Background: Biosynthesized silver nanoparticles (AgNPs) are widely used in various biomedical applications. However, limited reports are currently available about the safety of biofabricated AgNPs using Cinnamomum zeylanicum bark extracts. Objectives: The current study is aimed to assess the potential toxicity of biosynthesized AgNPs (using C. zeylanicum) by subacute oral administration in experimental rats. Methods: AgNPs were biofabricated using methanol extract of C. zeylanicum bark and characterized by scanning electron microscopy (SEM) and atomic force microscopy (AFM). Twenty-four Rattus norvegicus female rats were divided into 4 groups (6 animals per group) as follows: Group-I was the control, while groups II, III, and IV were given 0.85, 1.76 and 3.53 mg/kg doses of AgNPs, respectively for 14 consecutive days. After 14 days of oral administration of AgNPs, serum levels of malondialdehyde (MDA), superoxide dismutase (SOD) and catalase (CAT) were measured using an ELISA technique. Serum concentrations of urea, creatinine, aspartate transaminase (AST) and alanine transaminase (ALT) were also determined as well as histopathologic features of the liver and the kidney. Results: AgNPs did not induce any changes in mean body weight, biochemical parameters (AST, ALT, urea and creatinine), oxidative stress biomarkers (MDA, SOD and CAT) and histopathologic features (of the liver and kidneys) of the treated groups when compared to control group. Conclusions: Our findings suggest that the oral administration of biogenic AgNPs (biofabricated using methanol extract of C. zeylanicum) to rats at a specific dose is relatively safe and does not show any signs of toxicity.

Background. The number of reproductive-aged women with cancer, who desire child bearing, has increased with improvements in cancer detection and treatment. Cancer treatments have the potential to cause germline mutations that might increase the risk of cancer in the progeny of cancer patients. the aim of the study was to assess the feasibility of reducing the long-term side effects of Etoposide on the progeny of rats using Glutaxim. material and methods. Forty-five white outbred female Wistar rats, 2.5-month-old, were divided into 3 groups. Group I consisted of 15 intact rats. Group II comprised 15 rats treated with cytostatic drug (the control group). Group III consisted of 15 rats treated with Glutoxim® (Glutayil-Cysteinyl-Glycine, Pharma Vam Ltd., Russia) at a dose of 50 μg/kg 5 days before and 5 days after receiving cytostatic drug. results. An increase in the number of fetuses with external hemorrhages and pathological changes in internal organs was found in the progeny of female rats receiving Etoposide 3 months before mating. The progeny experienced a decrease in the rate of formation of sensory-motor reflexes, ability to learn and adaptive behavior. All studied parameters did not differ from background values in the progeny of female rats treated with combination of Etoposide and Glutoxim. conclusion. Glutaxim is the effective drug for correction of pathological changes in the progeny of female rats receiving cytostatic drugs.

