Abstract:The present study was designed to investigate the radio protective efficacy of ethanolic extract of leaves of the Averrhoa carambola by studying the following parameters: radioprotective effect before and after radiation, estimating the Total Antioxidant Status, haematological parameters, assessing the histopathology of liver and kidney. Extract was given orally at a dose of 450 mg/kg, orally for 15 days prior to and 15 days after radiation (2 Gy). The rats were kept under observation for 15 days for any sign of radiation sickness, morbidity, behavioural toxicity, urination and defecation pattern or mortality, which showed no changes. The haematological parameters showed a significant decrease in the irradiated rat when compared with the control group. Antioxidant activity revealed from the data obtained in the present study showed that A.carambola leaves ethanolic extract possess good antioxidant activity. Histopathological changes in the irradiated liver and kidney sections showed improved cellular architecture when compared to the normal. The present study suggests that supplementation with Averrhoa carambola has potent antioxidant activity and act as probable radioprotector against gamma radiation induced oxidative damage.

Introduction. The overwhelming number of publications contains only data on the content of individual antioxidants, but not on the overall antioxidant activity of the blood in patients with vibration disease.The aim of the study was to determine the total antioxidant activity of blood serum in patients with vibration disease.Materials and methods. Th e main group consisted of 30 people diagnosed with “Vibration disease” of 1 degree (n=21) and 2 degrees (n=9). Th e control group consisted of 30 clinically healthy men, comparable in age with the main group (p=0.66). Th e total activity of antioxidant systems of blood plasma was evaluated photometrically using the test system “Total antioxidant status-Novo” (“Vector-best”, Russia).Results. The indicator of the total antioxidant status (TAS) was 1,038±0.232 mmol/l in the examined main group, against 1,456±0.225 mmol/l in the examined control group (p<0.000001). Th e coefficient of variation (CV) in patients with vibration disease was 22.35%, 1.45 times higher than in the control group (15.45%). In the main group there was a positive correlation between age and TAS (R=0.525), in the control group there was no such relationship (R=0.095). Th e degree of decrease depended on the severity of vibration disease.Conclusions. 1. The development of vibration disease is accompanied by a decrease in the antioxidant status of blood serum. 2. Th e degree of decrease in the antioxidant status of blood serum correlates with the severity of vibration disease. 3. Reduction of TAS can serve as a pathogenetic justification of the need to include drugs and/or biologically active additives with antioxidant activity in therapy

Reducing the use of antibiotics in livestock in order to contain antibiotic resistance and studying natural substance additives are key to sustainability. Among the various biological activities of plant extracts, antioxidant activity plays an important role. The present study assesses the total antioxidant activity and antioxidant reserves using the Kit Radicaux Libres test (KRL™ Kirial International, Couternon, France). One hundred and sixty piglets (Topics × Tempo) weaned at 28 days of age were divided into four dietary treatment groups that were fed a commercial diet (the control group, C); 500 mg/kg Boswellia extract (BOS); 200 and 50 mg/kg Uncaria and Tanacetum extracts (UT) respectively; and 225 mg/kg of an antioxidant plant extract mixture (AOX). The blood antioxidant activity of the piglets was measured using the KRL test and the reserves were analyzed on whole blood samples after hydrolysis with glucosidase, sulfatase and glucuronidase. No significant differences were observed in growth performance. The delta KRL values of the whole blood showed a significantly higher total antioxidant status of the piglets from the BOS and AOX groups than the UT and C groups (+30.7 BOS; +27.7 AOX vs. +17.81 UT +13.30 C; p = 0.002) between 18 and 28 days post-weaning. The delta KRL values of red blood cells (RBCs) showed a significantly higher total antioxidant status of the piglets from the AOX groups than the UT and BOS groups (+22.2 AOX; vs. +9.90 UT +9.4 BOS; p = 0.016) between the two sampling times. Reserves of UT and AOX were higher than C and BOS for all enzymes, glucosides, sulphates, and glucuronides. The biological KRL test proved to be an extremely sensitive tool to evaluate the piglets’ antioxidant status. Determining the antioxidant reserve also provides a better understanding of the real antioxidant status of pigs.

