Pre-calving feeding of rumen-protected rice bran to multiparous dairy cows improves recovery of calcaemia after calving

Dairy cows can have different degrees of hypocalcaemia around calving. Lowering dietary Ca availability before calving can prevent it. Rice bran, treated for lower rumen degradability of phytic acid can reduce dietary availability of Ca. During 3 periods of 3 weeks, 113 multiparous cows calved in a single close-up group, which was fed first a control diet, then 140 g/kg DM of rumen-protected rice bran, and at last the control diet again. Cows joined the group 3 weeks before expected calving date and left it at calving. Blood samples were taken weekly before parturition and 0, 6 and 12 h after calving, as well as 3 and 28 d in lactation. Serum was analysed for Ca, Mg, and P. Rice bran introduction produced a transient serum Ca decrease. Rice bran feeding reduced serum P and its withdrawal reduced serum Mg. Serum Ca at calving, nadir of serum Ca and serum Ca the first 3 d after calving was higher in cows calving during rice bran feeding. Serum P decreased less and recovered faster after calving when cows had been fed rice bran. Rumen-protected rice bran reduced dietary availability of Ca and induced adaptation of Ca metabolism resulting in improved Ca and P homoeostasis at calving.

  Background: Nitrate and nitrite poisoning is associated with pasture intake that has high nitrate levels and leads to acute methemoglobinemia. Pasture may accumulate nitrate under certain conditions, such as excessively fertilized soil or en­vironmental conditions that enhance the N absorption (rain preceded by a period of drought). After ingestion of plants, this substrate reaches the rumen and, in physiological conditions, is reduced to nitrite and afterward to ammonia. The aim of this study was to evaluate changes in cholinesterase activities and oxidative stress caused by subclinical poisoning for nitrate and nitrite in cattle fed with Pennisetum glaucum in three different fertilization schemes. Materials, Methods & Results: In order to perform the experimental poisoning, the pasture was cultivated in three dif­ferent paddocks: with nitrogen topdressing (urea; group 1), organic fertilizer (group 2) or without fertilizer (group 3; control). Nitrate accumulation in forage was evaluated by the diphenylamine test. After food fasting of 12 h, nine bovine were randomly allocated to one of the experimental groups and fed with fresh forage (ad libitum) from respective pad­dock. In different time points from beginning of pasture intake (0, 2, 4, 6 and 9 h) heart rate and respiratory frequency were assessed, as well as mucous membrane color and behavioral changes. Blood samples from jugular vein into vials with and without anticoagulant were collected. From blood samples, serum nitrite levels, acetylcholinesterase (AChE) and butyrylcholinesterase (BChE) enzyme activity were evaluated, as well as oxidative stress through the following param­eters: levels of nitrate/nitrite (NOx), thiobarbituric acid reactive substances (TBARS) and reactive oxygen species (ROS), beyond the antioxidant system by enzyme activity measurement of catalase (CAT) and superoxide dismutase (SOD). The diphenylamine test was positive to group 1 and 2, so that the pasture presented 3.16 mg/kg, 2.98 mg/kg and 1.67 mg/kg of nitrate for group 1, 2 and 3, respectively. In addition, cows from group 1 demonstrated increased (P < 0.05) nitrite levels in serum, compared to other groups, and greater heart rate after 9 h (P < 0.05). The AChE and BChE activity in group 1 showed significant increase (P < 0.05) at 4 and 6 h (AChE), and 4 and 9 h (BChE) compared to group 3. Also, NOx levels were lower at 6 and 9 h (P < 0.05) and at 9 h (P < 0.05) for animals of group 1 and 2, respectively, when compared to group 3. Furthermore, in the group 1 levels of ROS and TBARS were significantly higher (P < 0.05) after 2 and 4 h, and 6 and 9 h compared to other groups, respectively. The CAT activity increased significantly (P < 0.05) with 2 and 4 h of the experiment, but on the other hand, decreased at 6 and 9 h in group 1. Nevertheless, the animals from group 2 presented only a significant reduction in this enzyme activity at 9 h. Furthermore, SOD activity was reduced in animals of groups 1 (P < 0.05) at 4, 6 and 9 h, compared to other groups. Discussion: It was concluded that the nitrate and nitrite poisoning by pasture intake cultivated and fertilized with urea leads to increased levels of serum nitrite, as well as the cholinesterase activity and causes oxidative stress in cattle. It is conjectured that the cholinesterase activity and oxidative stress may assist in understanding the pathophysiology of changes caused by poisoning.Keywords: plant toxicology, poisoning, methemoglobin, cholinergic system, oxidative stress.

