INTESTINAL MICROFLORA ON THE BACKGROUND OF BACTERIAL VAGINOSIS ON VULVOVAGINAL CANDIDIASIS IN OVERWEIGHT AND OBESE WOMEN

The aim of our work was to study the intestinal microbiome in obese women and further develop differentiated patterns of exposure to the intestinal microbiota to improve metabolism and reduce excess weight. Materials and methods: Surveyed 120 overweight and obese women. For comparison, 60 women without overweight and obesity were examined. Group I – women with vulvovaginal candidiasis and overweight and obesity (60 women); group II – women with vulvovaginal candidiasis without overweight and obesity (30 women); group III – women with bacterial vaginosis and overweight and obesity (60 women); ІV – women with bacterial vaginosis without overweight and obesity (30 women). The study of the intestinal microbiocenosis included the determination of the species and quantitative composition of the microflora. Quantitative indicators of intestinal microflora were studied by seeding 1 ml from each dilution on differential diagnostic media: Endo, Ploskireva, ICA (bismuth – agar sulfide) to detect pathogenic enterobacteria; ZhSA (yellow – salt agar) for determination of staphylococci. To study the hemolytic activity of bacteria was used agar with 5% erythrocyte content of sheep. The presence of bifidobacteria in the test material was studied on Blaurok medium, and lactobacilli – on MRS medium. Statistical analysis of the obtained research results was performed using standard computer packages “Data Analysis” Microsoft Excel for Windows 2002. Results: Based on the results of studies, we can summarize the nature of changes in the intestinal microbiome on the background of vulvovaginal candidiasis and bacterial vaginosis in women with overweight and obesity. In all women with excess body weight and obesity, intestinal microflora disorders have been registered, which are characterized by the replacing the lactobacilli and bifidum bacteria to opportunistic pathogens. A significant increase in Firmicutes phylum microorganisms and a significant decrease in Bacteroidetes phylum were found. The obtained data prove that changes in the species composition of the intestinal microbiota play an important role in the pathogenesis of obesity. Conclusions: The results indicate the feasibility of monitoring the intestinal microbiota in women with vulvovaginal candidiasis and bacterial vaginosis, especially in the presence of excess body weight and obesity to determine the degree of violations of its components, and timely correction of the detected changes.

Dietary methionine restriction is associated with improved health outcomes and an increase in lifespan in animal models. We have previously shown that an increase in dietary methionine induces alteration in the intestinal microbiome. The composition of the intestinal microbiota is a determinant of health and we, therefore, hypothesized that dietary methionine restriction would also induce changes in the murine microbiome. After one month on a methionine-restricted diet, five-month-old male and female C57BL/6 mice had decreased levels of serum methionine, without changes in body weight. We identified a decrease in the hepatic methylation status of animals fed a methionine-restricted diet compared to controls. This decrease was not associated with changes in DNA or protein methylation in the liver. In males, we saw an increase in families Bacteroidaceae and Verrucoccaceae (mostly A. mucinophila) and a decrease in Rumminococcaceae in animals fed a methionine-restricted diet compared to controls. In females, Bacteroidales family S24-7 was increased two-fold, while families Bacteroidaceae, Verrucoccaceae, Rumminococcaceae, and Rikenellaceae were decreased compared to controls. In summary, feeding a methionine-restricted diet for one month was associated with significant and sex-specific changes in the intestinal microbiome.

ObjectivesTo investigate the (1) time trends in body mass index (BMI) and (2) relationship between media use and body weight status among adult women in Nigeria. We hypothesise that higher frequency of media use is associated with higher likelihood of being overweight and obesity among adult women.Study designCross-sectional.SettingUrban and rural settings in Nigeria.ParticipantsAdult non-pregnant women aged between 15 and 49 years.MethodsData were derived from Nigeria Demographic and Health Surveys conducted in 2003, 2008 and 2013. The outcome variable was excess body weight (overweight and obesity), and main explanatory variables were frequency of reading newspaper, listening to radio and television (TV) viewing. Datasets were merged to perform pooled analysis, and were analysed using bivariate and multivariable regression techniques.ResultsOf the 69 401 participants, 16.2% had a BMI of 25.0–29.9 kg/m2(95% CI 15.8 to 16.6) and 6.6% had >30 kg/m2(95% CI 6.3 to 6.9). Between 2003 and 2013, the prevalence of overweight women increased by 4.1%, and that of obesity by 2.2%. Overall, radio was the most popular media followed by TV and newspaper. Respectively, 15.6% and 11.7% of the women reported using radio and TV almost every day and 30.6% and 25.1% at least once a week. In multivariable analysis, watching TV almost every day and at least once a week were associated with, respectively, 1.6 and 1.2 times higher odds of being overweight, and 2.7 and 1.5 times higher odds of being obese compared with those who never used radio. Similarly, significant associations were observed for newspaper and radio use as well.ConclusionThe prevalence of overweight and obesity is noticeably high among Nigerian women and has been increasing steadily over the past decade. A statistically significant association exists between BMI and the use of newspaper, radio and TV. Further studies are required to develop a better understanding of the mechanisms that underlie this relationship.

