Suvarna Bhasma” (gold calx) (SB) prepared as per Ayurveda, a practicing medical system, is prescribed as a medicine in several ailments including conditions like oligozoospermia and asthenoszoospermia in male infertility. SB is prepared from pure metal gold by one of the processing methods detailed in Ayurveda classics. Gold along with other elements was reported in normal human seminal plasma. . In human pathological semen samples, the level of gold was less than normal. Gold may have a role in physiological activity of sperm as shown in case of other elements. The present study supports the hypothesis that the presence of gold is essential for sperm motility. Here we studied the effect of SB on male reproductive organs and semen from epididymis of rat. Present study was performed using Charles foster strain albino rats. Rats were divided into two groups of ten each namely, Group 1 (control) and Group 2 (study). Animals in study group were orally given a fixed amount of SB for 15 days. At the end of the study treated animals showed increase in body weight (<0.05) and testicular weight. In treated animals total sperm count (<0.05) and percentage of sperm motility were increased in epididymal fluid. Histological study showed increase in interstitial area of testes (<0.001), proliferation and branching of the epithelial layer of seminal vesicle. In the Study (gold treated) Group, the increased gold level in genital system may be responsible for the increase in sperm production and motility.

Abstract Background Intimacy-related allergic reactions, including anaphylaxis, are under-reported due to social stigma, lack of awareness, and misdiagnosis. The differential diagnosis for intimacy-related anaphylaxis is extensive and includes systemic human seminal plasma allergy, exercise-induced anaphylaxis, asthma exacerbation, latex allergy, and transference of food or drug allergens through saliva or seminal fluid. Case presentation Two adolescents met on a popular dating phone application. One individual had a long-standing history of asthma and peanut allergy. Although they never kissed, the male with peanut allergy received fellatio, while the other male had eaten peanut butter before they met. During fellatio, the peanut allergic male developed respiratory symptoms, used his bronchodilator, and collapsed. He remained unconscious despite aggressive interventions by emergency personnel called to the site. The clinical history and autopsy results suggested anaphylaxis to peanut allergen exposure from the intimate exposure as the cause of death. Discussion and conclusions To date, nearly all reported cases of intimacy-related anaphylaxis involve symptomatic women. This is the first report of intimacy-related anaphylaxis involving men who have sex with men and the first report of potential allergen transfer from oral mucosa to a patient receiving fellatio. Based on the paucity of published cases, death from intimacy-related anaphylaxis is exceedingly rare. Post-mortem analysis is inherently difficult, as an elevated tryptase level has myriad potential causes; nevertheless, the authors suggest that intimacy-related anaphylaxis due to peanut allergy is the most likely diagnosis. With increasing popularity of relationship applications, especially amongst stigmatized populations, this case highlights the importance of allergy awareness and patient education to decrease risk, particularly in the adolescent population, who are already at increased risk of severe anaphylaxis. Especially amongst those participating in intimate activities, disclosure of one’s allergies warrants discussion, as the outcome can be fatal. Our case demonstrates the crucial need for increased advocacy in food allergy, education around intimacy-related anaphylaxis, and the importance of allergy awareness and prevention across all populations.

Currently, the molecular mechanisms underlining male infertility are still poorly understood. Our previous study has demonstrated that PIWI-interacting RNAs (piRNAs) are downregulated in seminal plasma of infertile patients and can serve as molecular biomarkers for male infertility. However, the source and mechanism for the dysregulation of piRNAs remain obscure. In this study, we found that exosomes are present in high concentrations in human seminal plasma and confirmed that piRNAs are predominantly present in the exosomal fraction of seminal plasma. Moreover, we showed that piRNAs were significantly decreased in exosomes of asthenozoospermia patients compared with normozoospermic men. By systematically screening piRNA profiles in sperms of normozoospermic men and asthenozoospermia patients, we found that piRNAs were parallelly reduced during infertility. At last, we investigated the expression of some proteins that are essential for piRNAs biogenesis in sperms and therefore identified a tight correlation between the levels of spermatozoa piRNA and MitoPLD protein, suggesting that the loss-of-function of MitoPLD could cause a severe defect of piRNA accumulation in sperms. In summary, this study identified a parallel reduction of piRNAs and MitoPLD protein in sperms of asthenozoospermia patients, which may provide pathophysiological clues about sperm motility.

In light of recent research, there is increasing evidence showing that extracellular semen components have a significant impact on the immune reaction of the female partner, leading to the tolerogenic response enabling the embryo development and implantation as well as further progress of healthy pregnancy. Seminal plasma glycoproteins are rich in the unique immunomodulatory glycoepitopes that may serve as ligands for endogenous lectins that decorate the surface of immune cells. Such interaction may be involved in modulation of the maternal immune response. Among immunomodulatory glycans, Lewis type antigens have been of interest for at least two decades, while the importance of T/Tn antigens and related structures is still far from understanding. In the current work, we applied two plant lectins capable of distinguishing glycoepitopes with terminal GalNAc and Gal to identify glycoproteins that are their efficient carriers. By means of lectin blotting and lectin affinity chromatography followed by LC-MS, we identified lactotransferrin, prolactin inducible protein as well as fibronectin and semenogelins 1 and 2 as lectin-reactive. Net-O-glycosylation analysis results indicated that the latter three may actually carry T and/or Tn antigens, while in the case of prolactin inducible protein and lactotransferrin LacdiNAc and lactosamine glycoepitopes were more probable. STRING bioinformatics analysis linked the identified glycoproteins in the close network, indicating their involvement in immune (partially innate) processes. Overall, our research revealed potential seminal plasma ligands for endogenous Gal/GalNAc specific lectins with a possible role in modulation of maternal immune response during fertilization.

Aim: The major aim of this study is to compare Zinc (Zn) concentration present in human seminal plasma of asthenospermia (motility issues) and normospermia semen samples for identification of clinical relevance. Materials and Methods: Semen samples of normospermia (N=75), asthenospermia (N=75) were collected and semen analysis report has been done by standard world health organization protocol. Zn concentration was evaluated by using atomic absorption spectroscopy (AAS) for both the groups by using standard protocol. Results: Independent sample T-test on fertile and infertile men (asthenospermia and normospermia group) reveals that the zinc concentration is statistically insignificant (P=0.367). Zinc concentration was found to be high in normospermia (mean 2.5 mg/ml) over asthenospermia (mean 0.90 mg/ml) samples. Conclusion: Zn being an essential element for normal functioning for fertilization, its absence or less concentration may lead to infertility. We found high concentration of Zn in normospermia which reflects normal conditions for fertilization and less concentration in asthenospermia reflecting infertile conditions.