Background: Protium heptaphyllum (Aubl.) March resin is accepted as an analgesic, healing, expectorant, anti-inflammatory, antinociceptive and gastroprotective. However, there are no specific studies with leaves of this plant. Objective: This study identified the chemical composition and biological activity of crude ethanolic extract (EE) and ethyl acetate (EA) fraction from P. heptaphyllum. Methods: Mice were intoxicated with acetaminophen (PCM; 250 mg/kg), subsequently treated with EE and EA (100 mg/kg) for 7 days via gavage. Plasma analyzes and liver and kidney homogenates of the animals were performed. Results: The extract and fraction showed the presence of phenols and flavonoids. Three main flavonoids were identified by HPLC-UV, LC-MS/MS fractionation, quercetin-3-β-D-glycoside, myricetin and quercetin. For total antioxidant potential tests, EE presented EC50 of 75 μg/mL and 100 μg/mL for EA. ALT, AST and ALP enzymes activity increased in the PCM-exposed plasma, but EA decreased these activities at control levels for ALT and AST. Animal glycemic levels decreased with EE and EA, which may be due to a possible hypoglycemic effect of the plant. In liver tissue, SOD was unchanged, CAT, GSH and ASA decreased in the PCM group, but EA was able to reverse these effects. TBARS and PC were reduced by both treatments. In renal tissue, EA fraction recovered GST activity, GSH levels were restored by EE and EA and PC was reduced by treatments. Conclusion: This antioxidant effect may be due to the presence of the flavonoids identified which are already well known for their antioxidant properties.

Some evidence suggests the neuroprotection of estrogen provided by the antioxidant activity of this compound. The main objective of this study was to determine the level of estradiol and its correlation with the activity of antioxidant enzymes, total antioxidant status and ferritin from ischemic stroke subjects. The study population consisted of 30 patients with acute ischemic stroke and 30 controls. There was no significant difference between estradiol in stroke and control group. The activity of superoxide dismutase and level of ferritin was higher in stroke compared with control group (P<.05,P<.001, resp.). There was no significant correlation between estradiol and glutathione peroxidase, glutathione reductase, catalase, total antioxidant status, and ferritin in stroke and control groups. We observed inverse correlation between estradiol with superoxide dismutase in males of stroke patients (r=−0.54,P=.029). Our results supported that endogenous estradiol of elderly men and women of stroke or control group has no antioxidant activity.

The aim of this study was to investigate the biological and chemical activity on two species of fungi of the genus Penicillium isolated from wastewater. On the selected species of fungi the different antioxidant activity assays were carried out: DPPH free-radical scavenging activity, total antioxidant activity, Fe2+- chelating ability and Fe3+- reducing power. Total phenol content was also determinate for ethanolic extract of mycelia. Penicillium chrysogenum ethanolic extract contained higher total phenolic content and better total antioxidant capacity as well as ferrous ion chelating ability. Penicillium fumiculosum ethanolic extract showed higher DPPH free-radical scavenging activity, as well as reducing power. Based on the obtained results it can be concluded that two types of fungi are potential new sources of natural antioxidants.

Objective: To compare the therapeutic effects of acupotomy trinity lysis and traditional acupotomy on cervical spondylotic myelopathy. Methods: A total of 205 patients with cervical spondylotic myelopathy of liver and kidney deficiency syndrome were randomly divided into the experimental group (105 cases) and the control group (100 cases). The experimental group was relaxed with acupotomy in three positions: Heaven (tian), Human (ren) and Earth (di). Traditional acupotomy was used to relax Ashi acupoints of the affected vertebra in the control group. One treatment was conducted in one week, and the duration of one course of treatment was three weeks. The VAS, JOA score and NDI index were observed after treatment.  Results: Before and after treatment, the total treatment efficiency of the treatment group was 95.23%, and that of the control group was 80.00%, there was significant difference between the two groups, P<0.05; Before operation, there was no significant difference in JOA score, NDI index score, and VAS score between the treatment group and the control group (P>0.05); there was no significant difference after 1 week (P>0.05), but there were significant differences between the two groups 2 weeks and 3 weeks after operation (P<0.05). Conclusion: Acupotomy trinity lysis is a safe, effective and economical treatment for cervical spondylotic myelopathy.

