Purpose: To evaluate the 90 day sub-chronic toxicity of aqueous extract of Gmelina arborea leaves in Wistar rats. Methods: Rats were submitted to repeated daily oral administration of extract (250, 62.5 and 15.62 mg/kg) of Gmelina arborea leaves. The control groups were given distilled water and the rats were monitored for any toxicity symptoms as well as body and organs weights, water and food intake changes. The biochemical, haematological and histolopathological parameters were analysed. Results: The 90 days administration of the aqueous extract did not produce any toxicity signs or mortality. In addition, no significant alteration in water or food intake by the rats was observed. Although there were no changes in the body weights, significant decrease in the weight of the kidneys of the rats was observed at 250 mg/kg. Biological parameters as well as the histopathology of liver and kidneys were not significantly affected. Significant decreases were noted in glucose level at the three dose levels. In addition, significant difference in the levels of transaminases, glucose and platelets were observed. Conclusion: The 90-days subchronic toxicity test on Gmelina arborea did not produce any toxic effects. This confirms the safety of the plant leaves by traditional medicine practitioners. Obesity is a chronic disorder of global prevalence and associated with morbidity and mortality. This pathology is a real public health problem. The work was undertaken to evaluate the antiobesity efficacy of aqueous extract of Kemite in cafeteria diet induced obese Wistar rats for a period of 28 days. Aqueous extract of Kemite (AEK) was prepared by hot extraction method. Female Wistar rats weighing 124-170 g were divided into different groups i.e. Normal control, cafeteria control and aqueous extract of Kemite at dose of 200 mg/kg bw. The antiobesity activity is estimated in terms of body weight gain, food intake, serum triglycerides (TG), Total cholesterol (TC), LDL cholesterol (LDL-C), HDL cholesterol (HDL-C), VLDL cholesterol (VLDL-C), blood glucose (BG), ASAT and ALAT activities, atherogenic index, coronary risk index and liver and fat pad weights. Results showed Cafeteria diet fed rats for 28 successive days significantly increased the body weight, food intake, ASAT and ALAT activities, liver and fat pad weights, atherogenic index, coronary risk index TG, TC, LDL, VLDL, BG and not influenced HDL levels. Rats treated with extract for 28 successive days along with cafeteria diet reversed the effects induced by cafeteria diet. In conclusion, this study revealed that AEK may be a natural and safe remedy for the prevention and control of obesity.

The tubers of Hydnophytum formicarum Jack. have been used for the treatment of rheumatism, liver and intestinal diseases in the traditional medicine of Vietnam. The 50% ethanolic extract of H. formicarum (EEHF) has been proved anti-inflammatory and analgesic effects as well as hepatoprotective activity at the oral doses of 100 and 200 mg/kg in Swiss albino mice. The aim of this work was to study on the subacute oral toxicity of the EEHF at the doses of 100 and 200 mg/kg for 30 and 60 consecutive days in both sexes of mice. The observations were undertaken on the mortality, the body weights, the toxic signs, hematological and biochemical parameters. At the end of the experiment, the histopathology of liver and kidneys was studied. The results showed that the 100 mg/kg of EEHF did not cause any toxic signs in mice. There was no significant difference of hematological and biochemical parameters between the treated animals and the controls. Histopathological examinations of livers and kidneys showed no significant difference suggesting no morphological disturbances in treated mice. In conclusion, the results indicated that the EEHF did not cause any subacute toxicity in mice at the dose of 100 mg/kg, providing the evidence of the safety of health products from this medicinal plant.

Aim and Objectives: Demonstrating the exact quantity of Moringa oleifera (MO) that will cure any hepatocyte diseases and the one that can harm the hepatocytes of the Adult Wistar rats. Methods: Twenty (20) adult Wistar rats (both sex) were used for the study (caged based on same sex to avoid mating and pregnancy) and were randomly assigned into four groups (n=5); A-Control, B-250 mg/body weight (BW) of MO, C-500 mg/BW of MO, and D-750 mg/BW of MO orally. Result: The crude aqueous extract of MO Lam, shows insignificant increased in BW at the 1st week of administration which latter dropped little by little doing the weeks of the administration in groups (B, C, and D) rats, by comparing (p<0.05) to the control group after MO administration, the organ (Kidneys) shows a significant difference between the kidneys (left and right kidneys) in relation to the control group rats. MO increases the weight of the animals morphologically by comparing the weight of the animals before and after administration. Histological sections shows a normal Glomerulli, Peri-Glomerular Space, Convoluted Tubules, and Interstitium, after administration of the Crude Aqueous Extract of MO lame in terms with the control group rats. Conclusion: MO is a good herb that has no damage effect on the body and hepatocytes but of more beneficial.