Objective. Overweight is one of the health problems that often occur in children and adolescents throughout the world, both in developed and developing countries. Study results in USA showed yearly increase of overweight prevalence in children aged 2–19 years old. In addition, based on the results by the National Health and Nutrition Examination Survey 2009–2010 in United States, the highest percentage of overweight and obesity by age group was found at 12–19 years old (33.6%). Basic Health Research/Riskesdas’s data in 2013 showed the prevalence of overweight in adolescents aged 16–18 years old in Indonesia reached the highest value of 11.5%. Moreover, most teenagers see their body images as a match between self’s and others’ ideal perception. Teenagers with positive body image tend to be more confident and also easier to get along with other people, especially their peers. In this study, researcher aimed to investigate the relationship between teenagers who have excess body weight with body image, whether it had positive or negative impact.Methods: This was a cross-sectional study. Data collection was done from December 2017 to January 2018. Data collected were anthropometric measurements (weight and height) and the King College London Body Image Questionnaire's filled by participants. From 400 subjects who filled the questionnaires, a total of 350 participants matched the inclusion criteria and were analyzed. Chi square test was done as data analysis.Results: Chi-square analysis for excess body weight status in relation to body image scores showed no relationship (p=1,000).Conclusions: There was no significant relationship between excessive body weight and body image.

Introduction. Obesity is a disorder of energy balance, which leads to excessive accumulation of fat. In recent years, many important discoveries were made in this field, including the discovery of hormones produced by adipose tissue and the identification of many of the central and peripheral pathways of energy balance.Objective. To study the levels of hormones that affect appetite and metabolism in women with obesity baseline and after weight loss while taking sibutramine.Materials and methods. The study included 56 women aged 42,9±9,5 years, with a BMI of 34,6±6,1 kg/m2. All patients underwent clinical, laboratory and instrumental examination. Hormonal study included determination of serotonin, leptin, ghrelin, endothelin-1, adiponectin.Results: In women with obesity we identified hyperleptinemia and increased serotonin levels. The decrease in body weight in patients receiving sibutramine was accompanied by lower levels of serotonin, leptin, ghrelin, endothelin-1, and increase of adiponectin.Conclusions: Obese patients have significantly elevated levels of leptin, serotonin, ghrelin compared to women of normal weight. Sibutramine treatment leads to a decrease in serotonin, leptin, ghrelin and is more effective in women with a BMI less than 36,5 kg/m2.

The obesity paradox for survival among individuals with type 2 diabetes has been observed in some but not all studies. Conflicting evidence for the role of overweight and obesity in all-cause mortality may largely be a result of differences in study populations, epidemiological methods, and statistical analysis. For example, analyses among populations with long-term prevalent diabetes and the accrual of other chronic health conditions are more likely to observe that the sickest participants have lower body weights, and therefore, relative to normal weight, overweight and even obesity appear advantageous. Other mortality risk factors, such as smoking, also confound the relationship between body weight and survival, but this behavior varies widely in intensity and duration, making it difficult to assess and effectively adjust for in statistical models. Disentangling the potential sources of bias is imperative in understanding the relevance of excess body weight to mortality in diabetes. In this review, we summarize methodological considerations underlying the observed obesity paradox. Based on the available evidence, we conclude that the obesity paradox is likely an artifact of biases, and once these are accounted for, it is evident that compared with normal body weight, excess body weight is associated with a greater mortality risk.

