The focus of the presented study is the in vitro anti-oxidant activity and anti-diabetic potential of water extracts from the following four herbal substances, not traditionally used for treatment of diabetes mellitus – leaves of Sambucus ebulus L. and Prunus mahaleb L., and flowering stems of Cichorium intybus L. and Satureja kitaibelii Wierzb. ex Heuff. The water extracts are obtained through ultrasonication. The extract of S. kitaibelii stands out due to its highest values in all studied indicators – total phenolic content, scavenging potential (DPPH, ABTS) and α-glucosidase inhibitory activity which was six times higher than acarbose. The extract of C. intybus also showed significant α-glucosidase inhibitory activity compared to acarbose. The flowering stems of both species are promising sources of biologically active substances for blood sugar control in diabetes mellitus.

Rosa canina fruits have been used traditionally for the treatment of diabetes mellitus and its complications. The aim of current study was to evaluate the in vitro mechanism of action of R canina in managing diabetes mellitus. Cell proliferation and cytotoxicity assay were performed on pancreatic β-cells, βTC6. The protective activity of the extract on streptozotocin-induced death in βTC6 cells was studied. The effect of R canina on the metabolism of glucose in HepG2, a hepatocellular carcinoma cell line, was evaluated. The effect of the extract on glucose diffusion across the dialysis membrane, which is a comfortable model for assessing cellular glucose absorption, was evaluated. The results obtained from current study confirmed that R canina extract can act as a growth factor for pancreatic β-cell line providing a novel mechanism for the observed antidiabetic effect of this natural agent. Further preclinical studies are necessary to evaluate the perfect mechanism of action of R canina in diabetes mellitus.

Diabetes mellitus is a multifunctional disorder which is increasing rapidly in most parts of the world. It has been estimated that up to one third of patients with diabetes mellitus use some form of complementary and alternative medicine. One plant that has received the most attention for its anti-diabetic properties is Momordica charantia commonly referred to as bitter gourd. In this study, bitter gourd was extracted in various media i.e. ethanol, water, citric acid, sodium carbonate and sodium chloride. The samples were evaluated for their in-vitro ability to inhibit α-amylase and α-glucosidase enzymes activity. Ethanol extract was observed to have maximum inhibition against these enzymes. The IC<sub>50</sub> values recorded for inhibition of α-amylase activity were 81.34, 87.56, 83.89, 97.71 and 83.12 μg/ml respectively for crude extracts of ethanol, water, citric acid, Na<sub>2</sub>CO<sub>3</sub> and NaCl. Similarly, the ability of extracts to inhibit α-glucosidase was reflected in the result pattern ethanol&lt;Na<sub>2</sub>CO<sub>3</sub>&lt;NaCl&lt;aqueous&lt;citric acid (i.e. 72.81&lt;75.66&lt;80.15&lt;81.22&lt;83.85 μg/ml). Therefore, ethanol extract has potential applications in developing drug therapy for the treatment of diabetes using bitter gourd.

Background. The leaves of Bersama abyssinica are used for the treatment of diabetes mellitus in folk medicine system of Ethiopia. The present study was done based on the traditional claim of B abyssinica for the treatment of diabetes mellitus. Methods. The α-amylase inhibition and antioxidant activities of B abyssinica extracts were evaluated by using 3,5-dinitrosalicylic acid method and diphenyl-2-picrylhydrazyl assay model, respectively. Blood glucose lowering activity of the extracts was studied in 4 animal models; normoglycemic, oral glucose loaded, and streptozotocin-induced diabetic mice models. Results. Among the extracts, the crude extract showed the highest α-amylase enzyme inhibition activity with an IC50 of 6.57 μg/mL. The water fraction showed the strongest antioxidant activity with an IC50 of 3.43 μg/mL. The crude extract at doses of 200, and 400 mg/kg showed significant ( P < .05) hypoglycemic activity in normoglycemic mice. All doses of the crude extract significantly ( P < .05) reduced blood glucose levels of oral glucose-loaded mice. In streptozotocin-induced diabetic mice models, both the crude and solvent fractions showed a significant ( P < .05) blood glucose lowering effect as compared with the negative control group post 8 hour treatment. Conclusion. The results demonstrated the beneficial biochemical effects of B abyssinica extract by inhibiting α-amylase and scavenging the free radicals. The crude extract and solvent fractions of B abyssinica had significant blood glucose lowering effect in all animal models.

