Swine wastewater (SWW) is a residue from pig farming that presents a high load of nutrients and organic matter. The appliance of organic matter in soil alters the microbial dynamic and may suppress soilborn phytopathogens. This study aimed at evaluating the inhibition on mycelial growth of Sclerotinia sclerotiorum and Sclerotium rolfsii in vitro under SWW doses. Hereupon, three kilograms of a soil classified as red dystroferric latosol was collected and sieved. Half of it was autoclaved. SWW was incorporated at doses of 0 mL, 2.5 mL, 5 mL, 10 mL and 20 mL in both soil conditions, autoclaved and not autoclaved. Afterwards, 130 grams of each soil was separately put into Petri plates above what a thin layer (≅ 5 mL) of Water-Agar (2%) medium was carefully spread over. Above this agar layer, one disk (6 mm diameter) of pure mycelium from each fungal grown in Potato Dextrose Agar medium was individually placed on the center of each plate. Daily evaluations on mycelial growth measuring were taken and ended when mycelium in control plates (without SWW addition) reached plate borders. Results indicated that in autoclaved soil condition, the inhibition was proportional to the dose, what is to say that the higher the dose the less the mycelial growth. In not autoclaved soil there was no significant difference among treatments, suggesting stimuli on suppression effect for both pathogens caused by SWW. In addition, the confirmed potential of SWW as a suppressor of S. sclerotiorum and S. rolfsii leads to promising investigations on other phytopathogens hard to control.

Nine isolates of Trichoderma spp. were isolated from different agro- ecological regions of Nepal viz; Jumla, Palpa, Chitwan, Tarahara, Banke, Illam and Salyan and screened against Sclerotium rolfsii Sacc. Adreded soil borne phytopathogen causing collar rot of chickpea in chickpea; In-vitro efficacy of nine fungal antagonist (Trichoderma spp.) against Sclerotium rolfsii were screened. Pot experiment was done to find out the effective management of S. rolfsi through Tricoderma using different methods i.e. Seed treatment, soil drenching and soil application. All the tested isolates of Trichoderma spp. were found effective on mycelial growth inhibition and sclerotial parasitization of S. rolfsii. Trichoderma isolated from Palpa district showed maximum growth inhibition (%) of pathogen periodically after 48(93.78%), 72(96.00%), 96(97.96%) and 120(100.00%) hours of inoculation. Parasitized sclerotium showed minimum sclerotial germination on agar plates. Moreover, Trichoderma species isolated from Palpa districts showed second best percent mycelial growth inhibition periodically at 72(25.00%), 120(29.16%), 168(29.16%) and 216(29.16%).In pot experiment at 40 days after sowing, Seedling height was maximum in soil drenching with 30g per 100ml of water (22.27cm) and Mortality percentage of seedlings was least or highest disease control was observed in seed treated with 109cfu/ml (0.000%).

ABSTRACTThis study aims to determine the digestibility of dry matter and organic matter level waste silage mixed vegetables and Gliricidia leaves were tested in vitro. This study used a completely randomized design with 3 treatments and 3 replications. Each of these treatments is R0 (Gliricidia leaves 100%), R1 (Gliricidia leaves 70% + 30% silage vegetable waste), and R2 (Gliricidia leaves 40% + 60% silage vegetable waste). Data were analyzed using analysis of variance (ANOVA) and further testing using the test Honestly Significant Difference (HSD). The results of ANOVA showed that the mixture of vegetable waste silage was highly significant (p <0.05) on dry matter digestibility and significantly (p <0.05) on the digestibility of organic matter. It can be concluded that the mixed of vegetable waste silage and Gliricidia leaves can improved digestibility of dry matter and organic matter, treatment of 40% and 60% Gliricidia leaves plus waste vegetable produce silage dry matter digestibility and percentage of organic matter is best (72,24% and 68,19%).Keyword: Silage vegetable waste, gliricidia leaves, dry and organic matter digestibilityABSTRAKPenelitian ini bertujuan untuk mengetahui kecenaan bahan kering dan bahan organik tingkat campuran silase sampah sayur dan daun gamal yang diuji secara in vitro. Penelitian ini menggunakan Rancangan Acak Lengkap dengan 3 perlakuan dan 3 ulangan. Masing-masing perlakuan tersebut ialah R0 (daun gamal 100%), R1 (daun gamal 70% + silase sampah sayur 30%), dan R2 (daun gamal 40% + silase sampah sayur 60%). Data yang diperoleh dianalisis dengan menggunakan analisis sidik ragam (ANOVA) dan uji lanjut menggunakan uji Beda Nyata Jujur (BNJ). Hasil analisis sidik ragam menunjukkan bahwa campuran silase sampah sayur berpengaruh sangat nyata (p<0,05) terhadap kecernaan bahan kering dan berpengaruh nyata (p<0,05) terhadap kecernaan bahan organik. Dapat disimpulkan bahwa perlakuan campuran silase sampah sayur dan daun gamal dapat meningkatkan kecernaan bahan kering dan bahan organik, perlakuan 40% daun gamal dan 60% silase sampah sayur menghasilkan persentase kecernaan bahan kering dan bahan organik yang terbaik yaitu (72,24% dan 68,19%).Kata kunci : Silase sampah sayur, daun gamal, kecernaan bahan kering, dan bahan organik.

