Role of sugars and proteins in development of desiccation tolerance in fresh and shade-dried onion seeds

In a 2-year field study, the umbels of open-pollinated onion cv. ‘Pusa Red’ were tagged on the day of anthesis and were sampled at different intervals until harvest maturity. Each umbel was cut in half and bulked with others in the respective replication. Seeds were immediately taken from half of the umbels for analysis of seed water content, leachate conductivity, seed capsule chlorophyll, seed dry weight and seed germination. They were also subjected to rapid desiccation to identify the stage at which desiccation tolerance occurred. The remaining umbel halves were shade dried and subjected to the same quality analyses except chlorophyll content. With seed maturation, there were rapid declines in chlorophyll content, leachate conductivity and seed water content, whereas seed dry weight and germination increased. In fresh seeds, maximum germination occurred around 50 days after anthesis, when physiological maturity and maximum seed weight were attained. In shade-dried seed, maximum germination occurred at 42 days after anthesis. This coincided with drastic reductions in seed capsule chlorophyll, leachate conductivity and seed water content in fresh seeds. At this stage, reducing sugar concentration declined and non-reducing sugar concentration increased many fold in shade-dried seeds. SDS-PAGE analysis of the heat-stable proteins extracted from the seeds also showed prominent bands in the lower molecular weight region at 42 DAA. Therefore, the shade-dried seeds attained membrane stability at 42 DAA. In cases of adverse weather conditions or disease attack, seed umbels can be harvested as early as 42 DAA and shade dried without compromising seed germination. Desiccation tolerance in fresh onion seeds occurred around 46 DAA and was a gradual event.

The Tabernas badlands in semiarid south-east Spain is one of the driest regions in Europe with a mean annual precipitation of c. 240 mm. The landscape is deeply dissected, with canyons, ramblas and sparsely vegetated eroded badland slopes. The vegetation is predominantly a biological soil crust consisting of different types of lichen-rich communities, one of the more conspicuous being dominated by Diploschistes diacapsis (Ach.) Lumbsch. This lichen is mainly restricted to the north- facing slopes, where it forms extensive whitish carpets and probably plays an important role in preventing erosion of the slopes and allowing plant colonization. South-facing slopes are much more eroded and generally lack vegetation. %The photosynthetic performance of north (shade) and south-facing (sun) populations of D. diacapsis was studied to determine if these different populations showed any adaptations to the microclimatic conditions of their individual habitats. The response of CO2 exchange to light intensity, temperature and water content was measured under controlled conditions in the laboratory. Dry weight-based net photosynthetic rates were higher in the southern-exposed population but quantum efficiency, and light compensation points were similar. Thallus weight per unit area (LMA) was considerably higher for shade specimens but maximum water content and optimal water content were very similar and chlorophyll content on a dry weight basis was also similar. Chlorophyll content on an area basis was higher in the northern-exposed population and always much larger than those reported in other studies on the same species (up to 8 times larger) with the result that NP values on a chlorophyll basis were relatively low. The larger LMA meant that shade thalli stored more water per unit area which should ensure longer active periods than sun thalli. The results support a strategy pair of high NP and short active time versus low NP and long active time, both having been reported for other soil crust species. However, the visibly larger biomass of the shade D. diacapsis suggests that the lichen is at the limit of its adaptability in these habitats.

The effectiveness of alkaline hydrogen peroxide as a suitable choice of pretreatment for the conversion of millet husk to reducing sugars using cellulase enzyme for hydrolysis and subsequent ethanol production was determined. The effects of three variables on reducing sugar production from millet husk were determined using one factor at a time (OFAT) method namely; peroxide concentration, pretreatment time and pretreatment temperature. From the results, it was observed that a significant (P<0.05) amount of reducing sugars were lost during pretreatment of millet husk. The untreated group which was only physically pretreated (milled) however yielded a significantly higher (P<0.05) reducing sugar concentration of 10.67mg/ml after enzymatic hydrolysis while the highest reducing sugar concentration of 4.82mg/ml was obtained using 0.375%v/v peroxide concentration for 60minutes at 250C. Therefore, pretreatment of biomass with alkaline hydrogen peroxide may be more suitable for feedstock with high lignin contents than millet husk.

Variations in the apparent desiccation tolerance of the moss Tortula ruralis were traced to several causes. When the moss was kept continuously hydrated for several days, desiccation tolerance tended to decrease ("dehardening"). Conversely, daily episodes of drying and rehydration induced hardening so that the moss could tolerate rapid drying to a tissue water content of 0.008 g∙g dry weight−1.Different clumps of moss showed very different degrees of desiccation tolerance. The least tolerant samples suffered severe damage after rapid drying, whereas the most tolerant suffered little damage, and did not deharden during 3 days continuous hydration.

