Isolation and identification of a novel LCI like antibacterial protein from Bacillus sp. MD-5 reveals its potential application in controlling Staphylococcus aureus in food industry

Food fermentation is one of various food processing techniques that has sufficient benefits of nutrition values, and also contains lactic acid bacteria which potentially inhibit pathogenic bacteria, thus prolong shelf life of  products. Bakasang is a traditional fermented food from North Sulawesi since many years ago. Reported research of bakasang previously had described that lactic acid bacteria was the dominant isolates and therefore current research  aimed to isolate and identify the lactic acid bacteria which associated during fermentation day 1 and day 15, respectively. Raw materials used were 5 kg intestine and liver of skipjack brought from local market Bersehati Manado. The intestine and liver of skipjack were washed and smashed and mixed with 10% salt  and 5% rice  from weight of the samples and then filled into bottle to be fermented for 15 days. Every 3 days (1,3,6,9,12,15), the samples were collected and analyzed for total lactic acid bacteria by using Total Plate Count Method on de Mann Rogosa Sharpe Agar after incubation at 37°C for 24 h. The colonies  grown were transferred to Tryptic Soy Broth and followed by streaking them on Tryptic Soy Agar and the free growing colony on agar medium were isolated into slant agar which were used for biochemical test such as Gram’s staining, motility test, catalase test, oksidase test, H2S test, IMVIC test (Indole, Methyl Red, Voges Proskauer, Citrate) and carbohydrate fermentation. The results showed that Lactobacillus sp., Bacillus sp., Eubacterium sp., and Bifidobacterium sp. All these four bacteria were distributed from day 1 to day 15 of the fermentation process© Fermentasi bahan pangan merupakan salah satu dari sekian banyak teknik pengolahan makanan yang mempunyai banyak manfaat dari kualitas gizi, mengandung bakteri asam laktat sehingga menghambat bakteri patogen sehingga daya simpan lebih panjang. Bakasang merupakan makanan fermentasi tradisional masyarakat Sulawesi Utara yang sudah ada sejak lama. Penelitian yang telah dilakukan terhadap bakasang menghasilkan informasi bahwa terdapat bakteri asam laktat pada bakasang sehingga menjadi tujuan untuk mengisolasi dan identifikasi bakteri asam laktat selama proses fermentasi 1-15 hari. Bahan baku bakasang ialah jeroan (usus dan hati) ikan cakalang Katsuwonis pelamis sebanyak 5 kg yang diambil dari pasar Bersehati Manado. Sampel jeroan dibersihkan kemudian dihancurkan, ditambahkan garam 10% dan nasi 5% kemudian difermentasi selama 15 hari dengan mengambil tiap-tiap sampel setiap 1, 3, 6, 9, 12, dan 15 untuk dihitung jumlah bakteri asam laktat dengan menggunakkan metode Total Plate Count pada media de Mann Rogosa Sharpe Agar dan koloni yang tumbuh di tumbuhkan  kembali pada media Tryptic Soy Broth  dan digores kembali pada media Tryptic Soy Agar, koloni yang tumbuh digores pada media slant agar yang selanjutnya diidentifikasi bakteri asam laktat berdasarkan uji biokimia yaitu uji pewarnaan Gram, uji motility, uji katalase, uji oksidase, uji H2S dan uji IMVIC (Indole, MethylRed, Voges Proskauer, Citrate). Hasil menunjukkan bahwa selama proses fermentasi berlangsung terdapat 4 genera bakteri asam laktat sesuai yaitu Lactobacillus sp., Bacillus sp., Eubacterium sp., dan Bifidobacterium sp., ke 4 genera ini tersebar pada fermentasi hari 1 sampai hari ke 15©

AbstractIn the past few decades, there have been remarkable advances in our knowledge of gold nanoparticles (AuNPs) and synthesizing methods. AuNPs have become increasingly important in biomedical and industrial applications. As a newly implemented method, AuNPs are being used in nanopackaging industries for their therapeutic and antibacterial characteristics as well as their inert and nontoxic nature. As with other NPs, AuNPs have privileges and disadvantages when utilized in the food sector, yet a significant body of research has shown that, due to the specific nontoxic characteristics, AuNPs could be used to address other NP flaws. In this mini review, we present synthesizing methods, food industry applications, and mechanisms of action of gold nanoparticles. Regarding the investigations, gold nanoparticles can play a major role to reduce microbial load in foodstuff and therefore can be implemented in food packaging as an effective approach.