Aim: Deformities of the jaw and face are often caused by infection, inflammation, and cystic and neoplastic pathological conditions. Defects with various aetiologies should be repaired promptly using the most appropriate approach to reconstruct the anatomical form. To treat defects, bone grafts with various combinations have been used. In particular, combinations including cellular products to enhance osteogenic properties have been implemented. In this study, we aimed to investigate the effects of different materials and cells on bone defects by using mesenchymal stem cells (MSCs), which are thought to have a positive effect on healing, demineralized bone graft (DMB) and platelet-rich plasma (PRP). Methodology: We used 55 female rats weighing between 200-250 g, four of which were used to obtain platelet-rich plasma. The remaining animals were divided into five groups. Group I (n = 6) was the operative control group, Group II (n = 24) was given DMB, Group III (n = 24) was given DMB+PRP, Group IV (n = 24) was given MSC+DBG and Group V (n = 24) was given DMB+PRP+MSC applied to rat tibial defects (10 mm x 3 mm x 2 mm). Results: Statistically significant differences were observed in bone osteoblastic activity in tibia defects among the groups (p<0.05). Conclusion: Bone regeneration was significantly improved in groups where MSCs were used in combination with DMB and PRP.   How to cite this article: Erdoğmuş Z, Gülsün B. Aims: This study was aimed to find out anti-fertility effects of Andrographis paniculata (AP) plant. Study design & Methodology: A prospective case control animal study with 85 female and 30 male rats (Total 115 rats) was done in the Department of Pharmacology of Dhaka Medical College and Bangladesh Medical College, Dhaka, Bangladesh from July 2002 to December 2003. The total 85 female rats were grouped into case study or exposure group (N=60 female rats) exposed to water extract of AP & control or non exposure group (N=25 female rats, 30 male rats also considered as control group) Exposure group (water extract) of female were again subdivided in 3 groups according to duration of (A.P) exposure (N=20 in each group) e.g. 4 weeks, 6 weeks and 8 weeks. Mating schedules were done after the completion of scheduled duration of exposure with A.P (Dose was 1 gm/kg). Results: In group I percentage of infertility was 33.33% in 4 weeks exposure, 50% in 6 weeks exposure & 100% in 8 weeks exposure respectively. In control group (Group II) percentage of infertility was 0%. In case study group of female rats the value of FSH was 1.20 1U/L (4 weeks), 1.12 1U/L (6 weeks), 1.00 1U/L (8 weeks), LH 0.78 1U/L (4 weeks), 0.70 1U/L (6 weeks), 0.64 1U/L (8 weeks), Estrogen 45.30 pg/ml (4 weeks), 44.80 pg/ml (6 weeks), 44.20 pg/ml (8 weeks) and Progesterone 4.84 nmol/L (4 weeks), 4.72 nmol/L (6 weeks) and 3.80 nmol/L (8 weeks). In non exposure group the value of FSH was 1.23 1U/L, LH 0.80 1U/L, Estrogen 47.05 pg/L and Progesterone 5.50 nmol/L. In exposure group all the values were lesser than the normal hormonal value. Conclusion: This study suggests that due to lower level of hormone, female rats have promising percentage of infertility with AP. Further study is needed with rat as well as clinical trial with human being. Nusantara Bioscience 12: 55-58. Menopause is the transition from productive to non-productive times due to reduced estrogen and progesterone and has an impact on improving cardiovascular problems. The fulfillment of estrogen needs in menopause can use a type of phytoestrogens from black soybeans. The purpose of research is to prove the administration of black soy extract (Glycine soja) can increase the levels of estradiol in the rat blood (Rattus norvegicus) ovariectomy. Design research uses post-test only control group design. Twenty-five female rats were randomly divided into 5 groups (n = 5 tails per group), negative controls, positive and 3 treatments. Ovariectomy was performed in a group of positive controls and treatment. Thirty-day post ovariectomy rats were given appropriate group treatment (negative control, positive, and treatment with the administration of black soy extract dose of 50, 100, 150 mg/200  g/day for 30 days). The estradiol test used the Enzyme-linked immunosorbent assay (ELISA). Anova's test results with a significant degree of α = 0.05 established that the p-value of 0.000 was less than α = 0.05, meaning there was a significant effect of giving black soy extract on increased levels of estradiol in the rat blood ovariectomy. Post - Hoc Tukey testing shows increased the highest estradiol levels in the dose treatment group 100 mg/200  g/day. This research proves that the administration of black soy extract (Glycine soja) can increase the levels of estradiol in the rat blood (Rattus norvegicus) ovariectomy.

Abstract. Benhouda A, Benhouda D, Yahia M. 2019. In vivo evaluation of anticryptosporidiosis activity of the methanolic extract of the plant Umbilicus rupestris. Biodiversitas 20: 3478-3483. Umbilicus rupestris (Crassulaceae) is a medicinal plant used in general traditional medicine to cure inflammation and irritation of the skin. The present research is aimed to evaluate the antiparasitic activity of the methanolic extract of the plant URMeOH of U. rupestris against the Cryptosporidium infection in immunocompetent and immunosuppressed rats experimentally infected. Twenty-one female rats were divided into two groups. Control group (group I) and experimental group (Group II). The group I was further divided into three equal groups (normal group infected and immunosuppressed infected group). The experimental group was divided into two immunosuppressed and four equal groups and two immunocompetent infected. Cryptosporidium oocysts were isolated from bovine species stools and used to infect rats. Experimental subgroups received URMeOH two as dose 100mg/kg b.w. and 200 mg/kg b.w. and continued until 15 days. Two weeks after the administration of URMeOH, feces of rats were examined for the detection of Cryptosporidium oocysts by Ziehl-Neelsen and immunofluorescence techniques, the animals were sacrificed; their small intestines were processed and examined for the detection of pathological lesions after histopathological study. In addition, the activity of myeloperoxidase (MPO) was measured in sections of the jejunum. Concerned the results, we observed a statistically significant (P<0.001) increase in the number of oocysts of Cryptosporidium in the stool for sub infected immunosuppressed groups and an increase of MPO activity compared to the corresponding subgroups immunocompetent subgroups. The URMeOH could remove Cryptosporidium oocysts from feces and intestinal sections subgroup infected immunocompetent rats receiving URMeOH. Moreover, the oocysts were significantly reduced in all other subgroups experimental infected compared to infected control subgroups. Intestinal sections in all subgroups received URMeOH revealed a more or less normal architecture. In addition, the reduction of MPO activity level was also detected in all experimental subgroups.