Gentiana lutea L. (yellow gentian, Gentianaceae) is an important medicinal plant under protection as endangered species in most European countries. The aim of this work was to evaluate variation in seed mass, seed water content, and seed germination among 56 wild accessions of G. lutea. The effect of gibberellic acid (GA3), putrescine, moist chilling, and level of ripeness of seeds on subsequent germination was also investigated. Seeds of G. lutea showed physiological dormancy (final germination percentages ranged from 0% to 11%, depending on the accession) and GA3 enhanced seed germination drastically in all the accessions. The highest germination (99%) of GA3-treated seeds was reached at 15 °C. Final germination percentage and germination rate (as expressed by mean germination time), as well as seed mass and seed water content, varied significantly among accessions. In general, 1 year moist chilling did not significantly enhance G. lutea seed germination. For most accessions, no significant differences were found between fully ripe seeds and less ripe seeds for seed water content, seed mass, and seed germination. Applications of GA3 were always most effective than those of putrescine for increasing seed germination.

ABSTRAK<br />Salah satu faktor yang berpengaruh terhadap viabilitas benih<br />sambiloto (Andrographis paniculata Nees) adalah waktu panen.<br />Berdasarkan permasalahan tersebut kegiatan ini dilakukan dengan tujuan<br />untuk mengetahui pengaruh umur panen terhadap viabilitas benih serta<br />hubungannya dengan produksi terna sambiloto. Percobaan dilakukan di<br />KP. Cimanggu dan Laboratorium, Balai Penelitian Tanaman Obat dan<br />Aromatik, dari Maret 2005 – Maret 2006. Percobaan disusun dalam<br />rancangan acak kelompok (RAK) dengan 10 perlakuan stadia umur<br />panen dan 4 ulangan. Stadia umur panen yang diuji yaitu 18, 21, 22, 23,<br />24, 25, 26, 27, 28, dan 29 hari setelah antesis (HSA). Variabel yang<br />diamati yaitu mutu benih (daya berkecambah benih, kecepatan<br />berkecambah), pertumbuhan tanaman (tinggi tanaman, jumlah cabang),<br />serta produksi terna (bobot basah tanaman, bobot kering daun, dan bobot<br />kering batang). Hasil penelitian menunjukkan bahwa: (1) Umur panen<br />benih berpengaruh terhadap daya berkecambah benih dan kecepatan<br />berkecambah benih sambiloto; daya berkecambah dan kecepatan<br />berkecambah tertinggi didapatkan pada umur panen benih 22 dan 21 HSA<br />(67,00 dan 55,00)%; sedangkan daya berkecambah yang terendah<br />diperoleh pada umur panen 18 HSA (23,50)%, (2) Umur panen benih<br />berpengaruh terhadap tinggi tanaman dan jumlah cabang pada tanaman<br />umur 1 bulan. Tinggi tanaman dan jumlah cabang tertinggi berturut-turut<br />didapatkan pada perlakuan umur panen benih 27 dan 26 HSA (39, 63<br />dan 36,58 cm serta 16,71 dan 16,61 buah); dan (3) Umur panen benih<br />berpengaruh terhadap produksi terna (bobot basah tanaman, bobot kering<br />daun, bobot kering batang) pada umur 3 bulan. Bobot basah tanaman,<br />bobot kering daun, serta bobot kering batang tertinggi didapatkan pada<br />perlakuan umur panen benih 27 HSA (291,25, 28, 27 dan 28,86)g. Bobot<br />basah tanaman, bobot kering daun, serta bobot kering batang terendah<br />didapatkan pada perlakuan 18 HSA (217,09, 22,10 dan 20,24)g. Umur<br />panen benih tidak berpengaruh terhadap jumlah cabang pada umur 3<br />bulan.<br />Kata kunci : Sambiloto, Andrographis paniculata Nees, umur panen,<br />viabilitas, produksi, Jawa Barat<br />ABSTRACT<br />The influence of harvesting time on the seed viability and<br />the  relationship  with  herb  yield  of  king  bitter<br />(Andrographis paniculata Nees)<br />One of the main factors influencing the viability of king bitter<br />(Andrographis paniculata Nees) is appropriate harvesting time. Based on<br />this problem a research was conducted to study the relationship between<br />seed maturity and seed viability, and herb yield of king bitter. The research<br />was conducted at Cimanggu Experimental Station and in the laboratory of<br />Indonesian Medicinal and Aromatic Crops Research Institute (IMACRI)<br />from March 2005 to March 2006. The experiment was arranged in a<br />randomized block design with 10 seed maturity stages and 4 replications.<br />Maturity seed tested was 18, 21, 22, 23, 24, 25, 26, 27, 28 and 29 days<br />after anthesis. The observations were made on seed quality (percentage of<br />seed germination, and rate of seed germination), plant growth (plant<br />height, number of branches) and herb yield (fresh herb weight, leaf dry<br />weight, and stem dry weight). The results of experiment indicated that (1)<br />seed maturity affected seed germination, and rate of seed germination of<br />king bitter; the highest seed germination and germination rate were<br />achieved by seeds harvested at 21 and 22 days after anthesis, (2) stage of<br />seed harvesting affected on the plant height and number of branches at 1<br />month after planting. The highest plant height and number of branches<br />were found on the treatments of seeds harvested at 26 and 27 days after<br />anthesis, they were 39.63 and 36.58 cm and 16.71 and 16.61; (3) seed<br />maturity also affected herb production, such as wet weight of plants, dry<br />weight of leaf and dry weight of steam at 3 moths after planting. Wet<br />weight of plant, dry weight of leaf and dry weight of stem were achieved<br />at the treatments of seeds harvested at 27 days after anthesis, they were<br />291.25, 28.27 and 28.86 g. The lowest of wet of plant weight, dry weight<br />of leaf and dry weight of stem were found on the seeds harvested at 18<br />days after anthesis. They were 217.09, 22.10 and 20.24 g. This study aims to determine optimum time of action of the enzyme α-amylase and glucoamylase needed in hydrolyze of starch from waste processed cassava (Manihot esculenta Crantz var. Lahumbu). This research was conducted through three main stages, namely the gelatinization, liquefaction and saccharification. The method was used method are liquefaction and  saccharification. The variation time of the stage liquefaction: 12; 24; 36; 48; 60; and 72 minutes and the saccharification stage are: 9; 18; 27; 36; 45; 54; and 63 hours. The results showed that the optimum time required for stage liquefaction using α-amylase enzyme is 48 minutes on the condition of a temperature of 80 oC with a value of 0.09% amylose levels were measured using UV-Vis spectrophotometer. The optimum time required for saccharification step using a glucoamylase which is 54 hours on the conditions of a temperature of 50oC with the amount of reducing sugar concentration of 9.186 g/L as measured using a UV-Vis spectrophotometer.

