Antioxidant Activity of Dendrobium lasianthera and Geodorum densiflorum

Many antioxidants are obtained from plants that contain secondary metabolic compounds such as alkaloids, flavonoids, saponins, tannins, etc. Thus, the search for plants that are rich in natural antioxidants is conducted continuously, including orchids. Dendrobium lasianthera and Geodorum densiflorum are orchids having secondary metabolites that are thought to have antioxidant activity. This study aims to determine the antioxidant activity of the ethanolic extracts of these two types of orchids using the DPPH method (1,1-diphenyl-2-picrylhydrazine). The results showed that the IC50 of the ethanolic extract of the leaves of D. lasianthera was 288.24 ppm and the stem was 26.27 ppm. The IC50 of the ethanol extract of leaves of G. densiflorum was 277.65 ppm and of the tubers was 197.48 ppm. Finally, the leaves have negligible antioxidant activity, but the stems and tubers had activity ranging from weak to very strong.

Antioxidants have an important role in delaying the oxidation process and preventing the occurrence of degenerative diseases in the body. Antioxidants consist of synthetic and natural; the use of synthetic antioxidants in a relatively long period is hazardous so that the use of natural antioxidants is considered more potential and safe for the body. The process of extracting secondary metabolites from the leaves of bangun-bangun (Coleus amboinicus L.) is carried out by the maceration method with methanol, then partitioned in stages with n-hexane, chloroform, and ethyl acetate. This research intends to identify secondary metabolites and test antioxidant activity from the ethyl acetate fraction with the DPPH method. Phytochemical screening results of ethyl acetate fraction showed the different secondary metabolite groups such as alkaloids, phenolics & polyphenols, flavonoids, coumarin, and triterpenoids. Antioxidant activity (IC50) obtained is 64.97 with a healthy category, so that the potential to be used as a natural antioxidant.

Kenaf (Hibiscus cannabinus L.) is an environmentally friendly natural fiber-producing plant that can produce diversified products, such as paper, wallcover, car interior, geotextile, soil safer, fiber drain, particle board, and plastic reinforcement as well as biofuel industry raw materials. Kenaf plant is a plant that has been only part of the fibers of the stem alone so that many unprofitable side products such as kenaf leaves after harvesting only as a waste. In Africa the leaves of kenaf are widely used as medicine This study aims to analyze phytochemical compounds and study the antioxidant activity of the kenaf leaf. The study started by doing phytochemical test and analyzing antioxidant in kenaf leaf and then testing its antioxidant activity with DPPH method. The results showed that the leaves of kenaf contain secondary metabolites of phytochemical compounds such as flavonoids, alkaloids, steroids, saponins, carbohydrates and tannins and have antioxidant activity that is quite good that is 44.4813 ppm.

Parkia speciosa (petai) pods are rarely used and considered as waste despite their phenolic and flavonoid content. Phenolic and flavonoid content in plants are known to exhibit antioxidant activity. Antioxidants are known to potentially act agains radical and mediate the damage by structural stabilization. This research was conducted to analyze the antioxidant potential of petai pods using DPPH method. Analysis showed the IC50 of petai pods ethanolic extract was 75.72 ppm, which indicate strong acting antioxidant. Standardization of extracts were determined to manage the quality of extract, including the analysis of density, water content, ash values, and quantification of total phenolic and flavonoid content. Research outcome indicate the extract retain its quality during storage and use which confrmed to standardization requirements of water content and ash values. Total phenolics and flavonoid obtained were 272.45 mg/gGAE and 243.2029 mg/gQE respectively. Based from this research, petai pods ethanolic extract present strong antioxidant activity which was contributed by high existing content of flavonoid and phenolic compounds.

