Routine analysis of blood parameters requires high precision. Therefore a significant number of methods and recommendations has been developed to ensure the obtaining of precise results. The aim of this study was to investigate whether the type of anticoagulant affects the values of biochemical parameters in the blood of dairy cows in relation to the values obtained from the serum. The study was carried out on 20 healthy cows of Holstein- Friesian breed in the second month of lactation. The blood was taken by venipuncture from v.coccigea. There were used five types of vacutainers as follows: for the serum separation, with heparin, EDTA, citrate, and fluoride. Samples from each cow were taken in all five types of vacutainers respectively. There was determined the concentration of: albumin, total protein, glucose, Ca, P, BHB, NEFA, urea, cholesterol, triglycerides, ALT, AST, AP, GGT and bilirubin. The results of the analysis of samples that contained anticoagulants were compared statistically with analysis in serum that was considered to be the control one. It was also calculated the percentage of deviation in average values of concentration of metabolites obtained from samples with different anticoagulants in regard to the serum value. The results of the investigation show that anticoagulants influence the values of biochemical parameters in the blood of cows. In the samples where heparin was used as anticoagulant there was found higher albumin level (deviation 4.1%) as well as total protein (1.4%), but lower value of alkaline phosphatase activity relative to the serum (-33%). In the samples in which EDTA was used there were significantly lower levels of total protein (-5.8%), Ca (-49.6%), P (-17.7%), AP (-32%) and higher value AST (10.6%) compared to serum. In the samples where citrate or fluoride were used as anticoagulants there were found lower values of total protein, albumin, glucose (only citrate), Ca, P, BHB, NEFA, urea (only citrate), cholesterol, AP and GGT (only fluoride) and lower bilirubin when using citrate or higher when using fluoride, in comparison to the serum. Deviations of biochemical parameters measured from blood samples that were in citrate or fluoride were as follows: total protein (-5.3%, -5.2%), albumin (-7%, -5.7%), glucose (only citrate -6.2%), Ca (-55%, -82%), P (-29%, -24%), BHB (-53%, -80%), NEFA (-62.1%, -79 , 4%), urea (only citrate, -25.5%), cholesterol (-28.6%, -28.4%), AP (-38%, -32%), and GGT (only -17 fluoride, 8%) and higher bilirubin (-22.8%, 64.4%). For routine clinical biochemical analysis of blood of cows it is the most reliable to use the samples with heparin as an anticoagulant, because the use of other anticoagulants gives large variations in the values of biochemical parameters in relation to serum.

Background: In this study, the effects of White Muscle Disease (WMD) on certain blood parameters, MDA and GSH levels in liver and thyroid tissues and the thyroid hormones were investigated in sheep. Methods: Sixteen lambs (8 with WMD, 8 controls) of similar age, 3 to 50 days old, selected from the same region were used in the study. Blood samples were taken from all animals. One ml Vitamin E + Se were injected (I/M) to the lambs with WMD and blood and tissue samples were collected after the treatment as well. The serum Ca, P, T3, T4, albumin, globulin, total protein, glutathione (GSH) values were analyzed by an auto-analyzer. Malondialdehyde (MDA) and GSH amounts of the liver and thyroid tissues were also determined. Result: The low Ca ion amount in lambs with WMD was found to have increased after injections of vitamin E + Se. P levels followed the same pattern. Total protein, albumin and globulin levels increased after treatment. It was determined that there was a decrease in T3 levels and an increase in T4 levels in patient lamb sera. The levels of GSH in both the liver and thyroid tissues were found to have decreased and statistical significance (at what level) was only found for the liver GSH levels. MDA levels in the hepatic and thyroid tissues were found to have slightly increased in the WMD group. In conclusion, serum differences in Ca, P, albumin, globulin, total protein, T3, T4, GSH, GSH and MDA levels were statistically significant between the groups.

