1. Ninety-one families containing 140 children under 4 years of age at enrolment were studied. At 1-monthly intervals, the children were weighed and measured and qualitative information about feeding habits was obtained. Weighed dietary surveys for periods of 5 d were made at approximately 8 months, 20 months, 3 years and 5 years of age; there were forty-six failures in 260 surveys attempted.2. At the four specified ages, mean energy intakes were 3.75, 5.03, 5.82 and 6.75 MJ (896, 1203, 1392 and 1613 kcal)/d respectively. At 8 months, milk products provided on average, 43% of the total energy intake and commercial baby foods provided 17%. These values decreased to 21 and 2% respectively at 20 months and there was little further change thereafter.3. Average intakes of energy and of most nutrients met recommended intakes (Department of Health and Social Security, 1969). Iron intakes were marginal, and vitamin D intakes were low.4. The energy intake of boys was significantly higher than that of girls at 3 years of age, but the boys were not significantly heavier. The energy intake of children from ‘manualworker’ families was higher than that from ‘non-manual’ families. Similarly, the energy intake of children from larger families was higher than that of children from smaller families.5. Energy intakes were correlated with body-weights and with rates of gain in weight. Irrespective of body-weight, ‘big eaters’ at 20 months tended to be ‘big eaters’ at 3 years also.

AbstractObjectives:To validate the use of supermarket receipts as an index of fat and energy intake in a population that buys most of its food from supermarkets.Design:Cross-sectional, prospective dietary survey – feasibility study.Setting:Households situated within a 20-mile radius of a large (Tesco) supermarket in Leeds.Subjects:Two hundred and fourteen households who spend ≥=60% of their food purse in (Tesco and other) supermarkets.Results:Mean daily household purchase of fat, energy and percentage energy from fat contained in food from supermarkets were 185 g, 19.2 MJ and 35.9%. Mean daily household intakes of fat and energy were 190 g and 20.7 MJ, and 35% of energy was derived from fat. Mean household size was 2.4 persons. The association between the amount of fat and energy purchased from supermarkets and the amount of fat and energy consumed by households was strong. 0.90 MJ (95% confidence interval (CI): 0.8–1.0) of energy were consumed for every 1 MJ purchased from supermarkets and 0.76 g (95% CI: 0.64–0.87) of fat were consumed for every 1 g of fat purchased.Conclusions:The results show a strong association between estimates of the intakes of fat and energy and percentage energy from fat using 4-day food diaries and 28 days of receipts, in populations who buy most of their food from supermarkets. They also show that the fat content of total food purchases from supermarkets is 35.9% energy from fat compared with 33% energy from fat recommended by the Department of Health. This preliminary research indicates the feasibility of and potential for utilising large quantities of readily available data generated from supermarket checkouts in dietary surveys.

Dry adult Merino ewes were fed at daily or weekly intervals on oat grain at levels which provided 4.0, 3.0, or 2.0 lb starch equivalent (S.E.) per sheep per week. The experimental periods were 223, 223, and 181 days respectively. Differences between the mean body weights of the ewes a t the three levels of feeding were highly significant (P < 0.001). Body weight varied only slightly and no losses occurred in ewes fed weekly at the rate of 4.0 lb S.E. per head. Ewes fed daily a t this level had a significantly greater mean body weight (P < 0.001) a t the conclusion of the 223-day experimental period. The addition of a sodium chloride supplement did not improve the body weight or wool production of ewes fed weekly a t the level of 4.0 lb S.E. The mean body weight of ewes fed at the levels of 3.0 or 2.0 lb S.E. per head declined over the first 12 and 18 respectively and thereafter remained relatively constant. There were no significant differences at these levels of feeding between groups fed daily and weekly in respect of body weight, wool production, or survival rates. Ewes fed a t the level of 4.0 lb S.E. grew significantly more wool than those given 3.0 lb S.E. (P < 0.001). Losses were negligible in all groups except those fed a t the level of 2.0 lb S.E. In these groups there were few deaths in the first 16 weeks, but in the subsequent 10 weeks losses totalled 17.1 per cent. Ewes fed a t the level of 2.0 lb S.E. consumed their rations at a significantly slower rate (P < 0.01) than did those fed a t the level of 4.0 lb S.E.

1. Children (405), initially of average age 11.5 years, recorded all food and drinks consumed for three consecutive days (with an interview on the fourth day) on five separate occasions over a 2-year period. Food tables (Paul & Southgate, 1978) enabled nutrient intakes to be calculated. The data collected were found to be of high reliability (Hackett et al. 1983).2. The mean energy intakes showed broad agreement with other recent British dietary surveys but were well below those recorded in the prewar study by Widdowson (1947) and the FAO/WHO (1973) recommended levels. They were slightly below the current Department of Health and Social Security (DHSS; 1979) recommended intakes.3. Over the 2-year period, the energy intake of the boys increased by 13% compared with an increase of only 7% in the girls.4. The iron and vitamin A intakes of all groups of children were low compared with current recommendations (DHSS, 1979). This seems to be a result of falling energy intake. Mean calcium intakes were also only marginally in excess of the recommended intake, and those of most of the girls would fall below the recommendation if the proposal to end the fortification of flour (DHSS, 1981) is implemented.