Zearalenone (ZEN) and its metabolites, alpha-zearalenol (α-ZEL) and beta-zearalenol (β-ZEL), are ubiquitous in plant materials used as feed components in dairy cattle diets. The aim of this study was to confirm the occurrence of ZEN and its selected metabolites in blood samples collected from different sites in the hepatic portal system (posthepatic–external jugular vein EJV; prehepatic–abdominal subcutaneous vein ASV and median caudal vein MCV) of dairy cows diagnosed with mastitis, ovarian cysts and pyometra. The presence of mycotoxins in the blood plasma was determined with the use of combined separation methods involving immunoaffinity columns, a liquid chromatography system and a mass spectrometry system. The parent compound was detected in all samples collected from diseased cows, whereas α-ZEL and β-ZEL were not identified in any samples, or their concentrations were below the limit of detection (LOD). Zearalenone levels were highest in cows with pyometra, where the percentage share of average ZEN concentrations reached 44%. Blood sampling sites were arranged in the following ascending order based on ZEN concentrations: EJV (10.53 pg/mL, 44.07% of the samples collected from this site), ASV (14.20 pg/mL, 49.59% of the samples) and MCV (26.67 pg/mL, 67.35% of the samples). The results of the study indicate that blood samples for toxicological analyses should be collected from the MCV (prehepatic vessel) of clinically healthy cows and/or cows with subclinical ZEN mycotoxicosis. This sampling site increases the probability of correct diagnosis of subclinical ZEN mycotoxicosis.

Phosphoproteomics is a cutting-edge technique that can be utilized to explore adipose tissue (AT) metabolism by quantifying the repertoire of phospho-peptides (PP) in AT. Dairy cows were supplemented with conjugated linoleic acid (CLA, n = 5) or a control diet (CON, n = 5) from 63 d prepartum to 63 d postpartum; cows were slaughtered at 63 d postpartum and AT was collected. We performed a quantitative phosphoproteomics analysis of subcutaneous (SC) and omental (OM) AT using nanoUPLC-MS/MS and examined the effects of CLA supplementation on the change in the phosphoproteome. A total of 5919 PP were detected in AT, and the abundance of 854 (14.4%) were differential between CON and CLA AT (p ≤ 0.05 and fold change ± 1.5). The abundance of 470 PP (7.9%) differed between OM and SC AT, and the interaction treatment vs. AT depot was significant for 205 PP (3.5% of total PP). The integrated phosphoproteome demonstrated the up- and downregulation of PP from proteins related to lipolysis and lipogenesis, and phosphorylation events in multiple pathways, including the regulation of lipolysis in adipocytes, mTOR signaling, insulin signaling, AMPK signaling, and glycolysis. The differential regulation of phosphosite on a serine residue (S777) of fatty acid synthase (FASN) in AT of CLA-supplemented cows was related to lipogenesis and with more phosphorylation sites compared to acetyl-coenzyme A synthetase (ACSS2). Increased protein phosphorylation was seen in acetyl-CoA carboxylase 1 (ACACA;8 PP), FASN (9 PP), hormone sensitive lipase (LIPE;6 PP), perilipin (PLIN;3 PP), and diacylglycerol lipase alpha (DAGLA;1 PP) in CLA vs. CON AT. The relative gene expression in the SC and OM AT revealed an increase in LIPE and FASN in CLA compared to CON AT. In addition, the expression of DAGLA, which is a lipid metabolism enzyme related to the endocannabinoid system, was 1.6-fold higher in CLA vs. CON AT, and the expression of the cannabinoid receptor CNR1 was reduced in CLA vs. CON AT. Immunoblots of SC and OM AT showed an increased abundance of FASN and a lower abundance of CB1 in CLA vs. CON. This study presents a complete map of the SC and the OM AT phosphoproteome in dairy cows following CLA supplementation and discloses many unknown phosphorylation sites, suggestive of increased lipid turnover in AT, for further functional investigation.