Imidacloprid (IMI), a newer neonicotinoid insecticide, induces oxidative insult to hepatocytes due to the formation of reactive metabolites during hepatic metabolism. The present study aimed to determine the potentiating effect of arsenic (As) on IMI-induced hepatic damage in Wistar rats. Rats, randomly divided into eight groups with six in each, were subjected to daily oral administration for 28 days. Group I served as control; group II received IMI at the dose rate of 16.9 mg/kg body weight; groups III, IV, and V received As at the dose rate of 50, 100, and 150 ppb, respectively, in drinking water; groups VI, VII, and VIII received both IMI (16.9 mg/kg) and As in drinking water at the rate of 50, 100, and 150 ppb, respectively. Repeated oral administration of IMI or As resulted in significant ( p < 0.05) elevation of plasma phosphatases, transferases, hepatic malondialdehyde, and advanced oxidation protein product levels, but significantly ( p < 0.05) decreased levels of total proteins, thiols, and activities of antioxidant enzymes that indicate oxidation-induced hepatotoxicity. These findings were further corroborated by histological alterations in hepatic tissue of IMI or As-administered rats. The coadministration of both IMI and As in rats produced more severe alterations in these parameters in hepatic tissue. Reduced antioxidant indices and increased hepatic damage biomarkers with pronounced histopathological alterations in hepatic tissue after combined exposure to toxicants indicate potentiating toxic effect of As on IMI-induced hepatotoxicity.

The prevalence of parasitic infections such as malaria, which leads to decrease in hematological indices, the major cause of anemia, constitutes a serious health challenge in many developing countries such as Nigeria. This study investigated the effect of methanol seed extract of Aframomum melegueta on selected hematological indices of 2,4-dinitrophenylhydrazine (2,4-DNPH)-induced anemic rats model. The toxicity study and qualitative phytochemical screening of the extract were carried out using standard procedure. Twenty Wistar rats were grouped into five of four rats each (n = 4). Group I: Normal control; Group II: Negative control; Group III: administered 20 ml/kg b.w. of Astifer (Standard drug); Group IV and V were administered 200 and 400 mg/kg b.w. of the extract, respectively. The animals of Groups II to V were induced with 2,4-dinitrophenylhydrazine (20 mg/kg b.w.) once daily for seven consecutive days; their blood samples were collected by ocular puncture into heparinized capillary tubes for hematological analysis and animals with packed cell volume (PCV) ≥ 30% reduction were considered anemic for the study. The result of the qualitative phytochemical analysis showed that the methanol extract tested positive to alkaloids, carbohydrate, saponins, flavonoids, steroids, terpenoids, and anthraquinones. Acute toxicity and lethality studies on methanol extract showed an oral LD50 equal or less than 5000 mg/kg b.w. in mice. The rats administered 20 ml/kg b.w. of Astifer showed significant (P < 0.05) increase in PCV, hemoglobin, red blood cell (RBC) count, white blood cell (WBC) count, and neutrophils count compared with the normal control animals. The rats administered 200 mg/kg b.w. of A. melegueta showed significantly (P < 0.05) higher PCV and WBC count; non significant (P > 0.05) decrease in hemoglobin count, RBC, platelet, neutrophils and lymphocyte count compared with the normal control animals. The rats administered 400 mg/kg b.w. of A. melegueta showed significant (P < 0.05) increase in hemoglobin, platelet, erythrocyte sedimentation rate (ESR) and neutrophils count; non significant (P > 0.05) lower PCV, RBC, WBC count, and lymphocyte count compared with anemic rats administered with 0.3 ml of normal saline. It can be concluded that Aframomum melegueta seed has beneficial immunological and hematological properties in Wistar rats and possessed erythropoietic potentials at minimal dose that support its use for treating anemia.

