The cashew sector has taken a great economic interest, due to the strong global demand for raw nuts. However, in Burkina Faso, the productivity of orchards still remains low, a consequence of a lack of breeding program on the species. In order to increase yields and improve the quality of the raw nut, efficient clones must be developed. Surveys were carried out and a primary in situ collection was made from which a core collection of 15 trees was extracted. An agro-morphological description of the selected trees and an analysis of the structuration of the variability within them were carried out. The study revealed appreciable agro-morphological diversity through shape, size and color of apple, type of inflorescence, fruiting period and the health of trees. A structure of diversity based on agronomic criteria led to identify 5 groups of trees, characterized by an average weight of the nut between 6.64 and 8.32 g, a kernel rate between 28.10 and 31.26% and a yield per tree between 34.27 and 104.42 kg. Performing trees will be erected as heads of clones for the production of grafted plants.

Abstract The objective of this work was to find practical procedures to overcome methodological drawbacks encountered during studies on sunflower downy mildew. Techniques for recovering living isolates of Plasmopara halstedii from the field and for the preservation of infected leaf samples for further molecular analysis were developed. A Polymerase Chain Reaction (PCR)-based test for the detection of P. halstedii in sunflower leaves and a method to remove azoxystrobin from fungicide-treated seeds are proposed. In situ-inoculations of pre-germinated seeds allowed the recovery of living isolates from the field. Three sample-preservation methods were evaluated (silica, heating and lyophilization) resulting in high yield and quality of the DNA extract. It was detected the presence of the pathogen in symptomless leaves through PCR using molecular markers based on expressed sequence tags. A treatment using sodium hypochlorite is recommended for the removal of azoxystrobin from fungicide treated seeds.

Cassava provides food security for millions of people worldwide. In Cote dIvoire, it is the second most important food crop. The objective of this study is to evaluate the structure and genetic variability of certain qualitative traits in targeted cassava landracesin order to identify the most widespread cassava landraces. Thus, prospecting missions were carried out in different regions of Cote dIvoire to characterize cassavalandraces, from which 180 cassava landrace accessions were characterized using14 morphological markers. Multiple Correspondence Analysis (MCA) showed that among the 14 markers, 10 were relevant fordescribing the total variability within accessions. In addition, the Hierarchical Ascending Classification (HAC) grouped the Ivorianlandraces into five homogeneous groups and also revealed that this morphological diversity is not structured according to the landracesgeographical origin. The study highlighted 131 distinct cultivars among the 180 accessions characterized. Moreover, of the 131 distinct cultivars, four were the most widespread. These are the local cultivars: Yace, Six mois, Blêbou and Trogla. Thus, the study has definitively shown that breeders have a wide choice of parents for developing improved cassava cultivars adapted fromthe populations found in Cote dIvoire.

Several methods of Deoxyribonucleic acid (DNA) extraction have been applied to extract bacterial DNA. The amount and the quality of the DNA obtained for each one of those methods are variable. The study aimed to evaluate bacterial DNA extraction using conventional boiling method followed by alcohol precipitation. DNA extraction from Gram negative bacilli was extracted and precipitated using boiling method with further precipitation by ethanol. The extraction procedure performed using the boiling method resulted in high DNA yields for both E. coli and K. pneumoniae bacteria in (199.7 and 285.7μg/ml, respectively) which was close to control method (229.3 and 440.3μg/ml). It was concluded that after alcohol precipitation boiling procedure was easy, cost-effective, and applicable for high-yield quality of DNA in Gram-negative bacteria.

The aim of this study is to develop a relocatable modelling system able to describe the microbial contamination that affects the quality of coastal bathing waters. Pollution events are mainly triggered by urban sewer outflows during massive rainy events, with relevant negative consequences on the marine environment and tourism and related activities of coastal towns. A finite element hydrodynamic model was applied to five study areas in the Adriatic Sea, which differ for urban, oceanographic and morphological conditions. With the help of transport-diffusion and microbial decay modules, the distribution of Escherichia coli was investigated during significant events. The numerical investigation was supported by detailed in situ observational datasets. The model results were evaluated against water level, sea temperature, salinity and E. coli concentrations acquired in situ, demonstrating the capacity of the modelling suite in simulating the circulation in the coastal areas of the Adriatic Sea, as well as several main transport and diffusion dynamics, such as riverine and polluted waters dispersion. Moreover, the results of the simulations were used to perform a comparative analysis among the different study sites, demonstrating that dilution and mixing, mostly induced by the tidal action, had a stronger effect on bacteria reduction with respect to microbial decay. Stratification and estuarine dynamics also play an important role in governing microbial concentration. The modelling suite can be used as a beach management tool for improving protection of public health, as required by the EU Bathing Water Directive.
