Abstract Advanced glycation end-products (AGEs) deteriorate bone strength. Among over 40 species identified in vivo, AGEs other than pentosidine were roughly estimated as total fluorescent AGEs (tfAGEs) due to technical difficulties. Using LC-QqTOF-MS, we established a system that enabled the quantitation of five AGEs (CML, CEL, MG-H1, CMA and pentosidine) as well as two mature and three immature enzymatic crosslinks. Human bone samples were collected from 149 patients who underwent total knee arthroplasty. Their clinical parameters were collected to investigate parameters that may be predictive of AGE accumulation. All the analytes were quantitated and showed significant linearity with high sensitivity and precision. The results showed that MG-H1 was the most abundant AGE, whereas pentosidine was 1/200–1/20-fold less abundant than the other four AGEs. The AGEs were significantly and strongly correlated with pentosidine, while showing moderate correlation with tfAGEs. Interestingly, multiple linear regression analysis revealed that gender contributed most to the accumulation of all the AGEs, followed by age, tartrate-resistant acid phosphatase-5b and HbA1c. Furthermore, the AGEs were negatively correlated with immature crosslinks. Mass spectrometric quantitation of AGEs and enzymatic crosslinks is crucial to a better understanding of ageing- and disease-related deterioration of bone strength.

Near infrared reflectance (NIR) spectroscopy is a rapid and cost-effective method for the measurement of organic constituents of agricultural products. NIR is widely used to measure feed quality around the world and is gaining acceptance in Australia. This study describes the development of an NIR calibration to measure crude protein (CP), predicted in vivo dry matter digestibility (IVDMD) and neutral detergent fibre (NDF) in temperate pasture species grown in south-western Victoria. A subset of 116 samples was selected on the basis of spectral characteristics from 461 pasture samples grown in 1987-89. Several grass and legume species were present in the population. Stepwise multiple linear regression analysis was used on the 116 samples to develop calibration equations with standard errors of 0.8,2.3 and 2.2% for CP, NDF and IVDMD, respectively. When these equations were tested on 2 independent pasture populations, a significant bias existed between NIR and reference values for 2 constituents in each population, indicating that the calibration samples did not adequately represent the new populations for these constituents. The results also showed that the H statistic alone was inadequate as an indicator of equation performance. It was confirmed that it was possible to develop a broad-based calibration to measure accurately the nutritive value of closed populations of temperate pasture species. For the resulting equations to be used for analysis of other populations, however, they must be monitored by comparing reference and NIR analyses on a small number of samples to check for the presence of bias or a significant increase in unexplained error.

AbstractFifty-one Churra da Terra Quente ewes (4–7 years old) were used to analyse the potential of real-time ultrasound (RTU) to predict the amount of internal adipose depots, in addition to carcass fat (CF). The prediction models were developed from live weight (LW) and RTU measurements taken at eight different locations. After correlation and multiple linear regression analysis, the prediction models were evaluated by k-fold cross-validation and through the ratio of prediction to deviation (RPD). All prediction models included at least one RTU measurement as an independent variable. Prediction models for the absolute weight of the different adipose depots showed higher accuracy than prediction models for fat content per kg of LW. The former showed to be very good or excellent (2.4 ⩽ RPD ⩽ 3.8) for all adipose depots except mesenteric fat (MesF) and thoracic fat, with the model for MesF still providing useful information (RPD = 1.8). Prediction models for fat content per kg of LW were also very good or excellent for subcutaneous fat, intermuscular fat, CF and body fat (2.6 ⩽ RPD ⩽ 3.2), while the best prediction models for omental fat, kidney knob, channel fat and internal fat still provided useful information. Despite some loss in the accuracy of the estimates obtained, there was a similar pattern in terms of RPD for models developed from LW and RTU measurements taken just at the level of the 11th thoracic vertebra. In vivo RTU measurements showed the potential to monitor changes in ewe internal fat reserves as well as in CF.