Objective: This study aimed to verify the effect of oral siper-metrin exposure to decrease serum estradiol 17b levels and increased malondialdehyde (MDA) in the uterus level of female Wistar strain rats (Rattus norvegicus).Materials and Methods: The method of this study was true experimental post test only control group in vivo using 24 female rats, divided into 3 groups treated by administering a dose of 5, 10 and 20 mg/kg sipermetrin for 28 days and one control group. Then blood samples were taken from the heart for measurement of serum estradiol 17b levels by ELISA and uterine organs were taken for measurement of Malondialdehyde (MDA) with spectro-photometry method.Results: The results of the measurement of serum estradiol 17b and uterus malondialdehyde (MDA) levels of female Wistar strain rats (Rattus norvegicus) showed an opposite pattern, where there was a decline in serum estradiol 17b levels and an increase in uterus malondialdehyde (MDA) level. There was a significant difference (p=0.000<alpha) in 17b estradiol serum and uterus Malondialdehyde (MDA) levels of female rats between control group and group exposed to sipermetrin treatment for 28 days.Conclusions: Oral sipermetrin exposure can decrease serum levels of estradiol 17b and increase uterine levels of malondi-aldehyde (MDA) of female Wistar strain rats (Rattus norvegicus).

<p>The objective of the present work was investigating the preliminary phytochemical screening and hepatoprotective activity of methanol extract of the leaves of <em>Artocarpus hirsutus</em>.<strong></strong></p><p>Group I served as vehicle control, Group II served CCL<sub>4 </sub>(2ml/kg, s.c.), Group III served as standard Silymarin (50 mg/kg, p.o.)  Group IV and V served as methanolic extracts of <em>Artocarpus hirsutus</em><strong> </strong>(MEAH) at the dose level (250 and 500 mg/kg, p.o.). The degree of protection was determined by measuring level biochemical marker like alanine aminotransferase (ALT), aspartate aminotransferase (AST), alkaline phosphatase (ALP), direct bilirubin, total bilirubin and Cholesterol. The histopathology study also showed the hepatic protection of extracts. The preliminary phytochemical screening was performed to find out the phytoconstituents responsible for the activity.<strong></strong></p><p>The marker biochemical level such as ALT, AST, ALP, Direct bilirubin, Total bilirubin and Cholesterol were significantly raised in CCL<sub>4 </sub>treated rats when compared with the normal group (p&lt;0.05), but the MEAH (500 mg/kg, bw) treated rats exhibited maximum depletion. The histopathology study also showed the hepatic protection of extracts. Preliminary phytochemical screening showed the presence of glycosides, flavonoids, Tannins, triterpenoids, carbohydrates and steroids.</p><p>The results of <em>in vivo</em> hepatoprotective activity showed that the methanol extract of <em>Artocarpus hirsutus</em><strong> </strong>exhibit significant hepatoprotective activity. This might be due to flavonoids and tannins; which was confirmed their presence in phytochemical tests.  </p>

The study was conducted to investigate the effects of caffeine treatment during organogenesis period to the fetal birth weight, using rat (Rattus norvegicus) as the animal model. Thirty-six primipararat obtained from Unit Pengembangan Hewan Percobaan, Gadjah Mada University (UPHP-GMU), 3 month old, 165-200 g body weight, were divided into 6 groups, consisted of 6 rats each. Six of the ratshave been selected based on the estrous cycles, and only rat with regular estrous were use for theexperiment. The rat then were mated, and during day 6-14 of the pregnancies were treated orally withcaffeine diluted in aquadest in dosage: placebo (1 cc aquadest) for group I (control), and 5.4, 10.8, 16.2,21.6, and 27 g/200 g body weight/day for treatment groups II-VI respectively. The pregnant rat bodyweights were determined at day 6 of pregnancies for calculating the caffeine treatment dosages. At day 20thof the pregnancies all of the pregnant rats were caesarotomized, and all of the fetuses were removed and weighed. The results showed that all of the treatment groups have significantly lower birth weightcompare to the groups control group. More over, fetal obtained from the treatment groups also showedserious subcutaneous hemorrhagic.Keywords: organogenesis, Rattus norvegicus, birth weight
