Application of a Modified Culture Medium for the Simultaneous Counting of Molds and Yeasts and Detection of Aflatoxigenic Strains of Aspergillus flavus and Aspergillus parasiticus

Molds and yeasts from 91 samples of feed and raw materials used in feed formulation were enumerated on a new culture medium to which a beta cyclodextrin (β-W7M 1.8-cyclodextrin) had been added. This medium was compared with other media normally used in laboratories for the routine analysis of fungi, such as Sabouraud agar, malt agar supplemented with 2% dextrose, and potato dextrose agar. When a t test for paired data (0.05 significance level, 95% confidence interval) was applied, no statistically significant differences between the results obtained with the new culture medium and those obtained with the other media used to enumerate molds and yeasts were found. For the evaluation of contamination due to aflatoxin for all of the samples, Sabouraud agar and yeast extract agar, both supplemented with 0.3% β-W7M 1.8-cyclodextrin, and APA (aflatoxin-producing ability) medium were used. Aflatoxin was detected in 21% of the feed samples and in 23% of the raw-material samples analyzed, with maximal amounts of 2.8 and 6.0 μg of aflatoxin B1 per kg, respectively, being detected. In any case, the aflatoxin contents found exceeded the legally stipulated limits. The t test for paired data (0.05 significance level, 95% confidence interval) did not show statistically significant differences between the results obtained with the different culture media used for the detection of aflatoxins. The advantage of the new medium developed (Sabouraud agar with 0.3% β-W7M 1.8-cyclodextrin)is that it allows simultaneous fungal enumeration and determination (under UV light) of the presence of aflatoxin-producing strains without prior isolation and culture procedures involving expensive and/or complex specific media and thus saves work, time, and money.

The purpose of the research was to know the affect of many factors which affected to the production volume in PT. Kelola Mina Laut Gresik. What the price of raw materials was and the used of raw materials partially and simultan eously affected on the production volume. The analysis tool which used was a model of multiple linear regression. Hypothesis testing used t test and F test, both at the significant level 5%. Based of the analysis of research on PT Kelola Mina Laut Gresik. Partially, inventory raw material price had not significant effect on the production volume, consumption of raw material inventory affected significantly of the production volume. Inventory of raw material price and the use of raw material simultan eously affect significantly to the production volume.

One of the technological innovations nowadays is the development of industrial processing of agricultural raw material, manufacturing process leading to lower risks – «Low/No/Reduced» and «Better-for-you». This may be done by light processing of raw materials and use of traditional technologies, i.e. fermentation of craft beverages by natural symbiotic cultures Oryzamycesindichiand Medusomycesgisevi (kombucha). The recent years saw a worldwide interest to a horticultural crop called Hippophaerhamnoides (sea-buckthorn) as it is eco-friendly and healthful. Its fruits have a special smell and taste, are reach in carotene, carotenoids, tocopherols, ascorbic acids, polyphenols. The composition is the reason why it is used for foodstuff, i.e. pastry, dairy products, unclarified juice. Production of beverages fermented by natural symbiotic cultures and local orchard products is a new direction in Russia. The supplies are limited which means that the research is relevant. The aim of the research is to study the possibility of usage ofjuice of Hippophaerhamnoides fruits in the new technology of soft drinks production fermented by Oryzamycesindichi and Medusomycesgisevi. A single nutritious substrate for Oryzamycesindichi and Medusomycesgisevi symbiotic cultures cultivation was proved impossible to be produced. Still the possibility to replace polyphenols of the Medusomycesgisevi by those of the sea-buckthorn juice must be analyzed.Sea-buckthorn juice added in a culture medium fermented by Oryzamycesindichi and Medusomycesgisevi symbiotic cultures increases biosynthetic activity of acidforming bacteria. This tangibly improves organoleptic properties of the fermented soft beverages produced. Besides, the technology of Oryzamycesindichi and Medusomycesgisevi production presupposes no clarification which makes the process of the Hippophaerhamnoides use significantly easier. The research helped to identify that the sea-buckthorn juice used in a must fermented by Medusomycesgisevi has many advantages. Namely more comprehensive utilization of carbohydrate, a higher level of reduction of organic acids, and decrease of the time of technological process (24 hours less).