The research objectives were to identify the secondary metabolite components, total phenolic content and determine the antioxidant activity of the ethanol extract of red betel leaf (Piper crocatum Ruiz & Pav.). The extraction process was carried out by materation using ethanol as a solvent. Determination of total phenolic content was carried out colorimetrically with Folin-Ciocalteu reagent measured at a maximum wavelength of 765 nm. Determination of antioxidant activity using the DPPH method measured by spectrophotometry at a maximum wavelength of 517 nm. The results of phytochemical screening of the ethanolic extract of red betel leaf contain secondary metabolites, including flavonoid, phenolic, tannin, alkaloids, steroids, and triterpenoids. The total phenolic content of the red betel leaf ethanol extract was 0.949±0.003 mg GAE/g d.w. and has antioxidant activity (IC50) 84,656 including strong category as an antioxidant. The study about antioxidant activity test of coffee parasite leaves (Loranthus ferrugineus Roxb.) by 1,1-diphenyl-2-pikrilhidrazyl (DPPH) method has been done. The aim of this study was to determine the content and activity of secondary metabolites, flavonoids and antioxidant, which found in the coffee parasite leaves. Dry powder of parasite coffee leaves (Loranthus ferrugineus Roxb.) as much as 0.5 kg were macerated by 2 L of ethanol solvent at room temperature for 4 x 24 hours, then mixed and filtered. Ethanol filtrate was evaporated at 30-40°C by using a rotary evaporator to obtain the crude extract of coffee parasite leaves. The results of the phytochemical screening showed positively that the extract was containing alkaloid, flavonoids, saponins, tannins and steroid compounds.The results of the antioxidant activity test by using DPPH method was obtained that the amount of antioxidant activity of the samples of ethanol extract had a very strong antioxidant activity against radical DPPH 0.05 mM, with IC50 values was obtained 6.063 ppm. Whereas, for comparison of ascorbic acid was about 3.127 ppm.

Antioxidants are substances that can provide endogenous protection and exogenous oxidative stress by capturing free radicals. Many plants are efficacious as antioxidants, namely plants that contain polyphenols, especially flavonoids, so many are formulated as natural antioxidants. Plants such as Muntingia calabura, Syzygium cumini, Ocimum basilicum, and Eleutherine bulbosa contain polyphenol compounds, especially flavonoids which are efficacious as natural antioxidants. This research aimed to study antioxidant activity derived from some potential plants using the DPPH method by calculating the IC50 value of each plant extract. This research method starts from the determination process to prove the validity of the plants used, the extraction process using the maceration method with 70% ethanol solvent, then the antioxidant activity of extracts from each plant was carried out using the DPPH method. This research starts from the determination process to ensure the correctness of the plants used, then the extraction process is carried out using the maceration method with 70% ethanol solvent. After that the antioxidant activity was determined from the four plants using the DPPH method to see the strongest IC50 value among the four plants. IC50 is the concentration of the sample to inhibit 50% of free radicals. The results of IC50 values from ethanol extract of M. calabura leaves, Syzygium cumini leaves, Ocimum basilicum leaves and Eleutherine bulbosa bulbs, were 18.72; 63,84; 141.59 and 173.15 ppm. Ethanol extract of M. Calabura has a smaller IC50 value of 18.72 ppm which has a very strong and most powerful antioxidant from the ethanol extract of Syzygium cumini, Ocimum basilicum and Eleutherine bulbosa. Keywords : Antioxidant, Muntingia calabura, Syzygium cumini, Ocimum basilicum, Eleutherine bulbosa, DPPH Method