Overweight is a state that exceeds the relative body weight of a person as a result of the accumulation of nutrients, especially carbohydrates, fats, and proteins. This condition is caused by an imbalance between energy consumption compared to the needs or the use of energy. This study aims to determine the relationship of consumption patterns of sweet drinks to excess body weight teenagers in SMP IT IQRA  Bengkulu 2018.  This study is a cross-sectional research design. A sample is all teenagers who have overweight (overweight and obesity) as much as 57 teenagers. consumption of sweet drinks was collected by interviews using Food Frequency Questionaire (FFQ). The result showed that there was no contribution of consumption of sweet drinks to overweight teenagers (p-value 0.590). The habit of consuming sweet drinks does not contribute to increased body weight in teenagers who have excess body weight. To prevent overweight is recommended to consume balanced nutrition and avoid foods and beverages high in glucose.Berat badan berlebih adalah suatu keadaan yang melebihi dari berat badan relatif seseorang sebagai akibat penumpukan zat gizi terutama karbohidrat, lemak dan protein. Kondisi ini disebabkan oleh ketidakseimbangan antara konsumsi energi dibandingkan kebutuhan atau pemakaian energi. Penelitian ini bertujuan untuk mengetahui hubungan pola konsumsi minuman manis terhadap berat badan berlebih pada remaja di SMP IT IQRA’ kota Bengkulu tahun 2018. Jenis penelitian ini adalah observasional dengan pendekatan cross sectional. Sampel adalah seluruh remaja yang mempunyai berat badan berlebih (overweight dan obesitas) yang diambil dengan metode Total sampling yaitu sebanyak 57 remaja. konsumsi minuman manis dikumpulkan dengan wawancara menggunakan Food Frequency Questionaire (FFQ). Hasil penelitian menunjukkan bahwa tidak ada kontribusi konsumsi minuman manis terhadap berat badan berlebih pada remaja (p value 0.593). Kebiasaan mengonsumsi minuman manis tidak berkontribusi untuk meningkatkan berat badan pada remaja yang memiliki berat badan berlebih. Untuk mencegah berat badan berlebih disarankan  mengonsumsi gizi seimbang dan menghindari makanan dan minuman yang tinggi glukosa.

Abstract Background: Weaning is one of the most stressful periods that cause gastrointestinal tract dysfunction and diarrhea in calves. HNa and Gln were reported to exert beneficial effects on promoting growth performance, decreasing diarrhea incidence, and modulating intestinal microbiota in animals. Therefore, this study investigated the effect of HNa and Gln combined supplementation on growth performance, diarrhea incidence, serum parameters, intestinal microbiome, and metabolites of weaned calves.Results: In Exp. 1, 40 calves at 51±3 days of age with similar body weight (66.82±4.31 kg) were randomly assigned to 4 treatments fed with a basal diet (NC group), and a basal diet supplemented with 100 mL of 1%, 3%, or 5% HNa+1% Gln, twice daily, respectively. In a 21-day trial, calves on the 5% HNa+1% Gln group had higher ADG and lower fecal score and diarrhea incidence than those in the control group (P < 0.05). In Exp. 2, 20 calves at 51±3 days of age with similar body weight (69.37±6.28 kg) were randomly assigned to 2 treatments fed with a basal diet (NC group) and a basal diet supplemented with 100 mL of 5% HNa+1% Gln, twice daily (H+G group, the dose was obtained from Exp. 1). In a 21-day trial, calves supplemented with HNa and Gln had higher final BW and ADG, serum IgG concentration and GSH-Px and T-AOC activities, but lower fecal score, diarrhea incidence, as well as serum DAO, D-lac, TNF-α, and MDA concentrations compared to NC group (P < 0.05). Analysis of intestinal microbiota indicated that supplemented with HNa and Gln significantly increased the abundance of phyla Firmicutes and genus of Bifidobacterium, Lactobacillus, Olsenella, Ruminiclostridium 9, Howardella, and uncultured organism, whereas the abundance of phyla Bacteroidetes, genus of Helicobacter and Lachnoclostridium were decreased as compared with NC group. Moreover, untargeted metabolomics analysis revealed that supplemented with HNa and Gln altered 18 metabolites and enriched 6 KEGG pathways (primary fatty acid biosynthesis) compared to the NC group. Conclusions: This study showed that combined supplemented with HNa and Gln could decrease diarrhea of weaned calves, which may be associated with improved intestinal microbial ecology and altered metabolism profile.

