Tropical black bolete, Phlebopus portentosus, provides various nutritional benefits and natural antioxidants to humans. In this study, the chemical composition, phenolic compounds, and antioxidant and antimicrobial activities of fresh mushroom samples and samples stored for a period of one year using different preservation methods (drying, brining, and frozen) were investigated. The results indicated that the brining method significantly reduced the protein and fat contents of the mushrooms. The polyphenol and flavonoid contents of the frozen sample were not significantly different from that of the fresh sample. The results revealed that an inhibition value of 50% (IC50) for the 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay of the extract of the dried and frozen samples was not statistically different from that of the fresh sample. The IC50 value of 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid (ABTS) assay and ferric reducing antioxidant power (FRAP) value in the extract of the frozen sample were not found to be significantly different from those of the fresh sample. Furthermore, the lowest degree of antioxidant activity was found in the extract of the brined sample. Additionally, the antimicrobial activities of the extracts of the fresh and frozen samples were not significantly different and both extracts could have inhibited the growth of all tested Gram-positive bacteria and Pseudomonas aeruginosa.

The aim of this study was to determine the chemical variability of wormwood extracts as affected by the growing region. Antioxidant and antimicrobial activities were also investigated. The essential oil composition variability ofA. absinthiumL. aerial parts collected from four different Tunisian regions was assessed by gas chromatography (GC/FID) and by gas chromatography mass spectrometry (GC/MS). In addition, total polyphenols, flavonoids, and condensed tannins as well as antioxidant, antibacterial, and antifungal activities of methanolic extract and essential oils were undertaken. Chromatographic analysis of wormwood essential oils showed the predominance of monoterpene hydrocarbons represented mainly by chamazulene. RP-HPLC analysis of wormwood methanolic extract revealed the predominance of phenolic acids. Antiradical activity was region-dependant and the methanolic extract of Bou Salem region has the strongest activity (CI50=9.38±0.82 µg/mL). Concerning the reducing power, the methanolic extract of Bou Salem, Jérissa, and Boukornine regions was more active than the positive control. Obtained results of antimicrobial activities showed that wormwood essential oil is endowed with important antibacterial activity which was strongly related to the organoleptic quality of oil which appeared strongly region-dependant.A. absinthiumL. EOs investigated are quite interesting from a pharmaceutical standpoint because of their biological activities.

Objective: To screen phytochemical compounds and study antioxidant and antimicrobial activities of Pluchea indica (L.) Less (PI) leaves extracts for assessment the moisturizing lotion bar formulation containing it’s as a bioactive ingredient. Methods: PI leaves were extracted in different solvents. Crude extracts were screened phytochemical compounds and evaluated antioxidant and antimicrobial activities before formulating to antibacterial moisturizing lotion bar. Results: 95% ethanolic extract displayed high antioxidant activities. DPPH radical scavenging capacity with IC50 was 6.80±0.04 mg/ml (61.37±0.41%) and the ferric reducing ability power was 381.20±0.10 mg AAE/100 gDW. Additionally, 95% ethanolic extract served as a broad spectrum bacteriostatic agents against gram-positive and gram-negative species with an inhibition zone of 6.50±0.00-46.67±0.58 mm. It also was simultaneously as a broad spectrum bactericidal agents (MIC and MBC value of 25-200 mg/ml) by broth micro-dilution method. The screened phytochemical constituents (saponins, tannins, flavonoids, terpenoids, steroids, and alkaloids) in PI leaves extract may be responsible for high biological activities. Thus, 95% ethanolic extract was the most appropriate bioactive ingredient for formulated the product development. Conclusion: The developed moisturizing lotion bar with 0.0041% of 95% ethanolic extract was the most eligible formula that showed good appearance, odor, color, homogeneity, after feel and stability. This extract-containing lotion bar formulation achieved a high level of customer satisfaction by sensory evaluation and development of this specific formulation should be subject to further clinical investigation.

