Obesity is a chronic disorder of global prevalence and associated with morbidity and mortality. This pathology is a real public health problem. The work was undertaken to evaluate the antiobesity efficacy of aqueous extract of Kemite in cafeteria diet induced obese Wistar rats for a period of 28 days. Aqueous extract of Kemite (AEK) was prepared by hot extraction method. Female Wistar rats weighing 124-170 g were divided into different groups i.e. Normal control, cafeteria control and aqueous extract of Kemite at dose of 200 mg/kg bw. The antiobesity activity is estimated in terms of body weight gain, food intake, serum triglycerides (TG), Total cholesterol (TC), LDL cholesterol (LDL-C), HDL cholesterol (HDL-C), VLDL cholesterol (VLDL-C), blood glucose (BG), ASAT and ALAT activities, atherogenic index, coronary risk index and liver and fat pad weights. Results showed Cafeteria diet fed rats for 28 successive days significantly increased the body weight, food intake, ASAT and ALAT activities, liver and fat pad weights, atherogenic index, coronary risk index TG, TC, LDL, VLDL, BG and not influenced HDL levels. Rats treated with extract for 28 successive days along with cafeteria diet reversed the effects induced by cafeteria diet. In conclusion, this study revealed that AEK may be a natural and safe remedy for the prevention and control of obesity.

Objective:Traditionally prepared infusions and decoctions are commonly used in the management of diabetes mellitus, in southern Nigeria; one of such is the aqueous extract of the sclerotia of Pleurotus tuberregium (“usu” milk). In this study, the effects of the extract on the body weights, tissue/ organ weights, fasting blood glucose, blood/plasma lipid profiles and atherogenic indices were investigated in normal and alloxan-induced diabetic rabbits.Methods:Diabetes mellitus was induced by the injection of alloxan (120 mg/kg body weight) via the marginal ear vein. The extract was administered orally at 100, 200 and 300 mg/kg to normal and diabetic rabbits; while metformin was administered at 50 mg/kg. The crude extract was analyzed by gas chromatography, coupled to flame ionization detector.Results:Thirty-one known flavonoids were detected, consisting mainly of isoquercetin (28.5%), luteolin (24.3%), quercetin (18.8%) and kaempferol (11.3%). Sitosterol (82.0%) and stigmasterol (12.5%) were the most abundant of the seven phytosterols detected. Compared to the diabetic control, the treatment significantly (p<0.05) lowered the weights of the kidney and liver, as well as the levels of blood glucose and triglyceride, plasma VLDL, LDL and non-HDL cholesterol, atherogenic index of plasma, cardiac risk ratio, atherogenic coefficient and Castelli’s risk index II. It, however, significantly (p<0.05) increased plasma HDL cholesterol, without significantly affecting blood total cholesterol levels.Conclusion:This study showed that the extract was hypoglycemic, and improved lipid profile and atherogenic indices, thus highlighting its cardioprotective potential, thereby supporting its use in the management of diabetes mellitus.

The toxicological effect of the aqueous extract of fresh, dried and boiled berries of Solanum aculeastrum Dunal at 1, 10 and 25 mg/kg body weight was investigated in male Wistar rats for 28 days. The parameters used were the body weight of the animals and absolute weights of the organs, haematological parameters, renal and liver functional endpoints. The animals gained appreciable weight and showed no signs of clinical toxicity. The dried (DB), boiled dried (BDB), fresh (FB) and boiled fresh berry (BFB) extracts reduced (p < .05) the heart-, liver-and spleen-body weight ratio of the animals whereas that of the lung was not altered. The kidney and testes-body weight ratios were specifically altered by the different extract. All these were not accompanied by any histomorphological changes. The extracts did not alter (p > .05) the levels of RBC, Hb, PCV and albumin of the animals. The platelets were decreased by the DB and FB whereas BFB increased this parameter. The FB and BFB at all the doses also reduced the mean corpuscular haemoglobin (MCH) and mean corpuscular haemoglobin concentration (MCHC) of the animals. With the exception of the FB where the creatinine and chloride levels decreased, other extracts did not alter the level of these kidney parameters. Only FB increased the levels of uric acid and urea. All the extract decreased the serum alanine aminotransferase (ALT) of the animal. The levels of total protein, globulin, total and conjugated bilirubin were not altered by DB and BDB whereas these indices were increased by FB and BFB. The DB and BDB increased the serum alkaline phosphatase (ALP) activity whereas FB decreased the activity of the enzyme. In contrast, DB and BDB decreased the serum aspartate aminotransferase (AST) activity of the animals whereas FB and BFB increased the activity of the enzyme. The FB and BFB also increased the levels of potassium, magnesium and phosphorus of the animals. Overall, the alterations in the biochemical parameters by the various extracts of S. aculeastrum berries at these doses indicated that the normal functioning of these organs may be adversely affected. However, drying and boiling might reduce the toxic effect of the berries.