For part 1 see Abst. 1564, Vol. 40. 2. The relation between digestibilities of organic matter in vivo and in vitro did not differ significantly between the institutes at Hoorn and Wageningen. No difference was found in digestibility of the cell wall constituents in relation to the lignin content, nor was there a significant difference in the relation between the percentage of digestible cell contents and the percentage of cell contents in forage from well managed pastures. Twelve forages of highly digestible perennial ryegrass deviated from these farm forages at Wageningen, showing smaller excretion of bacterial and endogenous residue. Comparison of the 2 lignin procedures showed that 72% sulfuric acid lignin gave consistent residual standard deviations of digestibility of cell wall constituents; permanganate lignin gave smaller errors in forages at Wageningen but greater errors in forages at Hoorn. The sensitivities of 11 isolates of Phytophthora infestans to dimethomorph were examined at all stages of the asexual life cycle and when inoculated onto potato leaf discs. In vitro zoospore encystment and cyst germination were highly sensitive to dimethomorph with 50% reduction of mycelial growth and cyst germination (EC50) values for most isolates <0.20 μg/ml, whereas direct sporangia germination and in vitro hyphal growth and sporulation were less sensitive (means of 0.45 and 0.22 μg/ml, respectively). Zoosporogenesis was not significantly inhibited at the maximum dimethomorph concentration examined, 10 μg/ml. Significant differences (Fisher's least significant difference, P = 0.05) in the EC50 values were present between isolates for all stages of the asexual life cycle, except direct sporangia germination and zoosporogenesis. Sensitivity ratios between the least- and most-sensitive isolates were 6.11, 12.14, 12.36, and 10.56 for hyphal growth, in vitro sporulation, zoospore encystment, and cyst germination, respectively. Application of dimethomorph at 1,000 μg/ml to potato leaf discs at 24 or 48 h before inoculation completely inhibited symptom incidence for most isolates, whereas application after inoculation generally was not significantly different from the untreated control, regardless of concentration. Sporulation from leaf discs treated with dimethomorph at 24 or 48 h after inoculation was completely inhibited for all isolates with dimethomorph at 1,000 μg/ml, even when symptom incidence was not significantly reduced.

Abstract Ammonium bicarbonate was evaluated for efficacy against southern stem rot and Sclerotinia blight of peanut in Georgia and Virginia, respectively. In vitro studies indicated the material provided little inhibition of mycelial growth by Sclerotinia minor and Sclerotium rolfsii, and negligible inhibition of mycelial growth of Rhizoctonia solani AG-4. However, ammonium bicarbonate did effectively inhibit formation of sclerotia by S. rolfsii in vitro. In the field, it was phytotoxic when applied as a granule or as a foliar spray and in general was not effective in controlling disease or increasing pod yield.

