Laportea interrupta also called as Hen’s Nettle is a primitive plant that is known for its property of causing itch after its touch. This plant has been found useful in treating pre maternal health, anti-pyretic and anti-inflammatory action. Previously conducted scientific studies revealed that it is having potential of anti- microbial activity. It has also been proved to have anti-oxidant property as it contains flavonoid contents especially rutin and gallic acid. Extraction of the plant phyto constituent was by maceration process. Healthy Albino rats of wistar strain of 150 to 250 gm were used as experimental animal. Gentamicin induced renal injury and Paracetamol induced nephrotoxicity are the two methodologies followed in this experiment to induce toxicity in rats and then it is followed by ethanolic extract of the plant of low, mid and high dose. Biochemical analysis, Histopathological studies and statistical studies are also performed. There was significant decrease in the gentamicin-induced nephrotoxicity. Observation of change in body weight serum creatinine, serum albumin and serum protein were assessed for kidney function test. Histological and pathological study of kidney tissue was observed to identify the changes in the degree of cellular damage and healing effects in this research work.

Cubeba Officinalis is traditionally effective in the treatment of various kidney ailments, and the main adverse effect of tacrolimus is nephrotoxicity. There is no documented evidence about the ameliorative potential of Cubeba Officinalis in tacrolimus induced nephrotoxicity. The main endeavor of the study was to determine the nephroprotective activity of ethanolic extract of Cubeba Officinalis dried fruits against tacrolimus induced nephrotoxicity in Wistar albino rats. The Cubeba Officinalis dried fruits were collected from the local market, and Male albino rats weighing 200-250 g were used for the study. The dose of is lower 200mg/kg, higher dose 400 mg/kg of test drug (EECO) was used, and silymarin is used as the standard at the dose of 20 mg/kg. The animals were divided into five groups, six animals each, which is started prior to oral administration of tacrolimus and continued with the fourteen days tacrolimus treatment. After the whole period of study, the rats were sacrificed, and histopathological studies and biochemical estimations were carried out. The BUN values  were decreased from 33.60±3.84 in nephrotoxic rats to 28.27±2.48 (200mg/kg) and 20.70±0.81 (400mg/kg),creatinine levels from 1.645±0.21 to 0.926±0.19 (200mg/kg) and 0.638±0.07(400 mg/kg),uric acid levels from 1.822±0.249 to 1.092±0.306 (200 mg/kg) and 0.806±0.181 (400 mg/kg) sodium, potassium and chloride levels from 1.607± 0.091, 2.548± 0.293 and 259.8±6.42 to 1.302± 0.169 , 1.023±0.174 and 134.7±9.138 (200mg/kg of EECO) and 0.586±0.092 , 0.831±0.174 and 130.2±2.29 (400mg/kg of EECO). The Ethanolic extract of cubeba officinalis was found to be effective in treating the nephrotoxicity in tacrolimus induced nephrotoxicity.

Background: The therapeutic value of Syzygium cumini (S. cumini) has been documented in traditional medicine for the treatment of many diseases and ailments. Various preparations of this plant have been made and used especially for liver inflammatory conditions in livestock. Further, many liver diseases in humans are inflammatory conditions, which are caused by alcohol intake. This study sought to examine the effect of S. cumini on ethanol-induced hepatotoxicity in Wistar albino rats. Methods: Twenty-five rats were divided into five groups of five rats each. The first group was control and the other four were administered ethanol at varying doses to induce liver and kidney damages. Two doses of the S. cumini extract were administered at a concentration of 200 mg/kg or 400 mg/kg. Silymarin was administered to the last group at 10 mg/kg. The liver and kidney tissue samples were collected and preserved for histological analyses and the rat sera were analyzed for the associated biochemical biomarkers. Results: Histopathological analyses revealed pyknotic nuclei and distortion in the arrangement of the hepatocytes in extract-treated groups. The kidney tissue samples showed signs of interstitial bleeding and aggregation of lymphocytes in the peri-glomerular areas. The analyses of the biochemical parameters revealed that there were significant increases in the aspartate aminotransferase (AST), alanine transaminase (ALT), Urea and creatinine in the sera of the groups treated with the extract compared to those of the controls (P<0.05). Conclusion: The S. cumini extract caused elevation of serum hepatic and renal biomarkers at 400 mg/kg and did not have a hepatoprotective effect.

The present study was aimed to evaluate the nephroprotective activity of ethanolic extract of Carissa carandas Linn. Leaves (EECC) in Gentamicin-induced nephrotoxicity in Wistar albino rats. The renal damage was induced by Gentamicin (80mg/kg body weight, i.p.). Nephroprotective activity was investigated by the administration of EECC at two different doses (100 and 200mg/kg body weight, p.o) for 28 days and by assessing serum parameters, renal oxidative stress markers and histopathological studies. Gentamicin-treated animals showed an increase in serum creatinine, uric acid, urea, and malondialdehyde (MDA) levels and decrease in total protein, reduced glutathione (GSH), and catalase(CAT) compared to normal control animals, which indicates severe nephrotoxicity. Histopathological studies of kidney Gentamicin-treated animals showed extensive acute tubular necrosis and peri-tubular inflammation. Administration of EECC showed a significant improvement (p<0.05) in biochemical and oxidative stress markers compared to the disease group. EECC treated groups showed better histological appearance when compared to the disease group. Ethanolic extract of Carissa carandas Linn. Leaves showed significant nephroprotective activity against gentamicin-induced acute kidney injury.

