygen-filled plastic separately and taken direcly to lab Fish Disease Control and Environmental Center, Tateli, Department of Marine and Fisheries of North Sulawesi province, for examination the presence of bacteria. Isolation of bacteria were conducted by taking samples from gill and kidney. This study was carried out from April - July 2013. Identification of the presence of bacteria was done through a series of observations of colony morphology and gram staining of bacteria, followed by a series of biochemical tests: oxidase test , catalase test , test TSIA , H2S production test , indole test , motility test , citrate test, and test O/F. It could be concluded that 22.22 % of farmed nile tilapia in Lake Tondano were infected with the Aeromonas sp with the following percentage from each site: Paleloan village 7.40% , 7.40% Toulimembet village, and 7.40% Eris village. A preliminary morphological screening and isolation of bacterial colony from latex industrial wastewater was carried out. Bacteria colonies from latex processing wastewater were isolated from a local latex processing industry. It was found that 17 bacterial isolates had been purified grown on nutrient agar under 35˚C. The colonies were then purified and morphologically indicated via Gram staining and motility test. After morphological observation, it was identified that out of 17 isolates, 9 isolates were Gram positive and 8 isolates were Gram negative. There are 11 out of 17 colonies were rod-shaped bacterial colonies, while the other 6 colonies were cocci-shaped bacteria. There were 11 colonies of gliding bacteria, three colonies were non-motile bacteria and the other three colonies were flagellated bacteria. This study is only limited to morphological observation as the main aim of this study was to investigate the potential occurrence of viable growth in treated latex processing wastewater. The bacterial colonies were classified base on their morphological properties shown. This study has classified several genera such as Staphylococcus, Escherichia, Thiobacillus, Arthrobacter and other Genus. The growth curve of 17 isolates studied and the chemical oxygen demand were determined.

<p>The use of probiotic bacteria is one of the methods in aquaculture to control infectious diseases. This study aims to obtain candidates probiotic bacteria from the coral reef and tiger grouper aquaculture environment (<em>Epinephelus fuscoguttatus</em>). Steps of the study included of: 1) isolation of candidates probiotic bacteria, 2) selection of candidates probiotic bacteria: inhibition test and co-culture methods, 3) enzymatic test, 4) the growth of bacteria test 5) pathogenicity test and 6) the molecular identification of probiotic bacteria. The results showed that of 124 isolates in test zone of inhibition, the four isolates with the widest zone of inhibition of isolate K7, K8, K21, and T4, a total of 51 isolates on co-culture methods, the four isolates were able to suppress the growth of pathogenic <em>Vibrio</em> bacteria, isolates K15, K68, K55, and T36. The results of hydrolyzing the highest protease amylase and lipase enzyme, respectively isolates K15 and T36; isolates T41 and K8; isolates K55 and K68. Candidates probiotic bacteria reached the exponential phase at 16‒18 h. The results of pathogenicity test isolate K7, K8, T36, and T41 provided the highest survival that can be used as candidates probiotics in the tiger grouper.</p> <p>Keywords: selection, probiotics, tiger grouper</p>

<p>In the present study, microbiological quality of biscuits from street side bakery shops at Dehardun city, India was conducted. A total 11 samples of bakery biscuits were collected randomly and analysed for their microbiologically quality by standard plate count method. Pure cultures of bacterial isolates were prepared by streaking on nutrient agar medium. Bacterial isolates were further studied for morphological characters, culture characters on nutrient agar medium and biochemical testes in laboratory. All samples studies were found contaminated by a variety of bacteria. The highest bacterial load was 2.2 ×105 cfu/g and lowest was 0.1×103 cfu/g, respectively. Morphological observation, culture characters and results of biochemical tests of bacterial isolates were compared with standard results of known bacteria. The bacterial isolated were identified as Alcaligenes faecalis, Bacillus cereus, Micrococcus luteus, Pseudomonas aeruginosa, Streptococcus lactis and Staphylococcus aureus, respectively. Among these, two isolates i.e., A. faecalis and P. aeruginosa were gram negative and other isolates were gram positive bacteria which suggested the poor hygienic conditions inside the bakery during preparation and storage of biscuits. Gram positive bacteria might be contaminated during storage and by bad handling. These food spoiling bacteria might be responsible for food-borne infection and diseases. Therefore, care should be taken while purchasing such products and should be purchased from certified bakery shop.</p>

This study was described for the nature of the pathpgenesis of bacteria Pasteurella multocida which was isolated from infected man made comparison between these bacteria and those from infected farm animals. The percentage of Pasteurella multcida diagnosed bacteria from animals and human was 29.4% and 16.9% respectively. Comparing to other culture media Pasteurella multocida selective agar medium was characterized by its selectivity and sensitivity and then was attempt for biotyping species and subspecies of isolated Pasteurella from animals and human samples were successfully achieved. Pathogenicity test was performed on mice, only nine human isolatetes and twenty-one animal isolates from Pasteurella multocida were virulent. Todistinguish between the pathogenesis of human and animal isolates, one isolated from human and animal were chosed, in addition to the standared strain. The mice had been experimentally infected by three different ways, I/P, I/T, I/Eye. The results were showed that Pasteurella multocida can produce lesions as fibrinous suppurative pneumonia in lungs, liver and spleen which were detected histopatho logically. However the animal isolates were more virulent than human or standared strain.