As an industrial organic solid waste, oil residue, with wide sources and low price, but could not find an effective way to use currently. In addition, with the development of mushroom industry and rising price of traditional raw-materials, it is very necessary to find the substitute raw material to reduce production cost of mushroom cultivation. This study attempts to produce the strains of Flammulina velutipes by taking use of oil residue to replace part of the traditional raw material, there are two groups in the experiment: the test group is adding 5%, 10%, 15%, 20%, 40% of the oil residue to replace the culture medium of cottonseed hull and wheat bran; the control group is the traditional culture medium, then comparatively observed the growth of the mycelium. The results showed that it can promote the growth of the mycelium and shorten the cycle of seed production to add the oil residue in the test. But the growth rate of mycelium will be hindered when the concentration exceeds a certain range.The optimal culture medium to add the oil residue is as follows: 15% of the oil residue, 64% cottonseed hull, 20% coarse wood chips, 0% wheat bran , 1% lime. Oil residue contains a lot of the nutritional components to benefit the growth of edible fungi, and without the heavy metal pollution, can replace the culture medium of cottonseed hull and wheat bran and other traditional materials, reduce cost of production the strains, a fair-sized economic efficiency can be received, the environment can be protected.

Gelidium latifolium is one of red seaweed types potentially can be developed as an industrial raw material. Since Gelidium is currently taken from ocean, the availability of seaweed from aquaculture is necessary to overcome the small number of its availability in nature. In Indonesia, G. latifolium cultivation has not been carried out so that domestication is required. The use of macro and micro nutrients in growth media is essentially needed for the domestication process. Domestication requires fast media and place for growth. The purpose of this study is to determine the growth of biomass and the survival of G. latifolium in different culture media. The study was conducted in a semi-outdoor research laboratory. The method used in this research is laboratory experimental method and Completely Randomized Design (CRD) with the treatment applied using 3 types of culture media (Urea: Za: TSP) by comparison (A) 100: 50: 50% (2 g.L-1), (B) 75: 75: 50% (2 g.L-1) and (C) 75: 50: 75% (2 g.L-1), with 3 replications. The seaweed was kept in 10 L of water in aeration equipped aquarium and filled with 10 g of G. latifolium on each treatment. The best growth rate of G. latifolium biomass is 5.67± 0.58 g and 100±0% are survived in C culture medium with a concentration of 75% Urea: 50% ZA: 75% TSP (2 g.L-1).

Salt in the main need for humans, In the process of shrinkage of salt products become the expected result is always increased because in the islands of Madura who have high salt income can be utilized. Light focus can be used through convex convex lens to increase sea water shrinkage so as to accelerate evaporation as well Get shrinkage of salt water salt raw material quickly. From observations made in May - June 2017 can be obtained data focusing light through convex convex lens is by focusing the sun on convex lens that is directed at sea water, so it can shrink the water faster. Acquisition of water depreciation using the tool obtained 95,000 cc / 95 liters, while 47.000 cc / 47 liters obtained from without tools. From this study it can be concluded that the value obtained yields a positive value, where the use of the tool from the volume of each volume filled 100 liters with a ratio of 1: 1 from the analysis obtained from using a tool faster 56% using tools with upper 14322.90 Of the 99% confidence interval received. So from the shrinkage using a faster tool, the depreciation of sea salt raw materials with convergent convex lenses can be applied directly as a technological reasoning.

The use of physical methods of processing, in particular ultraviolet radiation, is used in the food industry for the purpose of disinfection of raw materials, semi-finished products and finished products, which increases the storage time. The process of processing food products with ultraviolet light quickly proved itself. This method does not change the taste of the product, increases the shelf life and sales, and is a non-thermal method of deactivating pathogens. The wide use of the bactericidal effect of UV rays for food products is limited by their low penetration capacity, so the UV spectrum can be used mainly for sur-face sterilization, provided that the deep layers of the material do not contain microflora. The effectiveness of UV radiation depends on the following factors: the source and the UV dose; the sensitivity of microorganisms to UV light; the composition and physical properties of the product. Object of research: model media containing native microflora of dried fruit raw materials "dried Apri-cots". The research was carried out using model media simulating the proper-ties of the raw material surface, excluding its protective mechanisms and allow-ing to reveal the true dynamics of inhibition of native microflora. Native micro-flora of dried fruits implies a set of microorganisms. Literature sources have shown the possibility of the presence of such types of microorganisms as E. coli, Salmonella, Candida, Pseudomonas, spore forms of microorganisms on the surface of raw materials. During the experiment it was determined that the treatment duration of 5 min reduced the initial contamination of 2 order pro-cessing in 10 minutes – 2.6 the order in 15 minutes – 2.7 order and 20 minutes for 3 orders of magnitude.