AbstractAdriamycin (ADR), a cytotoxic antineoplastic drug, is used in the treatment of various solid tumors. However, its efficacy continues to be challenged by significant toxicities including nephrotoxicity. In the present study, the effects of N-acetyl cysteine (NAC) and vitamin E, known antioxidants, were investigated on ADR-induced peroxidative damage in rat kidney. Adult male albino rats of Wistar strain were administered ADR as a single dose (10mg/kg body weight, i.v.). Histopathological studies indicated that ADR-treated kidney sections show focal tubular necrosis and casts. ADR-injected rats showed a significant decline in the activities/levels of enzymic antioxidants (superoxide dismutase, catalase, glutathione peroxidase, glutathione reductase, glucose-6-phosphate dehydrogenase and glutathione-S-transferase) and non-enzymic antioxidants (thiols, vitamin C and vitamin E) with high malondialdehyde levels. The extent of nephrotoxicity was evident from the increased activities of urinary marker enzymes (alkaline phosphatase, lactate dehydrogenase and γ-glutamyltransferase). Treatment with NAC and vitamin E (50mg/kg b.w., i.p.) 1day prior to ADR administration maintained near normal activities of the enzymes, significantly reduced lipid peroxidation and prevented the necrosis caused by ADR, thereby proving to be an effective thiol replenishing agent and antioxidant.

Abstract Objective The present study was performed to evaluate the ethanolic extract of leaves of Acacia catechu (A. catechu) for its effect on streptozotocin (STZ)-induced diabetes mellitus (DM) and its renal complications in male Wistar albino rats. Materials and Methods Male Wistar albino rats were grouped into control (A), STZ-induced DM (B), STZ-induced DM rats with A. catechu orally of 75 mg/kg body weight (kbw) for 35 days (C), with each group having six rats (n = 6) weighing between 200 to 250 g each. Group A receives only water, orally; group B receives a single dose of STZ at 45 mg/kbw intraperitoneal administration (IP); group C receives STZ IP and oral A. catechu for 35 days. On the 36th day, animals were euthanized, the kidney tissues were analyzed for biochemical parameters, such as GOT (glutamic oxaloacetic transaminase), GPT (glutamic pyruvic transaminase), oxidative stress assessment parameters, and histopathological studies. Results In group C rats, activities of the enzymes were nearer to group A when compared with group B. Histopathological findings were also suggesting that renal toxicity were observed at a lesser extent in group C. Conclusion The ethanolic extract of A. catechu signified as nephroprotective effect. The present data could provide adequate confirmation of the efficacy of ethanolic extract of leaves of A. catechu for further experimental studies on a standardized formulation.

Thiamethoxam (TMX), a second-generation neonicotinoid insecticide, is one of the most widely used insecticides in Algeria. The present study assessed the effects of repeated subchronic exposure to the commercial formulation of thiamethoxam (Actara®, 25% WG) in albino male rats. The toxic effects of thiamethoxam (TMX) were studied biochemically and histopathologically. Twenty-eight male albino rats weighing between 226 and 243 g were randomly assigned to four groups. One group served as control, and the other three were served as experimental groups administered a neonicotinoid thiamethoxam (TMX; 26, 39 and 78 mg/kg/day) for 6 weeks. The effects of the insecticide on various biochemical parameters were evaluated at 2, 4 and 6 weeks. Histopathological studies were carried out in the liver, kidney, cerebellum and hippocampus at the end of the experiment. Changes in biochemical parameters glucose, ALT (alanine aminotransferase), AST (aspartate aminotransferase), γGT (gamma-glutamyltransferase) ALP (alkaline phosphatase) urea and creatinine were observed in treated-groups in a dose dependent manner when compared to the control. Histopathological alterations were more intense in male rats from the TMX high dose group than those from group 2 and 3. Based on these results, subchronic oral administration of thiamethoxam altered the biochemical parameters, which correlated with histopathological changes in the liver kidney and brain.