Flaxseed meal (FM) is rich in the plant lignan secoisolariciresinol diglucoside (SDG) which is converted to the mammalian lignans enterodiol and enterolactone (EL) by ruminal microbiota. Feeding FM to dairy cows increases linearly EL concentration in milk but enterodiol is not detected. The objectives of the study were to determine the length of time to obtain peak EL concentration in the milk of dairy cows fed 20% FM and the length of time to return to EL baseline level in milk when cows are switched from high to low intake of flax SDG. A total of 12 multiparous lactating Holstein cows were assigned randomly to one of two feeding regimens: the control (CO) diet was fed for 6 weeks or the FM diet was fed from week 0 to 3 inclusive and then cows were switched to the control diet from week 3 to 6 inclusive. Milk samples were taken weekly for EL analysis. There was a significant interaction between feeding regimen and week for milk concentration of EL as a result of higher concentration of EL from week 1 to 3 for cows on the FM regimen compared with those on the CO regimen. Concentrations of milk EL on the FM regimen maintained uniform high levels from week 1 to 3 and they decreased significantly from week 3 to 4 when the CO diet was reintroduced in week 3. This study suggests that the conversion of SDG to the mammalian lignan EL and the transfer of EL to the mammary gland are well established after one week of feeding 20% FM in the diet of dairy cows and that milk concentration of EL returns to baseline level after one week of FM deprivation.

Background Arterial catheters are potential sources of nosocomial infection. Objective To investigate use of a closed blood conservation device in preventing catheter-related bloodstream infections in children after cardiac surgery. Methods Children with an indwelling arterial catheter after cardiac surgery were randomly assigned to 2 groups: a control group with a conventional 3-way stopcock in the catheter system and an interventional group with the conservation device in the catheter system. Catheter tips, catheter intraluminal fluid, and blood samples obtained from the catheter and peripherally were cultured for microbiological analysis. RESULTS Intraluminal fluid contamination was significantly lower (P = .03) in the interventional group (3 of 147 catheters) than in the control group (10 of 137 catheters). The 2 groups did not differ significantly in the rate of tip colonization (9 of 147 vs 12 of 137; P = .40) or in the number of catheter-related bloodstream infections (0 of 147 vs 2 of 137; P = .21). Conclusion Use of a closed blood conservation device could decrease the incidence of catheter-related contamination of intraluminal fluid.

We hypothesized that lameness has an impact on milk flow traits. The aim of the current study was therefore to investigate the relation between lameness and milk flow traits in dairy cows. For this study 73 healthy and 55 cows with lameness were selected. Lameness was diagnosed by a local specialized veterinarian, according to the standard procedure. The blood samples were collected during clinical examination. The milking properties of cows were evaluated twice in a row—during evening and morning milking. The selected cows in the current lactation did not receive veterinary treatment, and correct hoof trimming was not performed at least four weeks before the experiment. The measurements were taken by two electronic mobile milk flow meters (Lactocorder®®, WMB AG, Balgache, Switzerland). Milk flow data were processed using LactoPro 5.2.0 software (Biomelktechnik Swiss). Cortisol concentration was measured with the automated analyzer TOSOH®® AIA-360 (South San Francisco, CA, USA). We found out that milk flow traits can act as biomarkers of lameness in dairy cows. We determined that the milk yield in the first minute of healthy dairy cows was 1.77 kg higher than that of lame cows. The electrical conductivity during the initial time of milking of healthy cows was 0.24 mS/cm lower than that of the lame group. The milking duration of LA cows was 1.07 min shorter and the time of incline in milk flow from 0.5 kg/min till the reach of the plateau phase was longer. The risk of lameness was most clearly indicated by an increase in blood cortisol concentration; if its blood level in cows exceeds 1 µg/dL, the risk of identifying lameness increases 4.9 times.

The objective of this study was to evaluate nutritional and energetic value of rice by-products, with or without phytase, using growing pigs. Thirty-six male pigs were housed in individual metabolic cages. Total collection of feces and urine was carried out in two periods of ten days: five days for adaptation and five days for collection. A randomized blocks design was used, considering the sampling period as a block, with five treatments and seven replicates. Two control diets (with and without phytase - Phy) were used in the digestibility calculations, the latter in order to evaluate the enzyme influence on energy digestibility of the tested ingredients. The control diet was replaced by 30% of one of the ingredients: defatted rice bran (DRB) with and without Phy and dephytinised defatted rice bran (DDRB). The use of Phy in the control diet did not influence DRB+Phy energy digestibility. Relative to DRB+Phy, dephytinised defatted rice bran had higher contents of ME and digestible protein but lower digestible P and Ca. Phy supplementation increased Ca and P utilization of DRB and improved energy and protein digestibility. The DRB without Phy showed the lowest digestibility coefficients for all responses. Metabolizable energy, digestible protein, phosphorus and calcium of DRB, DRB+Phy and DDRB were respectively, 2140, 2288 and 2519kcal kg-1; 79.25, 92.41 and 107.10g kg-1; 1.62, 3.41, and 2.11g kg-1 and 2.80, 3.79 and 2.90g kg-1.