Corynespora cassiicola threatens soybean and cotton production in Brazil. The objective of this study was to evaluate cultural and morphological aspects of C. cassiicola isolated from soybean and cotton of different Brazilian regions, in culture media. The isolates were grown in PDA (Potato Dextrose Agar) and V8 juice agar media. The characteristics evaluated were: color, aspect, and growth rate of mycelia, as well as production and dimension of conidia, and number of septa per conidium. Culture media and isolates were compared using the Kruskal-Wallis or Tukey’s test at 5% significance level. The mycelia of the isolates were predominantly dark gray and light brown. C. cassiicola isolates grew better in V8 juice agar medium, presenting a higher mycelial growth rate. In PDA medium, the production of conidia was higher in isolates from cotton, compared with soybean isolates. There was great variation in the production of conidia in V8 juice agar medium, regardless of the host origin. Conidia length and width varied for isolate and culture medium. The isolates of C. cassiicola coming from cotton presented a higher number of septa per conidium when grown in PDA medium. The morphological aspects of C. cassiicola vary depending on the host of origin and the culture medium.

The purpose of this research is to know about working capital management applied, and its influence on profitability and risk. The research object is trading company moves in import & distribute chemical raw material. The research used analysis descriptive method, and the hypothesis was testing by simple linier regression, correlation, and determination. The result of the research shows that the effect of the implementation of working capital management on the change of the net working capital with tend to rise has a profitability level of 10.4% lower than the net working capital change with tend to go down of 46%, but instead on the risk level, the net working capital change with tend to rise has a risk level of 43.8% higher than the change in net working capital with tend to go down of 0.3%.Based on  t test, the result shows that the net working capital change influence  is not significant  to profitability and risk.

This study aims to determine and analyze the influence of Service Quality and Customer Satisfaction on Customer Loyalty in Jabodetabek Commuter Line. The sample of this study was 50 people. Methods of collecting data by distributing questionnaires. Data analysis using the analysis used is simple linear regression, t test and coefficient of determination. The results showed 1) Service Quality has a positive and significant effect on Customer Loyalty in Jabodetabek Commuter Line, with a significance level of 0.048; and supported by the results of hypothesis testing with a t-count value of 4.433 > t-table value of 1.95, with a significance of 0.048 or < 0.05; 2) Customer Satisfaction positive and significant effect on Customer Loyalty in Jabodetabek Commuter Line, with a level significance of 0,000; and supported by the results of hypothesis testing with a t-count value of 4,969 > t-table value of 1.95, with a significance of 0,000 or < 0.05, 3) Service quality and Customer Satisfaction have a positive and significant effect on Customer Loyalty in Jabodetabek Commuter Line, with a significance level of 0,000. This means that the hypothesis H0 is rejected and Ha is accepted so that it can be concluded that service quality and customer satisfaction together have a positive and significant effect on customer loyalty in Jabodetabek Commuter Line.

This study has been aimed to find out the correlation emotional intelligence and Rhtype. The simple sizes were 35 undergraduate and graduate of Abadan Oil College with the cluster sampling.The correlation between emotional intelligence and P/N blood groups was 0.085 and it does not accept by standard statistics. We enter the other variable as emotional intelligence groups. The correlation between Rh type and emotional intelligence groups was 0.136 and this correlation in 0/2 significance level had been reported significant.The result was tested by t test, and analysis shown significance difference.it was found that scattering of Rh type was not equal on EI.As well as positive groups can receive upper scores of EI than negative ones and negatives groups have less scores of EI the other. In fact most positives have scores between "114-148", means that they can perch in the medium and high groups of EI.