Background. The steady growth of obesity in the children’s age group determines the need for integrated modern approaches to diagnosis and therapy.Objective. To determine the body composition of adolescents with different body mass for the quantitative analysis of the internal environment of the organism using the method bioimpendancemetria.Design and methods. To determine the composition of the body, 121 adolescents aged 10–17 years were examined, the average age was 13.9 years (± 2.1). Evaluation of anthropometric data and determination of BMI at the 1st stage of the study allowed to identify 3 groups of patients: group 1 — adolescents with normal body weight, 40 people (33 %), group 2 — overweight, 48 people (40 %) and group 3 — obese, 33 adolescents (27 %). At stage 2nd, the analysis of the body structure using computer impedance, which allows to determine the composition of the body in a percentage.Results. When assessing the structure of the body, in group with normal body weight, the content of adipose tissue corresponds to the normal value. In adolescents overweight and obesity in 100 % of cases there was an increased content of fat mass. In the group of adolescents with normal body weight, the percentage deviation of AKM is 16–17 %, in the group with excess body weight 12–40 %. Insufficient as well as excessive percentage of AKM causes hunger. The higher AKM in full adolescents, the more difficult the process of weight loss. During the evaluation of the main metabolism it was found that in the group of obese patients the indicators exceeded those in the groups with normal and overweight.Conclusion. The use of bioimpedance analysis allows to create an optimal set of sequential effects aimed at correcting the fat mass, water composition and muscle mass of the patient, which determines a more directed and effective weight loss and the possibility of dynamic control.

Obesity has been linked with poorer vitamin D status. The aim of this work was to analyze the changes in vitamin D status and body weight of 61 young, overweight/obese women following two different weight control programs. The study subjects were randomly assigned to one of two slightly hypocaloric diets: diet V, in which the consumption of greens and vegetables was increased, or diet C, in which the consumption of cereals (some of which were enriched with vitamin D) was increased. Dietary, anthropometric, and biochemical data were collected at the start of the study and at 2 weeks. At the beginning of the study, when taking into account only those women with a vitamin D intake below that recommended, obese women had a significantly lower mean serum 25(OH)D concentration than those who were lighter. Dietary intervention led to a greater reduction in energy intake among group C subjects; their weight was also that which was most reduced. In addition, the vitamin D intake and serum 25(OH)D values were increased only in group C women. Excess body weight was associated with deterioration in vitamin D status, especially when the intake of this vitamin was lower than that recommended. Subjects with higher serum vitamin D at the beginning of the study lost more weight than those subjects with lower initial values. Diet C was associated with a greater weight loss than diet V, and led to a greater increase in vitamin D intake and serum 25(OH)D levels. This might be of interest in the improvement of health in people trying to lose weight.

BACKGROUND Digital health promotion programs tailored to the individual are a potential cost-effective and scalable solution to enable self-management and provide support to people with excess body weight. However, solutions that are personalised, theory- and evidence- based and widely accessible are still limited. OBJECTIVE This study aimed to develop a digital behaviour change program, Choosing Health, that could identify modifiable predictors of weight loss and maintenance for each individual and utilise these to provide tailored support. METHODS We applied an Intervention Mapping protocol to design the program. This systematic approach to develop theory- and evidence-based health promotion programs consisted of 6 steps: development of (1) a logic model of the problem, (2) model of change, (3) intervention design and (4) production, (5) the implementation plan, and (6) evaluation plan. The decisions made during the Intervention Mapping process were guided by theory, existing evidence, and our own research (including four focus groups, N=40, expert consultations, N=12 and interviews, N=11). The stakeholders included researchers, public representatives (including individuals with overweight and obesity), and experts from the variety of relevant backgrounds (including nutrition, physical activity, and healthcare sector). RESULTS Following a structured process, we developed a tailored intervention that has potential to reduce excess body weight and support behaviour changes in people with overweight and obesity. The Choosing Health intervention consists of tailored personalised text messages and email support that correspond with theoretical domains potentially predictive of weight outcomes for each participant. Intervention content includes behaviour change techniques to support motivation maintenance, self-regulation, habit formation, environmental restructuring, social support and addressing physical and psychological resources. CONCLUSIONS Use of an Intervention Mapping protocol enabled the systematic development of the Choosing Health intervention and guided the implementation and evaluation of the program. Through the involvement of different stakeholders, including representatives of general public, we were able to map out program facilitators and barriers while increasing ecological validity of the program, to ensure that we build an intervention that is useful, user friendly, and informative. We also summarised lessons learnt for the Choosing Health intervention development and for other health promotion programs. CLINICALTRIAL This is an Intervention Mapping study which is currently evaluated through a Randomised Controlled Trial.
