Aim. Defining the peculiarities of forming the cannabinoid substances in modern monogamous varieties of hemp is of both theoretical and practical importance. Methods. Modern monogamous varieties of hemp bred in Hlukhiv, Zolotonosha and specimens receive from France; were used for the chromatographically analysis of «damp» and dried in the shadow plant samples was conducted by the method of thin layer chromatography (TLC). Results. It was founding, that in the initial phase of growth and development the Ukrainian hemp varieties with the practical absence of neutral compounds and the French varieties with the sufficient content of these substances formed only natural acids. During the generative phase of growth and development, the varieties from France formed not only natural acids but also neutral compounds. The Ukrainian varieties demonstrated only insufficient increase of natural acids. Conclusions. In the initial phase of growth and development, the investigated hemp varieties containing both large and insignificant amount of cannabinoid substances formed only natural acids. Separate plants demonstrated the decrease of natural acids up to their total absence, which proves their biological interconnections and possible forming cannabidiol, tetrahydrocannabinol and сannabinol by way of fermentative decarboxylation the respective acids. Aim. Modern researches identified a significant number of individual cannabinoid compounds: neutral – KBD, THC, KBN and acids – KBDK, THCC. Schultz et al., R. Mechoulam believe that cannabidiolic acid – KBDK has an advantage over the other cannabinoids in the nature. It is the main component of the cannabinoid substances. Defining acids in the sorts by the value of Rf by the method of the thin layer chromatography (TLC), with the absence of "witnesses" for them is of interest. Methods. TLC method was applied to the samples (leaves, perianth, buds) of the domestic and French varieties of hemp. KBD, THC and KBN were defined by "witnesses" – KBDK and THCC by the value of Rf. Results. By the results of the chromatographic analysis, it was found that the compound with the value 0.059–0.060 of Rf was related to cannabidiolic acid while compound with the value 0.131 of Rf belonged to tetrahydrocannabinolic acid. In the chromatogram, the defined acids occupied the lowest border by the distribution front of the mobile phase. Conclusions. The study showed that during the plant vegetative growth and development only cannabidiolic acid was biosynthesised: Rf – 0.059–0.060; the French variety Futura 77 also had tetrahydrocannabinolic acid: Rf – 0.131. During the biological maturity of domestic hemp varieties seeds the neutral compounds were virtually absent. Biosynthesis of acids amplified. Abstract Background Cannabidiol (CBD), the non-psychotropic compound from Cannabis sativa, shows positive results on controlling several health disturbances; however, comparable data regarding additional chemical from C. sativa, such as cannabidiolic acid (CBDA), is scarce due to its instability. To address this limitation, a stable CBDA analogue, CBDA methyl ester (HU-580), was synthetized and showed CBDA-like effects. Recently, we described that HU-580 increased wakefulness and wake-related neurochemicals. Objective To extend the comprehension of HU-580´s properties on waking, the c-Fos and NeuN expression in a wake-linked brain area, the hypothalamus was evaluated. Methods c-Fos and NeuN expression in hypothalamic sections were analyzed after the injections of HU-580 (0.1 or 100 μg/kg, i.p.). Results Systemic administrations of HU-580 increased c-Fos and neuronal nuclei (NeuN) expression in hypothalamic nuclei, including the dorsomedial hypothalamic nucleus dorsal part, dorsomedial hypothalamic nucleus compact part, and dorsomedial hypothalamic nucleus ventral part. Conclusion HU-580 increased c-Fos and NeuN immunoreactivity in hypothalamus nuclei suggesting that this drug might modulate the sleep–wake cycle by engaging the hypothalamus.