Contamination of meats and meat products with foodborne pathogenic bacteria raises serious safety issues in the food industry. The antibacterial activities of phosphorous-fluorine co-doped TiO2 nanoparticles (PF-TiO2) were investigated against seven foodborne pathogenic bacteria: Campylobacter jejuni, Salmonella Typhimurium, Enterohaemorrhagic E. coli, Yersinia enterocolitica, Shewanella putrefaciens, Listeria monocytogenes and Staphylococcus aureus. PF-TiO2 NPs were synthesized hydrothermally at 250 °C for 1, 3, 6 or 12 h, and then tested at three different concentrations (500 μg/mL, 100 μg/mL, 20 μg/mL) for the inactivation of foodborne bacteria under UVA irradiation, daylight exposure or dark conditions. The antibacterial efficacies were compared after 30 min of exposure to light. Distinct differences in the antibacterial activities of the PF-TiO2 NPs, and the susceptibilities of tested foodborne pathogenic bacterium species were found. PF-TiO2/3 h and PF-TiO2/6 h showed the highest antibacterial activity by decreasing the living bacterial cell number from ~106 by ~5 log (L. monocytogenes), ~4 log (EHEC), ~3 log (Y. enterolcolitca, S. putrefaciens) and ~2.5 log (S. aureus), along with complete eradication of C. jejuni and S. Typhimurium. Efficacy of PF-TiO2/1 h and PF-TiO2/12 h NPs was lower, typically causing a ~2–4 log decrease in colony forming units depending on the tested bacterium while the effect of PF-TiO2/0 h was comparable to P25 TiO2, a commercial TiO2 with high photocatalytic activity. Our results show that PF-co-doping of TiO2 NPs enhanced the antibacterial action against foodborne pathogenic bacteria and are potential candidates for use in the food industry as active surface components, potentially contributing to the production of meats that are safe for consumption.

El cáncer es la segunda causa de muerte en el mundo, y específicamente en Chile el cáncer colorrectal es el único que presenta un aumento sostenido de la mortalidad en la última década. La búsqueda de nuevos agentes quimioterapeúticos anticancerígenos ha propuesto a los microorganismos extremófilos como una fuente potencial para obtener moléculas citotóxicas, que induzcan apoptosis en las células tumorales. Las condiciones extremas del continente antártico y las presiones selectivas por el espacio y los nutrientes que se producen entre los microorganismos del rizobioma de la planta Deschampsia antarctica Desv sugirieron como hipótesis que las bacterias rizosféricas aisladas en la Antártica secretan al sobrenadante de cultivo moléculas bioactivas que inhiben la invasión y proliferación de líneas tumorales humanas de origen colorrectal mediante un mecanismo apoptótico. En este sentido, el objetivo general del trabajo fue identificar y caracterizar a moléculas bioactivas con acción antinvasiva y antiproliferativa, además, determinar el mecanismo inhibitorio de la proliferación en líneas tumorales humanas de origen colorrectal. Los resultados del primer objetivo específico demostraron que los sobrenadantes de cultivo de los aislados rizosféricos antárticos K2 y MI disminuyeron la viabilidad de la línea celular de adenocarcinoma colorrectal LoVo en el ensayo de reducción metabólica del MTT. Además, como los sobrenadantes no tuvieron efecto en la viabilidad de las bacterias E. coli y Staphylococcus aureus, y tampoco en los hongos unicelulares Candida albicans y Saccharomyces cerevisiae, el resultado indicó que la actividad antiproliferativa fue selectiva hacia la línea celular LoVo.