Purpose: To evaluate the 90 day sub-chronic toxicity of aqueous extract of Gmelina arborea leaves in Wistar rats. Methods: Rats were submitted to repeated daily oral administration of extract (250, 62.5 and 15.62 mg/kg) of Gmelina arborea leaves. The control groups were given distilled water and the rats were monitored for any toxicity symptoms as well as body and organs weights, water and food intake changes. The biochemical, haematological and histolopathological parameters were analysed. Results: The 90 days administration of the aqueous extract did not produce any toxicity signs or mortality. In addition, no significant alteration in water or food intake by the rats was observed. Although there were no changes in the body weights, significant decrease in the weight of the kidneys of the rats was observed at 250 mg/kg. Biological parameters as well as the histopathology of liver and kidneys were not significantly affected. Significant decreases were noted in glucose level at the three dose levels. In addition, significant difference in the levels of transaminases, glucose and platelets were observed. Conclusion: The 90-days subchronic toxicity test on Gmelina arborea did not produce any toxic effects. This confirms the safety of the plant leaves by traditional medicine practitioners. AbstractObjective. The hypothalamic arcuate nucleus proopiomelanocortin (POMC) and neuropeptide Y (NPY) circuitries are involved in the inhibition and stimulation of the appetite, respectively. The aim of this study was to investigate the effects of one-month lasting high-intensity exercise on the POMC mRNA and NPY mRNA expression in the above-mentioned brain structure and appetite and food intake levels.Methods. Fourteen male Wistar rats (250±50 g) were used and kept in the well-controlled conditions (22±2 °C, 50±5% humidity, and 12 h dark/light cycle) with food and water ad libitum. The rats were divided into two groups (n=7): 1) control group (C, these rats served as controls) and 2) exercised group (RIE, these rats performed a high-intensity exercise for one month (5 days per week) 40 min daily with speed 35 m/min. The total exercise time was 60 min. The body weight and food intake were recorded continuously during the experiments.Results. The results showed relative mRNA expression of POMC and NPY estimated in the hypothalamic arcuate nucleus. There were no significant differences in the NPY and POMC mRNAs expression levels and food intake between C and RIE groups.Conclusions. The present data indicate that one-month regular intensive exercise did not alter the levels of NPY and POMC mRNAs expression (as two important factors in the regulation of appetite) in the hypothalamic arcuate nucleus and food intake suggesting that this type of exercise itself is not an appropriate procedure for the body weight reduction.

The objective of this study is to examine the effect of maternal and weaning intact protein- and amino acid-based diets on regulation of food intake, intake regulatory hormones, and body weight in dams and their male offspring. Pregnant Wistar rats were allocated to two groups (n = 12) and were fed either an intact protein diet (IPD) or mixed amino acid diet (AAD) from day 3 of gestation throughout gestation and lactation. Male offspring were weaned to either an IPD or AAD for 18 weeks. Food intake (FI) and body weight (BW) were measured weekly. Results: In dams, the AAD group had lower FI and BW in the post-partum period compared with the IPD group. In pups born to AAD dams, birth weight and BW were lower. However, the percentage of fat and lean mass were not affected. Food intake was influenced by maternal diet and was higher in pups born to IPD dams throughout post-weaning. Short-term FI in response to protein preloads was lower in pups born to AAD dams in 1 h. Fasting plasma concentrations of glucose, insulin, and ghrelin were not influenced by either maternal or weaning diet. However, peptide YY (PYY) was higher in pups born to IPD dams at weaning. Conclusions: The physicochemical properties of proteins fed during pregnancy and lactation had determining effects on the body weight and development of food intake regulatory systems in offspring. Maternal AAD resulted in lower BW in dams and lower birth weight and post-weaning BWs in pups compared with maternal IPD which was consistent with their lower FI.