Heavy metal toxicity contributes to liver and kidney dysfunction and damage through oxidative stress mechanisms; however, from previous studies, extracts from the Vernonia amygdalina plant have shown to possess potent antioxidant properties. This study was aimed at uncovering the potential ameliorative effects of ethanolic extract from Vernonia amygdalina plant in heavy metal toxicity-induced liver and kidney dysfunction. For this study, 44 Sprague Dawley rats were divided into three groups. The control group received a basal diet and water only while the treatment groups received varied dosages of the heavy metals. The copper (Cu) and lead (Pb) groups had five subgroups. The Cu only and Cu recovery subgroups were administered with 16 mg/kg Cu intraperitoneally daily for 14 days, whereas the Pb only and Pb recovery subgroups were administered with 13 mg/kg Pb intraperitoneally daily for 14 days. Subsequently, the Pb only and Cu only subgroups were sacrificed. The three Pb and Cu treatment subgroups received oral graded doses (100 mg/kg, 200 mg/kg, and 300 mg/kg) of the extract for 21 days. The Cu recovery and Pb recovery subgroups were left to recover for 21 days. After histological examinations, the Pb and Cu pretreatment groups showed evidence of focal necrosis accompanied by inflammatory cell infiltrations. The serum levels of liver biomarkers AST, ALT, and GGT, as well as urea and creatinine, were significantly elevated (P=0.01) following copper and lead exposure. Upon posttreatment of the rats with the extract, the physiological levels of the biomarkers were restored and tissue architecture of the organs improved. Thus, the ethanolic extract of Vernonia amygdalina is capable of ameliorating the effects of heavy metal toxicity through potent antioxidative mechanisms.

Objectives: The objectives of this study were to determine the antioxidant activity and cardioprotective activity of bangun-bangun leaves ethanolic extract.Methods: Bangun-bangun leaves ethanolic extract was obtained by maceration process. The antioxidant activity test was performed by 2,2’-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) free radical scavenging method with various concentrations of extract. The absorbance was measured by visible spectrophotometric method and calculated the inhibitory concentration (IC50) value for antioxidant activity analysis. Cardioprotective activity test was performed by measuring the cardiac troponin T (cTnT) level, creatine kinase-muscle/brain (CK-MB) level, and histology of the heart tissue. Animals induced with doxorubicin at the 8th day and the 9th day, bangun-bangun leaves ethanolic extract was administered from the 1st day to the 9th day with various doses of extract.Results: Bangun-bangun leaves ethanolic extract had IC50 value of 57.79 μg/mL. Difference dose of bangun-bangun leaves ethanolic extract shows difference cardioprotective activity. Bangun-bangun leaves ethanolic extract at dose 300 mg/kg bw did not differ significantly to the positive control group and normal group. The higher the dose of an extract the greater the decrease in cTnT and CK-MB levels and increase protection against heart damage.Conclusion: Bangun-bangun leaves ethanolic extract had strong antioxidant and had cardioprotective activity.

SummaryBackground:Oxidative stress may be involved in the pathogenesis of every human disease. To understand its possible role in benign prostatic hyperplasia (BPH), we measured the overall oxidative status of patients with BPH and the serum activity of the high density lipoprotein (HDL)-related antioxidant enzymes paraoxonase 1 (PON1) and arylesterase (ARE).Methods:Fifty-six urology outpatient clinic patients with BPH (mean age 64±8.6 years) were prospectively included in the study. Forty volunteer healthy controls from the laboratory staff (mean age 62±10 years) were enrolled for comparison. Serum total antioxidant status (TAS), total oxidant status (TOS), PON1, ARE, and HDL levels were measured by commercially available, ready-to-use kits.Results:Serum TAS and HDL levels were significantly lower in the BPH group than in the control group (P=0.004 and P=0.02, respectively). No significant between-group differences were observed for TOS levels or PON1 and ARE enzyme activities (P=0.30, P=0.89, and P=0.74, respectively). In the BPH group, the calculated parameters PON1/HDL and ARE/HDL were significantly higher (P=0.02 and P=0.04, respectively).Conclusions:Our findings agree with the previous reports of impaired oxidant/antioxidant balance in BPH patients. The activities of HDL-related enzymes between groups with significantly different HDL levels may be deceptive; adjusted values may help to reach more accurate conclusions.

Introduction. The sources of Reactive Oxidative Species (ROS) in the organism are the respiratory processes occurring in cells catalyzed by different enzymes. Operation of ROS is balanced by antioxidants, the compounds; although present in low concentrations, they significantly inhibit the degree of oxidation of particular molecules. The Aim of the Study. The aim of this study was to assess the changes in the integrated antioxidant system under the influence of radon therapy in osteoarthritis patients. Material and Methods. Observation included 35 patients suffering from degenerative joints and disc disease (mean age 56.5 years) undergoing radon water therapy and control group that consisted of 15 osteoarthritis patients (mean age 54.2) without contact with radon water. Before therapy and after 18 days of treatment, serum total antioxidant status (TAS) was assessed with the use of standard colorimetric assay. Results. In the study group, we observed trends to increase TAS concentration, whereas, in the control group, TAS concentration was decreasing. Conclusions. (1) Radon waters treatment influenced the level of TAS of osteoarthritis patients treated with the radon water. (2) The change in TAS concentrations in the study group may be the result of low doses of ionizing radiation, but further studies on larger patient’s groups are demanded.