Objective: Thiazolidinediones have favorable influences on surrogate markers of atherosclerosis such as adiponectin, and arterial stiffness in diabetic patients. However, it is not well known whether these beneficial effects occur in subjects without diabetes, such as prediabetes or the non-diabetic metabolic syndrome (MetS). The present study was therefore designed to evaluate the effectiveness of the insulin-sensitizing agent rosiglitazone on circulating adipocytokine levels and brachial-ankle pulse wave velocity (baPWV) in non-diabetics. Design and methods: Ninety-nine subjects with prediabetes or non-diabetic MetS were randomly assigned to either rosiglitazone or an untreated control group (50 and 49 subjects respectively). The rosiglitazone group was treated daily for 12 weeks with 4 mg rosiglitazone. All subjects received a 75 g oral glucose test (OGTT) before and after treatment. In addition, baPWV, together with the levels of adiponectin, resistin, and high sensitivity C-reactive protein (hsCRP) were determined. Results: Rosiglitazone treatment significantly increased circulating adiponectin levels (P < 0.001) relative to the control group (P = 0.21). Plasma resistin levels were unchanged in both the rosiglitazone-treated and -untreated groups, but baPWV and hsCRP were significantly decreased (P < 0.001 and P = 0.003 respectively) in the rosiglitazone group only. Multiple linear regression analysis showed that changes in plasma adiponectin and baPWV were significantly affected by rosiglitazone treatment. Conclusions: These data suggest that rosiglitazone may have an anti-atherogenic effect in subjects with prediabetes or non-diabetic MetS.

The rapid global urbanization of the past century poses several challenges for planners and policy makers. In particular, the conflation of social and urban issues must be understood to create sustainable and livable urban places. In this regard, it was our aim to model and understand the relationship between urban characteristics and peoples’ perceived quality of urban life (QoUL) using statistical analysis and geospatial modeling. We selected objective variables representing urban characteristics based on literature and used principal components analysis to develop uncorrelated components. These components served as the independent variables in a multiple linear regression analysis. The subjective, dependent variables were extracted from a QoUL survey. Results indicated that only the Education/Income component is related to QoUL (R2 of 0.46). Using only single independent variables in a linear model explained 46% of the total variance—over 10% higher than any previously determined relationship between objective variables and subjective QoUL. Furthermore, we found that subjective high QoUL and subjective low QoUL were not strongly correlated, indicating that they are affected by different objective variables, respectively. This suggests that future efforts of increasing QoUL need to define their goals more precisely, as measures for increasing perceptions of high QoUL are likely different from measures for decreasing perceptions of low QoUL.

Background. Research has confirmed the involvement of oxidative stress (OxS) in allergic contact dermatitis whilst other inflammation-related biomarkers have been less studied.Objective. To evaluate systemic levels of selected inflammatory markers, OxS indices and adipokines as well as their associations in allergic contact dermatitis.Methods. In 40 patients, interleukin- (IL-) 6, monocyte chemoattractant protein (MCP-1), and IL-10 levels were measured in sera with the Evidence Investigator Cytokine & Growth factors High-Sensitivity Array, total peroxide concentration (TPX) and total antioxidant capacity (TAC) by means of spectrophotometry, and the plasma concentrations of adiponectin and leptin by the quantitative sandwich enzyme immunoassay technique.Results. TNF-αlevel (P< 0.01) and TPX (P< 0.0001) were increased whilst IL-10 (P< 0.05) and TAC (P< 0.0001) were decreased in the patients as compared to controls. Correlation and multiple linear regression analysis identified both, TPX and TAC (inversely), as possible independent markers for evaluating allergic contact dermatitis. Adiponectin level in patients was increased (P< 0.0001), but neither adiponectin nor leptin correlated significantly with the biomarkers of inflammation or OxS.Conclusion. OxS parameters, especially TPX and OSI, reflect the degree of systemic inflammation associated with allergic contact dermatitis in the best way. The relation between OxS and adiponectin level warrants further studies.

AbstractThere are limited data on the association between Fe overload and leucocyte telomere length (LTL), known as a useful biomarker of the replicative ageing of cells. The aim of the study was to evaluate associations between Fe-status biomarkers and LTL. A cross-sectional study included 1174 men and women aged 50–79 years who provided blood samples for assays of Fe-status biomarkers including ferritin, transferrin saturation (TSAT), total Fe-binding capacity (TIBC) and relative LTL. They were free of hepatitis, potential infection or Fe deficiency. In multiple linear regression analysis adjusted for potential confounding variables, log-transformed LTL was positively associated with TIBC (adjusted coefficient estimate for its highest quartile: 0·17 (se 0·03), P<0·001) and inversely associated with TSAT (adjusted coefficient estimate for its third and fourth quartiles: −0·09 (se 0·03), P<0·01). These associations were consistent after additional adjustment for serum concentrations of high-sensitivity C-reactive protein, alanine transaminase and aspartate transaminase. In particular, participants with not only abnormally high concentrations (>45 %) but also with high-normal concentrations (35–45 %) of TSAT had shorter LTL compared with those with low-normal concentrations (<30 %) (P<0·05). We also observed that less-active or obese persons with high TSAT concentrations had shorter LTL than others. Our findings that cellular ageing is influenced not only by Fe overload but also by high-normal concentrations of TSAT support the hypothesis regarding the detrimental effects of labile Fe, which has a potent pro-oxidant activity in the body.