Cannabidiolic acid (CBDA) is the main phytocannabinoid in fiber and seed-oil hemp (Cannabis sativa L.) plants, but its potential health-related capabilities have been masked for years by a greater scientific interest towards its neutral derivative cannabidiol (CBD). This review aims to collect from the literature and critically discuss all the information about this molecule, starting from its biosynthesis, and focusing on its bioactivity, as an anti-inflammatory, anti-emetic, anti-convulsant, and anti-cancerogenic drug. Furthermore, in the awareness that, despite its multiple bioactive effects, currently poor efforts have been made to achieve its reliable purification, herein, we propose a relatively simple, fast, and inexpensive procedure for its recovery from pollen of industrial hemp cultivars. Spectroscopic and spectrometric techniques allowed us to unequivocally identify pure isolated CBDA and to distinguish it from the constitutional isomer tetrahydrocannabinolic acid (THCA-A).

Genetic markers can be used in seeds and in plants to distinguish drug-type from fiber-type Cannabis Sativa L. varieties even at early stages, including pre-germination when cannabinoids are not accumulated yet. With this aim, this paper reports sequencing results for tetrahydrocannabinolic acid synthase (THCAS) and cannabidiolic acid synthase (CBDAS) genes from 21 C. sativa L. varieties. Taking into account that THCAS- and CBDAS-derived enzymes compete for the same substrate, the novelty of this work relies in the identification of markers based on both THCAS and CBDAS rather than THCAS alone. Notably, in our panel, we achieved an adequate degree of discrimination (AUC 100%) between drug-type and fiber-type cannabis samples. Our sequencing approach allowed identifying multiple genetic markers (single-nucleotide polymorphisms—SNPs—and a deletion/insertion) that effectively discriminate between the two subgroups of cannabis, namely fiber type vs. drug type. We identified four functional SNPs that are likely to induce decreased THCAS activity in the fiber-type cannabis plants. We also report the finding on a deletion in the CBDAS gene sequence that produces a truncated protein, possibly resulting in loss of function of the enzyme in the drug-type varieties. Chemical analyses for the actual concentration of cannabinoids confirmed the identification of drug-type rather than fiber-type genotypes. Genetic markers permit an early identification process for forensic applications while simplifying the procedures related to detection of therapeutic or industrial hemp.

Abstract Gene copy number (CN) variation is known to be important in nearly every species where it has been examined. Alterations in gene CN may provide a fast way of acquiring diversity, allowing rapid adaptation under strong selective pressures, and may also be a key component of standing genetic variation within species. Cannabis sativa plants produce a distinguishing set of secondary metabolites, the cannabinoids, many of which have medicinal utility. Two major cannabinoids—THCA (delta-9-tetrahydrocannabinolic acid) and CBDA (cannabidiolic acid)—are products of a three-step biochemical pathway. Using whole-genome shotgun sequence data for 69 Cannabis cultivars from diverse lineages within the species, we found that genes encoding the synthases in this pathway vary in CN. Transcriptome sequence data show that the cannabinoid paralogs are differentially expressed among lineages within the species. We also found that CN partially explains variation in cannabinoid content levels among Cannabis plants. Our results demonstrate that biosynthetic genes found at multiple points in the pathway could be useful for breeding purposes, and suggest that natural and artificial selection have shaped CN variation. Truncations in specific paralogs are associated with lack of production of particular cannabinoids, showing how phytochemical diversity can evolve through a complex combination of processes.

