In Vitro Antimicrobial and Antioxidant Activities of Organic and Aqueous Extracts of Bacillus clausii KP10

Humans and plants are experiencing different kinds of diseases. Many of these diseases are mainly caused by microorganisms like fungi, bacteria and viruses that adversely affect their hosts. Bioactive compounds produced by bacteria can be used to treat the infections caused by these microorganisms. Free radicals that can cause serious damage to the cell can be repressed by antioxidants that are produced by bacteria as secondary compounds. The current case study was therefore planned to investigate the antimicrobial and antioxidant potentials of the organic and aqueous extracts of Bacillus clausii KP10 isolated recently by our group from hot springs of Tatta Pani, Poonch, Azad Kashmir. The antimicrobial activities were assessed using well diffusion method against two Gram positive bacteria; B. subtilis, and S. aureus, and two Gram negative bacteria; E. coli and P. multocida and three fungal strains; A. niger, A. parasiticus and A. ustus. Antioxidant activities of the extracts were done by DPPH free radical scavenging assay. In our experiments the maximum zones of inhibition were shown by ethanolic extract against P. multocida (21 mm), S. aureus (13 mm) and methanol soluble fraction showed zone of inhibition against A. ustus (21 mm) as compared to the standard ciprofloxacin for bacteria and as antifungal agents with terbinafine. In our studies n-hexane fraction of B. clausii KP10 showed maximum 88.67% antioxidant activity. Finally, results were analyzed statistically through analysis of variance (ANOVA). Hence it is proved that the bioactive fractions obtained from B. clausii KP10 are good source of antimicrobials and antioxidants and they can be used as medicines in future for the respective treatments.

This study aimed to investigate the phytochemical, antimicrobial, and antioxidant activities of Carica papaya L. seeds extracts collected from Al Hudaydah city, Yemen (Latitude: 14°47′52″ N Longitude: 42°57′16″ E) during the months of January and February 2021. The seed was extracted with different solvents by the cold percolation method. The disk diffusion method was employed to assess the antibacterial activity of the seed extract against six bacterial and four fungal strains. Spectrometric methods were employed to calculate the total alkaloids, anthocyanin, flavonoid, phenolic, quinones, saponin, steroids, terpenoid, tannin, and phenols contents, as well as the antioxidant activities. Antibacterial and antifungal activity tests exhibited that the selected microorganisms are highly sensitive to the ethanolic and methanolic extracts of C. papaya L. seeds, followed by chloroform, water, and n-hexane extracts. The samples also demonstrated a significant DPPH, FRAP, and APTS radical scavenging activity. Additionally, the preliminary phytochemical analysis revealed the presence of flavonoid, terpenoid, saponin, alkaloids, steroids, quinones, anthocyanin, tannin, and phenols, all of which potentially contribute to the antimicrobial activities of C. papaya L. seeds.

The propolis, an extremely complex resinous material, exhibits valuable pharmacological and biological properties, mainly attributed to the presence of polyphenols. The composition of propolis depends on time, vegetation, and the area of collection. Total flavonoid and polyphenol contents of aqueous extracts of propolis samples from different areas of Algeria, determined by using aluminum chloride and Folin–Ciocalteu colorimetric methods, were in the range of 3.047 ± 0.004–5.273 ± 0.013 mg/g and 96.833 ± 0.027–458.833 ± 0.0005 mg/g crude extract of propolis, respectively. This study examined the antioxidant and antimicrobial activities of propolis. Aqueous extracts of propolis were obtained in order to evaluate their antioxidant activities by using 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical scavenging, β-carotene and electrochemical assays. All tested propolis samples had relatively strong antioxidant activities, which were also correlated with the total polyphenol and flavonoid content present. The percentage of inhibition of lipid peroxidation of linoleic acid emulsion during 24 h varied between 86.04 ± 0.42 and 90.60 ± 3.77% among the tested samples. The highest DPPH radical scavenging activity was observed by ABAL (Ain Abassa Aqueous Extract) with IC50 = 8.49 ± 5.07 10−5 μg/ml, and the lowest was observed by SAL (Setif Aqueous Extract) with IC50 of 21.16 ± 0.0001 μg/ml. The most important antibacterial activity was obtained with Ain Abassa extract; the zones of inhibition obtained for this excerpt vary from 15.22 to 15.5 mm. Followed by the Setif extract with areas of 12.33 to 12.75 mm, the Tizi-Ouzou extract with areas of 10.11 to 11.11 mm. This study will bring an innovation for further studies with regard to the antioxidant and antibacterial properties of the aqueous extracts of propolis. This study corroborates that Algerian propolis is a rich source of natural antioxidants, properties which could be used in the prevention of different diseases, both in humans and in animals.