ABSTRAK Sistem antioksidan merupakan bagian dari sistem kekebalan tubuh yang mampu menghambat reaktifitas radikal bebas. Sumber alami antioksidan salah satunya adalah buah yaitu buah mangga Arumanis. Proses fraksinasi dengan pelarut dietileter dilakukan untuk menyari senyawa-senyawa yang bersifat semipolar yang diharapkan memiliki potensi sebagai antioksidan. Penelitian ini bertujuan untuk mengetahui aktivitas antioksidan fraksi dietil eter ekstrak etanol buah mangga arumanis (Mangifera indica L.) dengan Metode DPPH. Ekstraksi dilakukan dengan metode maserasi menggunakan pelarut etanol 70%, kemudian dipartisi cair-cair menggunakan pelarut dietil eter. Penentuan aktivitas antioksidan menggunakan metode DPPH dengan membuat seri konsentrasi fraksi dietil eter ekstrak etanol buah mangga Arumanis yaitu 12,5; 25; 50 dan 100ppm. Vitamin C digunakan sebagai larutan pembanding dengan seri konsentrasi 0,625; 1,25; 2,5 dan 5 ppm. Hasil penelitian menunjukkan bahwa fraksi dietil eter ekstrak etanol buah mangga arumanis memiliki aktivitas antioksidan dengan nilai IC50 sebesar 75,22 ppm dan 1,18 ppm untuk Vitamin C. Fraksi dietileter ekstrak etanol buah mangga Arumanis memiliki potensi lemah sebagai antioksidan jika dibandingkan dengan Vitamin C. Source of natural antioxidants is a fruit that is mango Arumanis. Process of fractionation with solvent diethylether to extraction the compounds semipolar and expected to have potential as antioxidants. This study aims to determine the antioxidant activity of diethyl ether fraction of ethanol extract Arumanis mango (Mangifera indica L.) with DPPH method. Extraction used ethanol 70% by maceration method, then partitioned using a liquid-liquid partition with diethylether as a solvent. Determination of antioxidant activity using DPPH method with a series of concentrations of diethyl ether fraction of ethanol extract Arumanis mango ie 12.5; 25; 50 and 100ppm. Vitamin C used as a reference solution with a concentration series of 0,625; 1.25; 2.5 and 5 ppm. The results showed that the diethyl ether fraction of ethanol extract Arumanis mango have antioxidant activity with IC50 value of 7.22 ppm and 1.18 ppm for Vitamin C. Diethylether fraction of ethanol extract Arumanis mango has weak potential as a antioxidant, if compared with Vitamin C. Antioxidants are compounds that can delay, reduce, slow down or inhibit oxidation reactions from free radical reactions. Antioxidants donate electrons to unstable free radicals so that these free radicals can be neutralized to not interfere with the body's metabolic processes. Red seaweed Galaxaura rugosa has potential as an antioxidant. The study aimed to determine the antioxidant activity of the ethanol extract of red seaweed Galaxaura rugosa against DPPH free radicals based on the IC50 value. Phytochemical testing and determination of antioxidant activity were carried out using the DPPH method (2,2-diphenyl-1-picrylhydrazyl), which was carried out quantitatively using a UV-Vis spectrophotometer. The results of the phytochemical screening of red seaweed ethanol extract contained alkaloids, flavonoids, saponins, steroids, and polyphenols and the IC50 value of red seaweed ethanol extract was 4.59 ppm, while the positive control for Vitamin C was 6.64 ppm. Penelitian ini bertujuan untuk menentukan aktivitas antioksidan ekstrak etanol rumput laut merah Galaxaura rugosa terhadap radikal bebas DPPH berdasarkan nilai IC50. Pada penelitian ini dilakukan pengujian fitokimia dan penentuan aktivitas antioksidan menggunakan metode DPPH (2,2-difenil-1-pikrilhidrazil) yang dilakukan secara kuantitatif menggunakan spektrofotometer UV-Vis. Hasil dari skrining fitokimia ekstrak etanol rumput laut merah mengandung senyawa alkaloid, flavonoid, saponin, steroid dan polifenol serta nilai IC50 ekstrak etanol rumput laut merah sebesar 4,59 ppm sedangkan kontrol positif Vitamin C sebesar 6,64 ppm. ABSTRACT Aplysina sp., is one of the marine biota, which forms the coral reefs that contain active compounds, whose percentage of activity is greater than the compounds produced by terrestrial plants. This study aims to analyze the antioxidant activity of the sponge Aplysina sp. Sponge sample Aplysina sp., obtained from the waters of the Lembeh Strait, Bitung. This research is an experimental laboratory by testing the ethanol extract of sponge Aplysina sp., with the DPPH method [1,1-diphenyl-2-picrilhidrazil] to analyze antioxidant activity using a spectrophotometer UV-Vis. The results of this study showed that the ethanolic extract of the sponge Aplysina sp. Lembeh straits waters have antioxidant activity in each concentration test. Penelitian ini merupakan eksperimental laboratorium dengan pengujian terhadap ektrak etanol spons Aplysina sp.dengan metode DPPH [1,1-difenil-2-pikrilhidrazil] untuk menganalisis aktivitas antioksidan dengan menggunakan sprektrofotometer UV-Vis. Hasil penelitian ini memperlihatkan ekstrak etanol spons Aplysina sp.di perairan selat lembeh mempunyai aktivitas antioksidan disetiap konsentrasi pengujiannya. The free radical is an unstable molecule because contains one or two unpaired electrons. The antioxidant substance is a simple way to decrease the illness caused by free radicals. Cawat hanoman (Bauhinia aculeata L.) was known to contain tannin components one of the benefits as an antioxidant. This research aims to determine the antioxidant activity of the B. aculeata stem tested by qualitatively used thin-layer chromatography (TLC) and quantitatively using the DPPH method. Bauhinia aculeata stem was extracted using a maceration extract method with 96% ethanol. Antioxidant activity test was done qualitatively by eluent of ethyl acetate : methanol : purified water (6 : 2 : 1) using TLC and quantitatively using the DPPH method. The result of antioxidant activity from 96% ethanol extract of B. aculeata stem qualitatively showed the presence of yellow spots on a purple background at TLC after syringed DPPH 0.5 mM and quantitative test that resulted in an IC50 of 21.862 �g/mL. These results indicate that 96% ethanol extract of B. aculeata has very strong antioxidant activity.