Environmental conditions, including the availability of mineral nutrients, affect secondary metabolism in plants. Therefore, growing conditions have significant pharmaceutical and economic importance for Cannabis sativa. Phosphorous is an essential macronutrient that affects central biosynthesis pathways. In this study, we evaluated the hypothesis that P uptake, distribution and availability in the plant affect the biosynthesis of cannabinoids. Two genotypes of medical “drug-type” cannabis plants were grown under five P concentrations of 5, 15, 30, 60, and 90 mg L–1 (ppm) in controlled environmental conditions. The results reveal several dose-dependent effects of P nutrition on the cannabinoid profile of both genotypes, as well as on the ionome and plant functional physiology, thus supporting the hypothesis: (i) P concentrations ≤15 mg L–1 were insufficient to support optimal plant function and reduced photosynthesis, transpiration, stomatal conductance and growth; (ii) 30–90 mg L–1 P was within the optimal range for plant development and function, and 30 mg L–1 P was sufficient for producing 80% of the maximum yield; (iii) Ionome: about 80% of the plant P accumulated in the unfertilized inflorescences; (iv) Cannabinoids: P supply higher than 5 mg L–1 reduced Δ9-tetrahydrocannabinolic acid (THCA) and cannabidiolic acid (CBDA) concentrations in the inflorescences by up to 25%. Cannabinoid concentrations decreased linearly with increasing yield, consistent with a yield dilution effect, but the total cannabinoid content per plant increased with increasing P supply. These results reveal contrasting trends for effects of P supply on cannabinoid concentrations that were highest under <30 mg L–1 P, vs. inflorescence biomass that was highest under 30–90 mg L–1 P. Thus, the P regime should be adjusted to reflect production goals. The results demonstrate the potential of mineral nutrition to regulate cannabinoid metabolism and optimize pharmacological quality.

Introduction. When creating the Simmental beef breed of cattle during many years carried out researches to study the effectiveness of the use of meat Simmentals of foreign selection in crossbreeding with cows of Simmental breed of domestic selection. Were created the herds of the desirable type by breeding "in themselves" animals, that meet the requirements of the target standard, formed a genealogical structure. Now carry out the work with related groups of cattle of the created Simmental meat breed, which are the basis for the creation of further lines, but the growth and development of animals of different related groups have not been sufficiently studied. Therefore, the study problems of growth and development of repair heifers of different related groups has not only theoretical, but also practical importance. The purpose of our researches was to study the dynamics of linear growth and weight development of heifers of different related groups of the created Simmental meat breed. Research materials and methods. The researches was carried out on heifers of different related groups of the created Simmental meat breed in PJSC "Dniprovske" of Boryspil district of Kyiv region. To analyze the growth and development of heifers were formed groups of animals depending on their belongings to related groups of 15 heads in each: I Group – Metz 5290; II – Achilles 369; III – Abricot 58311; IV – Huxla 19223; V – Hercules 8942 and VI – Signal 120. Research results. It was determined, that heifers of the II group, which belong to the related group of Achilles 369, by live weight at all ages of periods surpass to peers. Thus, at 18 months of age the advantage of heifers of the II group by live weight in comparison with to peers of the I, III, IV, V and VI groups was 4.1; 3.0; 5.0; 14.1 and 11.4 kg. In heifers of related groups Hercules 8942 and Signal 120, the coefficients of variability of live weight at birth in which were the lowest, and in the following age periods they little changed, therefore at 18 months of age in these heifers they were the highest, comparison with analogues, and was to 8.4 and 9.2%, respectively. When studying the exterior of heifers of different related groups it was determined, that newborn heifers of the I group, which belong to the related group of Metz 5290, surpass their peers in height indicators. Their the height at the withers and the height at the sacrum were 74.0 and 79.1 cm, which is 0.6; 1.3; 2.1; 3.2 and 2.6 cm and is 0.3; 1.2; 2.9; 3.6 and 3.2 cm more than in analogues of the II, III, IV, V and VI groups, respectively. The best indicators of latitudinal measurements had the heifers of the III group, which belong to the related group Abricot 58311. In heifers of this group the width of the chest was 17.6 cm, width in the hip joints – 21.8 cm, width in the hips – 18.1 cm, and width in the ischial tubercles – 12.6 cm. The depth of the chest and the