The aim of this study is to investigate in vitro antioxidant and antibacterial activities of the aqueous and hydroethanolic extracts for aerial parts of Herniaria hirsuta. Extracts were screened for their possible antioxidant activities by three tests: DPPH free radical-scavenging, reducing power, and molybdenum systems. The screening of antibacterial activity of extracts was individually evaluated against sixteen bacteria species using a disc diffusion method. Flavonoids, total phenols, and tannins content were performed for both extracts. It shows higher content in the hydroethanolic extract. The hydroethanolic extract showed a significant antioxidant activity for the three methods studies to the aqueous extract, but nonsignificant results compared to the reference (BHT). However, both extracts have negative effect on the strains studies for the antibacterial activity.


Medicinal plants are play significant impact in the personal medicine for most of the people all over the world as an alternative live saving medicines and most of their medicinal properties are well known for anticancer activity. The different extracts of Holoptelea integrifolia (H.integrifolia) leaves, stem bark and fruits were studied as a potent natural source of antimicrobial, antioxidant and wound healing potential. This work was carried out to evaluate antimicrobial, antioxidant and anti-inflammatory activity of different extracts of H.integrifolia. The antimicrobial activity of the H.integrifolia ethanolic extract was studied against five fungal and bacterial strains by utilizing the agar well diffusion method and MIC. Among several strain, the ethanolic extract of fruit has shown higher antimicrobial inhibition zone as 9.25-16 mm compare to other two extracts of stem and leaves as 10- 13.25 mm and 6-10.2 mm respectively. The antioxidant activities for different extract were also determined by DPPH free radical assay, Hydroxyl Radical Scavenging and Nitric Oxide Radical Scavenging Activity method. The anti-inflammatory activity also estimated basedon formalin induced paw edema method on Wistar albino rats. The different extracts of leaves, stem bark and fruit parts of Holoptelea integrifolia were estimated for in vivo anti-inflammatory activity against the animal model of female Wistar albino rats. The results of anti-inflammatory activity revealed that the Ethanol extracts showed vital and dose-dependent anti-inflammatory effects. Our findings revealed that aerial parts of H.integrifolia contais potential antimicrobial, antioxidant and anti-inflammatory compounds, which expose the medicinal potential of the selected plant could be a significant drug candidates against microbial, oxidative and inflammation-related pathological processes as a future alternative medicine.


Annona muricata is one of the important herbal plant that are widely used to treat antidiabetic, anti-inflammatory, insecticidal, antimalarial, anticancer, antibacterial and antioxidant activities. In the present study, extraction procedure was carried out in few step processes method. The extract contains high percentage of steroid, alkaloid, flavonoid, phenolic and saponin. The extract was used to study the antioxidant activity; antibacterial activities and anticancer activity by standard methods. The antioxidant activity was studied by using radical scavenging DPPH, FRAP and H2O2 method and disc diffusion method. The results of antibacterial activity of the following bacteria’s such as Pseudomonas, Staphylococcus, Bacillus, E.coli shows the maximum zone of inhibition against Staphylococcus aureus which is around 1.7 cm. The anticancer activity was carried out by MTT assay using Hep-G2 as cell line and results are reported in the paper.

The aril (mace) of Myristica fragrans, known as Dok-Chan, is a spice that has long been used for treating stomach discomfort, peptic ulcer, and nausea. It is an ingredient in many remedies in Thai traditional medicine, e.g., Ya-Hom-Thep-Bha-Jit, Ya-Hom-Nao-Wa-Kot, and Ya-That-Bun-Job, which are used to treat dyspepsia and other gastrointestinal tract symptoms. The aqueous and ethanolic extracts of mace were used for all tests. Anti-H. pylori activities were determined by the disc diffusion method and agar dilution. Anti-inflammatory activity was determined by the LPS-induced nitric oxide (NO) inhibition in a RAW264.7 cell line, and cytotoxicity was determined against gastric cancer cell lines (Kato III) using the sulphorhodamine B (SRB) assay. The DPPH radical scavenging and ABTS radical cation decolorization assays were used to determine the antioxidant activities. The result found that the ethanolic extract of mace exhibited antimicrobial activity against H. pylori ATCC 43504 and six clinical strains with MIC values of 125–250 μg/ml. The aqueous extract MICs against H. pylori ATCC reference strain and six clinical strains were 500 μg/ml compared with 0.5 μg/ml for the positive control, clarithromycin. The inhibitory effect of LPS-induced NO release and cytotoxic activity of the ethanolic extract had IC50 values of 82.19 μg/ml and 26.06 μg/ml, respectively, and the EC50 values for the DPPH and ABTS antioxidant assays were 13.41 μg/ml and 12.44 μg/ml, respectively. The mace extract also had anticancer properties. In conclusion, the ethanolic mace extract had anti-H. pylori, anti-inflammatory, antioxidant, and anticancer activities. These data support further preclinical and clinical investigation to see if the mace extract could have a role in treating patients with dyspepsia, peptic ulcers, and possibly gastric cancer.