Chitosan, a natural polysaccharide, has been previously proposed as an elicitor in plants to prevent pathogen infections. The present study aimed to analyze the effect of chitosan solution and chitosan nanoparticles treatment applied on the grapevine variety Sousão with respect to the phenolic composition, antioxidant potential and antibacterial activity of its individual grape components. Grapevine plants of selected lines were sprayed with chitosan solution and chitosan nanoparticles, and ethanolic extracts of stems, seeds and skins were prepared from grapevines treated and not treated with chitosan. Total phenolic, anthocyanin and tannin contents were studied, and the identification of the individual phenolic compounds was performed by HPLC-DAD. The antimicrobial susceptibility method was performed using the Kirby-Bauer disc diffusion method against multidrug-resistant bacteria. Overall, there was small increase in the concentration of phenolic compounds, antioxidant and antimicrobial activities in grape components treated with chitosan solution. Seed extracts showed the highest antioxidant and antimicrobial activities. The studied individual components obtained from chitosan-treated grapevines could represent an added value due to the increased antioxidant and antibacterial potentials. The phenolic compounds found in components may be used in food and pharmaceutical industries as natural food preservers and antibiotic adjuvants.

In recent years, indole derivatives have acquired conspicuous significance due to their wide spectrum of biological activities—antibacterial, antiviral, and anticonvulsant. This compound is derived from naturally grown plants. Therefore, synthesis of a novel “3-(Naphthalen-1-ylimino)indolin-2-one” compound (2) and its analysis using UPLC systems along with antimicrobial assessment was the aim of the current study. Isatin was used as a parent drug for synthesizing compound (2). Liquid Chromatographic analysis was performed using a C18 BEH column (1.7 μm 2.1 × 50 mm) by UPLC systems. Degradation studies were carried out to see whether acid, base, thermal, and oxidizing agents had any impact on the synthesized molecule in stress conditions (100 °C). A lipid-based self-nanoemulsifying formulation was developed and selectivity, specificity, recovery, accuracy, and precision were measured as part of the UPLC system’s validation process. Antimicrobial studies were conducted using gram-positive and gram-negative bacteria. The standard samples were run with a concentration range of 5.0–100.0 μg/mL using the isocratic mobile phase comprising of methanol/water (70/30 %v/v) at 234 nm; good linearity (R2 = 0.9998) was found. The lower limits of detection (LOD) and quantitation (LOQ) of the method were found to be 0.81 μg/mL and 2.5 μg/mL, respectively. The coefficients of variation were found to be less than 2%. The antimicrobial study suggests that compound (2) has a substantial growth effect against gram-negative bacteria. It was successfully synthesized and applied to measure the concentrations in lipid-based dosage form, along with potent antimicrobial activities.

Lectins are a diverse group of proteins of non-immune origin that interact specifically with glycans. Owing to their specificity, they can mediate various cellular and molecular recognition processes. To explore information on biological activities of lectins from Penicillium duclauxii, P. proteolyticum and P. griseoroseum, they were investigated for their antioxidant and antimicrobial activities. Penicillium sp. lectins exhibited moderate antioxidant activity. P. duclauxii, P. proteolyticum and P. griseoroseum lectins inhibited DPPH with an IC50 value of 71.42, 75.04 and 82.11 ?g/mL, respectively. P. duclauxii, P. proteolyticum and P. griseoroseum lectins inhibited the hydrogen peroxide radical with IC50 values of 198.57, 209.76 and 215.31 ?g/mL, respectively. P. duclauxii and P. proteolyticum lectins exhibited potent antibacterial activity against Gram-negative bacteria. P. griseoroseum lectin inhibited only Gram-positive bacteria. Penicillium sp. lectins did not exhibit antifungal activity. The biological potential of Penicillium sp. lectins will help to understand their biomedical applications. This is the first report on the antioxidant and antimicrobial activities of purified lectins from Penicillium sp.