AbstractBackgroundHabb-e-Suranjan(HS) is a Unani pharmacopoeial formulation used for treatment of rheumatoid arthritis. The present study was carried to evaluate the toxic potential of HS in albino Wistar rats.MethodsThe subchronic was carried out at the dose levels of 2440 mg/kg of body weight in both sexes. The rats were observed carefully for any behavioral change for next 24 hours (hr) after the administration of the drug and daily twice thereafter till the completion of experimentation. Water/feed consumption and body weight were recorded on weekly basis. The rats were sacrificed on 91st day after overnight fasting. Blood was collected for hematological and biochemical analysis. The rats were dissected and organs were observed for any morphological changes. Organ samples were collected for histological studies.ResultsThere were no incidences of mortality/morbidity found in any of the treated rats.HScaused no significant difference in the body weights as well as relative organs weights of treated groups when compared with the control groups. There were no significant changes found in any biochemical, hematological and histopathological parameters that could indicate any signs of toxicity.ConclusionsThe study suggests thatHSis safe and did not induce any adverse effect in rats.

The present study aimed to investigate the sub-chronic toxicity of “Phong thap dan” (PTD) tablets through oral administration in experimental animal. The sub-chronic toxicity was evaluated by the WHO recommendation in Wistar rats at doses of 0.72 g/kg/day (equal to recommended human dose) and 2.16 g/kg/day (3 times as high as recommended human dose). In the sub-chronic experimental group, the PTD was administered orally daily for 8 consecutive weeks. In the evaluation of sub-chronic toxicity, there were no behavioral and physiological change or sign of toxicity. The result of the hematological and biological parameters after administration of PTD tablets showed no change. The histopathology analysis of livers and kidneys indicated that no significant difference was observed between the exposed and unexposed rat groups. In conclusion, “Phong thap dan” tablets did not produce sub-chronic toxicity in Wistar rats.

Background: G protein-coupled bile acid receptor (TGR5) is involved in a number of metabolic diseases. The aim of this study was to identify the role of TGR5 after Roux-en-Y gastric bypass (GBP). Methods: Wild type and TGR5 knockout mice (tgr5-/-) were fed a high-fat diet (HFD) to establish the obesity model. GBP was performed. The changes in body weight and food intake were measured. The levels of TGR5 and peptide YY (PYY) were evaluated by RT-PCR, Western blot, and ELISA. Moreover, the L-cells were separated from wild type and tgr5-/- mice. The levels of PYY in L-cells were evaluated by ELISA. Results: The body weights were significantly decreased after GBP in wild type mice (p<0.05), but not tgr5-/- mice (p>0.05). Food intake was reduced after GBP in wild type mice, but also not significantly affected in tgr5-/- mice (p>0.05). The levels of PYY were significantly increased after GBP compared with the sham group (p<0.05); however, in tgr5-/- mice the expression of PYY was not significantly affected (p>0.05). After INT-777 stimulation in L-cells obtained from murine intestines, the levels of PYY were significantly increased in L-cells tgr5+/+ (p<0.05). Conclusion: Our study suggests that GBP up-regulated the expression of TGR5 in murine intestines, and increased the levels of PYY, which further reduced food intake and decreased the body weight.

Goldthioglucose-obese mice cannot adjust their food intake to meet the increased energy requirements due to cold. At all ambient temperatures above 15°C the spontaneous running activity of these animals is less than that observed for nonobese controls. Activity of obese mice is maximal at 19°C and minimal at 15°C or lower. Body weights decrease during exposure to cold. In contrast to that of obese mice, running activity of nonobese controls is maximal at an ambient temperature of 25°C but nearly ceases at 15°C or lower. The food intake of these animals increases in the cold and remains elevated even at temperatures at which activity decreases. The body weight of nonobese controls is either maintained constant or increases during exposure to cold air.