Carrot juice is a critical source of vitamins, selenium, and β-carotene, which is suggested to protect from Rheumatoid Arthritis (RA). The present study aimed to show the effect of carrot juice supplementation compared to selenium tablet supplementation, so our study includes (44) blood samples belonging to young men with RA. All patients aged (20-45) years, 44 blood samples were obtained before treatment at week (0), twenty-four men with RA were supplied with fresh carrot juice. The other group of 20 patients was given an artificial selenium supplement for 21 days as well, and the results were analyzed.The samples were collected from Kirkuk hospital, and external specialized clinical from October/2019 to September /2020; experimental groups were divided into three groups: Group 1 : (44) men Rheumatoid arthritis (RA) before treatment, Group 2: (24) men with RA+ Carrot juice, Group 3: (20) men with RA +Selenium Tab, We reach to following results: the patients who have RA consumption Carrot juice and patients take up Se tab. show significant decrease respectively in RBCs, WBCs, ERS, and RF compared with the Rheumatoid arthritis group, so as the results show a significant decrease in Leptin, IL-6, C-Reactive Protein, and TNF-α concentrations in comparison with the Rheumatoid arthritis group. In contrast, we found a significant increase in GSH, Selenium concentrations, and VD3 in men with RA administration carrot juice and patients' consumption Se tab. Respectively compared with the RA group and, finally, our finding shows no difference in Ceruloplasmin in experimental groups.

Routine analysis of blood parameters requires high precision. Therefore a significant number of methods and recommendations has been developed to ensure the obtaining of precise results. The aim of this study was to investigate whether the type of anticoagulant affects the values of biochemical parameters in the blood of dairy cows in relation to the values obtained from the serum. The study was carried out on 20 healthy cows of Holstein- Friesian breed in the second month of lactation. The blood was taken by venipuncture from v.coccigea. There were used five types of vacutainers as follows: for the serum separation, with heparin, EDTA, citrate, and fluoride. Samples from each cow were taken in all five types of vacutainers respectively. There was determined the concentration of: albumin, total protein, glucose, Ca, P, BHB, NEFA, urea, cholesterol, triglycerides, ALT, AST, AP, GGT and bilirubin. The results of the analysis of samples that contained anticoagulants were compared statistically with analysis in serum that was considered to be the control one. It was also calculated the percentage of deviation in average values of concentration of metabolites obtained from samples with different anticoagulants in regard to the serum value. The results of the investigation show that anticoagulants influence the values of biochemical parameters in the blood of cows. In the samples where heparin was used as anticoagulant there was found higher albumin level (deviation 4.1%) as well as total protein (1.4%), but lower value of alkaline phosphatase activity relative to the serum (-33%). In the samples in which EDTA was used there were significantly lower levels of total protein (-5.8%), Ca (-49.6%), P (-17.7%), AP (-32%) and higher value AST (10.6%) compared to serum. In the samples where citrate or fluoride were used as anticoagulants there were found lower values of total protein, albumin, glucose (only citrate), Ca, P, BHB, NEFA, urea (only citrate), cholesterol, AP and GGT (only fluoride) and lower bilirubin when using citrate or higher when using fluoride, in comparison to the serum. Deviations of biochemical parameters measured from blood samples that were in citrate or fluoride were as follows: total protein (-5.3%, -5.2%), albumin (-7%, -5.7%), glucose (only citrate -6.2%), Ca (-55%, -82%), P (-29%, -24%), BHB (-53%, -80%), NEFA (-62.1%, -79 , 4%), urea (only citrate, -25.5%), cholesterol (-28.6%, -28.4%), AP (-38%, -32%), and GGT (only -17 fluoride, 8%) and higher bilirubin (-22.8%, 64.4%). For routine clinical biochemical analysis of blood of cows it is the most reliable to use the samples with heparin as an anticoagulant, because the use of other anticoagulants gives large variations in the values of biochemical parameters in relation to serum.