A weight-reducing potential has been ascribed to high dietary fibre intake. To investigate the practical reliability of this hypothesis, fifty-three moderately overweight females (BMI > 27.5 kg/m2) on reduced energy intake (1200kcal/day) were treated for 24 weeks with a fibre supplement on a randomly, double-blind, placebo-controlled basis. The fibre was administered as an initial dose of 6 g and a maintenance dose of 4 g. Body weight and blood pressure were recorded weekly during the first 3 months and thereafter every second week. Blood samples were drawn at start and at end of the study. Initial body weights were 75.6 ± 1.6 kg in the fibre group versus 75.5 ± 1.6 kg in the placebo group. After treatment, mean weight loss in the fibre group was 8.0 kg versus 5.8 kg in the placebo group (p < 0.05). Systolic and diastolic blood pressures were significantly reduced in both groups without differences between the groups. Serum concentrations of cholesterol, triglycerides and uric acid were significantly reduced in the group with reduced energy intake, whereas no additional effect was observed when fibre was supplemented. Serum concentrations of potassium and sodium did not change significantly. The results suggest that a dietary fibre supplement in combination with a hypocaloric diet is of value as an adjunct in the management of overweight.

Previously, we have found that the addition of L-ascorbic acid to chitosan enhanced the reduction in body weight gain in guinea pigs fed a high-fat diet. We hypothesized that the addition of L-ascorbic acid to chitosan would accelerate the reduction of body weight in humans, similar to the animal model. Overweight subjects administered chitosan with or without L-ascorbic acid for 8 weeks, were assigned to three groups: Control group (N = 26, placebo, vehicle only), Chito group (N = 27, 3 g/day chitosan), and Chito-vita group (N = 27, 3 g/day chitosan plus 2 g/day L-ascorbic acid). The body weights and body mass index (BMI) of the Chito and Chito-vita groups decreased significantly (p < 0.05) compared to the Control group. The BMI of the Chito-vita group decreased significantly compared to the Chito group (Chito: -1.0 kg/m2 vs. Chito-vita: -1.6 kg/m2, p < 0.05). The results showed that the chitosan enhanced reduction of body weight and BMI was accentuated by the addition of L-ascorbic acid. The fat mass, percentage body fat, body circumference, and skinfold thickness in the Chito and Chito-vita groups decreased more than the Control group; however, these parameters were not significantly different between the three groups. Chitosan combined with L-ascorbic acid may be useful for controlling body weight.

Background: G protein-coupled bile acid receptor (TGR5) is involved in a number of metabolic diseases. The aim of this study was to identify the role of TGR5 after Roux-en-Y gastric bypass (GBP). Methods: Wild type and TGR5 knockout mice (tgr5-/-) were fed a high-fat diet (HFD) to establish the obesity model. GBP was performed. The changes in body weight and food intake were measured. The levels of TGR5 and peptide YY (PYY) were evaluated by RT-PCR, Western blot, and ELISA. Moreover, the L-cells were separated from wild type and tgr5-/- mice. The levels of PYY in L-cells were evaluated by ELISA. Results: The body weights were significantly decreased after GBP in wild type mice (p<0.05), but not tgr5-/- mice (p>0.05). Food intake was reduced after GBP in wild type mice, but also not significantly affected in tgr5-/- mice (p>0.05). The levels of PYY were significantly increased after GBP compared with the sham group (p<0.05); however, in tgr5-/- mice the expression of PYY was not significantly affected (p>0.05). After INT-777 stimulation in L-cells obtained from murine intestines, the levels of PYY were significantly increased in L-cells tgr5+/+ (p<0.05). Conclusion: Our study suggests that GBP up-regulated the expression of TGR5 in murine intestines, and increased the levels of PYY, which further reduced food intake and decreased the body weight.

AbstractBody weight is an indispensable parameter for determination of contrast medium dose, appropriate drug dosing, or management of radiation dose. However, we cannot always determine the accurate patient body weight at the time of computed tomography (CT) scanning, especially in emergency care. Time-efficient methods to estimate body weight with high accuracy before diagnostic CT scans currently do not exist. In this study, on the basis of 1831 chest and 519 abdominal CT scout images with the corresponding body weights, we developed and evaluated deep-learning models capable of automatically predicting body weight from CT scout images. In the model performance assessment, there were strong correlations between the actual and predicted body weights in both chest (ρ = 0.947, p < 0.001) and abdominal datasets (ρ = 0.869, p < 0.001). The mean absolute errors were 2.75 kg and 4.77 kg for the chest and abdominal datasets, respectively. Our proposed method with deep learning is useful for estimating body weights from CT scout images with clinically acceptable accuracy and potentially could be useful for determining the contrast medium dose and CT dose management in adult patients with unknown body weight.