Introduction: The aim of this study was to monitor the occurrence of bacterial agents causing mastitis, forms of mastitis and antibiotic resistance in 300 ewes on a farm in Eastern Slovakia. Material and methods: During the milking season, were performed three complex investigations including clinical examination, California Mastitis Test and laboratory analysis of milk samples. The investigations and milk samples were taken in three phases; the beginning (April), the middle (June) and the end (September) of the milking season. Results: Of all the samples (806), 225 (28.0%) were positive for bacterial pathogens. The highest incidence of mastitis (33.3%) was recorded in September, while April (23.8%) and June (25.3%) had lower incidence. The samples from September showed the highest incidence of subclinical mastitis (20.1%), with 13.2% being an acute form of mastitis. Coagulase-negative staphylococci (CNS) were identified in 61.7% of the positive samples. Especially, S. chromogenes, S. epidermidis and S. schleiferi were most frequently isolated.  Staphylococcus aureus was the causative agent in 20.0% of the positive samples and caused acute or subclinical mastitis in the affected ewes. The tested bacteria showed very high resistance to novobiocin (59.5%) and penicillin (51.4%) and high resistance to amoxycillin (35.1%). We found that 80% of Staphylococcus aureus bacteria tested for antibiotic resistance were resistant to novobiocin and 70% were resistant to penicillin. Of all tested CNS, 56.5% were resistant to novobiocin, 39.1% to penicillin, and 34.7% to amoxicillin. Conclusion: Proper isolation and identification of the causative organism play a significant role in the prevention and control of the intramammary infection. In our study, a combinations of Streptomycin, Ciprofloxacin and Tetracycline were the most effective antibiotics for the control of mastitis.

Antibiotik adalah bahan obat yang sangat memegang peranan penting dalam menanggulangi penyakit infeksi. Senyawa antimikroba dapat diperoleh dari tumbuh-tumbuhan, dan mikroba. Senyawa antimikrob yang dihasilkan oleh mikroba memiliki keunggulan dibandingkan dengan antibiotik sintetik karena memiliki sifat yang lebih efektif, sebab targetnya spesifik serta toksisitasnya rendah. Tujuan dari penelitian yang telah dilakukan ialah untuk mengisolasi bakteri dari tanah yang mampu menghasilkan senyawa antimikrob, serta untuk menguji aktivitas penghambtannya terhadap  pertumbuhan Escherichia coli dan Staphylococcus aureus. Metode yang telah digunakan dalam penelitian ini ialah isolasi, purifikasi, dan seleksi bakteri dari sampel tanah dengan metode uji penghambatan sedangkan bakteri Bacillus sp. Merupakan bakteri pembanding. Berdasarkan hasil penelitian yang diperoleh terdapat 2 isolat yang berpotensi memiliki aktivitas antimikroba yaitu isolat 1 dan isolate 4. Isolat 1 lebih berpotensi menghambat E. coli dengan indeks hambat 4.0 mm dibandingkan dengan penghambatan S. aureus dengan indeks hambat 3.1 mm. Sedangkan isolat 4 lebih berpotensi menghambat S. aureus dengan indeks hambat 2.8 mm dibandingkan dengan penghambatan terhadap E. coli dengan indeks hambat 1.4 mm.

Abstract Background: Staphylococcus aureus is one of the important superbugs distributed throughout the world. It causes minor skin infections to severe complications including nosocomial infections in both hospitals and community settings. These strains have multi-drug resistant property. Hence, they are difficult to manage which increase health-related costs and simultaneously intensifying the need for new antibiotics. The extent of Methicillin-Resistant Staphylococcus aureus (MRSA) in children is largely unknown. The study determines the current status of S. aureus and MRSA causing various infections in pediatric patients visiting International Friendship Children’s Hospital (IFCH). Methods: A cross-sectional study was conducted among patients visiting a hospital. Various clinical specimens were aseptically collected and processed according to standard microbiological procedures. Isolation and identification of S. aureus were done by microscopy, mannitol fermentation, and coagulase positivity. All identified S. aureus isolates subjected to in-vitro antibiogram by Kirby-Bauer disc diffusion technique adopting Clinical and Laboratory Standards Institute (CLSI) guideline. Isolates resistant to cefoxitin were considered to be MRSA. Whereas, isolates produced D-shaped inhibition zone around clindamycin when kept near erythromycin were considered to be Inducible Clindamycin Resistant (ICR). Results: 672 various types of clinical samples were processed from the microbiology laboratory from June and November 2015. Out of 300 culture positive samples, 52 (17.3%) were S. aureus isolates, among them 39 (75.0%) were found to be MRSA. The D-test showed that Macrolide-Lincosamide-Streptogramin-B (MLSB) phenotype was 15.4%. Conclusion: The study shows the MRSA occurrence is prevalent in pediatric patients and newer classes’ drugs are found more effective than β-lactam drugs to treat S. aureus infection. However, restriction on the indiscriminate use of such drugs may be an effective strategy to control the drug resistance.