Serum profile is generally considered as comprehensive data which directly reflected animal health and their potential resistance to environmental, nutritional and pathological stress. The present study aimed to provide physiological reference values for selected biochemical parameters in Noi chickens, a famous Vietnamese native breed, at 56 days old. The collected blood samples of 355 Noi birds (164 males and 191 females) were used to evaluate biochemical serum profile. Glucose, total protein, albumin, globulin, albumin/globulin ratio, triglycerides, cholesterol, creatinine, and uric acid in the serum were measured. The variation in these values arising from different sexes was also investigated. The results indicated that there were no differences (P>0.05) in the assessed indices between male and female Noi chickens. In overall for the whole group, these values were obtained as 250.86 mg/dL, 7.34 g/dL, 3.40 g/dL, 3.94 g/dL, 1.75, 148.54 mg/dL, 190.86 mg/dL, 0.44 mg/dL, and 1.44 mg/dL, respectively. The relationship among the biochemical parameters showed relatively low coefficients (r=0.79 in maximum). The females obtained higher correlation coefficients between levels of total protein and glucose (r=0.42) as well as between total protein and globulin (r=0.79), compared to the males. It was concluded that the male and female chickens performed similar values of serum indices. The contribution of this study might expand the knowledge on the biochemical profile and improved breeding strategies of Vietnamese indigenous Noi chickens.

In this study, we report the characterisation of a novel centrifugation and spectrum-integrated veterinary clinical analyser, the AmiShield<sup>TM</sup>, which has been developed for the multiplex measurement of biochemical, electrolyte and immunoassay parameters in a point-of-care testing environment. The aims of this study were to evaluate the analytical performance of the AmiShield<sup>TM</sup> and to compare it with six reference instruments using clinical blood samples. Two hundred and four canine and 120 feline blood samples collected from veterinary teaching hospitals were analysed in parallel using the AmiShield and appropriate reference instruments. All results were evaluated separately for canine and feline specimens. The instrument’s analytical performance was evaluated initially for short- and long-term precision, bias, and observed total error using quality control material. This was followed by comparison of clinical specimens on the AmiShield analyser in parallel with the Vitros and Hitachi for biochemical parameters, VetScan and SNAPshot for total bile acids, and VetLyte and Biolyte for electrolytes. Overall, the AmiShield analyser’s performance met the standards of the American Society for Veterinary Clinical Pathology for total allowable error for most analytes, and can be considered suitable for use in veterinary clinical practices. Using canine samples, excellent correlation coefficients (r ≧ 0.92) were identified for 14 analytes of various categories including glucose, total protein, aspartate aminotransferase, alanine aminotransferase, alkaline phosphatase, total bilirubin, amylase, blood urea nitrogen, creatinine, phosphorus, Na<sup>+</sup>, K<sup>+</sup>, Cl<sup>–</sup> and total bile acid, while good correlations (0.91 ≧ r ≧ 0.80) were recorded for albumin (r = 0.91). Bland-Altman difference plots also showed agreement (greater than 95% within Limits of Agreement) for glucose, total protein, albumin, alanine aminotransferase, alkaline phosphatase, total bilirubin, amylase, blood urea nitrogen, creatinine, Na<sup>+</sup>, K<sup>+</sup>, Cl<sup>–</sup> and total bile acid between AmiShield and the reference instruments. However, aspartate aminotransferase and phosphorus exhibited higher outliers, implying potential problems associated with matrix interferences such as lipemic samples, which warrant further study. This study demonstrates that the AmiShield compares favourably with standard reference instruments, and the new device generated data of high quality for most analytes in clinical canine and feline samples. The capability of reliably measuring multi-category analytes in one device using minute amounts (170 μl) of whole blood and short turn-around times (&lt; 15 min) underlines the high potential of the device as a good alternative in-house diagnostic application.