The aim of this research was to investigate the effects of extract ethanol of pegagan (Centella asiatica) after treated with restraint stress by measuring the eight arms radial maze performance of rats. Pre-test and post-test controlled groups design was applied in this research. Male Wistar rats (Rattus novergicus) with the mean age of 8 weeks and the body weight of 150-200 grams which used for sample were divided randomly into 4 groups, each group consisted of 6 rats, i.e. (i) control group (without any treatment), (ii) stress group (it was given by restraint stress for 2 hours/day for each rat), (iii) pegagan group (it was given by 0.3 mg/g BW/day/rat extract ethanol of pegagan), and (iv) pegagan and stress group (it was given by 0.3 mg/g BW/day/rat extract ethanol of pegagan and restraint stress for 2 hours/day for each rat). The treatments were given for 21 days. Within 12 days for each pre-treatment and post-treatment, a test on the eight arm radial maze was conducted on individual rat to observe its performance. The assessment of rat performance in the eight arms radial maze test was conducted based on error type B. Kruskal-Wallis and Mann-Whitney tests with SPSS for Windows 16 version were applied to analyze statistically the difference between four groups. Kruskal-Wallis test was used to show the significant performance level difference between four groups of rats with p=0.001, while Mann-Whitney test was used to determine the significant difference between stress group and pegagan group (p=0.001), also stress group and pegagan and stress group (p=0.001). The result of research showed that there was no significant difference between control group and stress group (p=0.051), control group and pegagan group (p=0.143), control group and pegagan and stress group (p=0.143), also pegagan group and pegagan and stress group (p=0.952). It was concluded that extract ethanol of pegagan improved the performances of rats on the eight arms radial maze after treated with restraint stress.

Background. Herbal medication is a worldwide and ancient practice, mostly in developing countries, where a large part of the population is involved in this practice. Hence, studies must be conducted to evaluate their safety and efficiency to avoid or prevent toxicological risks due to their usage. In Togo, Carissa spinarum is a medicinal plant belonging to Apocynaceae family, used as an aphrodisiac or to heal some ailments including malaria, sickle cell anemia, hypertension, pain, and asthma. Notwithstanding its several ethnomedicinal benefits, just a few toxicological data associated with its chronic use are available. Objective. Therefore, this study aims to assess the toxicity of an ethanolic root extract of Carissa spinarum in Wistar rats. Methods. The 90-day oral toxicity process following OECD TG 408 guidelines is used. Male Wistar rats received Carissa spinarum root hydroethanolic extract at 500 and 1000 mg/kg for 90 days by oral gavage. Body weight changes, hematological and blood biochemical parameters, organ weight changes, malondialdehyde as a lipoperoxidation marker expressed according to tissue proteins, and histopathology of vital organs were assessed. Results. No signs of toxicity or mortality were observed during the 90 days experiment. Hematological parameters have not shown any treatment-related abnormalities. According to biochemical parameters, an increase in the chloride ion level was observed at 1000 mg/kg p < 0.01 . There was no significant difference between the treated groups and the control group concerning the malondialdehyde concentration, body weight, and organ relative weight. No changes in necropsy and histopathology of vital organs associated with extract treatment were observed. Conclusion. The results indicated that an ethanolic root extract of Carissa spinarum does not cause adverse effects, which can lead to Wistar rats’ death after 90-day oral administration at 500 and 1000 mg.

Abstract Modern treatment interventions for kidney stones are wrought with side-effects, hence the need for alternative therapies such as plant-based medicines. We have previously documented through in vitro studies that statistically optimized aqueous extract of Tribulus terrestris (Zygophyllaceae family) possesses antiurolithic and antioxidant potential. This provides strong scientific foundation to conduct in vivo efficacy and preclinical safety studies to corroborate and lend further proof to its ability to prevent and cure kidney stones. The preventive and curative urolithiatic efficacy in experimentally induced nephrolithiatic Wistar rats, along with preclinical toxicity was evaluated following oral administration of statistically optimized aqueous extract of T. terrestris. Treatment showed augmented renal function, restoration of normal renal architecture and increase in body weight. Microscopic analysis of urine revealed excretion of small sized urinary crystals, demonstrating that treatment potentially modulated the morphology of renal stones. Tissue enzymatic estimation affirmed the antioxidant efficacy of treatment with reduced free radical generation. Significant upregulation of p38MAPK at both the gene and protein level was noted in hyperoxaluric group and interestingly treatment reversed it. Acute oral toxicity study established the Median Lethal Dose (LD50) to be greater than 2000 mg/kg body weight (b.wt.) No observed adverse effect level (NOAEL) by repeated oral toxicity for 28 days at 750 mg/kg b.wt. was noted. This study lends scientific evidence to the safe, preventive and curative potential of statistically optimized aqueous extract of T. terrestris at a dose of 750 mg/kg b.wt. and suggests that the extract shows promise as a therapeutic antiurolithic agent.
