Defence response of tomato seedlings to oxidative stress induced by phenolic compounds from dry olive mill residue

Barley (Hordeum vulgare) accumulates phenolic compounds (PhCs), which play a key role in plant defense against environmental stressors as antioxidants or UV screening compounds. The influence of light and atmospheric CO2 concentration ([CO2]) on the accumulation and localization of PhCs in barley leaves was examined for two varieties with different tolerances to oxidative stress. PhC localization was visualized in vivo using fluorescence microscopy. Close relationships were found between fluorescence-determined localization of PhCs in barley leaves and PhC content estimated using liquid chromatography coupled with mass spectroscopy detection. Light intensity had the strongest effect on the accumulation of PhCs, but the total PhC content was similar at elevated [CO2], minimizing the differences between high and low light. PhCs localized preferentially near the surfaces of leaves, but under low light, an increasing allocation of PhCs in deeper mesophyll layers was observed. The PhC profile was significantly different between barley varieties. The relatively tolerant variety accumulated significantly more hydroxycinnamic acids, indicating that these PhCs may play a more prominent role in oxidative stress prevention. Our research presents novel evidence that [CO2] modulates the accumulation of PhCs in barley leaves. Mesophyll cells, rather than epidermal cells, were most responsive to environmental stimuli in terms of PhC accumulation.

Alperujo—a two-phase olive mill waste that is composed of olive vegetation water and solid skin, pulp, and seed fragments - is a highly valuable olive by-product due to its high content in phenolic compounds. In this study, we assessed whether β-cyclodextrin (β-CD), which is used to extract and protect alpejuro phenolic compounds (hydroxytyrosol-O-glucoside, tyrosol, caffeic, and p-coumaric acids) could impact on their bioaccessibility (i.e., the percentage of molecule found in the aqueous phase of the digesta) and uptake by intestinal cells, by using an in vitro digestion model and Caco-2 TC7 cells in culture, respectively. Our results showed that β-CD did not change the bioaccessibility of the selected phenols. Hydroxytyrosol-O-glucoside and caffeic did not cross Caco-2 cell monolayers. Conversely ferulic acid, identified as the main caffeic acid intestinal metabolite, was absorbed through intestinal cell monolayers (~20%). Interestingly, β-CD moderately but significantly improved the local absorption of tyrosol and p-coumaric acid (2.3 + 1.4% and 8.5 ± 4.2%, respectively, p < 0.05), even if their final bioavailability (expressed as bioaccessibility × absorption by Caco-2 cells) was not modified (16.2 ± 0.6% vs. 16.8 ± 0.5% for tyrosol and 32.0 ± 3.2% vs. 37.2 ± 3.2% for p-coumaric acid, from pure alperujo and alperujo complexed with β-CD, respectively). Overall, our results show that β-CD is an interesting extraction and storage agent for phenolic compounds that does not alter their in vitro bioavailability.

Industrial oil olives have a fundamental economic importance for many countries around the Mediterranean Sea. However, during oil olive production, large amount of polluted waters known as olive mill wastewaters (OMW) are generated. Many studies have been undertaken to find efficient treatment systems and several techniques have been tested, but there have been few studies investigating sand filter or macrophytic plants for this type of effluent. The objective of this work is to study of the suitability of macrophytic plants to treat olive mill wastewater (OMW) after their passage through sand filter. The experimental pilot consists of a sand filter followed by a planted system. The sand filter is filled with 50 cm of sand and 10 cm of gravel in the top and the bottom of the filter. The alimentation (4 cm/j) is done sequentially following one day wet /three days dry cycle. In order to activate the degradation processes in the sand filter, OMW are diluted at 50% by domestic wastewater. The second step of the treatment consists of a tank (1x1x1 m3) filled with 20 cm of gravel and 60 cm of soil planted with a mixture of aquatic plants (Phragmites australis, Typha latifolia and Arundo donax) at a density of 25 young plants/m2 and irrigated two times in the week by 20L of preliminary filtered OMW. The obtained results show that the sand filter ensures a neutralization of the OMW pH which passes from 4.79 to 7.68. The enrichment of sand by the micro-organisms makes it possible to have an important mineralization of the organic matter. After ten weeks of operation of this system without clogging sign, the rate of abatement of the TSS, total COD, dissolved COD and phenolic compounds is about 70%, 79%, 76% and 81% respectively. The preliminary outputs by the planted system show a good adaptation of the tested plants to the preliminary treated OMW. The pilot allows an elimination of 94% of TSS, 99.7% of total COD, 99.5% of dissolved COD and 95% of phenolic compounds. With regard to OMW treatment efficiency, the results obtained agree with important role of macrophytes for maintaining the aquatic plants treatment capability particularly for wastewater with high organic matter.