The purpose of this research is to know whether the aqueous extract of purple nutsedge(Cyperus rotundus) have an effect on the growth and chlorophyll content of upland riceseedling of Inpago 8 varieties. This research was conducted at Botanical Laboratory ofBiology Department Faculty of Mathematics and the Natural Sciences University ofLampung from September to October 2017. The experiments were carried out in a completerandomized design with the main factors being aqueous extract of purple nutsedge with 5 level : 0% v / v (control), 5% v / v, 10% v / v, 15% v / v, and 20%v/v. Parameters measuredwere shoot length, fresh weight, dry weight, relative water content, and total chlorophyllcontent of rice seedling. Homogeneity of variance was determined by the Levene test at 5%significant level. Analysis of variance and Tukey HSD test is done at 5% significant level.The results showed that the concentration of extract correlated quadratic with shoot lengthwhere the maximum shoot length was 3.9 cm at 10% extract concentration. Fresh weight andrelative water content maximum were 73.21 mg and 76% respectively at concentrations of14% and 10%. The minimum total chlorophyll content was 0.47 mg / g tissue at aconcentration of 11%. There was no significant effect on the dry weight of the seedling.From the results of the study, it was concluded that the extract of purple nutsedge at aconcentration below 14% was a growth stimulator, but it was an inhibitor of chlorophyllbiosynthesis.Keyword: Chlorophyll, Growth, Upland rice variety Inpago 8, Purple nutsedge

Organic wastes from Swiettenia marcophylla L, Artocarpus heterophyllus L, Mangifera indica L, and Annona muricata L were prepared by grinding into 0.1875, 0.3750, 0.7500 mm of particle size and delignified by 2% NaOH at 80°C for 90 minutes. Acid dilution hydrolysis process with H2SO4 1% was performed at 150°C for 120 minutes in a closed reactor. The effect of particle size and delignification on and reducing sugar concentration were investigated. The result showed (1) leaves that can be used as raw material to produce hydrogen should have 38–49% cellulose and hemicellulose. (2) Reducing sugar concentration increased with particle size reduction and delignification. (3) the best result with the highest reducing sugar concentration was achieved by 0.1875 mm particle size with delignification on Annona muricata L.

Several management practices were imposed on four Jerusalem artichoke (Helianthus tuberosus L.) accessions to determine changes in tuber yield, sugar content and the percentage of fructose and glucose in the hydrolyzed soluble sugar extracts. The fructose concentration in the reducing sugars declined from 82.3 to 74.2% from 6 September to 24 October. Storage of tubers at 2 C for up to 11 wk reduced the mean level of fructose from 78.2 to 68.0% and had a variable effect on reducing sugar concentration. Supplemental irrigation lowered the reducing sugar concentration significantly, while fertilization had no affect on any quality parameters. The Morden accession, M6, produced the best combination of high tuber yield (37.4 t/ha), reducing sugar concentration (16.3%) and fructose concentration in the reducing sugars (77.4%). Although a long period of growth was desirable to produce maximum yields of both tubers and total sugar, it was accompanied by a reduction in the ratio of fructose to glucose. This crop does not appear to require a high level of management to produce a good quality carbohydrate product.
