Assuring Growth Inhibition of Listerial Contamination during Processing and Storage of Traditional Greek Graviera Cheese: Compliance with the New European Union Regulatory Criteria for Listeria monocytogenes

The current microbiological regulatory criteria in the European Union specify a maximum Listeria monocytogenes population of 100 CFU/g allowable in ready-to-eat foods provided the product will not exceed this limit throughout its shelf life. The aim of this study was to validate the manufacturing method for traditional Greek Graviera cheese produced from thermized milk. Initial challenge experiments evaluated the fate of inoculated L. monocytogenes (ca. 4 log CFU/ml, three-strain cocktail) in thermized Graviera cheese milk (TGCM; 63°C for 30 s) in the presence and absence of a product-specific starter culture (SC) in vitro. Milk samples were incubated for 6 h at 37°C and then for 66 h at 18°C. Experiments were conducted to evaluate the fate of a cocktail of three nonpathogenic L. monocytogenes and L. innocua indicator strains inoculated (ca. 3 log CFU/g) in Graviera cheeses commercially manufactured from TGCM+SC. Cheeses were brined, ripened at 18°C and 90% relative humidity for 20 days, and stored at 4°C for up to day 60 under vacuum. In TGCM, L. monocytogenes increased by ca. 2 log units, whereas in TGCM+SC L. monocytogenes growth was retarded (P < 0.05) after a ca. 1-log increase within 6 h at 37°C. Populations of Listeria indicator strains did not grow in TGCM+SC cheeses at any stage; they declined 10-fold in fresh cheeses within 5 days and then survived with little death thereafter. Thus, growth inhibition but not inactivation of potent natural Listeria contaminants at levels below 100 CFU/g occurs in the core of traditional Greek Graviera cheese during fermentation, ripening, and storage.

Yogurt is a nutritious food that is regularly consumed in many countries around the world and is widely appreciated for its organoleptic properties. Despite its contribution to human dietary requirements, yogurt in its traditional recipe is a poor source of fat-soluble vitamins. To respond to consumer demands and further increase the nutritional value of this product, this work aimed to fortify yogurt with vitamin E by using emulsification as the method of encapsulation. The effects of thermal processing and chilled storage on the physicochemical stability of the yogurt-based beverage was investigated. Vitamin E was only minorly affected by bulk pasteurization at 63 °C for 30 min and remained stable during storage at 4 °C for 28 days. Fortified samples showed increased in vitro antioxidant activity compared with non-fortified samples. Lactic acid bacterial counts were above the minimum recommended levels (>106 cfu/g) after processing and storage. In conclusion, this work has demonstrated that emulsification can be an effective strategy for developing yogurt-based products fortified with fat soluble vitamins.

This chapter reflects on the election of Donald Trump and the vote of the British electorate in favour of ‘Brexit’ from the European Union. While we refrain from judging the outcomes of these votes, we do discuss concerns pertaining to the lack of truthfulness in both campaigns. After rehearsing the lies on which the Trump and Brexit campaigns were based, we consider different explanations as to why these campaigns were nevertheless successful, and where this leaves the argument for epistemic democracy. Particularly worrisome are tendencies towards ‘epistemic insouciance’, ‘epistemic malevolence’, and ‘epistemic agnosticism’. We also consider the problematic influence of social media in terms of echo chambers and filter bubbles. The core argument in favour of epistemic democracy is that the pooling of votes by majority rule has epistemically beneficial properties, assuming certain conditions. If these assumptions are not met, or are systematically corrupted, then epistemic democracy is under threat.

ABSTRACT Here, we report the complete genome sequences of three Bacillus amyloliquefaciens strains isolated from alfalfa, almond drupes, and grapes that inhibited the growth of Listeria monocytogenes strain 2011L-2857 in vitro. We also report multiple gene clusters encoding secondary metabolites that may be responsible for the growth inhibition of L. monocytogenes.

ABSTRACT: Buffalo milk is rich in nutrients and can serve as a substrate for the proliferation of microorganisms. Thus, the objective of the present study was to evaluate the growth kinetics of Salmonella Typhimurium and Listeria monocytogenes in buffalo milk under different processing and storage conditions. Samples of raw and pasteurized milk were inoculated with 1 CFU of each bacterium, separately and together, per 25 mL. After contamination, samples were stored at 8 °C or 37 °C, and bacterial counts were performed at 24, 48, and 168 h. In addition, the accompanying microbiota growth, pH, and the effect of these variables on the growth kinetics of microorganisms were monitored. The pathogens tested were able to proliferate under most conditions tested, reaching high titers throughout the experimental period. At 37 °C, there was a decrease in pH and an increase in the accompanying microbiota that interfered with the microbial growth curve. It was also observed that pasteurized milk subjected to 8 °C provided better conditions for the multiplication of bacteria. Therefore, it was concluded that care throughout the production chain, storage, and commercialization of milk must be adopted to guarantee the microbiological safety of this food.

This introductory chapter outlines the core argument of the book: that as Russia ramped up its hybrid war on the West starting around 2007, politics in Western countries has become more similar to politics in the vulnerable “lands in between.” Russia’s hybrid war on the West has contributed to political polarization by promoting extremist parties and creating a sense that every election presents voters with a “civilizational choice” between Russia and the West or authoritarianism and democracy. Paradoxically, many of the leaders that rise to the top in these conditions are those who find ways to profit from both sides. They benefit from the sponsorship of pro-Russia and pro-Western interests to enrich themselves in the process. The plan of this book is simple. It starts with exploring the nature of Russia’s hybrid war on the West and the West’s delayed response. Then it shows how this conflict shapes the politics of the lands in between, Central and Eastern European member states of the European Union, and core Western countries.

The heterogeneity of economic performances in the EU member states is one of the main reasons for the existence of a “core-periphery” relationship. The goal of this research is to examine various economic indicators to reveal possible divisions between the EU members. This issue emphasized the contribution of rich “core” countries to the imbalances in poorer “peripheral” EU members. By applying cluster methodology and considering the most recent data, two groups of countries were identified, the first comprising 11 countries that form the “centre” or the “core”, and the rest of the EU forming the “periphery”. Considering differences between these countries is necessary and justified for discussions about the future development of the EU that will involve differences between member states.