Wastewaters from the olive oil industry are a regional environmental problem. Their phenolic content provides inherent toxicity, which reduces the treatment potential of conventional biological systems. In this study, Sulfate Radical based Advanced Oxidation Processes (SRbAOPs) are compared with advanced oxidation processes (namely Fenton’s peroxidation) as a depuration alternative. Synthetic olive mill wastewaters were submitted to homogeneous and heterogeneous SRbAOPs using iron sulfate and solid catalysts (red mud and Fe-Ce-O) as the source of iron (II). The homogenous process was optimized by testing different pH values, as well as iron and persulfate loads. At the best conditions (pH 5, 300 mg/L of iron and 600 mg/L of persulfate), it was possible to achieve 39%, 63% and 37% COD, phenolic compounds and TOC removal, respectively. The catalytic potential of a waste (red mud) and a laboratory material (Fe-Ce-O) was tested using heterogenous SRbAOPs. The best performance was achieved by Fe-Ce-O, with an optimal load of 1600 mg/L. At these conditions, 27%, 55% and 5% COD, phenolic compounds and TOC removal were obtained, respectively. Toxicity tests on A. fischeri and L. sativum showed no improvements in toxicity from the treated solutions when compared with the original one. Thus, SRbAOPs use a suitable technology for synthetic OMW.

Supplementation with bioactive compounds capable of regulating energy homeostasis is a promising strategy to manage obesity. Here, we have screened the ability of different phenolic compounds (myricetin, kaempferol, naringin, hesperidin, apigenin, luteolin, resveratrol, curcumin, and epicatechin) and phenolic acids (p-coumaric, ellagic, ferulic, gallic, and vanillic acids) regulating C. elegans fat accumulation. Resveratrol exhibited the strongest lipid-reducing activity, which was accompanied by the improvement of lifespan, oxidative stress, and aging, without affecting worm development. Whole-genome expression microarrays demonstrated that resveratrol affected fat mobilization, fatty acid metabolism, and unfolded protein response of the endoplasmic reticulum (UPRER), mimicking the response to calorie restriction. Apigenin induced the oxidative stress response and lipid mobilization, while vanillic acid affected the unfolded-protein response in ER. In summary, our data demonstrates that phenolic compounds exert a lipid-reducing activity in C. elegans through different biological processes and signaling pathways, including those related with lipid mobilization and fatty acid metabolism, oxidative stress, aging, and UPR-ER response. These findings open the door to the possibility of combining them in order to achieve complementary activity against obesity-related disorders.

Steam-explosion is a promising technology for recovering phenolic compounds from olive mill solid waste (OMSW) due to its high impact on the structure of the fibre. Moreover, the recovery of the phenols, which are well-known microbial inhibitors, could improve the subsequent biomethanization of the dephenolized OMSW to produce energy. However, there is a considerable lack of knowledge about how the remaining phenolic compounds could affect a long-term biomethanization process of steam-exploded OMSW. This work evaluated a semi-continuous mesophilic anaerobic digestion of dephenolized steam-exploited OMSW during a long operational period (275 days), assessing different organic loading rates (OLRs). The process was stable at an OLR of 1 gVS/(L·d), with a specific production rate of 163 ± 28 mL CH4/(gVS·d). However, the increment of the OLR up to 2 gVS/(L·d) resulted in total exhaust of the methane production. The increment in the propionic acid concentration up to 1486 mg/L could be the main responsible factor for the inhibition. Regardless of the OLR, the concentration of phenolic compounds was always lower than the inhibition limits. Therefore, steam-exploited OMSW could be a suitable substrate for anaerobic digestion at a suitable OLR.