AntecedentsIn previous works, referential thinking was predicted by clinical and dispositional variables such as social anxiety or vulnerability to depression (Rodríguez-Testal, Senín-Calderón & Fernández-Jiménez, submitted to revision).Objectives and hypothesesWe propose to find personality variables to characterize the emergence of referential thinking. We predict a greater referential thinking in subjects with a high sensitivity to punishment and higher scores on social anxiety.MethodsParticipants: 366 subjects selected from the general population, 66.6% women, mean age = 33.18 (SD = 12.79).MaterialsWe used the REF-scale (Lenzenweger et al., 1997) adapted to Spanish language, GHQ-28 (Goldberg, 1996), SPSRQ (Torrubia et al., 2001) and The Revised Self-Consciousness Scale (Scheier & Carver, 1985).It was applied a cross-sectional design and a correlation method. All the analysis were accepted at p < .05.ResultsThe multiple linear regression analysis showed the importance of the clinical variable of depression, public self-consciousness, and sensitivity to reward and punishment as predictors of referential thinking (34% of the variance explained). The discriminant analysis according to scores in referential thinking was significant (Lambda = .87, p = .001). The combination of the above variables correctly classified 85.1% of cases.ConclusionsSubjects more concerned about how they are perceived by others tend to a greater presence of self-references, although they don’t show a high score in social anxiety. Susceptibility to reward and high vulnerable to punishment are the personality variables that best predicted referential thinking.

Muscle lipid oxidation is stimulated by peroxisome proliferator-activated receptor (PPAR) δ or adiponectin receptor signalling. We studied human myocyte expression of the PPARδ and adiponectin receptor genes and their relationship to lipid parameters of the donors. The mRNA levels of the three adiponectin receptors, AdipoR1, AdipoR2, and T-cadherin, were highly interrelated (r≥0.91). However, they were not associated with GPBAR1, an unrelated membrane receptor. In addition, the adiponectin receptors were positively associated with PPARδ expression (r≥0.75). However, they were not associated with PPARα. Using stepwise multiple linear regression analysis, PPARδ was a significant determinant of T-cadherin (P=.0002). However, pharmacological PPARδ activation did not increase T-cadherin expression. The myocyte expression levels of AdipoR1 and T-cadherin were inversely associated with the donors' fasting plasma triglycerides (P<.03). In conclusion, myocyte expression of PPARδ and the adiponectin receptors are highly coordinated, and this might be of relevance for human lipid metabolism in vivo.

We investigated the beneficial effects and underlying mechanisms of Zhuanggu Guanjie (ZGGJ) pill in osteoporosis in vitro and in vivo. Bone marrow macrophages from 4–6-week-old mice were cultured in the presence of macrophage colony-stimulating factor (15 ng/mL) and receptor activator of nuclear factor-κB ligand (30 ng/mL). Osteoclast differentiation was determined by quantification of tartrate-resistant acid phosphatase activity. Gelatin zymography was used to detect the activity of matrix metalloproteinases in osteoclasts. Ovariectomized rats were administered orally with estradiol valerate or ZGGJ for 8 weeks. Blood was collected to measure serum indices. Tibiae were harvested to carry out bone microcomputed tomography scanning, histomorphological analysis, and bone strength determination. ZGGJ inhibited tartrate-resistant acid phosphatase activity, matrix metalloproteinase 9 expression, and bone resorption in vitro. At doses of 0.55, 1.1, and 2.2 g/kg, ZGGJ exerted significant osteoprotective effects including inhibition of bone turnover markers and improved tibia bone strength in ovariectomized rats. Microcomputed tomographic analysis showed that ZGGJ improved the trabecular architecture with increased connectivity density and trabecular thickness and decreased trabecular spacing. These results revealed that ZGGJ prevents bone loss induced by ovariectomy in rats and that inhibition of bone resorption is involved in the bone-protective effects of ZGGJ.