The damping off is the main disease that affects the beet crop during the seedling production. The aim of this study was to evaluate different salicylic acid (SA) concentrations for resistance induction against damping-off in beet seedling and its antifungal activity against Fusarium sp., in vitro condition. Treatment of beet seed was with SA solution by immersion during 5 minutes in the 0.5, 1.0, 1.5 and 2.0 mM concentrations and control (distilled water). It was used four replications with 20 cells by experimental unit. The experiment was carried out for 14 days in cultivate chamber with temperature (23 oC ± 2°C), lighting (12 hours photoperiod) and humidity (70% ± 10%) controlled. After this time, the germination, damping off incidence, seedling length and fresh mass matter weight were evaluated. It was evaluated also in the seedling tissue the phenylalanine ammonia-lyase (PAL), ?-1.3 glucanase and chitinase level enzymes. In the in vitro the SA was putted in PDA (potato-dextrose-agar) medium, where the Fusarium sp. mycelial growth was evaluated. The SA applied for seeds treatment didn’t had effect significant on damping off of beet seedlings, but it induced the activity of ?-1.3 glucanase enzyme, it being this higher in nine times when compared the treatment control. The SA acted in the Fusarium sp. in vitro control with fungitoxic action, suppressed mycelial growth in 28% if compared to control.

SUMMARYLeaves of olive trees (Olea europaea L.) of eight cultivars, Manzanilla, Picual, Hojiblanca, Edremit, Conservolia, Picholine-Marroqui, Chemlali and Frantoio, were collected in different seasons and years in Córdoba, Spain. Their chemical composition and in vitro organic matter digestibility values were determined. Differences in composition and nutritive value between cultivars, years and seasons were studied in Manzanilla, Picual and Hojiblanca, the three most important local cultivars. There were significant differences both between cultivars and seasons (P < 0·05) and a non-significant difference between years.The effect of various drying procedures (fresh, oven-dried, air-dried indoors or air-dried outdoors) on chemical composition, in vitro digestibility and in vivo digestibility in sheep was also examined. The in vivo organic matter digestibility of fresh leaves was significantly higher than that of dried leaves. No difference was found between leaves air-dried indoors and oven-dried leaves. However, leaves air-dried outdoors were significantly less digestible. Quick drying in an oven reduced the digestibility of crude protein and acid detergent fibres compared to air drying indoors. There was a significant decrease in digestibility of 11% when leaves were dried separated from the branches as compared to leaves dried on the branches.

Four plots of Gliricidia and two plots of Leucuena were harvested at three monthly intervals to monitor dry matter yield, chemical composition and in vitro organic matter digestibility. The plants had been established (Gliricidia from stakes, Leucaena from seeds) and were spaced 1m apart on rows and 1m between rows. Each plot size was 6m x 6m.  There was no significant difference (P>0.05) in the dry matter yield of the two browse species. Season did not have significant (P>0.05) effect on the dry matter yield of Leucaena but had on that of Gliricidia (P<0.01). The early dry harvest produced the lowest yield for both Gliricidia and Leucaena and these were significantly lower than the yield from other harvests.  Season had significant effect (P<0.01) on the chemical composition of Leucaena except on the percent Acid Detergent Fibre (ADF) and lignin but season had no effect on the organic matter digestibilities of the two browse species: Leucaena was higher in nutritive value than Gliricidia.

This study was carried out to determine chemical composition and in vitro dry matter and organic matter digestibility of range plant species of the area. Samples were collected from range lands at full maturity stage, dried under shade and subjected to chemical analysis and dry matter and organic matter in vitro digestibility for Vigna sunhum, Andropogan gayanus, Pennisetum Pedicelatum, Shoenfeldia gracilis and Stylothanis flavicans were determined. The data was analyzed via analysis of variance and differences among means were determined using Least Significant Difference (LSD) test. The results showed that the legumes (Vigna sunhum and Stylothanis flavicans) were significantly higher (P≤0.01) in their crude protein content and the percentage of crude protein respectively was 13.4% and 10.00% while the grasses were significant (P≤0.05) higher in Crude Fibre (CF), ash, Acid Detergent Fibers (ADF) and Neutral Detergent Fibers (NDF). Crude fiber % values were 41.78, 43.37 and 46.07% for Pennisetum Pedicelatum, Andropogan gayanus and Shoenofeldia gracilis, respectively. The acid detergent values were 53.85, 47.74 and 53.39 for the three weeds. (P≤0.05), while the ratio of neutral detergent was significantly higher (P≤.05) in grasses than in the two legume species; 87.22, 82.35 and 88.99% for the three grasses respectively. Similarly higher in vitro dry matter and organic digestibility coefficients were obtained from legume species than grasses. The study concluded that, the studied grasses were higher in fiber content and lower in their protein content but, legumes grasses (Vigna sunhum, Pennisetum Pedicelatum and Stylothanis flavicans) had high nutritive value and in vitro digestibility which may provide ruminants with the needed protein and energy.