ABSTRACT: In face of the few reports found in national literature analyzing the potential influence of parturition number in serum proteinogram and biochemical profile in the peripartum period of high yielding dairy cows, the aim of the present study was to comparatively evaluate the dynamics of these serum constituents’ concentrations in blood samples obtained from primiparous and multiparous Holstein cows, 60 and 30 days prepartum and in the day of parturition. Data were analyzed by repeated measures variance analysis (ANOVA) and differences between groups and moments were analyzed by Tukey’s test. Results were considered significant when P<0.05. Parity influenced levels of total protein, albumin, globulins, magnesium, cholesterol, which were higher in multiparous cows, as well as concentrations of ceruloplasmin, total calcium, chloride and alkaline phosphatase activity, which were higher in primiparous cows. Parturition influenced serum concentrations of ceruloplasmin (+58%), transferrin (-25%), haptoglobin (+33%), total protein (-17%), globulins (-25%), immunoglobulin A (-43%), immunoglobulin G (-24%), total calcium (-12%), inorganic phosphorus (-10%), chloride (+5%), sodium (+4%), cholesterol (-23%), triglycerides (-38.6%), as well as activities of aspartate aminotransferase (+14%) and alkaline phosphatase (+28%). A decrease in serum levels of total calcium, inorganic phosphorus, cholesterol and triglycerides was more pronounced in multiparous than in primiparous cows. These results demonstrate that the interpretation of proteinogram and serum constituents should take into consideration lactation number and the moment of parturition as relevant factors in high yielding dairy cows in the transition period.

The health status of cows is evaluated and depending on haematological and biochemical profile of blood. Nutrition is the main technological factor that can produce profound changes in the metabolic profile in animals (Dhiman et al., 1991; Khaled et al., 1999; Ingvartsen, 2006). Blood parameters analyze can lead to identify if there are errors in nutrition of lactating cows (Payne et al., 1970). The aim of this study was the evaluation of metabolic and biochemical changes that occur during colostrum period and in terms of number of lactations in cows. The biological material was represented by a total of 60 heads of dairy cows from a family farm from Sălaj County, Romania. The cows are all from Holstein breed and presented no clinical signs of any specific pathology. Blood samples were collected from the jugular vein of each cow and analyzed. 10 individuals from each of the six lactations have been randomly selected. Haematological and biochemical parameters showed variations depending on factors analyzed here. In lactation 1 Hb was 7.55±3.05 (g/dl), while in lactation 6 the value was 12.5±2.10 (g/dl). RBC ranged as follows: in lactation 1 - 28.50±2.05 and in lactation 6 - 30.02±2.05. Lymphocytes varied within very wide limits under the influence of lactation: in lactation 1 - 2.8±1.56 and in lactation 6 - 7.55±1.80. The number of lactations and lactation rank have influenced blood biochemical and hematological parameters in dairy cows. Biochemical parameters are influenced by post-partum day, showing the lowest values in the early days of colostral period and the highest in the last few days of the same period.

Background/Aims: Water-pipe smoking (WPS) is popular in the Middle East and is starting to gain popularity in several Western countries as well. It is widely and erroneously perceived to be less harmful than other forms of tobacco use. The reproductive adverse effects of cigarette smoking have been studied before with conflicting results, but data on the possible adverse reproductive effects of WPS are lacking. Here, we assessed the effects of nose-only exposure to mainstream WPS generated by commercially available honey-flavored "moasel" tobacco in mice. Methods: The duration of the session was 30 min/day for one month. Control mice were exposed to air. Twenty- four h after the last exposure, mice were killed and the testes and plasma removed for analysis. In testicular homogenates total protein, alkaline phosphatase activity, several indices of oxidative damage and Vascular Endothelial Growth Factor Receptor 2 (VEGFR2) were quantified. The plasma concentrations of leptin, testosterone, estrogen and luteinizing hormone (LH) were also measured. Histological analysis of testes and lungs was also conducted. Results: WPS caused statistically significant decreases in the plasma concentrations of leptin, testosterone, and LH, and in the concentrations of total protein and the antioxidant indices measured. A statistically non - significant decrease in VEGFR2 protein in the WPS - exposed mice compared to the control mice was also found. The body and testicular weights of mice exposed to WPS, as well as their testicular alkaline phosphatase activity and light microscopic histology, and plasma estrogen concentration were all not significantly affected by WPS. Conclusion: Further studies on the functional implications of these findings in mice exposed to WPS for longer durations are warranted.