The CB1 receptor antagonist, rimonabant, affects the endocannabinoid system and causes a sustained reduction in body weight (BW) despite the transient nature of the reduction in food intake. Therefore, in a multiple-dose study, female candy-fed Wistar rats were treated with rimonabant (10 mg/kg) and matched with pair-fed rats to distinguish between hypophagic action and hypothesized effects on energy expenditure. Within the first week of treatment, rimonabant reduced BW nearly to levels of standard rat chow-fed rats. Evaluation of energy balance (energy expenditure measured by indirect calorimetry in relation to metabolizable energy intake calculated by bomb calorimetry) revealed that increased energy expenditure based on increased fat oxidation contributed more to sustained BW reduction than reduced food intake. A mere food reduction through pair feeding did not result in comparable effects because animals reduced their energy expenditure to save energy stores. Because fat oxidation measured by indirect calorimetry increased immediately after dosing in the postprandial state, the acute effect of rimonabant on lipolysis was investigated in postprandial male rats. Rimonabant elevated free fatty acids postprandially, demonstrating an inherent pharmacological activity of rimonabant to induce lipolysis and not secondarily postabsorptively due to reduced food intake. We conclude that the weight-reducing effect of rimonabant was due to continuously elevated energy expenditure based on increased fat oxidation driven by lipolysis from fat tissue as long as fat stores were elevated. When the amount of endogenous fat stores declined, rimonabant-induced increased energy expenditure was maintained by a re-increase in food intake.

AbstrakPengurangan durasi tidur menurunkan kadar leptin dan meningkatkan kadar ghrelin sehingga merangsang nafsu makan dan meningkatkan kemungkinan terjadinya obesitas pada manusia. Pada tikus akan menyebabkan peningkatan asupan makanan tetapi terjadi penurunan berat badan yang disebabkan karena aktivitas yang tinggi pada tikus. Tujuan penelitian ini adalah untuk melihat pengaruh kurang tidur 24 jam, 48 jam dan 72 jam terhadap berat badan pada tikus Wistar jantan. Jenis penelitian adalah true experimental research dengan rancangan randomized post control group terhadap 14 ekor tikus Wistar yang dibagi atas kelompok kontrol, kelompok perlakuan 24 jam, 48 jam dan 72 jam. Tikus dikondisikan mengalami paradoxycal sleep deprivation dengan metode modified multiple platform. Asupan makanan diberikan ad libitum dan berat badan diukur setelah pengurangan durasi tidur selama 24 jam, 48 jam, dan 72 jam.Analisis data menggunakan uji Saphiro-Wilk Test dan One-Way ANOVA. Rerata berat badan setelah pengurangan durasi tidur 24 jam adalah 193,6±17,9 gram; setelah 48 jam 179,6±17,3 gram; dan setelah 72 jam 176,7±15,9 gram dibandingkan dengan kontrol 219.6±11,3 gram. Pengurangan durasi tidur 48 jam dan 72 jam dibandingkan dengan kontrol bermakna (p<0,05). Dapat disimpulkan bahwa terjadi penurunan berat badan pada pengurangan durasi tidur selama 48 jam dan 72 jam.Kata kunci: kurang tidur, berat badan, tikus wistarAbstractSleep deprivation lowers level of leptin and increases level of ghrelin which stimulates appetite and increases the likelihood of obesity in humans. The objective of this study was to examine the effect of sleep deprivation 24 hours, 48 hours and 72 hours on body weight in male Wistar rats. This type of research was a true experimental design research with post randomized control group on 14 Wistar rats were divided into control group, treatment group 24 hours, 48 hours, and 72 hours. Rats conditioned paradoxycal sleep deprivation experienced by the modified multiple platform method. Given ad libitum food intake and body weight were measured after sleep deprivation for 24 hours, 48 hours, and 72 hours. Analysis of the data using the Shapiro-Wilk Test and One-Way ANOVA. The mean of body weight after 24 hour sleep deprivation was 193.6±17.9 g, after 48 hours was 179.6±17.3 g, and after 72 hours was 176.7±15.9 g compared with control was 219.6±11.3 g. Sleep deprivation 48 hours and 72 hours compared with controls was significant (p<0.05). Background: G protein-coupled bile acid receptor (TGR5) is involved in a number of metabolic diseases. The aim of this study was to identify the role of TGR5 after Roux-en-Y gastric bypass (GBP). Methods: Wild type and TGR5 knockout mice (tgr5-/-) were fed a high-fat diet (HFD) to establish the obesity model. GBP was performed. The changes in body weight and food intake were measured. The levels of TGR5 and peptide YY (PYY) were evaluated by RT-PCR, Western blot, and ELISA. Moreover, the L-cells were separated from wild type and tgr5-/- mice. The levels of PYY in L-cells were evaluated by ELISA. Results: The body weights were significantly decreased after GBP in wild type mice (p<0.05), but not tgr5-/- mice (p>0.05). Food intake was reduced after GBP in wild type mice, but also not significantly affected in tgr5-/- mice (p>0.05). The levels of PYY were significantly increased after GBP compared with the sham group (p<0.05); however, in tgr5-/- mice the expression of PYY was not significantly affected (p>0.05). After INT-777 stimulation in L-cells obtained from murine intestines, the levels of PYY were significantly increased in L-cells tgr5+/+ (p<0.05). Conclusion: Our study suggests that GBP up-regulated the expression of TGR5 in murine intestines, and increased the levels of PYY, which further reduced food intake and decreased the body weight.