Abstract Objective The present study was aimed to evaluate the effect of ethanolic extract of Adhatoda zeylanica (EAZ) leaves on streptozotocin (STZ)-induced diabetes mellitus (DM) and its renal complications in male Wistar albino rats. Materials and Methods Adult male Wistar albino rats were randomly selected from a colony, divided into four groups, namely, A, B, C, and D, with each having six rats (n = 6) and each weighing between 200 and 250 g. Group A served as control and received only water per oral (p.o.). Group B, C, and D animals received a single dose of STZ at 45 mg/kg body weight (kbw) intraperitoneal administration (i.p.) on day 1 and observed for fasting blood glucose (FBG) to induce DM for next 72 hours. After the DM was induced, group B served as DM control, group C received the standard drug glibenclamide (GL) at 5 mg/kbw p.o. once daily, and group D received EAZ of 500 mg/kbw p.o. once daily for 35 days. After the observation period, the animals were euthanized, serum creatinine and blood urea, antioxidants in the kidney tissue homogenate, and histopathological studies were assessed to know the ameliorative effect of the test drugs. Results Renal parameters, such as serum creatinine, blood urea, antioxidants activities, in group D were nearer to the control when compared with groups B and C. Histopathological studies revealed that there was minimal renal damage in group D when compared with groups B and C. Conclusion Administration of ethanolic EAZ showed significant ameliorative effects on the FBG, biochemical, oxidative, and histopathological parameters on kidney tissues treated with STZ to induce DM.

<p><strong>Abstract </strong></p><p>Aim of this research work was to explore the anti-diabetic activity and acute toxicity of <em>Annona squamosa </em>L. leaves ethanolic extract in albino rats and mice respectively.</p><p>Diabetes was induced by Alloxan (120 mg/kg). Seven rats were taken in each group. Glibenclaimide (0.25 mg/kg) was taken as the standard drug. <em>A. squamosa</em> was administered orally in 100mg, 200mg and 400mg doses in three different groups of diabetes- induced rats. <em>A. squamosa</em> leaves extracts were found to have significant anti-diabetic activity.</p><p>Acute toxicity study was carried out on administration of 800mg/kg, 1600mg/kg and 5000 mg/kg body weight. No acute toxicity was observed at 800mg/kg and 1600mg/kg doses. At 5000mg/kg body weight dose 100% fatality was recorded within 24 hours.</p><p>Our research work revealed the safe and effective anti-diabetic activity of <em>Annona squamosa</em> ethanolic leaves extract. </p>

The present research work is aimed to investigate the anti-oxidant/neuroprotective role of Resveratrol in reversing the valproic acid induced autism in postnatal swiss albino mice. Separate 13 day old/ Post natal day (PND) 13 swiss albino mice of either sex into 5 groups, each group consists of six mice of either sex, groups namely Group I - Control, Group II - Resveratrol, Group III - Negative control, Group IV and V - Resveratrol treatment groups. On PND 14 administer single dose of Valproic acid (VPA) or Sodium Valproate (400 mg/kg, subcutaneously) to III, IV and V groups to induce autism. Treatment is given in two doses 10 mg/kg, intraperitoneally (i.p) as Low dose and 40 mg/kg, i.p as High dose from the 13th day to the end of study. Assessment of autism is done by different behavioral screening methods during PND 14 to 40. Treatment with resveratrol significantly decline the autism symptoms compared with negative control. At the end of study on PND 41 all the animals were sacrificed to assess the biochemical estimations like Anticholinesterase enzyme, Total Protein, antioxidant enzyme (Catalase, Superoxide and Glutathione) activity and cerebellar histopathological examination. Treatment with Resveratrol has shown a significant beneficial difference on behavioral alterations, oxidative markers, neurotransmitters, and restoration of the altered purkinje cells of autism. This research work we conclude that resvertrol have a potent anti-oxidant, neuroprotective, anxiolytic, learning and memory enhancing agent against valproic acid induced autism.

Objective: In the present study, the protective effect of Roflumilast (ROF, a selective phosphodiesterase (PDE-4) inhibitor) was investigated against cadmium (Cd)-induced nephrotoxicity in rats. Methods: A total of 24 rats were selected and randomly divided into four groups ( n = 6). Group 1 served as the control; groups 2–4 administered with CdCl2 (3 mg/kg, i.p.) for 7 days; groups 3 and 4 were co-administered with ROF in doses of 0.5 and 1.5 mg/kg, orally for 7 consecutive days. Nephrotoxicity was evaluated by measuring urine volume, urea and creatinine levels in urine and serum. Oxidative stress was confirmed by measuring malondialdehyde (MDA), glutathione (GSH), superoxide dismutase (SOD), and catalase (CAT) levels in kidney tissue followed by histopathological studies. Results: CdCl2 administration results in a significant ( p < 0.01) decrease in urine volume, urea, and creatinine levels in urine, as well as GSH, SOD, and CAT levels in renal tissue. In addition, Cd also produced significantly increased ( p < 0.01) urea and creatinine levels in serum and TBARS levels in renal tissues. Rats treated with ROF significantly ( p < 0.01) restore the altered levels of kidney injury markers, nonenzymatic antioxidant, as well as depleted enzymes in dose-dependent manner. An increased expression of NF-κB p65 and decreased expression of GST and NQO1 in the Cd only treated group were significantly reversed by high dose of ROF (1.5 mg/kg). Histopathological changes were also ameliorated by ROF administration in Cd-treated groups. Conclusion: In conclusion, ROF treatment showed protective effect against renal damage and increased oxidative stress induced by Cd administration.