In this study, some biochemical parameters of hair goat kids which had consumed excessive amounts of oak leaves and acorns in the autumn season and displayed toxicity symptoms during clinical inspections were evaluated. Blood samples were collected from the jugular veins of the goat kids. Glucose, AST, ALT, ALP, GGT, Na, K, Cl, cholesterol, and triglyceride levels of poisoned animals were found to be within the reported reference intervals. Creatinine, total protein, albumin, Ca, Mg and Fe levels, on the other hand, were found to be below the reference intervals. As a result, it was concluded that goats reflect the clinical findings of oak poisoning described in the literature, which courses with lowered amounts of mineral (Fe, Mg, Ca), TP, albumin and creatinine, yet without a significant change in other biochemical parameters. It can also be concluded that excessive consumption of oak by goats was capable of leading to toxicity.

AbstractAn influence of various humic-plant feed additives based on some herbs (nettle, chamomile, yarrow, perforatum), lucerne and humic materials on biochemical indices of Lohmann Brown (LB) layers blood plasma was estimated. Hens were housed in deep litter system, 20 birds in a group. Four groups were formed: control (C – standard feeding), and experimental, supplemented with preparations: E-1 herbal-humic, E-2 humic-herbal and E-3 – humic-lucerne. Hens were placed in the pens on the 16thweek of life, addition of preparations with standard food mixture started at the 22ndwk and lasted until 66thwk of life. Blood for analyses was collected four times in the following periods: 27, 37, 54 and 65thwk of life. The applied humic-plant preparations to a limited degree affected the values of examined biochemical parameters in serum: total protein (TP), albumins (Albs), glucose (Glu), urea, triacylglycerols (TAG), total cholesterol (TCh), alanine aminotransferase (ALT), aspartate aminotransferase (AST) and alkaline phosphatase (ALP). It is difficult to determine based on these study, which preparation is one the most active biologically, however is seems that humic-lucerne preparation affected the examined blood parameters to the highest degree. The reference values ranges in hens blood serum LB hens were proposed for: TP (43-65 g/l), Albs (15-22 g/l), urea (0.5-1.2 mmol/l), Glu 10-15 mmol/l), TCh (2.2-4.5 mmol/l), TAG (10-24 mmol/l), AST (4-12 U/l), ALT (150-280 mmol/l) and ALP (190-350 U/l).

This study is focus on the biochemical analysis to investigate the condition of different biochemical parameters for the Vitiligo patients. This biochemical analysis deals with the blood serum level of random blood sugar, bilirubin, urea, total protein, SGOT, SGPT, alkaline phosphatase, total cholesterol and uric acids. The increasing some of the biochemical parameters were statistically significant with the age groups. Random blood sugar analysis of the 125 patient's age group 31- 40, 41- 50 and above 50 significantly higher than the control group. No significant changes were observed in serum bilirubin, serum urea, serum creatinine, serum alkaline phosphatase (ALP) and serum asparate amino transferase (AST) level in different age group of Vitiligo patients. In this study, the serum total proteins were slightly decreased in the age group patients below 10 and 11-20 as compared to healthy controls. The average serum total protein level of the below 10 and 11-20 age group patients were 5.31 g/dl and 6.50 g/dl respectively, whereas the controls were 5.95 g/dl and 6.92 g/dl respectively. Serum cholesterol, serum triglycerides and serum uric acid levels were significantly changed in the age group 11-20 and 31-40 compared with the healthy control. The average serum cholesterol level, triglyceride (TG) level and uric acid level were 152.17 mg/dl, 90.27 mg/dl, 4.75 mg/dl and 173.40 mg/dl, 149.95 mg/dl, 5.54 mg/dl respectively for the patients of 11-20 and 31-40 years age group. The controls of these parameters for the 11-20 years and 31-40 years age groups were 150.57 mg/dl, 94.41 mg/dl, 4.28 mg/dl and 156.75 mg/dl, 101mg/dl, 4.77mg/dl respectively. These biochemical studies on Vitiligo indicated that there was no significant changed observed in some biochemical parameters in the different age group of Vitiligo patients but some biochemical parameters like total protein, total cholesterol, triglycerides (TG), uric acid and random blood sugar level were significantly changed in different age group of Vitiligo patients specially among the 11-20 and 31-40 years age group. This biochemical studies would be helpful to know details about the pathological condition of the Vitiligo patients associated with other disease.