Twelve adult (10 months old) castrated Korean black goats, with an average initial body weight of 24.98 ± 3.7 kg, were used in this experiment to determine their maintenance energy requirements. Dry matter intakes (g/d, p = 0.945) were not affected by energy levels, but metabolic energy intake (kcal/d, p < 0.002) and average daily gain (g/d, p < 0.001) were significantly increased at higher energy levels. Nutrient digestibility was similar in the treatments, but crude fat digestibility increased with the addition of protective fat powder (p = 0.001). The energy required for fattening the castrated Korean black goats was estimated using the correlation between metabolic energy intake per dietary body weight and average daily gain per dietary body weight. The Y-axis intercept value was calculated to be 108.76 kcal/kg BW0.75 (p < 0.05, r2 = 0.6036), which was the metabolic energy requirement for maintaining the lives of the fattening Korean black goats. The estimated energy requirements of the black goat can improve specification techniques, such as the energy level and the amount of feed supply required for domestic black goats.

Abstract ONC201, an anti-cancer DRD2 antagonist and ClpP agonist, is in Phase II trials for adult H3 K27M-mutant diffuse midline gliomas. In adults, the recommended phase 2 dose (RP2D) of 625mg ONC201 once a week has been established as a biologically active dose that is well tolerated. Radiographic regressions with single agent ONC201 have been reported in adult recurrent H3 K27M-mutant glioma patients. This multi-arm, dose-escalation and dose-expansion trial determined the pediatric RP2D of ONC201 administered as an oral capsule (Arm A) or liquid formulation (Arm E) in post-radiation H3 K27M-mutant glioma (Arm A) or in newly diagnosed DIPG (Arm B) patients. Molecular assessments include intratumoral ONC201 concentrations (Arm C) and CSF H3 K27M DNA levels (Arm D). Enrollment as of April 30, 2020 is complete in Arm A (22) and Arm E (26) and continues in Arm B (18/24), Arm C (5/12), and Arm D (22/24). The RP2D of weekly 625mg ONC201 scaled by body weight was confirmed when administered as a capsule or a liquid formulation as a single agent or in combination with radiation without dose-limiting toxicity. The most frequent adverse events regardless of attribution to the drug were predominantly low grade: ONC201 capsule alone was headache (54.5%), nausea (36.4%), and fatigue (36.4%); ONC201 liquid formulation was vomiting (31.8%), headache (22.7%), VIth nerve disorder (22.7%); ONC201 capsules in combination with radiation (Arm B) was headache (47.1%), vomiting (52.9%), nausea (41.2%). Pharmacokinetic analysis in plasma of Arm A patients revealed T1/2: 8.4h; Tmax: 2.1h; Cmax: 2.3ug/mL; AUC0-tlast: 16.4ug/mL, with similar exposure across body weights. In conclusion, when scaled by body weight the ONC201 capsule or liquid formulation alone or in combination with radiation were associated with safety and pharmacokinetic profiles in pediatric H3 K27M-mutant diffuse midline glioma patients that are similar to the experience in adults.

Abstract This research evaluated the effects of ensiled edamame soybean processing waste on ruminant intake and total tract digestibility. Waste from processing stored edamame was obtained (3 trips) and wilted (28%, 25%, and 37% DM), then treated with or without inoculant (Lactobacillus buchneri) and ensiled in 167 L plastic barrels (2 or 3 barrels/treatment from each trip) for 72 (Trip 1) or 69 (Trip 2 and 3) days. Pregnant ewes (n = 18; body weight = 55.5 ± 1.2 kg; 2 to 3 years old) were blocked by body weight and assigned randomly within a block to treatments within a trip, then each ewe was assigned to a barrel of silage. Ewes were housed individually in 1 × 1.5-m pens and offered silage for a 10-day adaptation period followed by 7 days of total feces and urine collection. Ewes were offered silage to allow for 10% orts and were offered 0.2% of their body weight of soyhulls and 32 g of mineral supplement/day to meet their predicted nutrient requirements for gestating ewes. Ewes consumed 1,616 ± 54 g DM/day (X ± SEM) or 2.9 ± 0.12% of their body weight and there was no effect (P ≥ 0.85) of inoculant treatment on DM intake (g/day or % of body weight). Dry matter digestibility was not affected (P = 0.98) by inoculant and averaged 55.7 ± 0.66%. Ewe average daily gain for the 17-day trial tended to be greater (P = 0.08) for the ewes offered the silage without inoculant (0.18 vs. 0.04 kg/d). Ensiling edamame processing waste yielded a feed that ewes consumed in adequate amounts to maintain their body weights over 17 days when also supplemented with soyhulls. The addition of silage inoculant had minimal effects on intake, digestibility, or ewe body weight change.