Abstract Background: Mesenchymal stem cells (MSCs) from human adipose tissue and bone marrow have a great potential for use in cell therapy due to their ease of isolation, expansion, and differentiation. Our intention was to isolate and promote in vitro expansion and differentiation of MSCs from human adipose and bone marrow tissue into cells with a pancreatic endocrine phenotype and to compare the potency of these cells together.Methods and Results: MSCs were pre-induced with nicotinamide, mercaptoethanol, B-27 and b-FGF in L-DMEM for 2 days and re-induced again in supplemented H-DMEM for another 3 days. Expression of five genes in differentiated beta cells was evaluated by Real-time PCR and western blotting and the potency of insulin release in response to glucose stimulation was evaluated by insulin and C-peptide ELISA kit.Quantitative RT-PCR results showed up-regulation of four genes in differentiated beta-islet cells (Insulin, Ngn-3, Pax-4 and Pdx-1) compared with the control. Western blot analysis showed that MSCs cells mainly produced proinsulin and insulin after differentiation but nestin was more expressed in pre-differentiated stem cells. Glucose and insulin secretion assay showed that insulin levels and C-peptide secretion were significantly increased in response to 10 mM glucose.Conclusions: Our study showed that both adipose and bone marrow stem cells could differentiate into functional beta-islet cells but it seems that adipose stem cells could be a better choice for treatment of diabetes mellitus according to their more safety and potency.

Diabetes mellitus is a disease that is characterized by the chronic presence of blood glucose levels caused by a defect in the secretion of insulin or in the action of this hormone in the body which must be treated integrally with a multidisciplinary approach. The natural treatment of this disease is a common practice around the world, especially in Latin America, there are several clinical studies, in vivo or in vitro assays that focus on assessing the hypoglycemic capacity of various natural products used empirically by the population for years for the phytotherapeutic treatment of the disease as well as the chemicals related to the mechanism of action that produces the hypoglycaemic effect. In the present article, a brief review of the evidence of the hypoglycemic capacity of some natural products for the alternative treatment of diabetes mellitus 2

In recent years, many studies of Momordica charantia (MC) in the treatment of diabetes mellitus (DM) and its complications have been reported. This article reviewed the effect and mechanism of MC against diabetes, including the results from in vitro and in vivo experiments and clinical trials. The common side effects of MC were also summarized. We hope that it might open up new ideas for further mechanism exploration and clinical application as well as provide a scientific theoretical basis for the development of drugs or foods derived from MC.

The aim of the present study was to develop colon targeted matrix tablets of Metformin HCl using various conc. of selected polymers such as HPMC, Ethyl Cellulose Guar gum and combination of the same. Tablets were prepared by direct compression method and both pre-compression and post- compression parameters for all batches shows in the acceptable ranges. Short term accelerated stability studies was performed according to ICH guidelines temperature of 400±20 and relative humidity of 75%±5% RH to study any physical changes and chemical decomposition of drug, no formulation shown any physical or chemical changes. The compatibility of drugs, polymers and excipients were determined by FT-IR Spectroscopy results showed that the drug was compatible with polymers and all excipients. Dissolution studies were performed for 12 hours study in 1.2 pH for first 2 hrs then in 7.4 pH for next 3hrs followed by 6.8pH phosphate buffer at the temperature of 37±0.50C at 100rpm. The dissolution data so obtained was fitted to various mathematical kinetic models and the drug release followed mixed order and Higuchi’s model. To study release mechanism of drug from matrices the data were fitted to Koresmeyer-Peppas model and the release. In –vitro release profile of Metformin HCl from various polymers showed that drug increasing the conc. of polymers resulted in reduction in the release rate of drug (MTF1 to MTF12). Formulation containing combination of E.C-G.G, HPMC-G.G and E.C-HPMC showed drug release profile for MTF-12 about 38.72% after 12 hrs, MTF-11 about 40.66% after 12 hrs, for MTF-10 about 45.45% after 12 hrs. This is an indicative of retardation of drug release when polymer combination was changed. Results showed that the tablets with higher binding concentration showed minimum drug release. Combination of polymers shows greater retarding of drug release.

Annona genus contains plants used in folk medicine for the treatment of diabetes. In the present study, an aqueous extract prepared fromAnnona macroprophyllata(Annonaceae, also known asA. diversifolia) leaves was evaluated on both the activity of yeastα-glucosidase (anin vitroassay) and sucrose tolerance in Wistar rats. The results have shown that the aqueous extract fromA. macroprophyllatainhibits the yeastα-glucosidase with an IC50 = 1.18 mg/mL, in a competitive manner with aKi= 0.97 mg/mL, a similar value to that of acarbose (Ki= 0.79 mg/mL). The inhibitory activity ofA. macroprophyllatawas reinforced by its antihyperglycemic effect, at doses of 100, 300, and 500 mg/kg in rats. Chromatographic analysis identified the flavonoids rutin and isoquercitrin in the most polar fractions ofA. macroprophyllatacrude extract, suggesting that these flavonoids are part of the active constituents in the plant. Our results support the use ofA. macroprophyllatain Mexican folk medicine to control postprandial glycemia in people with diabetes mellitus, involving active constituents of flavonoid nature.