AbstractPlasma xanthine oxidoreductase (XOR) activity is high in metabolic disorders such as diabetic mellitus, obesity, or overweight. Thus, this study investigated whether the XOR inhibitor, topiroxostat, affected body weight. Male db/db mice were fed standard diets with or without topiroxostat for 4 weeks. Body weight and food intake were constantly monitored, along with monitoring plasma biochemical markers, including insulin and XOR activity. Additionally, hepatic hypoxanthine and XOR activity were also documented. Single regression analysis was performed to determine the mechanism. Topiroxostat treatment suppressed weight gain relative to the vehicle without any impact on food intake. However, the weight of fat pads and hepatic and muscle triglyceride content did not change. Topiroxostat decreased the plasma uric acid and increased hepatic hypoxanthine in response to the inhibition of XOR activity. Plasma ketone body and free fatty acid were also increased. Moreover, fat weight was weakly associated with plasma XOR activity in the diabetic state and was negatively associated with ketone body by topiroxostat. These results suggested that topiroxostat amplified the burning of lipids and the salvage pathway, resulting in predisposing the body toward catabolism. The inhibition of plasma XOR activity may contribute to weight loss.

In the present study, the methanolic extract of Moringa oleifera leaves (MEMOL) was evaluated for antiobesity activity in rats. The antiobesity potential of MEMOL was studied against high fat diet-induced obesity (HFD) in rats. In this study, chronic administration of HFD in rats produced hypercholesterolemia (116.2 ± 0.27 mg/dL), which led to an increase in the body weight (225 gr), total cholesterol, triglycerides (263.0 ± 4.69 mg/dL), and attenuation in the levels of HDL (34.51 ± 2.20 mg/dL) as well as changes in body temperature of animals. Treatment of obese rats with MEMOL for 49 days resulted in a significant (P<0.001) change in body weight, total cholesterol, triglycerides, and LDL level along with a significant (P<0.001) increase in body temperature as compared to the HFD-induced obesity. MEMOL treated rats also showed a significant decrease in the level of liver biomarkers, organ weight, and blood glucose level. Further, rats treated with MEMOL (200 mg and 400 mg/kg) show reduced atherogenic index (1.7 ± 0.6 and 0.87 ± 0.76). The results indicate that the rats treated with Moringa oleifera (MO) have significantly attenuated the body weight without any change in the feed intake and also elicited significant thermogenic effect and to act as hypolipidemic and thermogenic property in obesity related disorders.