The metal complexes of Fe(III) and Cu(II) were prepared by using 2,4-dinitrophenyl hydrazine (DNPH) and thiocyanate (SCN) with stirrer refluxed for about 6 h. The prepared Fe(III) and Cu(II) complexes were characterized by elemental analysis, molar conductance, magnetic susceptibility and electronic spectrum, FT-IR spectral studies. The result suggested the octahedral geometry for Fe(III) and Cu(II) complexes. Powder X-ray diffraction indicate the crystalline nature of the metal complexes. The antimicrobial activities of the Fe(III) and Cu(II) complexes were tested with various micro organisms by disc diffusion method. The antimicrobial results indicate that the metal complexes are highly active with compared to the free ligand. The in vitro antioxidant activity of the free ligand and its metal complexes was assayed by radical scavenging activity (DPPH). The result proposed that Fe (III) and Cu(II) complexes exhibited strong antioxidant activity than that of the ligand.

Gas chromatography-mass spectrometry (GC-MS) analysis revealed the major components in black cumin essential oils which were thymoquinone (37.6%) followed by p-cymene (31.2%),α-thujene (5.6%), thymohydroquinone (3.4%), and longifolene (2.0%), whereas the oleoresins extracted in different solvents contain linoleic acid as a major component. The antioxidant activity of essential oil and oleoresins was evaluated against linseed oil system at 200 ppm concentration by peroxide value, thiobarbituric acid value, ferric thiocyanate, ferrous ion chelating activity, and 1,1-diphenyl-2-picrylhydrazyl radical scavenging methods. The essential oil and ethyl acetate oleoresin were found to be better than synthetic antioxidants. The total phenol contents (gallic acid equivalents, mg GAE per g) in black cumin essential oil, ethyl acetate, ethanol, and n-hexane oleoresins were calculated as11.47±0.05,10.88±0.9,9.68±0.06, and8.33±0.01, respectively, by Folin-Ciocalteau method. The essential oil showed up to 90% zone inhibition againstFusarium moniliformein inverted petri plate method. Using agar well diffusion method for evaluating antibacterial activity, the essential oil was found to be highly effective against Gram-positive bacteria.

Abstract-Medicinal plants represent an essential source of drugs and have played an important role in healthcaresystem.PyracanthacrenulataandZanthoxylum armatumhave been used as traditional medicine. The main aim of the study was to find the antibacterial activity of the selected plants against bacterialspecies:E.coliandPseudomonasaeruginosa. The solvents used for plant extraction were hexane, chloroform, ethanol and aqueous. The in vitro antimicrobial activity was performed by Agar disk diffusion method. The hexane and aqueous extracts showed moderate activity whereas theethanolicextractsshowedasignificantantibacterial activity. In the study Tetracycline was used as standard. The combined ethanolic extract of both the selected plant showed the synergistic effect on the bacterial strain tested. This leads to the conclusion that the combined effect can have possible application in the development of products as antimicrobial.

Background: Xylopia aethiopica is highly reputed for its numerous medicinal properties. In the present study, antioxidant, antimicrobial, and toxicity profile of methanol seed extract of Nigerian X. aethiopica in rat were evaluated. Methods: Phytochemical compositions were evaluated using standard procedures. The antibacterial study was carried out using agar well diffusion method, while antioxidant activities were evaluated by DPPH and FRAP assay. Twenty-five rats (5 each) were given 0, 75, 150, 300 and 600 mg/kg bwt of the extract orally for 28 days. Results: The extract had total phenolic and total flavonoid contents of 15.98±0.03mg GAE/g and 2.29±0.02 mg/g CE respectively. The extract had IC50 values of 52.45±3.05 µg/mL and 73.45±3.89 μg/mL in DPPH and FRAP assay respectively. The E. coli showed the highest susceptibility (20.27±0.90mm) while P. aeuruginisa showed the least (15.08±0.20mm). The MIC ranged from 25-50 µg/mL while MBC ranged between 50µg/mL and 100 µg/mL. In comparison with the control rats, the levels of serum creatinine, bicarbonate total proteins, albumin, and ALP were significantly higher in rat dosed 600 mg/kg bwt while urea decreases in rat dose 300 and 600 mg/kg. However, serum concentration of ALT, AST, bilirubin, Na+, K+ and Cl- compared favorably (P>0.05) with control at all doses. Conclusion: The study revealed the antioxidant and antimicrobial activities of Nigerian X. aethiopica, the extract at 75, 150 and 300 mg/kg/b.wt did not provoke toxic effects to the animals’ liver and kidney; however, caution should be exercised when using as a prolonged oral remedy at high doses.