girth of the chest behind the shoulder blades were the highest in heifers of the I group, which belong to the related group of Metz 5290. They were 32.2 and 79.6 cm, respectively. When analyzing the measurements of heifers at 18 months of age, it was determined, that the height indicators had advantage by heifers of the II group, which belong to the related group of Achilles 369. Their, the height at the withers and height at the sacrum were 124.5 and 129.8 cm, which is 0.3; 1.3; 2.5; 5.3 and 4.2 cm and is 0.9; 2.2; 4.1; 6.6 and 5.9 cm more than in analogues of the I, III, IV, V and VI groups, respectively. The best indicators of latitudinal measurements had the heifers of the III group, which belong to the related group of Abricot 58311, since they had increases of measurements width of the chest, width in the hip joints, width in the hips and width in the ischial tubercles for the period from birth to 18 months of age the highest comparison with peers. Such indicators as depth of the chest and the girth of the chest behind the shoulder blades were highest in heifers of the II group, which belong to the related group of Achilles 369. Their increases of measurements depth of the chest and the girth of the chest behind the shoulder blades for the period from birth to 18 months of age also were highest comparison with peers and were 35.5 and 94.2 cm, respectively. Conclusions. It was determined, that the animals of the related group of Achilles 369 at all ages of periods had a large live weight in comparison with their analogues. The lowest coefficient of variability of live weight at 18 months of age was in heifers of the related group Abrikot 58311 and amounted to 6.5%. Heifers of the related group of Achilles 369 had the highest indicators of the following measurements: height at the withers (124.5 cm), height at the sacrum (129.8 cm), depth of the chest (67.4 cm), girth of chest behind the shoulder blades (173.1 cm) and oblique length of body (146.4 cm). The highest latitudinal measurements were observed in the heifers of the related group Abrikot 58311. Thus, the width of the chest was 49.9 cm, the width in the hip joints was 42.2 cm, the width in the hips was 43.3 cm, and the width in the ischial tubercles was 29.2 cm. Heifers of the related group Metz 5290 had the highest half-girth croup (110.2 cm) and girth of metacarpus (18.4 cm). In general, heifers of all groups showed good energy of growth and a typical for beef cattle exterior.

Aim. To determine the change in the structure of the inzucht-offspring in the content of cannabinoid substances in plants of different varieties of hemp under the influence of directed plus and minus selections. Methods. Taking into account the lability of natural phenol-cannabinoids, their transformation and isomerization under the influence of various factors, the method of thin-layer chromatography (TLC) was used. Plants of varieties YuSO1 and Hlyana at the budding period were labeled and separated. Results. By selecting plants in inzucht offsprings for three years aimed at reducing the content of cannabinoids in the hemp straw YSO 1 and increasing the amount of these substances in the Hlyana variety showed that the increase in the content of cannabinoids was more active than reduction, indicating a dominant feature of cannabinoids. Under the condition of isolation, self-pollination and regardless of the plus and minus selections for the content of cannabinoids, for three generations there was a decrease in plant height and sterility of varieties. Conclusions. The results of chromatographic analyzes of inzucht generations by the content of cannabinoid substances under the influence of minus and plus selections indicate their dominant feature. Regardless of the minus and plus selections by the content of cannabinoids, provided that the isolation and self-pollination were carried out, the plant height declined. Introduction- HPTLC uses include phytochemical and biochemical analyzes, ayurvedic medicine quantification and quantification of active ingredients, formational fingerprinting and adulterant testing of formulations. HPTLC can be used as a simple tool for tracking the consistency of plant-based raw plant materials and formulations. The source of many biomarkers is Sammoha Loha Vati. The HPTLC method has been used to classify and measure the bark of leaves and branches. Aim & Objective- Identification and authentication of raw drugs used for Sammoha Loha Vati through HPTLC. Observations- 1. Stationary phase, 2. Mobile phase, 3. Optimization, 4. Preparation of Sample and its use, 5. Separation, 6. Detection. Conclusion and Finding- The use of HPTLC for the screening of pharmaceutical compounds for antimicrobial activities is emerging. Requirements for the testing of new incoming products and their integration into regulatory frameworks are of great significance for the future of HPTLC.
